1.Clinical observation of coronary artery bypass grafts in the same period of heart valve replacement on the treatment of coronary heart disease with heart valve disease

Objective To analyze and discuss the value and significance of coronary artery bypass grafts in the same period of heart valve replacement in the process of the coronary heart disease with heart valve disease treatment, in order to improve the effect of treatment and reduce the incidence of adverse events. Methods 60 cases with coronary heart disease with heart valve disease , which admitted in our hospital during February 2013-February 2015, were selected as the research objectaccording to random number table method and divided into the-experimental group and the control group , each group of 30 cases according to the principle of double blind control. The experimental group was given the coronary artery bypass grafts in the same period of heart valve replacement and the control group was given the coronary artery bypass grafts without heart valve replacement. The curative ef-fects of different schemes and effects on cardiac function after surgery of the two groups were compared. Results After statistical analysis found thatthe observation indexes of the control group was significantly increased compared with the experimental group, the difference was significant statistically significant (P<0.05). The postoperativecar-diac function indexes of the experimental group was significantly improved then control group , the difference was significant statistically significant (P<0.05). Conclusion Coronary heart disease with heart valve disease in the process of treatment can also take heart valve surgery combined coronary artery bypass surgery. By preoperative and postoperative symptomatic treatment, intraoperative protect myocardium, can effectively prevent and reduce the op-eration of various complications, which is worthy of clinical popularization and application.
2.Burdens of Spouse Caregivers of Patients with Dementia at Home

Objective: To explore the burden of spouse caregivers of patients with dementia at home and factors having influence.Methods:Forty-two patients with dementia were collected from psychiatric out-clinic consecutively. Trained investigators did home visit to collect data on severity of the disease (CDR, MMSE), pathological behavior (BEHAVE-AD), activity of daily living (ADCS-ADL) and burdens of their spouse as caregiver (CBI). The caregivers' anxiety, depression (Zung's Anxiety Scale and Depression Scale) and knowledge about dementia were also collected.Results:The caregivers felt big burden (BDI total score: 30.7?15.1). Nearly half of them (42.9%) were in depression and one quarter (23.8%) were anxious. They were lack of knowledge about dementia and unskillful in care-giving. 26.2% caregivers thought dementia is curable or equal to normal aging, 57.1% of them did not believe it is an improving disease. 3.The prevention and treatment effect of ω-3 fatty acids on intestinal mucosa in critical illness

Objective To study the prevention and treatment effect of ω-3 fatty acids on intestinal mucosa in critical illness. Methods Forty patients including severe trauma, infection shock were enrolled as experimental group, while 30 healthy people as control group. At the same time, the patients in expermental group were randomly divided into group A and group B(20 cases each). While the patients were. Treated with low calorie parenteral nutrition totally, those in group A received ω-3 fatty acids additionally. The plasma concentrations of dimnine oxidase (DAO), endotoxin were detected by spectrophotography, and TNF-α was detected by ELISA. Results After treatment the concentration of DAO, endotoxin, TNF-α in group A and that of endotoxin in group B decreased significantly (P<0.05 ). While there was no significant difference of endotoxin levels between group A and group B. After therapy, DAO and TNF-α levels in group A were sig-nificantly lower than those in group B (P<0.05 ). The concentrations of DAO and TNF-α in group B were also significantly higher than those in control group (P<0.05). Conclusion ω-3 fatty acids can prevent and treat critical intestinal mucosa effectively.
4.The effects of simulated family nursing on the cognitive function and activity of daily living in patients with Alzheimer's disease

Objective To evaluate the effects of simulated family nursing on the cognitive function and activity of daily living in patients with Alzheimer's disease.Methods Sixty-eight patients with Alzheimer's disease were assigned to routine nursing condition and simulated family nursing condition.The patients were assessed with Mini-Mental State Examination (MMSE) and Activity of Daily Living Scale (ADL) before intervention,and 3 months and 6 months after intervention.Results Compared with the control group,the patients' cognitive function (t=2.31,P=0.026) and activity of daily living (t=2.59,P=0.012) were improved significantly in the experimental group.Conclusion The simulated family nursing can improve the cognitive function and activity of daily living in patients with Alzheimer's disease.
5.Endoscopic implantation of 5-FU slow-release particles for advanced gastroenteric tumor

