1.Specific expression of nucleolin in cap-stage tooth germ of mouse

Objective To investigate the specific expression of nucleolin in the development of cap-stage tooth germ of mouse embryo,and detect the possible biological function of nucleolin. Methods In situ hybridization and immunohistochemistry were performed to detect the expression of nucleolin mRNA and protein in the development of tooth germ at embryo day 15(E15).Double staining for apoptotic cells and nucleolin was employed to explore the relationship between nucleoin and apoptosis in tooth germ. Results The expression of nucleolin mRNA and protein was mainly detected in the cervical loop,inner epithelium and the underlying dental papilla.Moreover,nucleolin protein was also located in the primary enamel knot and basement membrane.It was revealed by double staining that the fluorescence images of nucleolin and the location of apoptotic cells were not overlapped. Conclusion The expression pattern of nucleolin mRNA do not completely coincide with that of nucleolin protein at the cap stage of tooth germ.The location for nucleolin protein in the basement membrane suggests that nucleolin may be involved in the reciprocal interactions between the inner epithelium and dental papilla mesenchyme,subsequently affect the morphogenesis of tooth germ.
2.Clinical Observation of Modified Yanghe Decoction Combined with Analgesic Western Medicine in Relieving Cancerous Pain of Osseous Metastasis Patients

Objective To observe the therapeutic effect of modified Yanghe Decoction combined with analgesic western medicine on relieving cancerous pain and constipation and on improving the quality of life ( QOL) of osseous metastasis patients. Methods A total of 200 patients with cancerous pain were evenly randomized into treatment group and control group in the admission order. The control group was given oral use of oxycodone hydrochloride controlled-release tablets, and the treatment group was given modified Yanghe Decoction on the basis of treatment for the control group. Twenty-one days constituted one treatment course. After one treatment course, the effect on relieving pain and constipation and on improving QOL were evaluated in both groups. Results (1) The analgesic efficiency was 95.0 % in the treatment group, and was 81.0 % in the control group, the analgesic efficiency of the treatment group was better than that of the control group ( P<0.05) . ( 2) Constipation occurred in 92 cases of the treatment group and was relieved to various degrees in 88 cases after treatment, with an efficiency of 95.7%. In the control group, constipation occurred in 91 cases, but was not relieved after treatment. The treatment group had better effect on relieving constipation than the control group (P<0.05) . (3) The total effective rate on improving QOL was 89.0% in the treatment group and was 71.0% in the control group, the difference being significant (P<0.05) . (4) Both groups had 2 cases of drug-induced mild hepatic damage, and the damage disappeared after symptomatic treatment . Conclusion Modified Yanghe Decoction combined with analgesic western medicine shows good toxicity-reducing and efficiency-enhancing effect for the treatment of cancerous pain.
3.Clinical Efficacy of Chinese Traditional Medicine Combined Western Medicine in the Treatment of Cirrhosis with Ascites and Hypoalbuminemia

Objective To explore the clinical efficacy of Chinese traditional medicine combined with western medicine in the treatment of cirrhosis with ascites and hypoalbuminemia. Methods The trial group was treated with Chinese traditional medicine (modified decoction for Zhongman Fenxiao) combined with western medicine (drugs for protecting liver, diuretis, etc.). The control group was simply treated with Western medicine. Results After 4 weeks of treatment, the cure rate of the trial group was 43.75% and the total effective rate was 87.50%, significantly higher than that of the control group with 30.00% and 80.00%, respectively (P 0.05). 4.Clinical Observation of Tegafur,Gimeracil and Oteracil Potassium Combined with Oxaliplatin Assisting with Radiofrequency Ablation in the Treatment of Liver Metastases after Radical Operation of Rectal Can-cer

