1.Expression of adhesion molecule in placenta in patients with hypertensive disorders in pregnancy and its significance

Objective To observe the expression of intercellular adhesion molecule 1(ICAM-1)and vascular cellular adhesion molecuh 1(VCAM-1)in placenta in patients with hypertensive disorders in pregnancy,investigate their roles in patients with hypertensive disorders in pregnancy and correlation.Methods Fifty-two patients with hypertensive disorders in pregnancy(disease group)were collected,including 11 csses with gestational hypertension(GH),18 cases with mild pre-eelampsia(MP)and 23 cases with gevere pre-eclampsia(SP),26 cases of normal pregnant women as control group.Immunohistochemistry and image analytical technique were employed to show the expression of ICAM-1 and VCAM-1.Results The quantities of ICAM-1 and VCAM-1 expressed in the placental vascular endothelial cell were 78.85%(41/52)and 75.00%(39/52)in disease group,26.92%(7/26)and 23.08%(6/26)in control group,there was a significant difference between the two groups(P<0.01).and those in SP patients[100.00%(23/23)and 100.00%(23/23)]were higher than those in MP patients[77.78%(14/18)and 72.22%(13/18)]and GH patients[36.36%(4/11)and 27.27%(3/11)](P<0.05 or<0.01).The quantities of ICAM-1 and VCAM-1 expressed in the placental villous syncytlotrophoblast were cell 38.46%(20/52)and 44.23% (23/52)in disease group,100.00%(26/26)and 96.15%(25/26)in control group,there was a significant difference between the two groups(P<0.01),and those in SP patients[8.70%(2/23)and 13.04%(3/23)]were lower than those in MP patients[44.44%(8/18)and 50.00%(9/18)]and GH patients[90.91%(10/11)and 100.00%(11/11)](P<0.05 or<0.01).Conclusion Abnormal expression of ICAM-1 and VCAM-1 in the placental vascular endothelial and villous syncytiotrophoblast cell in patients with hypertensive disorders in pregnancy may participate in pathogenic process of vascular endothelial damage of hypertensive disorders in pregnancy.
2.Sixteen Cases with Tethered Cord Syndrome

Objective To analyse the pathogenesis,diagnosis and treatment of tethered cord syndrome(TCS) and to improve the knowledge of the disease.Method The clinical data of 16 cases with TCS who underwent surgical lysis were analyzed,retrospectively.Results Thirteen cases were followed-up for 6 months to 1 year.Three cases with handicap of lower limb function and aesthesia and rectum and bladder sphincter dysfunction got good outcome,and 2 cases showed no improvement,while 10 cases with no symptoms(didn′t) appear TCS.Conclusions It is necessary to diagnose and manage as early as possible.The tethered cord lysis can relieve the symptoms of some cases and control development of TCS.
3.Studies on extraction and antioxidation activity of phycobiliproteins from Porphyra haitanensis

Objective To extract phycobiliproteins from Porphyra haitanensis,find out the optimum extraction conditions and investigate the antioxidation activity of phycobiliproteins in vitro and anti-oxygen free radical in vivo.Methods Porphyra haitanensis was extracted with deioned-water at 40 volumes for 6 days and tissue-triturated for 2 minutes.The crude phycobiliprotein(CPP) was purified by precipitating with 45% ammonium sulphate and dialyzing.The scavenging activities of the crude phycobiliprotein on?OH of the system of 1,10-phenanthroline-Fe(Ⅱ)/H2O2 and O-2? of pyrogallol autoxidation were studied.And the mice were treated by gastric incubation with crude phycobiliprotein,at doses of 100,200,300mg?kg-1?d-1.The activities of erythrocyte catalase(CAT) and glutathione peroxidase(GSH-Px),serum superoxide dismutase(SOD) and the content of erythrocyte MDA in mice were determined on the 25th day after treatment.Results CPP could scavenge free radicals,and also can increase the activities of erythrocyte CAT,GSH-Px,serum SOD and decrease the content of MDA significantly.Conclusion The crude phycobiliprotein from Porphyra haitanensis has antioxidation activity in different ways.
4.Effect of preoperative glucose infusion on glucose transport-4 mRNA expression in skeletal muscle in patients undergoing thoracic surgery

