Journal of Chinese Physician 2001;0(08):-
Objective To observe the effect of heat stress on plasma cGMP and hemodynamics in animals with chronic heart failure. Methods Coronary arteries of twenty-five rabbits were ligated to set up the animal model of heart failure and other five rabbits only received thoractomy as sham group. Eight weeks after coronary artery ligation, they are randomly divided into heat stress (HS) group, control group, HS+L-NAME group, and non-HS+L-NAME group and non-HS+L-Arg group, each group containing 5 animals. Hemodynamic indices and plasma levels of NOS and cGMP were measured. Results Hemodynamics of all 25 rats received operation was poorer compared with the rats of sham group (P

Chinese Mental Health Journal 2017;31(2):127-132
Objective:To explore the relations of adolescents'behavioral problems to genetic and environmental factors.Methods:66 twin pairs aged 12 to 18 years in Chongqing were included in the study. The Youth Self-Report (YSR) was used to evaluate behavioral problems. The blood sample was collected to identify zygosity of twins.Genetic effects were evaluated by comparing the phenotypic correlationamong monozygotic twins(rMZ) and the phenotypic correlation among dizygotic twins(rDZ).Structural equation modeling (SEM) was performed to evaluate the relations of adolescents behavioral problems to the additive genetic factors (A),shared environment factors (C) and individual specific environmental factors (E).Results:MZ correlations of Social problems and Delinquency were less than DZ correlations(rMZ < rDZ).It prompted that data was not suitable for genetic analysis.MZ correlations(rMZ) of any others (including social withdrawal, somatic complaints,anxiety/depression,thinking problem, attention problems,self-destructive/identity problem)were less than 2DZ correlations (2rDZ).It prompted to fit the ACE model. The optimal model was AE model.Among the contribution of the additive genetic factors (A) and individual specific environmental factors (E)to the total variance, the genetic factors ranged from 0.52 to 0.57 and individual specific environmental factors ranged from 0.43 to 0.48 in anxiety/depression and self-destructive/identity problem. For social withdrawal, somatic complaints, thinking problem and attention problems, the genetic factors ranged from 0.17 to 0.45and individual specific environmental factors ranged from 0.55 to 0.83.Conclusion:There is no obvious relationship between genetic effects and adolescent social problems or delinquency.Other behaviors problems (social withdrawal, somatic complaints,anxiety/depression,thinking problem, attention problem,self-destructive/identity problem)are correlated with additive genetic factors and individual specific environmental factors.Additive genetic factors have high-correlation with anxiety/depression and self-destructive/identity problem, and individual specific environmental factors have high-correlation with other factors.

OBJECTIVE To investigate the role of TLR4 signaling pathway in secretory otitis media of rats.METHODS Rats SOM models were made by infusing FS and LPS to the middle otocysts. The rats were killed separately at the 3rd, 5th, 7th, 9th,11th and13th day(5 rats each time). And then the expressions of TLR4, iNOS, IL-8mRNA of middle ear mucosae were detected. NF-?B was also detected through the method of immunohistochemistry. RESULTS The expressions of TLR4mRNA, iNOS and IL-8mRNA in middle ear mucosae of SOM group were significantly higher than that of control group. Immunohistochemistry method showed that the activation of NF-?B in middle ear mucosae was obviously increased in SOM. CONCLUSION The expression of TLR4 was obviously increased in SOM and it activates the NF-?B, promotes the expression of downstream IL-8. That maybe the mechanism which cause SOM.

Aim To investigate the cytotoxic activity of HDAC inhibitor Trichostatin A (TSA)combined with anticancer drugs targeting DNA on T24 bladder cancer cell line. Methods MTT assay was used to detect the inhibitory rate of TSA alone or combined with ADM, MMC and DDP respectively on T24 bladder cancer cell in cancer cell proliferation by administering TSA alone or combined with ADM, MMC and DDP respectively.Jin′s equation was used to evaluate the efficacy of drug combination. Results The growth inhibitory rate of TSA combined with ADM, MMC and DDP respectively was in a concentration-dependent manner. The synergism of TSA combined with MMC was most significant. When administered in lower or moderate concentration, TSA combined with ADM or DDP in lower or moderate concentration demonstrated synergic effect too. Conclusion HDAC inhibitor TSA enhances the cytotoxic activity of anticancer drugs targeting DNA on bladder cancer cells and is promising to be used in chemotherapeutic regimens for advanced bladder cancer.

Objective To study the effects of mild hypothermia on cardiomyocyte contractility improvement after ischemia-repeffusion injury and on the preservation of well-functioning mitochondrial respiratory capability. Methods A total of 50 newborn SD rats 1 ~ 2 days after delivery were sacrificed and their hearts taken to preserved in 4 ℃ cold D-hanks buffer solution with 0.12% pancreatic proteinase and collagenase and then processed with 37 ℃ water bath to collect the cardiomyocytes cultured in DMEM medium with 10% FBS for 5 days. The cardiomyocytes of rats were subjected to ischemia/reperfusion,in vitro,by oxygen and glucose deprivation(OGD)/oxygen and glucose restoration(OGR).The cardiomyocytes of rats after ischemia/reperfusion were divided into three groups:control group, hypothemia group and normothermia group.Contractile frequency and velosity were determined before OGD and 0 h,0.5h,1 h,1.5 h and 2 h after OGR.Ultrastructure changes of cardiomyocytes and mitochondrion were observed under transmission electron microscope(TEM)0 h and 2 h after OGR as well as assessment ot respiratory rate and respiratory control rate(RCR)with Clark oxygen electrode in each group. All data were analyzed with statistical software of SPSS 13.0.Results Contractile function of cardiomyocytes in hypothermia group and normothermia group declined to nadir at 0 h after OGR(P =0.000)and the contractile function of cardiomyocytes in hypothermia group was improved one hour later, compared with the normothermia group(P =0.000).Obvious swelling of mitochondrion was observed under TEM in normothermia group with little alteration after OGR.The RCR assessments indicated respiratory function in normothermia group was impaired after OGR(P =0.000)and this may be responsible for contractility dysfunction. Conclusions Mild hypothemia used after ischaemia can optimize the contractility of cardiomyocytes after a normothermia OGR,and the well-functioning respiratory capability of mitochondrion may be preserved in this process.