OBJECTIVE:To observe therapeutic efficacy and safety of tegafur,gimeracil and oteracil potassium combined with oxaliplatin assisting with radiofrequency ablation(RFA)in the treatment of liver metastases after radical operation of rectal cancer. METHODS:A total of 130 patients with liver metastases after radical operation of rectal cancer were randomly divided into control group and observation group,with 65 cases in each group. After RFA one week,control group was given Oxaliplatin injection 130 mg/m2 intravenously,d1,more than 3 h. Observation group was additionally given Tegafur capsule,for consecutive 14 d,drug with-drawal of 7 d,as a treatment course. A treatment course lasted for 3 weeks,and both groups received 2 courses of treatment. Short-term efficacies of 2 groups were observed as well as complete ablation rate,local recurrence rate,survival rate and the occur-rence of toxic reaction. RESULTS:The total remission rate,disease control rate,complete ablation rate,1-year,2-year and 3-year survival rates of observation group were significantly higher than those of control group,and local recurrence rate was significantly lower than control group,with statistical significance(P<0.05). There was no statistical significance in the incidence of toxic reac-tion between 2 groups(P>0.05). CONCLUSIONS:Tegafur,gimeracil and oteracil potas siucn combined with oxaliplatin assisting with RFA shows significant therapeutic efficacy for liver metastase after radical operation of rectal cancer,and can significantly de-lay disease progression,reduce local recurrence rate and prolong survival time,and does not increase the occurrence of toxic reac-tion.

6.Prospective study of radical resection of breast cancer with preservation of intercostobrachial nerve

Objective To explore the clinical significance of preserving the intercostobrachial nerve (ICBN) in the radical resection of breast cancer. Method All of 140 cases of breast cancer with Ⅰ - Ⅲ a stage operable eligible,were randomly divided into the experimental group with 79 cases (7 cases were rejected, 1 case was lost) that patients received radical resection of breast cancer with preservation of ICBN,and the control group with 61 cases (2 cases were lost) that patients received routine radical resection of breast cancer based on the time of admission. Analyzed postoperative local recurrence and compared to sensory dysfunction of the two groups. Results There was no significant difference between the two groups of patients in the rate of postoperative local recurrence [0-4.2% ( 3/71 ) vs. 0-3.4% (2/59)] ( P > 0.05 ). The local sensory disturbance of the experimental group [2.8% (2/71 )-11.3% (8/71)] was significantly lower than the control group [54.2%(32/59)-71.2%(42/59)], there was significant difference between the two groups (P < 0.01 ). Conclusion Radical resection of breast cancer with preservation of ICBN is an ideal operation in breast cancer.
7.Clinical efficacy of open reduction and internal fixation plus radial head resection in treatment of radial head fractures

Objective To investigate the prognostic outcome of open reduction and internal fixation(ORIF)with radial head resection in treatment of comminuted radial head fractures so as to discuss the corresponding operation options.Methods A series of 23 patients with Mason type Ⅲ or Ⅳ radial head fractures were enrolled in the study including 11 males and 12 females.Eight patients(three with Mason type Ⅲ and five with Mason type Ⅳ)underwent radial head resection and 15(seven with Mason type Ⅲ and eight with Mason type Ⅳ)received open reduction and internal fixation including nail and plate fixation in eight and screw and K-wire fixation in seven.The outcomes were assessed in regard of range of motion,radiographic findings,visual analogous scale(VAS)of pain,patient satisfaction degree,disabilities of the arm,shoulder and hand score system(DASH)and the Broberg & Morrey elbow performance score system.Results The follow-up period ranged from 12 to 65 months(mean 35 months),which showed fracture healing in all patients,with no traumatic arthritis happened.The mean range of flexion and extension arc at the elbow of ORIF group and resection group was 132°and 79.4°(P<0.01)and the patient satisfaction was 9.1 and 8.0(P <0.05)respectively.According to the Broberg & Money elbow performance score system,the score was average 93.9 in the ORIF group,with excellent result in nine patients,good in five and fair in one,while the score was average 80.6 in the resection group,with excellent result in two patients,good in two and fair in four.There was statistical difference between two groups(P <0.01).The DASH score of the ORIF and resection group was 2.9 and 18.1 respectively(P<0.05).Conclusions ORIF has better elbow motion,elbow function,daily living and satisfactory outcomes than radial head resection and is recommended for treatment of Mason type Ⅲ or Ⅳ radial head fractures for repair of the radial head as far as possible.
8.Limping gait improvement by femoral lengthening in ankylotic hips and limb length discrepancy in young adults

