3.Screening and preliminary research of an actinomyces strain with antitumor effect

The apoptosis was observed under a fluoroscope when the tested tumor cells were treated with the supernatant derived from the liquid culture of an actinomyces strain AK 11, which had showed cell toxicity by means of MTT assay, and was further confirmed by the correspondent band of ”DNA ladder” in the agarose gel electrophoresis. In a bioassay employed nude mouse, the pure compound produced by the strain AK 11 showed tumor inhibition in rates of 52.01% and 33.64% corresponding to the dosages of 0.016 ?g?kg -1 and 0.008 ?g?kg -1 ip, respectively. According to the indexes IC 50 of human hepatocarcinoma SMMC7721 and stomach cancer MGC 803 cells, it was estimated that the inhibition effect of the pure compound was slightly stronger than that of Adriamycin. The results showed high antitumor effect of the tested compound from the strain AK 11. When the cells were treated with AK 11, it was found by flow cytometry analysis that cell division was ceased at S phase due to the DNA synthesis block .

5.Intrahepatic biloma formation and its significance after interventional treatment in patients with hepatocellular carcinoma

Objective To investigate the pathologic basis of intrahepatic biloma formed in the patients with hepatocellulr carcinoma (HCC) after interventional treatment and to assess its clinical significance. Methods Three hundreds and fifty cases of HCC were followed up with CT examination after TAE/PEI, 11 cases received percutaneous bilomography, 10 cases received percutaneous biopsy, and 1 case had surgical resection. All cases of biloma underwent external drainage of bile or ethanol ablation. Results 11 cases of biloma were found (11/350), and all of them displayed a cystic dilation (11/11), 4 cases showed a columnar dilation with indefinite border (4/11), and 3 cases accompanied with a “soft rattan sign” of intrahepatic bile ducts (3/11). The pathologic findings included necrosis on the wall of the biliary tract, extravasation of bile, and infiltration with inflammatory cells. After the external drainage or the ethanol ablation, jaundice had been decreased and symptoms relieved (9/11). Conclusions Biloma is one of the complications of the patients with HCC after TAE/PEI caused by the necrosis of the wall of biliary tract, the correct diagnosis depends on the imaging procedures and biopsy. It is helpful to the further treatment to distinguish biloma from residual nidus of carcinoma. External drainage, stent placement, and ethanol ablation may be the choice of management.

To explore the effects of angiotensin Ⅱ on plasminogen activator inhibitor 1 (PAI 1) gene expression in human glomerular mesangial cells.Glomerular mesangial cells (GMCs) were cultured from a healthy adult human kidney (unsuitable for renal transplantation) and then treated with angiotensin Ⅱ at concentration of 10 -9 ,10 -8 , 10 -7 and 10 -6 mol/L, respectively. The mRNA expressions of angiotensinⅡAT1 and AT2 receptor, and PAI I were examined with RT PCR and Northern blot analysis, respectively. Plasminogen activator activity in the supernatants of the GMCs was measured with fibrin plate method. After the GMCs were stimulated by angiotensin Ⅱ, the mRNA level of the AT1 receptor was markedly enhanced, but the AT2 receptor mRNA level was still undetectable as that in normal condition. When GMCs were treated with angiotensin Ⅱ for 48 hours, PAI 1 mRNA expression was increased in a dose dependent manner. The plasminogen activator activity was significantly reduced in the supernatants of the angiotensinⅡ treated GMCs. the results suggested that angiotensinⅡ promote PAI 1 gene expression of the GMCs possibly through its AT1 receptor.
7.The effect of vacuum-assisted drainage on granulation tissue formation of infected soft tissue explosive injury in pig

Objective To observe the effect of vacuum-assisted drainage (VAC) technique on the granulation tissue formation of infected soft tissue explosive injury in pig. Methods 16 wounds produced by explosion with electric detonators,which were fixed on the skin of the shoulders and hips of 4 small white pigs. The wounds were divided into 2 groups randomly: in group A the wounds were treated with conventional method,and in group B the wounds were treated with VAC set with a pressure of -15kPa. All wounds were infected on the third day after explosion. The depth of wounds was measured,and specimens were talcen from wound bed,immediately before treatment,and 1,3,6,9,14,19,and 24 days after treatment. The specimens were bistopathologieally studied with HE staining to assess the wound healing process of the two groups. Furthermore,immunohistochemistry for Factor Ⅷ related antigen and Ki67 was done to estimate the number of vascular endothelial cells and proliferating cells. 8.Establishment of rabbit model for acute retinal necrosis and experimental study of infectious pathway in opposite eye

Objective To establish the rabbit model for acute retinal necrosis(ARN) and investigate the possible route of the virus migrates from one eye to the other. Design Experimental study. Participants 41 chinchilla rabbits. Methods HSV-1 COS strain was inoculated into subretina in 41 rabbits to establish ARN animal model. Ganciclovir was used to treat by intraocular and/or intravenous injection 1 day or 3 days after virus inoculation. PCR test of blood samples was performed for HSV detection. Histopathological examinations were also conducted in some eyeballs, optic nerves and brains of the rabbits. Main Outcome Measures Results of blood PCR and histopathology. Results 10 rabbits appeared bilateral retina necrosis (10 to 18 days after virus inoculation) and 15 rabbits appeared central nervous system damages. PCR samples of 5 rabbits obtained before and after the contralateral retina damages appeared (9 to 19 days after virus inoculation) had all negative results. Histopathological damages were found in bilateral eyeballs, optic nerves and brains. Conclusion The ARN model can be established by injection of HSV-1 into subretina of rabbits. Blood samples PCR don't support the hypothesis of virus transmission through circulation system in bilateral ARN cases. Trans-optic chiasm transmission may be a possible route.
9. Research and design of comprehensive database for optimal decision-making in military medical service

Objective: To provide effective information resource for military medical support during strategic war, so as to provide a basis for the evidence-informed policy making in military medical service. Methods: A large quantity of multi-source and isomerism data were integrated and organized by the techniques of data warehouse and geography markup language. Multi-dimension subject models for decision-making in military medical commanding were constructed. Results: The integration of multi-source and isomerism data was achieved and comprehensive database was established for optimal decision-making in military medical service, which includes space data warehouse and its multi-dimension model for decision-making. The design of a 6-layer structure of data warehouse was also put forward. Conclusion: The database provides a comprehensive data platform for the optimal decision support of military medical service, and can achieve the decision support for the evidence-informed decision making in military medical service.

ObjectiveTo observe the therapeutic effect of early comprehensive rehabilitation and the value of intensity time (I/t) curve examine for peripheral facial paralysis.Methods89 patients with peripheral facial paralysis were treated by early comprehensive rehabilitation program in 1 week and were examined by I/t curve in 10~20 days.ResultsThe rate of the cure and excellent was 93.26%.Normal nerve detected by I/t curve were 62.92% and partial denervation were 37.08%.ConclusionPatients with peripheral facial paralysis who were treated by early comprehensive rehabilitation will have good prognosis. As a kind of examine ways, I/t curve can be used in clinic.
