Chinese Journal of Pathophysiology 1989;0(05):-
AIM: To investigate the ability of Chinese cobra snake venom-metalloproteinase(MT) to induce the histamine release from human mast cells and its potential mechanisms.METHODS: MT was purified from the snake venom by using heparin agarose and Superdex75 chromatography.Mast cells were dispersed from human lung, colon and tonsil tissues after digestion with collagenase and hyaluronidase. The dispersed mast cells were then challenged with MT,stimulus and control in LP4 tubes for 15 min at 37 ℃.A glass fibre-based fluorometric assay was used to measure histamine in the supernatants of dispersed mast cells.RESULTS: MT induced a dose-dependent release of histamine from human colon, lung and tonsil mast cells. As low as 0.03(mg/L) of MT was able to stimulate significant histamine release from human colon mast cells, but a minimum of 0.3 or 30 mg/L of MT was required to stimulate a similar level of histamine release from lung or tonsil mast cells, respectively. The release of histamine from colon and lung mast cells in response to MT was maximized at 12 min following the addition of the stimulus. This was quite different from the picture of the peak histamine release from tonsil mast cells,in which histamine release was maximized at 8 min following the addition of MT.Pretreatment of cells with metabolic inhibitors and pertussis toxin reduced dramatically histamine release from human colon, lung and tonsil mast cells by MT.In exogenous Ca~(2+) and Mg~(2+) free experiments, the release of histamine induced by MT was significantly decreased.CONCLUSION: Cobra snake venom MT induces human mast cells to release histamine through a G-protein-related mechanism, which may contribute to the pathogenesis of venomous snake bite.

Journal of Medical Research 2006;0(04):-
Objective To evalute Qiliqiangxin capsule for improving cardiac function and identifiy Qiliqiangxin capsule for directing cytoprotective effects against apoptosis of cardiomyocytes. Methods Sprague Dawley (SD) rats were tested as subjects and the model rats were built by anterior descending coronary deligation, then the survival rats were devided into 6 groups randomly:sham-operated group(sham,n=8),fed with normal saline of same volume instead;control group(control,n=7),fed with normal saline of same volume instead;Qiliqiangxin great dosage group(great,n=9),Qiliqiangxin 1.2g/(kg?d) treatment;Qiliqiangxin medium dosage group(medium,n=9),Qiliqiangxin 0.6g/(kg?d) treatment;Qiliqiangxin small dosage group(small,n=8 rats),Qiliqiangxin 0.3g/(kg?d) treatment;the captopril group(captopril,n=9),captopril 25mg/(kg?d) treatment.24 hours later, rats in sham operated group and model group were given normal saline, rats in treating group were given Qiliqiangxin, rats in the contrasting group were given captopril. After being treated for 4 weeks, all rats′ heart function was measruated by right flank arteria carotis communis arterial cannula. The left ventricle mass index was calculated and the expression of protein caspase-3 was tested by immunohistochemical method and Westernblot. Results Compared with control group, Qiliqiangxin capsule could reduce effectively BW,HMI,LVMI,increase LVEDP and LVSP,+dp/dtmax and -dp/dtmax.Immunohistochemical analysis and Westernblot analysis showed that adminstration of Qiliqiangxin significantly inhibited caspase-3 expressions in myocardial cells in a dose-dependent manner. Conclusion Qiliqiangxin significantly improved the CHF rat′s cardiac function and reduced the left ventricle mass index and heart mass index 4 weeks after being treated with relative high dose Qiliqiangxin. It might be partially attributed to the suppression of caspase-3 expressions.

Neurology Asia 2017;22(2):133-138
Objective: The aim of this study was to evaluate whether there is an association between Parkinson’s
disease (PD) and colorectal cancer in Taiwan. Methods: This was a case-control study using claim
data of the Taiwan National Health Insurance Program. There were 64,619 subjects aged 20-84 with
newly diagnosed colorectal cancer as cases and 64,619 randomly selected subjects without colorectal
cancer as controls from 2005 to 2011. Both cases and controls were matched by sex, age, comorbidities,
and index year of diagnosing colorectal cancer. Subjects who were diagnosed with PD within 5 years
of diagnosing colorectal cancer were excluded. The multivariable logistic regression model was used
to estimate the adjusted odds ratio (OR) and 95% confidence interval (95% CI) for risk of colorectal
cancer associated with PD. Results: After adjusting for confounding factors, the multivariable logistic
regression analysis revealed that the adjusted OR of colorectal cancer was 0.69 for subjects with PD
more than 5 years before index date (95% CI 0.59, 0.81), as compared with subjects without PD.
Conclusion: PD is associated with reduced odds of colorectal cancer. Further research is needed to
elucidate the mechanisms underlying our findings.

Tetradrine-tashionone II(A)-PLGA composite microspheres were prepared by the SPG membrane emulsification method, and the characterization of tetradrine-tashionone II(A) -PLGA composite microspheres were studied in this experiment. The results of IR, DSC and XRD showed that teradrine and tashionone II(A) in composite microspheres were highly dispersed in the PLGA with amorphous form. The results of tetradrine-tashionone II(A) -PLGA composite microspheres in vitro release experiment showed that the cumulative release amounts of tetradrine and tashionone II(A) were 6.44% and 3.60% in 24 h, and the cumulative release amounts of tetradrine and tashionone II(A) were 89.02% and 21.24% in 17 d. The process of drug in vitro release accorded with the model of Riger-Peppas. Tetradrine-tashionone II(A) -PLGA composite microspheres had slow-release effect, and it could significantly reduce the burst release, prolong the therapeutic time, decrease the dosage of drugs and provide a new idea and method to prepare traditional Chinese medicine compound.