Objective The purpose of this study was to examine the effect of preoperative glucose infusion on glucose transport-4 mRNA ( Glut-4 mRNA) expression in skeletal muscle before and after thoracic operation. Methods Twelve male ASA Ⅰ - Ⅱ patients undergoing elective radical surgery for esophagus cancer were randomly divided into 2 groups : (Ⅰ ) control group; (Ⅱ) glucose infusion group. Patients with hypertension, hyperlipidemia or diabetes mellitus were excluded. The patients in both groups were premedicated with phenobarbitol and atropine. Anesthesia was induced with midazolam 0.1 mg ? kg-1 , propofol 1.5 mg ?kg-1 , fentanyl 4?g?kg-1 and vecuronium 0.15 mg?kg-1 . The patients were mechanically ventilated after trachea! intubatioa Anesthesia was maintained with continuous iv infusion of propofol (80?g ? kg-1? min-1 ) and vecuronium (1.0 ?g?kg-1?min-1) supplemented with droperidol 0.1 mg?kg-1 and fentanyl 6 ?g ?kg-1 given in several boluses. In group Ⅱ 5 % glucose (0.25 g?kg-1) was infused 2 h before operation. Blood samples and 1 g of skeletal muscle were obtained while chest was being opened and closed for determination of plasma glucose and insulin levels and Glut-4 mRNA expression in muscle. The total RNA of the muscle cells was extracted by trizol one-step template. The Glut-4 mRNA amplification products were determined using RT-PCR with ?-action mRNA as an internal control. BACKGROUND: Stellate ganglion block (SGB) has become a technique frequently used in the field of pain management for its wide indication spectrum. Its ameliorative effect on cephal-facial pain may be achieved by regulating the cerebrovascular diastolic and systolic functions and intensifying cerebral perfusion.OBJECTIVE: To observe the influence of SGB on the blood velocity of anterior cerebral artery (ACA), middle cerebral artery (MCA), posterior cerebral artery (PCA) and vertebral artery with three-dimensional transcranial Doppler (TCD) in patients with caphal-facial diseases caused by is chemia or vascular spasm.DESIGN: A before-after control observation.SETTINGS: Department of Pain Management, Guizhou Provincial Orthopaedic Hospital; Department of Anesthesiology, the First Affiliated Hospital of Guangzhou Medical College; Department of Anesthesiology, the Second Affiliated Hospital of Sun Yat-sen University.PARTICIPANTS: Twenty patients of 18-50 years old were selected from the Department of Pain Management of Guizhou Provincial Orthopaedic Hospital from March 2004 to December 2005, including 6 cases of migraine,3 of cluster headache, 4 of muscle tension-type headache, 2 of nervous tinnitus and earache, and 5 of derangement of temporomandibular joint.METHODS: Unilateral SGB was performed by means of paratracheal anterior technique, with the following compound prescription: lidocaine (20 g/L)2.5 mL and dexamethasone 1 mg in normal saline with a total volume of 10 mL. SGB was performed once a day for 7 consecutive days as one course.MAIN OUTCOME MEASURES: The changes of blood velocity of ACA,MCA, PCA and vertebral artery were observed with three-dimensional TCD before SGB and at 30 minutes after SGB.RESULTS: All the 20 patients finished the treatment and entered the analysis of results. After treatment, the blood velocities of ACA, MCA and PCA of the blocked side were increased remarkably as compared with those before treatment [(76.20±3.83), (61.95±2.50) cm/s; (83.65±2.36),(77.55±2.69) em/s; (65.20±2.99), (58.70±2.2.35) cm/s; P < 0.01], but that of vertebral artery had no significant change before and after treatment [(47.20±2.04), (45.55±2.23) cm/s, P > 0.05]. CONCLUSION: The blood velocities of the blocked side are obviously increased after treatmeut, the increased blood velocities of ACA, MCA and PCA have important clinical significance for the blood supply of cerebral hemisphere, especially the anterior 2/3 part, but had little great influence on vertebral artery, indicating that SGB has great pertinence to the caphalfacial diseases caused by ischemia or vascular spasm, and can ameliorate the cerebral blood perfusion.
7.Effect of Slug knock-down on tumor formation and metastasis of HCT116 cells in nude mice