Objective To investigate the feasibility, safety and short-term efficacy of endoscopic implantation of 5-FU Slow-release Particles for advanced gastroenteric tumor. Methods During the endoscopy procedure,slow-releasing 5-FU agents were implanted densely into the tumors and infiltrated area. Forty-five to sixty pieces of agents ( each piece equivalent to 1.67 mg 5-FU)were injected, which containing an average dose of 100 mg 5-FU. Results A total of 13 advanced gastric cancer patients were enrolled into this study. Significant effects were observed in 3 patients and good effects in 8 patients, but 2 cases with no effects. The total effective rate was 84.62%. The endoscopy examination showed that the size of tumors reduced in various degree at 1 - 2months after the implantation. No hemorrhage or perforation was observed. Parameters of hepatorenal function and routine blood test were stable after implantation. Conclusion Endoscopic implantation of 5-FU Slow-release Particles can relieve the symptoms of patients and limit tumor growth in advanced gastroenteric tumor with no marrow and hepatorenal functional impair.
6.Preparation of 99Tcm-MAG3-VEGF125-136 polypeptide and its biodistribution and imaging in osteosarcoma mice

Objective To synthesize 99Tcm labeled VEGF fragment (VEGF125-136) and evaluate its biodistribution and imaging in the nude mice bearing human osteosarcoma,in order to find a probe to improve the early diagnosis and targeted therapy of osteosarcoma.Methods VEGF125-136 was synthesized by conventional solid phase synthesis method,and directly coupled with MAG3 to form MAG3-VEGF125-136,then MAG3-VEGF125-136 was labeled with 99Tcm.The osteosarcoma bearing nude mice were randomly divided into four groups by random number table and studied for 99Tcm-MAG3-VEGF125-136 distribution at 0.5,1,2,4 h,respectively.The other 10 osteosarcoma bearing nude mices were divided into the experimental group and competitive inhibition group,and underwent radionuclide imaging.The T/NT ratio was calculated.Data were analyzed using two-sample t test.Results The radioactive labeling rate was greater than 95%,radiochemical purity was still greater than 85% after 12 h in room temperature.The 99Tcm-MAG3-VEGF125-136 was cleared fast in blood and mainly excreted through the kidneys and liver.Tumor uptake of 99Tcm-MAG3-VEGF125-136 was highest at 1 h with (0.28±0.02) %ID/g.The T/NT ratios of experimental group at 0.5,1,2,4 h were 1.07±0.10,3.10±0.19,2.63±0.14,1.90±0.09,respectively.In competitive inhibition group,no obvious radioactivity concentration was observed in tumors.The T/NT ratios were 0.99± 0.05,1.06± 0.06,0.98±0.03,0.97±0.03 at 0.5,1,2,4 h respectively.The T/NT ratios were significantly different between the two groups at 1,2,4 h (t=22.52,26.36 and 22.31,all P<0.05),but no significant difference was observed at 0.5 h (t =1.44,P>0.05).Conclusion 99Tcm-MAG3-VEGF125-136 has high labeling efficacy,good stability and high specificity to osteosarcoma.It can be used as an agent for early diagnosis and therapy monitoring in osteosarcoma.
7.Comparison of the Main Components between Traditional Decoction and Dispensing Granule Decoction of Huanglian Jiedu Decoction

Objective To compare the contents of berberine,palmatine,jatrorrhizine,baicalin and geniposide between traditional decoction and dispensing granule decoction of Huanglian Jiedu Decoction.Methods A HPLC method was carried out on Diamonsil C18 column(250 ? 4.6 mm,5 ?m).The mixture of H2O(A),CH3OH(B)and 0.05 % H3PO4 solution(C)was used as the mobile phase for gradient elution.The column temperature was set up at 35 ℃ and the flow rate was l mL/min.The detection wavelength was 345nm for berberine,palmtine and jatrorrhizine,280 nm for baicalin,and 238 nm for geniposide.Results This method can be used to determine 5 components in Huanglian Jiedu Decoction at the same time and the method was rapid,sensitive and accurate.Conclusion The HPLC chromatograms of traditional sliced decoction of Huanglian Jiedu Decoction are similar to the dispensing granule decoction.The contents of 5 main components in dispensing granule decoction are higher than those in traditional sliced decoction.
8.Effects of Hawthorn Leaf Procyanidins on Over Expressions of ICAM-1 and E-selectin in Human Umbilical Vein Endothelial Cells Induced by TNF-α

Objective To observe the effects of hawthorn leaf procyanidins (HLP) on over expressions of ICAM-1 and E-selectin in human umbilical vein endothelial cells (HUVEC) induced by TNF-α,and clarify the mechanism of HLP's anti-inflammation effect. Methods HUVEC were cultured in vitro. MTT assay was used to detect cell viabilities. The expressions of ICAM-1 and E-selectin in HUVEC were detected by flowcytometry. Results Up to 200 mg/L, HLP showed no significant decrease in cell viabilities; the expression levels of ICAM-1 and E-selectin in the model group significantly increased, compared with that in the normal group; 10, 20, 30, 40, 50 mg/L HLP inhibited the expression elevations of ICAM-1 and E-selectin in concentration-dependent manner; and there were statistical significances in 40, 50 mg/L HLP groups, compared with the model group. Conclusion HLP can inhibit the over expressions of ICAM-1 and E-selectin of vascular endothelial cells induced by TNF-α, which possibly underlies HLP's anti-inflammation effect.
9.3D printed tissue engineering scaffolds combined with bone marrow mesenchymal stem cells transplantation to repair spinal cord injury

