1.Comparative analysis between HP-083/4 and rational used instrument on results of six items

Objective To compare the performance of HP-083/4 and rational used instrument on detecting six items.Methods The rational instruments were used as contrast instrument,HP-083/4 was the verified instrument.A total of 100 blood specimens and 100 urine specimens were collected,and the levels of antistreptolysin O(ASO),hypersensitive C reactive protein (hsCRP),D-di-mer(D-D),glycosylated hemoglobin(HbA1c),rheumatoid factor(RF)and urine microalbumin(mAlb)were detected.The regression equation and correlation coefficient(r)of the two methods were calculated,and the Kappa values(κ)were analyzed to evaluate the performance of HP-083/4.Results There was a good linear correlation (r >0.950)for the two methods in detecting the serum ASO,hsCRP,D-D,HbA1c,RF and mAlb,r were 0.991,0.995,0.970,0.957,0.980 and 0.967 respectively.Besides,they had good concordance(κ>0.6),theκ values were 0.830,0.957,0.601,0.720,0.920 and 0.694 respectively.Conclusion HP-083/4 is effec-tive in detecting ASO,hsCRP,D-D,HbA1c,RF and mAlb,which should be suitable for clinical application.
2.Corticosteroid?induced Osteonecrosis of the Femoral Head:Detection, Diagnosis, and Treatment in Earlier Stages

Objective: This review aimed to provide a current recommendation to multidisciplinary physicians for early detection, diagnosis, and treatment of corticosteroid?induced osteonecrosis of the femoral head (ONFH) based on a comprehensive analysis of the clinical literature. Data Sources: For the purpose of collecting potentially eligible articles, we searched for articles in the PubMed, Cochrane Library, Embase, and CNKI databases up to February 2017, using the following key words: "corticosteroid", "osteonecrosis of the femoral head","risk factors", "diagnosis", "prognosis", and "treatment". Study Selection: Articles on relationships between corticosteroid and ONFH were selected for this review. Articles on the diagnosis, prognosis, and intervention of earlier?stage ONFH were also reviewed. Results: The incidence of corticosteroid?induced ONFH was associated with high doses of corticosteroids, and underlying diseases in certain predisposed individuals mainly occurred in the first 3 months of corticosteroid prescription. The enhanced awareness and minimized exposure to the established risk factors and earlier definitive diagnosis are essential for the success of joint preservation. When following up patients with ONFH, treatment should be started if necessary. Surgical treatment yielded better results than conservative therapy in earlier?stage ONFH. The ideal purpose of earlier intervention and treatment is permanent preservation of the femoral head without physical restrictions in daily living. Conclusions: Clinicians should enhance their precaution awareness of corticosteroid?induced ONFH. For high?risk patients, regular follow?up is very important in the 1st year after high?dose prescription of corticosteroids. Patients with suspected ONFH should be referred to orthopedists for diagnosis and treatment in its earlier stage to preserve the joint.

4.Effect of in vitro and in vivo treatment with mitomycin C on activities of CYP2D1/2, CYP2C11, and CYP1A2 in rat liver.

AIMTo evaluate the effect of in vitro and in vivo treatment with mitomycin C (MMC) on activities of CYP2D1/2, CYP2C1 , and CYP1A2 in the liver of male rats.
METHODSUsing HPLC to determine the activities of the three isoenzymes in rat liver microsomes by detecting the specific metabolites of their substrates after treatment with inducers in vivo or inhibitors in vitro.
RESULTSIn vitro, MMC inhibited the activity of CYP2D1/2, CYP2C11, and CYP1A2 in dexamethasone-induced microsomes by (19 +/- 6)% (P < 0.05), (85 +/- 10)% (P < 0.01), and (36 +/- 6)% (P < 0.05), respectively, and decreased the activity of CYP1A2 in beta-naphthoflavone-induced microsomes by (58 +/- 6)% (P < 0.01). Rats were injected intraperitoneally with 20% of the LD50 of MMC for 3 or 6 d. The treatment showed no significant effect on microsomal activities of CYP2D1/2, CYP2C11 or CYP1A2.
CONCLUSIONMMC can inhibit the activities of CYP2D1/2, CYP2C11, and CYP1A2 in rat liver microsomes in vitro, but it showed no significant effect on the activities of the three isoenzymes in vivo.

5.Acupuncture inhibiting responses of spinal dorsal dorsal horn neurons induced by noxious dilation rectum and colon.

OBJECTIVETo study on mechanisms of acupuncture in relieving visceral pain.
METHODSIn SD rats CRD was used as noxious visceral stimuli. Activities of spinal dorsal horn wide dynamic (WDR) neurons of L1-L13 were recorded by extracellular microelectrode technique. Acupuncture was given at ipsi-lateral and contra-lateral Zusanli (ST 36) of the same segmental innervation of rectum and colon.
RESULTSVisceral noxious afferent could significantly activate spinal dorsal horn convergent neurons, and mechanical stimulation of contra-lateral body surface and hand acupuncture at Zusanli (ST 36) could inhibit this noxious response. When the spinal cord was acutely blocked, the inhibiting CRD effect of needling CRD effect of needling contra-lateral Zusanli (ST 36) completely disappeared.
CONCLUSIONAcupuncture and visceral noxious afferent signals converge and interact each other in spinal level, and acupuncture at acupoint can inhibit the spinal dorsal horn neuron respon se activated by visceral noxious afferent and this action needs the participation of the center above the spinal cord.

6.Study on composing mechanism by effect of disassembled compositions of Yiqi Huoxue decoction on platelet aggregation of health adults.

