1.INDUCTION OF APOPTOSIS OF HUMAN LIVER CARCINOMA CELLS BY TEA POLYPHENOLS

Tea polyphenols were tried to induce apoptosis of human cultured hepatocellular carcinoma cell line HepG-Ⅱ. MTT assay, DNA agarose gel electrophoresis, transmission electron microscopy , fluorescence decoration and DNA end labeling method (Tunel) were used to identify apoptosis. Having been treated by tea polyphenols in 250μg/ml , HepG-Ⅱcell apoptosis was induced. The induction of apoptosis was the dose dependent. Chromatin condensation, apoptotic body formation, fluorescence of yellow green and pellet were observed. Agarose gel electrophoresis analysis revealed DNA cleavages( DNA ladder). The results indicated that tea polyphenole could induce apoptosis of cultured human hepatocellular carcinoma cell line HepG-Ⅱ. It suggested that tea polyphenols could be used to treat human liver carcinoma.

4.Cervical adenoid basal cell carcinoma:a clinlcopathologic study of 12 patients

Purpose To investigate the clinicopathologic features of cervical adenoid basal cell carcinoma. Methods Clinical and pathological data were analyzed in 12 cases of cervical adenoid basal cell carcinoma and the relevant literatures were reviewed. Results The age of 12 cases of with cervical adenoid basal cell cancer patients ranged from 36~70 years ( mean:53. 3 years) , with a medi-an age of 51. 5 years. Amongst 12 patients who contained no gross definite lesion, 5 patients (41. 7%) had the symptoms of vaginal bleeding. 8 cases of patients underwent hysterectomy, while the other four received cervical conization. Cervical intraepithelial neopla-sia ( CIN) lesions were observed in all 12 cases. The tumor cells were small and uniform, with dark oval nuclei without conspicuous nucleoli and scanty cytoplasm. Tumor infiltrated into the stroma in nests and cords. Glandular differentiation within tumor nests were seen in 12 cases, including two cases of squamous cell differentiation accompanied by partial and minor interstitial edema. Peripheral palisading cells around tumor nests were also found. The infiltrating depth of tumor ranged from 0. 5~10 mm with the average 4. 12 mm. Cancer embolis were available in 2 cases with no lymph node metastasis. 10 cases followed up for 3~78 months with no recur-rence and metastasis, but 2 cases were lost. Conclusion Adenoid basal cell carcinoma is a rare uterine cervical tumor found in post-menopausal women, which are often accompanied with CIN. This tumor has a favorable prognosis and should be clearly separated from adenoid cystic carcinoma and other tumors which also have infiltrative growth pattern.
5.Study of pharmacokinetics of nicotine in local brain by using microdialysis and stable labeled isotope.

The paper is to report the study of pharmacokinetics of transdermal administered nicotine in the brain of freely moving rat by using microdialysis with stable labeled isotope as internal standard. The pharmacokinetic behavior of nicotine in Sprague Dawley rat brain was investigated after intranasal administration (3.75 mg). Brain fluid samples were collected by intracerebral microdialysis with DL-nicotine as internal standard. Concentrations of nicotine and DL-nicotine in the sample were measured by HPLC-MS/MS. Main pharmacokinetic parameters were calculated and analyzed by Das 2.0 pharmacokinetic software. The recovery of nicotine and the delivery of DL-nicotine were the same. The fate of absorption and distribution was two compartment model and the values of t1/2alpha was 170.31 min, t1/2beta was 263.30 min and the AUC(0-infinity) was 2.75 x 10(5) microg x L(-1) min separately. DL-nicotine can be used to calibrate the recovery of nicotine, and the new method of stable isotope microdialysis can be used to study the pharmacokinetics of freely moving rat. It will make sense for the treatment of addiction of tobacco and provide a new thought for the research of pharmacokinetics-pharmacodynamic combination.
6.The clinical significance of CYP2C19 genotype detection for antiplatelet therapy of cardiovascular and cerebrovascular diseases in the elderly

Objectives To investigate the clinical significance of CYP2C19 genotype detection for antiplatelet therapy of elder cardiovascular and cerebrovascular diseases(CCVD).Methods We enrolled all elderly patients with either cardiovascular or cerebrovascular disorders who received clopidogrel as mono drug or in combination with another antiplatelet drug aspirin as secondary prevention for more than 12 months in our hospital from January to August 2015.Somatotypes of CYP2C19 genotypes of all participants were assessed to analyze the relapse of cardiac-cerebral vascular diseases in these patients.Results A total of 250 patients were enrolled,including 179 male and 71 female,with average age of (85.2 ± 7.9) years.Among these patients,there were 97 (38.8%) cases with EM CYP2C19 genotypes,110 cases(44.0 %) with IM CYP2C19 genotypes,43 cases(17.2 %) with PM CYP2C19 genotypes.When treated with clopidogrel for antiplatelet in secondary prevention process,the rate of the relapse in cardiovascular event was 34.9% and higher in PM CYP2C19 genotype than in EM and IM CYP2C19 genotype (19.6 % and 15.5 %,respectively) (x2 =7.251,P =0.027).This phenomenon was similar to patients who received stent implantation(x2=6.393,P =0.041).However,no statistically significant difference was observed in the recurrence rate of cerebral vascular disease between three different genotypes(EM 29.9 %,IM 20.0 %,PM 27.9%,x2 =2.880,P =0.237).Conclusions Our results highlight that CYP2C19 genotype might be a potential guidance for secondary prevention of cardiovascular and cercbrovascular disorders among elderly patients.Clopidogrel may be less effective in patients with SM CYP2C19 genotype than those with EM or PM CYP2C19 genotype for secondary prevention of cardiovascular disease.
7.An algorithm of nodule detection based on high resolution CT images.