Objective:To explore the influence of RNAi mediated Slug silencing on the growth and metastasis of colon cancer in nude mice.Methods: HCT116 colon cancer cells use for 24 five-week-old nude mice implanted subcutaneously , established colon cancer xenograft model in nude mice ,all divided into blank control group ,negative control group and the experimental group ,each group had eight nude mices.All group were injected with saline , negative plasmid and lentivirus vectors respectively.Tumor growth was observed and draw tumor curved growth ,changes in tumor growth and lymph node metastasis between the groups were observed ,Slug gene and protein expression were detected by immunohistochemistry ,qRT-PCR and Western blot analysis.Results: Slug gene shRNA intervention group compared with the control group and negative control group ,tumor grew slower ,tumor mass was significantly reduced (3.08±0.31 vs 7.37±1.18,7.46±1.16,P<0.01),experimental group of lymph node-positive rate was 36.3%( 4 /11 ) ,compared to the negative control group 77.8% ( 14/18 ) and the control group was 68.4% ( 13/19 ) ( P<0.01 ).Conclusion: Targeted Slug RNA interference can significantly inhibit the growth of colon cancer in nude mice ,lymph node metastasis and the expression of the gene protein in cancer tissue ,Slug may be a potential molecular target for colon cancer gene therapy.

Objective:Preformed anti-human leukocyte antigen(HLA)specific antibodies are a major risk for antibody-mediated rejection.The aim of this study is to detect and analyze anti-HLA specific antibodies in sera of renal transplant candidates for evaluating the status of presensitization.Methods:A total of 3 500 patients awaiting renal transplantation in our hospital from 1998 to 2007 were included in the study.Panel reactive antibody(PRA)in sera of 694 candidates was detected by complement-dependent cytotoxicity(CDC)before March 2000,then the sera of other 2 806 recipients were screened by enzyme-linked immune absorbent assay(ELISA)alternatively.The polymorphism,specificity and relevant factors of anti-HLA antibodies were analyzed.Results:Only IgG type of antibodies specific to HLA classⅠcould be detected by CDC,whereas both anti-HLA classⅠand classⅡ of IgG class could be found out by ELISA.Lower PRA positive rate by CDC(8%)was shown when compared to that by ELISA(17%).Furthermore,multiple types of specific antibodies against HLA-A(20),B(37),CW(8),DR(14)and DQ(7)were determined by ELISA.Some types of the antibodies presented higher frequencies,such as anti-HLA-A2,24,68,23 and 32;B27,56,57 and 7;DR7,4,9,13,17 and 12;CW1,2,6,4 and 8.These high frequenies of anti-HLA antibodies were somewhat different from the distribution of HLA antigens in South China population.There were significantly different positive rates of anti-HLA antibodies between the male and the female,as the male were sensitized mainly through blood infusion and the female were sensitized by either blood infusion or pregnancy.Conclusion:Specific antibodies against HLA can be detected out by ELISA accurately,whereby to find high freguencies of the antibodies and to avoid donor-recipient mismatching at HLA-loci.Detection of preformed anti-HLA and reducion of HLA-mismatched blood infusion to reduce production of anti-HLA antibodies may be the valuable pathway to improve graft survival.