Objective:Through histological analysis, immunofluorescence staining, electrophysiological detection and Sensory and motor function evaluation to investigate the effects of 3D printed hydrogel scaffold combined with bone marrow mesenchymal stem cells (BMSCs) in promoting functional recovery of spinal cord injury.Methods:10% GelMA hydrogel and 10 6 U stem cell suspension were prepared into bioink of appropriate concentration to construct the biomimetic spinal cord scaffold through 3D printing platform. The scaffold was placed in the medium and cultured in an environment of 37 ℃ CO 2 incubator. The microstructure of the scaffolds and the distribution of BMSC in the scaffolds was observed by scanning electron microscope. CAM/PI staining and confocal microscopy were used to observe the survival of stem cells in the scaffolds and determine the biocompatibility of the scaffolds. The scaffolds were implanted into the subcutaneous tissues of the back of rats, and the subcutaneous tissues were determined by HE staining to detect the immunogenicity of the scaffolds. After the rat model of hemicytoma defect was made, stents were transplanted for treatment, and confocal microscopy was used to evaluate the regeneration of neurons and axons in local area of spinal cord injury. At the same time, BBB score was used to evaluate motor function, mechanical pain score was used to evaluate sensory function, and surface electrode detection method was used to evaluate electrophysiological recovery weekly. Results:The long spindle shaped BSMC were uniformly distributed in the scaffold with a loose reticular structure. The scaffolds had good biocompatibility, and the cell survival rate of the prepared scaffolds reached 96% after 24 hours of printing. After 28 days of subcutaneous transplantation, the immune rejection was mild and immunogenicity was low. It was shown that the regenerated spinal cord tissue in the treatment group was significantly increased compared with the control group, which was widely distributed with cells after 28 days by HE staining. It was confirmed that part of the regenerated spinal cord tissue was neurons by immunohistochemical staining.Compared with the injured group, the regeneration of neurons and axons in the treatment group were significantly increased by immunofluorescence staining and confocal microscopy. In the treatment group, the BBB score recovered to 10 points, while the control group only recovered to about 1 point in the first week, which was statistically significant. And it recovered to 17 in the fourth week, while the control group only recovered to about 4 point in the four week, which was statistically significant. The Angle of inclined plate support of the treatment group was restored to 40 degrees, while it was only restored to 22 degrees in the control group. The pain threshold of the treatment group decreased to 18.5 points, which was not statistically different from that of the control group. The latent recovery effect of electrophysiology in the treatment group was the same as that in the sham operation group and better than that in the control group.Conclusion:3D printing hydrogel scaffold with loose network structure is suitable for cell proliferation. It has well biological survival, low cytotoxicity and low immunogenicity, which promoted neurons and axons to recovery and extend so as to effectively promote the recovery of motor function, sensory function and neural signal transmission rate after spinal cord injury.
10.Effects of hawthorn leaf polymeric procyanidins on calcium mobilization in human umbilical vein endothelial cells

AIM:To observe the effects of hawthorn leaf polymeric procyanidins ( PPC) on calcium mobiliza-tion of vascular endothelial cells , and to study the underlying mechanism .METHODS: Free calcium in cultured human umbilical vein endothelial cells (HUVECs) was labeled with Fura-2.HUVECs were treated with ATP, a positive control drug, and PPC at concentrations of 12.5, 25 and 50 mg/L..The intracellular calcium concentrations were measured with a living cell microscope for 30 min.RESULTS:PPC concentration-dependently increased the intracellular calcium concen-tration of HUVECs .The intracellular calcium concentrations in 25 and 50 mg/L PPC groups were significantly higher than that in normal group (P<0.01).The dynamic manner of calcium concentration elevations elicited by PPC was a slow in -crease which happened after a latency time of several minutes , lasted for several minutes , and reached a plateau finally . This manner was quite different from that elicited by ATP , a typical SOC operator , hinting different mechanisms between them .Inhibiting the intracellular calcium release did not influence the effects of PPC;however , deleting extracellular calci-um, inhibiting the reverse mode of Na +-Ca2+exchange, or deleting extracellular sodium , restrained or even abolished the effects of PPC.CONCLUSION:PPC elicits calcium mobilization in vascular endothelial cells , which may be one of the mechanisms of the vascular modulatory activity of hawthorn procyanidins .This effect may be achieved through inducing the influx of sodium and then activating the reverse mode of Na +-Ca2+exchange.