Combining the speed of two-dimensional detection with the precision of three-dimensional detection, an automatic algorithm based on high resolution CT images is proposed to identify nodules in this paper. Nodule candidates are extracted by a two-dimensional convergence index (CI) filter, then a three-dimensional Hessian matrix detection filter is introduced to reduce false positive lung nodules, Experiments show that the algorithm is effective with a sensitivity of 90% and the false positive lung nodules per slice is 0.33.

8.Phenotypic and functional characteristics of T cells in tuberculosis pleurisy

Objective:To elucidate the phenotypic and functional characteristics of CD3+ CD4+ and CD3+ CD8+ T in pleural fluid of patients with tuberculous pleurisy.Methods:PBMCs and PFCs were isolated. The frequency of CD3+,CD3+ CD4+,CD3+ CD8+ T cells and the ratio of CD4/CD8 in PFCs were analyzed by FACS. The concentration of IFN-?,IL-2 and TNF-? in the pleural fluid were analyzed by ELISA.Production of IFN-?,IL-2 and TNF-? by CD3+ CD4+ and CD3+ CD8+ T cells was detected by FACS following stimulation with BCG.Results:Higher frequencies of CD3+ CD4+ T cells and CD4+ T/CD8+ T ratio were demonstrated in PFCs when compared with PBMCs from normal donors and TB patients.ELISA analysis demonstrated that significantly high levels of IFN-? and TNF-? were detected in pleural fluid.Further analysis by FACS indicated that IL-2,TNF-? and IFN-? were predominantly secreted by CD3+CD4+T cells but not by CD3+ CD8+ T cells.Conclusion:The frequencies of CD3+ and CD3+ CD4+ T cells are increased in PFCs. Large amounts of Th1 cytokines are detected,which are produced by CD3+ CD4+ T cells,and might play important roles against TB infection.
9.Monitoring of renal pelvic pressure and its siginifcance during flexible ureteroscopic lithotripsy

Objective To monitor the renal pelvic pressure and to investigate its clinical significance during retrograde flexible ureteroscopic lithotripsy (RFUL).Methods The data of renal pelvic pressure measured in 60 cases of RFUL with the mean irrigation pump speed and pressure of 30 ml/min and 30 mmHg were analyzed retrospectively.The influence factors of renal pelvic pressure and its correlation with postoperative fever were analyzed.Renal pelvic pressure was measured by baroceptor,which was connected to PHILIP-MP4 monitor IBP channel and ureteric catheter positioned in renal pelvis through a dual channel ureteral access sheath (UAS).The renal pelvic pressure data was collected and analyzed in every 2 seconds by computer.The 60 cases were divided into 3 groups according to their intra-pelvic pressure situations:normal pressure group(NP,IPPmax ≤30 mmHg),high pressure group(HP,IPPmax>30 mmHg,but high pressure duration≤ 10 min),and backflow pressure group(BP,IPPmax>30 mmHg and high pressure duration> 10 min).Results The baseline intra-pelvic pressure (IPP0) and max imum intra-pelvic pressure (IPPmax) were (13.2±5.6) mmHg and (95.6±2.3) mmHg respectively.IPP levels during the RFUL were significantly higher than the IPP0(P<0.001).There were 32,17 and 11 cases in NP,HP and BP groups,respectively.There were 6 cases with fever higher than 38.5 ℃ (10%),in which there were 1 case in NP,1 case in HP group and 4 cases in BP group.The postoperative fever rate in NP,HP and BP group were 3%,6% and 36% respectively,which were significantly different between groups(P<0.01).There were 12 cases with procalcitonin >0.1 ng/ml and 8 cases with procalcitonin >0.5 ng/ml,in which 2 cases in HP group and 6 cases in BP group.Conclusions RFUL would result in a temporal elevated intrapelvic pressure greater than 30 mmHg.Postoperative fever is relevant with renal perfusion pressure and perfusion time.It's necessary for the surgeons to adjust the perfusion pressure during operation.
10.Preliminary Study of Necroptosis in Cardiac Hypertrophy Induced by Pressure Overload.

The aim of this study was to observe whether necroptosis is involved in the process of cardiac hypertrophy induced by pressure overload. SD rats underwent transverse abdominal aortic constriction (TAC) operation for establishing cardiac hypertrophy model. The structure and function of the left ventricle of rats were evaluated via echocardiography, left ventricular mass index, the expression of markers of cardiac hypertrophy and histological detection. Real-time PCR and Western blot were used to measure the gene and protein expression of receptor interacting protein kinase 1 and 3 (RIPK1 and RIPK3, the necroptosis markers) respectively. Four weeks after TAC operation, rat model for cardiac hypertrophy was established. The experimental data showed that the gene and protein expressions of RIPK1 and RIPK3 in the rat heart hypertrophic tissues after TAC for 4 weeks were increased significantly compared with those in the sham group. HE staining showed cardiomyocytes injury and hypertrophy in the hearts of TAC rat models. By transmission electron microscope, we observed that mitochondria of cardiomyocytes were damaged seriously in the TAC models. Treatment with losartan used, the selective antagonist of angiotensin II type I receptor could improve the cardiac function of TAC rats. Moreover, losartan treatment decreased the expression of RIPK1 and RIPK3 in heart tissues of TAC rats. The results suggest that necroptosis occurrs in the process of cardiac hypertrophy with pressure overload, and losartan could alleviate the cardiac hypertrophy and inhibit necroptosis.
