1.Prevention and management of hypertension in the patients with renal diseases

Effective management for hypertension in renal diseases should be carried out according to the principles of 2007 ESH-ESC Practice Guidelines for the Management of Arterial Hypertension.The important role of kidney in the classification of hypertension,the corresponding therapy,the set of the target blood pressure,the choice of antihypertensive agents and combination therapy should be focused.Comprehensive approaches should be used for the treatment of hypertension in renal diseases to protect the target organs(including the kidney).Appropriate prevention and management strategies for hypertension in renal diseases should be disseminated and optimized.
2.Main methods of computational pharmacology and their application in research on traditional Chinese medicine pre-scriptions for treating chronic liver diseases

Traditional Chinese medicine (TCM)has certain advantages in the treatment of chronic liver diseases,especially chronic inflam-matory injury and liver fibrosis.However,the studies on medicinal components and action mechanism of TCM prescriptions are confronted with a great challenge.The computational pharmacology methods used in recent years,mainly including virtual screening technique,omics technologies,and network pharmacology approaches,as well as their application in TCM research,are presented;given the previous re-search work,the progress in research on active ingredients and action mechanism of TCM prescriptions using computational pharmacology methods is reviewed.It is thought that computational pharmacology techniques combined with conventional molecular biological and pharma-cological methods are an important research strategy for the material basis and action mechanism of TCMprescriptions for treating chronic liv-er diseases.

5.Expression of p19 Gene of Avian Leukosis Virus in Escherichia coli

Based on avian leukosis virus ( ALV) p19 gene terminal nucleotide sequence, a 82 bp double-stranded DNA fragment was chemically synthesized and cloned into the expression vector pGEMEX-1. The sequencing result indicated th at the cloned fragment was a correct version of the one designed both in nucleot ide sequence and in its open reading frame. The recombinant was used to transfor m E.coli BL21 (DE3). The cloned fragment was expressed as a fused protein wi th T7 gene 10 leader peptide and was shown to be 34 kD in size on SDS-PAGE gel when induced with 1 mmol/L IPTG. The expression product was able to bind immunol ogically to rabbit anti-ALV serum in Western-blot assay and is being tested to differentiate exogenous from endogenous ALV.
6.Study on correlativity of TCM Syndrome Types of Mild Congnitive Impairment with Metabolism of Free Radicals and Cholin Esterase

Objective:To study on symptom characteristics and TCM syndrome types of mild congnitive impairment(MCI),and reveal the relation of TCM syndrome types of MCI with metabolism of free radicals and cholin esterase.Methods:With reference to internationally acknowledged criteria for MCI diagnosis at present,44 cases of MCI were selected and 20 healthy persons were used as controls.Relative neuropsychological scales tests were conducted and TCM syndrome types were established.Serum acetycholin esterase(AchE),superoxide dismulase(SOD)activities,serum malondialdehyde(MDA)content and DNA defect rate in mitochondria of white cells were determined.Results:27 cases(61.4%)of deficiency syndrome,17 cases(38.6%)of coexistence of deficiency and excess syndromes and 93.18% patients of MCI had the symptoms of kidney-deficiency and hollow marrow.Serum MDA level and AchE activity and the DNA defect rate of mitochondria in white blood cells in the patients of deficiency syndrome and coexistence of deficiency syndrome and excess syndrome increased significantly and SOD activity and memory quotient significantly decreased as compared with the healthy persons.Memory quotient,serum MDA level and AchE activity in the patients of coexistence of excess and deficiency syndromes increased significantly and SOD activity decreased significantly as compared with the patients of deficiency syndrome.Conclusion:TCM syndrome types of the patient of MCI have objective substance basis,i,e.varying degrees of memory impairment and disturbances of metabolism of free radicals and AchE.
7.Clinical observation on insulin pump in the treatment of diabetes mellitus

The effects of continuous subcutaneous insulin injection (CSII, n=306) and multiple subcutaneous insulin injection (MSII, n=310)on hyperglycemia in patients with diabetes mellitus were compared. The results suggest that the effect of CSII is more prompt and efficient with lower incidence of hypoglycemia as compared with MSII.
8.Contrast-enhanced ultrusonography in the diagnosis of hyperechoic renal tumors