OBJECTIVETo investigate and explain the composing mechanism by observing enhancement property of disassembled compositions of Yiqi Huoxue decoction (YQHXD) on platelet aggregation of health adults.
METHODVenous whole blood was obtained from healthy human subjects and anti-coagulated, and then centrifuged to produce platelet-rich plasma (PRP) and platelet-poor plasma (PPP). Disassembled compositions of YQHXD and sodium chloride were placed in different translucent tubes containing PRP and PPP, calculated the maximum percents of platelet aggregation stimulated by platelet agonists: adenosine diphosphate (ADP), platelet activating factor (PAF) and arachidonic acid (AA).
RESULTThe maximum percents of platelet aggregation induced by ADP and PAF were largely improved by YQHXD, both astragalus membranaceus and Angelica Scinesis played the most important roles in the decoction, the herbs of Yiqi and Huoxue must work together to enhance platelet aggregation.
CONCLUSIONIn vitro study, YQHXD can markedly improve platelet aggregation of health adults, but the exact mechanism should be proved by further experiments.

7.Sero-and molecular epidemiology of the gene of different ORF of transfusion transmitted virus (TTV) in different population of China.

BACKGROUNDTo investigate the distribution of anti-TTV antibody and the different ORF gene in different populations of China.
METHODSThe antibody to TTV in sera collected from different population were detected by using ELISA and the different ORF genes were amplified with PCR.
RESULTSThe positive rates of TTV ORF1 DNA, ORF2 DNA and the antibody in various populations were as follows: 16.0% (12/75), 10.7% (8/75) and 25.3% (19/75) in paid blood donors; 10.0 (3/30), 16.7% (5/30) and 16.7% (5/30) in patients with hepatitis A, 47.5% (19/40), 42.5% (17/40) and 22.5% (9/40) in patients with hepatitis B; 42.9% (15/35), 37.1% (13/35) and 28.6% (10/35) in patients with hepatitis C; 20.0% (3/15), 26.7% (4/15) and 13.3% (2/15)in patients with hepatitis D; 16.7% (2/12),16.7% (2/12) and 33.3% (4/12)in patients with hepatitis E; 23.8% (5/21), 38.1% (8/21) and 23.8% (5/21) in patients with hepatitis G; 61.1% (11/18), 50.0% (9/18) and 44.4% (8/18) in patients with non A-G hepatitis, respectively. The positive rate of different ORF DNA had no significant difference. Significant differences were found in the positive rates of TTV DNA in various populations. There was no relationship between the TTV DNA and the antibody to TTV.
CONCLUSIONSThe antibody to TTV and TTV DNA were found in every population of China. There was no significant difference in the positive rates of TTV DNA between ORF1 and ORF2. The positive rate in patients with non A-G hepatitis was higher than those in the other populations.

OBJECTIVETo evaluate the inhibitory effect of recombinant adenovirus carrying human endostatin gene (Ad-endo) on the growth of human pancreatic carcinoma xenograft in nude mice.
METHODSThe expression of endostatin in human pancreatic carcinoma Capan-2 cells was examined by RT-PCR after infection with Ad-endo. The supernatants of Capan-2 cells were collected after 48 h of infection with Ad-endo as the conditioned medium for human umbilical vein endothelial cells (HUVECs), whose proliferation in vitro was assayed. Capan-2 cell xenografts were established to determine the antitumoral effects of Ad-endo in vivo. The intratumoral microvessel density (MVD) was evaluated using CD31 staining.
RESULTSThe expression of endostatin gene was detected by PT-PCR in infected Capan-2 cells. The conditioned medium from Ad-endo-infected cells significantly inhibited HUVEC proliferation (P<0.05). Ad-endo significantly suppressed the growth of Capan-2 tumor xenografts in nude mice (P<0.05), and the MVD decreased significantly in the treated tumor (P<0.05) as compared with that in the control group.
CONCLUSIONAdenovirus carrying human endostatin gene produces inhibitory effects on the growth of human pancreatic carcinoma tumors in nude mice.

10.The significance of serum anti-titin antibodies and anti-acetylcholine receptor antibodies in patients with myasthenia gravis

Objective To study the clinical significance of serum anti-Titin antibodies and thecorrelation between anti-Titin anbibodies and anti-acetyleholine receptor (AchR) antibodies in patientswith myasthenia gravis (MG). Methods We detected anti-Titin antibodies and anti-AchR antibodieswith enzyme linked immunosorbent assay (ELISA) in the sera from 81 cases of MG groups includingearly-onset MG, late-onset MG and MG with thymoma (MGT). The positive rates were compared amongthese groups. Results The positive rates of anti-Titin antibodies in sera of MG patients weresignificantly higher than controls (P<0.01). The mean titers and the positive rates of anti-Titin antibodieswere significantly higher in MGT (80%) and late-onset MG (69.4%) subgroups than in the early-onsetMG subgroup (25%). In contrast, the positive rates and mean titers of AchR antibodies in the early-onsetMG subgroup were higher than those in the late-onset MG subgroup. The detections of serum anti-Titinantibodies and AchR antibodies were consistent with the diagnosis of MG. Conclusion Anti-Titinantibodies are specific for MG. They often present in the sera of patients with MGT and late-onset MG.The positive rates and mean titers of AchR antibodies in early-onset MG patients sera are higher thanthose in late-onset MG patients. These features prompt fiu'ther studies on the pathogenesis of MG. Testingfor a combination of anti-Titin antibodies and AchR antibodies gives the higher sensitivity for thedetection of MG than each antibody alone.
