1.Establishment of an in vitro model of brain blood barrier

Objective To study the dynamic changes of transendothelial electrical resistance (TEER) on our in vitro model of brain blood barrier (BBB) made from primary culture of BALB/c mouse brain microvascular endothelial cells (BMVEC), and to explore the relation between TEER and BBB permeability, and search for the best culture condition. Methods BMVEC were isolated from BALB/c mouse and cultured on a transwell insert with special micro-pore. The cells were identified with immunohistochemical methods and electron microscope. TEER over BMVEC was measured after BBB model establishment for determining the 3H-Glucose permeability of BBB in vitro. Results BMVEC cultured in the transwell insert exhibited typical "flagstone" appearance and in a tight monolayer structure under electron microscope. Immunohistochemical detection of ZO-1 protein, a marker antigen of tight junction, showed smooth, continuous and tight junctions between confluent BMVEC. TEER over BMVEC monolayer increased to (346?10) ?/cm2 when the permeability for 3H-Glucose was decreased to the minimum. Conclusion BBB model in vitro made from primary culture of BMVEC in transwell has the basic characteristics of BBB in morphology, electrical resistance and permeability.
2.Effects of Glucagon Peptide-1 on Blood Pressure Variability in Diabetic Hypertensive Patients

Objective To investigate the role of glucagon peptide-1(GLP-1) in blood pressure variability (BPV) in diabetic hypertensive patients. Methods A total of 120 diabetic hypertensive patients were included in this study. GLP-1 was used for the treatment. The values of body mass index (BMI), fasting blood glucose, glycosylated hemoglobin, creatine and 24-hour ambulatory blood pressure (BP) monitoring were detected before and after treatment with GLP-1 for six months. The clinical indices were compared before and after treatment. Results The values of fasting blood glucose (mmol/L:7.12± 0.64 vs 9.19±2.78), glycosylated hemoglobin (%:7.00±0.14 vs 8.28±1.32) and BMI (kg/m2:19.30±3.24 vs 24.5±4.53 ) were de-creased to the normal level after GLP-1 treatment for six months (P<0.05). There were significant decrease in the 24-h sys-tolic BP (mmHg:135.02 ± 16.57 vs 139.52 ± 15.60), daytime systolic BP (mmHg:132.5 ± 14.6 vs 136.44 ± 14.24), 24-h systolic BP variability (mmHg:12.20 ± 1.44 vs 12.73 ± 1.66), daytime systolic BP variability (mmHg:11.11 ± 1.48 vs 11.74 ± 1.52 ), and daytime diastolic BP variability (mmHg:7.03±1.42 vs 7.43±1.45) after GLP-1 treatment for six months (P<0.05). Conclu-sion GLP-1 can decrease BPV in diabetic hypertensive patients.
3.Efficacy of anatomical locking plate combined with anchor nail in the treatment of Neer Ⅱ distal clavicle fracture

Objective To observe the efficacy of anatomical locking plate combined with anchor nail internal fixation and clavicular hook plate in the treatment of Neer Ⅱ distal clavicle fracture and their impact on shoulder function.Methods Eighty patients with Neer Ⅱ distal clavicular fracture treated in the Eighth People's Hospital in Shanghai from January 2015 to December 2016 were divided into observation group and control group,each group 40 cases.The observation group was treated with anatomical locking plate combined with anchor nail,the control group was treated with clavicular hook plate.The efficacy,operation time,intraoperative blood loss,fracture healing time,postoperative hospital stay,shoulder function score and incidence of complication were observed in the two groups.Results The excellent rate was in the observation group was higher than that in the control group (97.50%(39/40) vs.80.00%(31/40),χ2=2.477,P<0.01).The operation time,intraoperative blood loss and postoperative hospital stay in the observation group and the control group were (53.36±4.69) min vs.(54.19±5.13) min,(69.65±7.17) ml vs.(70.38±8.29) ml,(7.39±1.64) d vs.(7.42±1.97) d,the differences between the two groups had no statistical significance (P>0.05);the fracture healing time in the observation group was longer than that of the control group ((23.65±2.19) w vs.(26.9±3.46) w,P<0.01).The shoulder joint Constant-Murley score in the observation group was (86.02±5.19) points,control group (68.46± 6.94) points,the difference was statistically significant (P<0.01).Observation group showed 2 cases of pain around acromion,the incidence rate of complications was 5.00%(2/40);7 cases in the control group had pain around acromion,2 cases suffered from impingement syndrome and 1 cases had delayed union,the incidence rate of complications was 25.00% (10/40).Conclusion Anatomical locking plate combined with anchor nail internal fixation is effective in the treatment of Neer II distal clavicle fracture.It has the advantages of fast fracture healing,good functional recovery of shoulder joint and less complications.
4.Expressions of CD147 and EGFR protein and their relationship with metastasis and prognosis of gastric cancer

