BACKGROUND:Through compounding hydroxyapatite and poly-DL-lactide, the mechanical properties, physical and chemical properties of the implants can be enhanced.
OBJECTIVE:To detect the effect of hydroxyapatite/poly-DL-lactide composite internal fixation screws in canine femoral condyle cancel ous bone fracture repair.
METHODS:Forty-two beagle dogs were operated to bilateral femoral condyle fracture models. The left side was fixed using hydroxyapatite/poly-DL-lactide screws as experimental group;while the right side was fixed with pure poly-DL-lactide screws as control group. After 2, 4, 8, 12, 24, 36 and 48 weeks, conditions of fracture were observed using X-ray, femoral and screw specimens were observed histopathological y, and the bending strength and the average molecular weight were detected. The biological absorption rate, intensity decay rate and biodegradation rate were calculated.
RESULTS AND CONCLUSION:After 2-48 weeks, bilateral fractures were fixed wel and new bone grew wel , but the biological absorption rate of the screws in the experimental group was lower than that in the control group (P ≤0.01). In the first 2, 4 weeks, the bending strength of the experimental screws was higher than that of the control screw (P ≤0.05), but the biodegradable speed was slower in the former one (P ≤ 0.05 or P ≤ 0.01). The pathological changes were similar in the two groups. After 48 weeks, the fractures were healed and bone tissue reconstruction was completed. Compared with the pure poly-DL-lactide screws, the hydroxyapatite/poly-DL-lactide composite internal fixation screws have better fixation effects, biocompatibility, mechanical properties and biodegradability.

OBJECTIVETo study the effect of hydroxycamptothecin (HCPT) on apoptosis-inducing factor (AIF) expression and AIF translocation from mitochondria to the nucleus in human hepatocellular cancer cell SMMC-7721 during apoptosis.
METHODSAfter treatment with 80 mg/ml of HCPT, the cancer cells were stained with A0/EB to monitor their apoptosis. Their mitochondria was examined with electronmicroscopy and the AIF expression of the cells was tested by RT-PCR and Western blot. The translocation of AIF from mitochondria to the nucleus during apoptosis was analyzed by confocal microscopy.
RESULTSSMMC-7721 cells treated with HCPT showed chromatin condensation, nuclear fragmentation and mitochondria swelling. The mRNA and protein expression of AIF in treated and untreated SMMC-7721 cells were not significantly different. However, cells treated with 80 mg/ml HCPT for 6 h or 12 h showed massive translocation of AIF into the nuclei.
CONCLUSIONThese results show the important role the mitochondrial pathway of apoptosis plays in HCPT-induced tumor cell death, at least in SMMC-7721 cells.

Objective To investigate the usefulness of 99Tcm-MDP whole body bone scintigraphy (WBBS) in patients with synovitis, acne, pustulosis, hyperostosis, osteitis (SAPHO) syndrome. Methods 99Tcm- MDP WBBS was performed in 25 patients (6 males,19 females, mean age =(55.1 ±9.8) years)with SAPHO syndrome.Bone lesions were classified into five categories:anterior chest wall, spine, mandible, sacroiliac joint, and limbs. The typical scintigraphic manifestations of SAPHO syndrome were summarized and compared to other radiological imaging data. Results Among 25 patients,32% of cases (8/25)were associated with skin lesion; 48% ( 12/25 ) were pathologically diagnosed with chronic nonspecific bone inflammation by bone biopsy. On 99Tcm-MDP WBBS,abnormal metabolic foci at anterior chest wall were found in all cases, most of which located in the sternocostoclavicular region (96%,24/25 ),including sternoclavicular joints (60%,15/25),first costosternal junctions (48%,12/25),and manubriosternal junctions (44%,11/25 ).Only 20% of the patients (5/25) demonstrated the typical scintigraphic characteristic:“bull's head” sign. The second most frequent part was spine (44%,11/25).Appendicular skeleton was affected in 16% (4/25) patients.WBBS also demonstrated additional skeletal lesions in 68% (17/25 ) of the patients, mainly in first costosternal junctions (7 patients),sternoclavicular joints (6 patients),manubriosternal junctions (5 patients) and spine (4 patients). Conclusions Abnormal metabolic foci in sternocostoclavicular region and other imaging manifestations on 99Tcm- MDP WBBS can be used to diagnose, differentiate, and localize the insidious lesion and evaluate the lesion activity in patients with SAPHO syndrome.