Objective To investigate the value of the diagnosis of contrast-enhanced ultrasound(CEUS) in renal tumors. Methods Nineteen patients with hyperechoic renal tumors detected by CEUS underwent resection for suspected renal malignant and benign tumors. The characterization of hyperechoic renal tumors on CEUS was evaluated. Results Histopathologic results of resected lesions showed benign tumors in 11 patients with angiomyolipoma and malignancy in 8 patients with clear cell carcinoma. Six patients with angiomyolipoma showed rapid wash in contrast enhancement and slow wash out on all tumor and focal tumor (6/11). One patient with angiomyolipoma was observed in rapid wash in and wash out (1/11). Four patients with angiomyolipoma showed rapid wash in and negative contrast enhancement after rapid wash out(negative contrast enhancement compared to renal cortex,4/11). Five patients with clear cell carcinoma showed rapid wash in and wash out (5/8). Two patients with clear cell carcinoma showed rapid wash in and delayed wash out (2/8). One patient with showed no enhancement on CEUS (1/8). The pseudocapsule could be seen at 5 patients with clear cell carcinoma. Wash in enhancement and wash out on clear cell carcinoma were significant in comparison to these of angiomyolipoma (P<0.05). Wash in enhancement,delayed wash out and negative contrast enhancement were no significant between clear cell carcinoma and angiomyolipoma (P>0.05). Conclusions CEUS is useful for discriminating between benign and malignant renal tumor.
9.Application of contrast-enhanced ultrasound in the diagnosis of renal hematoma and active hemorrhage

Objective To explore the value of contrast-enhanced ultrasound(CEUS) in the diagnosis of hematoma and active hemorrhage of renal trauma. Methods Totally 28 patients with renal trauma were examined by conventional ultrasound and CEUS,respectively,including 24 cases caused after renal biopsy,4closed trauma. The detectability of renal hematoma and active hemorrhage with these two methods were compared. All patients were identified by CT or follow-up studies with ultrasound. Results The detectability of renal hematoma with conventional ultrasound and CEUS were 67. 86% (19/28), 92. 86%(26/28), respectively. There was statistically different for detectability in the diagnosis ( P <0.05), and the extent of hematoma was more obvious with CEUS. CEUS diagnosed 7 of 26 were renal hematoma with active hemorrhage,which were difficult to be detected with conventional ultrasound. For renal hematoma,the features of CEUS were no enhancement found in every phase; for renal hematoma with active hemorrhage,the contrast agents overflowed from injured blood vessels and formed irregular remarkable enhanced regions. Conclusions CEUS is useful in diagnosing hematoma and identifying the extent and active hemorrhage of renal trauma,in addition,CEUS is valuable in detecting complications after renal biopsy.
10.Screening and identification of HIV-1 gp41 C-helix mimotopes displayed on phages

Objective:To find small molecular leads for inhibition on early stage of HIV infection by identification and characterization of the HIV-1 gp41 C-helix mimotopes.Methods:For identification of the gp41 C-helix mimotopes,C7C phage display peptide library was biopanning by using a synthetic peptide N36 which was derived from the gp41 N-helix as target.After three rounds of screening,positive phage clones were identified by ELISA and sequenced.Results:16 of 26 phage clones were identified to bind with peptide N36,and 10 of them were sequenced.Every clone of ten clones contains at least two hydrophobic residues,which may dock into the hydrophobic pocket in the gp41 N-helix domain.9 of the 10 clones have a conservative sequence WW,which may mimic the W628 and W631 in C-helix to interact with the hydrophobic residues in the gp41 pocket.One clone expressing the conservative sequence named clone No.8(CYWWHRLHC) was selected for characterization.The binding between the clone No.8 and N36 was blocked by free peptide N36.And the binding between clone No.8 and peptide N36 was inhibited by peptide C34(IC 50=12.5 ?g/ml).Conclusion:The short circular peptides displayed on phages containing WW residues may mimic the conformational epitope of the HIV-1 gp41 C-helix to interact with the N-helix.This information may be useful for design of HIV-1 fusion inhibitors.