Objective To investigate the expressions of extracellular matrix metalloproteinase inducer molecule CD147 and epidermal growth factor receptor (EGFR) protein in gastric cancer and their relationship with the pathological behavior and prognosis.Methods Immunohistochemical staining was used to detect the expressions of CD147 and EGFR protein in 63 cases of gastric cancer and 40 cases of paracancerous normal tissues.The study was also combined with analysis of the pathological behavior and clinical follow-up survey of gastric cancer.Results The positive expression rates of CD147 and EGFR protein in gastric cancer were 77.8% (49/63) and 42.9% (27/63),respectively,which were higher than those in paracancerous normal tissues 25.0% and 0 (x2 =27.851,P =0.000;x2 =23.233,P =0.000).In gastric cancer,the expressions of CD147 and EGFR were closely related to the depth invasion (x2 =5.992,P =0.014;x2 =13.148,P =0.000),TNM stages (.x2 =8.481,P=0.004;x2=14.589,P=0.000),lymph node metastasis (x2 =5.270,P=0.022;x2=20.261,P =0.000) and prognosis of patients (x2 =5.606,P =0.018;x2 =4.982,P =0.026).The expression of CD147 was also significantly correlated with Ki-67 expression (x2 =10.481,P =0.001).CD147 expression was positively correlated with EGFR expression(r =0.309,P =0.014).Conclusion The expressions of CD147 and EGFR are closely related to carcinogenesis,metastasis and survival time of gastric cancer.Detecting the expressions of CD147 and EGFR may be prognostic indicators of gastric cancer.
5.Liraglutide promotes the reduction of blood pressure and drives the water and salt through in renal medulla of type 2 diabetes rats

Objective To observe the effects of glucagon like peptide-1 (GLP-1) analogues liraglutide on expressions of nitric oxide synthase (NOS) and cyclo-oxygen-ase (COX)2 in renal medulla of type 2 diabetes rats, and the mechanism of its lowering blood pressure and promoting excretion of water and salt in kidney. Methods Type 2 diabetes model rats were generated by high-fat and high-sugar feeding for 8 weeks followed by intraperitoneal injection of streptozotocin (STZ). Subse?quently, eighteen type 2 diabetes rats were divided into two groups: liraglutide treatment group (DMT) and diabetes group (DM). Twelve normal rats were divided into two groups: liraglutide treatment wild type group (WTT) and wild type group (WT). DMT and WTT groups were given liraglutide (200μg/kg) by subcutaneous injection, DM and WT groups were given equivalent normal saline by the same way. The levels of blood glucose and blood pressure were detected at 0, 2, 4 and 6 weeks after treatment in groups of rats. Samples of urine were collected for detecting ion concentrations (K+, Na+and Cl-) af?ter treatment for six weeks. Rats were sacrificed and blood samples were collected for detecting ion concentrations (K+, Na+and Cl-). The expression levels of NOS and COX2 mRNA and protein in renal medulla were detected by real-time PCR and Western blot assay. Results After treating with liraglutide, the values of blood glucose (F=5.933, P<0.05) and blood pres?
sure (F=22.070, P<0.05) were gradually decreased in DMT group. After treatment with liraglutide for 6 weeks, the values of blood glucose (mmol/L:12.78 ± 3.82 vs. 18.75 ± 1.68) and blood pressure (mmHg:119.98 ± 4.43 vs. 136.42 ± 4.48) were signifi?cantly decreased (P<0.05) in DMT group than those of DM group (P<0.05). There were no significant differences in the concentrations of K+, Na+and Cl-between the two groups. There were higher levels of K+(mmol/L:46.55 ± 6.43 vs. 33.13 ± 9.71), Na+(mmol/L:56.33±8.83 vs. 41.20±7.25) and Cl-(mmol/L:159.81±25.06 vs. 71.44±12.99) in urine in DMT group than those of DM group (P<0.05). The mRNA levels and protein expressions of NOS and COX2 in renal medulla were significant?ly increased in DMT group than those of DM group (P<0.05). Conclusion GLP-1 analogues liraglutide may enhance the expression of COX2 by increasing the expression of NOS to excrete water and salt, and decrease blood pressure.

8.Research on correlation between lung and large intestine based on meridian and acupoint palpation in patients with bronchial asthma.

OBJECTIVETo explore correlation between lung and large intestine and the two meridians under pathological condition in the view of meridian theory.
METHODSNinety-six cases of bronchial asthma were applied palpation at the running course of 12 regular meridians under the elbow and knees and back-shu points. And abnormal reactions were recorded, the affected meridians and back-shu points were discovered.
RESULTSThe abnormal reactions most frequently appeared on the Lung Meridian, followed by the Large Intestine Meridian, the Spleen Meridian, the Liver Meridian, the Stomach Meridian and the Triple Energizer Meridian. And the unusual reaction of the back-shu points most frequently appeared on Feishu (BL 13), and Dachangshu (BL 25) and Pishu (BL 21) followed as the next two.
CONCLUSIONThe existence of correlation between the Lung Meridian and the Large Intestine Meridians under pathological condition can be proved through meridian and acupoint palpation on bronchial asthma patients.

Objective To study the effects of found in inflammatory zone 1(FIZZ1) on the migration of cul- tured mouse vascular smooth muscle cell(VSMC) and the potential mechanism.Methods Using Boyden chamber, immunofluorescence and Western blot,the migratory effects,F-actin content and Akt/Akt1 expression were deter- mined in VSMC after stimulation by FIZZ1.Results FIZZ1 markedly increases the migratory ratio,F-actin con- tent and the Akt1 expression of VSMC.Ly294002,a PI3K inhibitor,attenuated migratory ratio,F-actin content and the Akt1 expression of VSMC promoted by FIZZ1 in a dose-dependent manner (10-30 ?mol/L).Conclusion Our data demonstrated FIZZ1 by activating PI3K/Akt1 pathway induced the expression of F-actin of VSMC,and promoted the migration of VSMC.