To investigate the clinical improvement of limping gait in patients with ankylotic hips and limb length discrepancy.Methods:From 1996 to 2005,12 patients with ankylotic hips and limb length discrepancy were treated by distraction osteogenesis with a mono-lateral external fixator and an intramedullary nail.The limb length discrepancy was 6.20-12.50 (median 8.45) cm.Limping gait was classified according to the recommendations of the American Academy of Orthopedic Surgeons/Hip Society and scored according to Harris:no limping scored 11 points,mild limping scored 8 points,moderate limping scored 5 points,while severe limping scored 0 points.Limping gait was severe in all patients pre-operatively and the total score was 0.Results:All patients were followed up for 30.00-46.00 (median 38.55) months,and all reported improvement in limping gait.The gain in length was 6.00-12.50 (median 8.20) cm,and the mean residual limb length discrepancy was 0-0.50 (median 0.20) cm.The total treatment time was 41.00-82.00 (median 61.50) weeks,the lengthening time was 14.00-38.00 (median 29.55) weeks.At the last follow-up,10patients had mild limping gait and 2 had moderate limping gait; the total score was 90.00.The median score was 7.50 (P25 was 8.00,P75 was 8.00).According to Wilcoxon signed rank test,the post-operative limping gait scores were significantly higher than pre-operative (P=0.001 ).Conclusion:Femoral lengthening can improve the limping gait significantly in ankylotic hips and limb length discrepancy.
9.A mete-analysis of the association of apoE gene polymorphisms and gallstone

Objective To evaluate the relationship between apoE gene polymorphisms and gallstone. Methods We included all the published studies on the association between apoE gene polymorphisms and gall-stone. A meta-analysis was employed to summarize all these studies, calculate the pooled OR and its 95% confidence interval (95% CI) , and test the overall effects. The Egger's publication bias analysis and sensitivity analysis were carried out to evaluate the reliability and stability of the meta-analysis. Results Eleven association studies between the apoE gene polymorphisms and gallstone fulfilled our inclusion criteria. There were 1248 patients with gallstones and 1660 controls. Remarkable heterogeneities were discovered in the allele ε4 and genotype E3/E4 of apoE between gallstone and control subjects in these studies (P<0. 05). Their ORs and 95%CIs were 1.32 (1.01, 1. 71), 1. 60 (1. 04, 2. 46), respectively (P<0. 05). The results of sensitivity analysis and publication bias analysis showed the reliability and stability of this meta-analysis. Conclusion apoE gene polymorphisms are associated with gallstone. Those with the alleleε4 or genotype E3 /E4 had a higher risk of suffering from gallstone.
10.Construction of anti-lipopolysaccharide single-chain phage antibody library in mice

Objective To construct an anti-LPS single chain phage antibody library in mice for further biomedical works. Methods Total RNA was extracted from splenic cells for reverse transcription after BALB/C mice had been immunized with pure LPS for 4 weeks. The designed primers were used to amplify the variable region genes of both heavy and light chain (VH,VL) with polymerase chain reaction. The VH and VL were then conjugated to form a single chain of variable fragment (ScFv) by a linker. The ScFv was cut by NotⅠ and SfiⅠ and then ligated into a pCANTAB5E phagemid vector which was digested with the same restriction enzymes. The ligated vector was then tranfected into the competent E.coli TG1 cells. Four TG1 clones were randomly selected to detect the exotic DNA. Results The titer of anti-LPS in murine sera was 1∶12 800. The concentration of total RNA was 12.3813 μg/ml. The length of the fragments were 340 bp for VH, 320 bp for VL and 800 bp for ScFv respectively. 1.9×107 clones were grown after transfection and one from the four randomly selected clones was identified to contain the exotic DNA. Conclusion A 4.75×106 murine anti-LPS single chain phage antibody library was successfully constructed.
