1.Ontology and associated data-based library resources integration model

After the application of associated data in digital library and in library resources integration was described, the necessity of library resources integration was analyzed, the semantic association process of library resources and the problems of library resources in research on semantic association were studied in terms of system construction, technical implementation and discovery algorithm. The semantic integration of library resources was achieved by de-signing the integration framework, ontology and associated data.
3.Expression and Antigenic Characterization of the Epitope-G1 of the Bovine Ephemeral Fever Virus Glycoprotein in Pichia pastoris

The epitope-G1 gene of Bovine ephemeral fever virus (BEFV) glycoprotein was synthesised by PCR and cloned into expression vector pPIC9K to construct recombinant plasmid pPIC9K-G1. Then the pPIC9K-G1 was linearized and transformed into Pichia pastoris GS 115. The recombinant P. pastoris strains were selected by a G418 transformation screen and confirmed by PCR. After being induced with methanol, an expressed protein with 26 kDa molecular weight was obtained, which was much bigger than the predicted size (15.54 kDa). Deglycosylation analysis indicated the recombinant G1 was glycosylated. Western blot and ELISA tests, as well as rabbit immunization and specificity experiments indicated that the target protein had both higher reaction activity and higher immunocompetence and specificity. The recombinant G1 protein could be used as a coating antigen to develop an ELISA kit for bovine ephemeral fever diagnosis.
4.Associaiton of irisin and vaspin with clinical presentations of metabolic syndrome in patients with type 2 diabetes mellitus

Objective To explore the changes in serum concentrations of irisin,vaspin and reactive oxygen species (ROS) in patients with type 2 diabetes mellitus (T2DM) and metabolic syndrome (MS),and to investigated the correlation of irisin and vaspin with clinical parameters of MS.Methods A total of 260 T2DM patients were enrolled.Age and gender were recorded,anthropometrics,biochemical parameters,and levels of irisin,vaspin and ROS in fasting serum were measured,and homeostatic model assessment of insulin resistance (HOMA-IR) calculated.Wilcoxon rank sum test,correlation analysis,Logistic regression analysis,multiple linear regression analysis and receiver operating characteristic (ROC) curve analysis were performed.Results Compared to T2DM patients without MS,T2DM patients with MS had lower serum level ofirisin [male:112.81 (86.96-191.84) μg/Lvs.156.23 (110.61-225.97) μg/L,female:141.09 (77.52-175.55) μg/L vs.172.15 (95.69-240.37) μg/L,P <0.01],higher levels of vaspin and ROS [male:1.13 (0.95-1.38) μg/Lvs.0.36 (0.21-0.82) μg/L,1 540 (1 250-1 860) kU/Lvs.1 020 (920-1 350) kU/L;female:1.52 (1.13-1.80) μg/Lvs.0.51 (0.47-1.08) μg/L,1 650 (1 320-1 940) kU/Lvs.1 120 (980-1 420) kU/L,P <0.01].In the T2DM patients,serum irisin level was negatively correlated with vaspin (r =-0.382,P < 0.01) and ROS (r =-0.410,P < 0.01),while vaspin was positively correlated with ROS (r =0.400,P < 0.01).Multiple linear regression analyses showed that irisin was significantly correlated with body mass index (BMI),waist circumference and triglyceride,while vaspin was correlated with gender,BMI,and waist circumference (all P < 0.05).Logistic regression analysis revealed that irisin,vaspin and ROS were all associated with MS (OR =0.77,95 % CI 0.608-0.978;OR=1.39,95% CI 1.252-1.539;OR=1.38,95% CI1.112-1.718,all P<0.05).ROC analysis demonstrated that irisin and vaspin had significant area under the curve (AUC =0.931,P <0.01;AUC =0.777,P < 0.01) for the prediction of MS.Conclusions Serum irisin level was significantly decreased,while vaspin and ROS were significantly increased in T2DM patients with MS.Irisin and vaspin were associated with clinical presentations of MS,suggesting that irisin and vaspin might be valuable predictors of MS.
5.Expression of OPG and RANKL at protein level in human periodontal ligament cells and the effect of l_?,25(OH)_2 vitamin D_3 on the secretion of OPG protein in vitro

Objective:To study the expression of osteoprotegerin(OPG)and receptor activator nuclearfactor kappa B ligand(RANKL)at protein level in human periodontal ligament cells(HPDLCs),and theeffect of 1?,25(OH)_2 vitamin D_3[1,25(OH)_2 vitD_3] on the secretion of OPG protein in vitro.Meth-ods:HPDLCs were harvested in vitro by sequential digestion with trypsin and collagenase.The expressionof RANKL in HPDLCs at protein level was tested by immunocyto-chemistry.Enzyme-linked immuno-adsordent assay(ELISA)was used to detect the OPG protein which was secreted into the culture mediumby HPDLCs cultured with and without 10~(-8) mol/L 1?,25(OH)_2 vitD_3 on the 0,2nd,4th,and 6th days,respectively.Results:RANKL protein was detected on the membrane and plasma of HPDLCs,and OPGprotein was secreted in the culture medium.The secretion of OPG protein was down-regulated by 10~(-8)mol/L 1?,25(OH)_2 vitD_3.Conclusion:HPDLCs have the bone metabolism system of OPG/RANKL,which works during the process of 1?,25(OH)_2 vitD_3 inducing HPDLCs.The conclusion has laid thegroundwork for the study on bone remodelling mechanisms of HPDLCs.
6.Autopsy study for maxillary nerve and its artery supply.

OBJECTIVETo observe the travel, divisions, and the lengths, diameters, branches, artery supplies of the main segments of maxillary nerve.
METHODSFifty formalin-preserved adult half-head specimens with intravascular injection of red color emulsion were used for the gross and microanatomical studies of maxillary nerve. The lengths, diameters, branches and artery supplies of four main segments of maxillary nerve were observed. SPSS 11.5 software was used to analyze the data.
RESULTSThe length and diameter of cranial middle fossa segment of maxillary nerve were (10.70 ± 1.31) mm and (4.01 ± 0.52) mm respectively, which was supplied by inferior-lateral cavernous sinus artery. The length and diameter of pterygopalatine fossa segment were (16.21 ± 1.80) mm and (3.27 ± 0.62) mm respectively, in which one zygomatic branch, one to three posterior superior alveolar nerves, two ganglion branches and tuberal descending branches; were given off, and the segment was supplied by foramen rotundum artery. The length and diameter of infraorbital segment were (25.73 ± 2.03) mm and (3.30 ± 0.52) mm and it gave off middle superior alveolar nerve (64%) and anterior superior alveolar nerve and was supplied by infraorbital artery. Facial segment gave off superior labial branches, internal and external nasal branches, inferior palpebral branches, buccal branch and zygomatic branch and these branches were supplied by infraorbital artery and superior labial and angular artery originating from facial artery.
CONCLUSIONSUnderstanding of travel and artery supply of maxillary nerve is helpful to regional anaesthesia and surgery for maxillary nerve. Foramen rotundum, sphenopalatine foramen and infraorbital nerve are important marks for endoscopic surgery in pterygopalatine fossa.

7.Potential predicting function of betatrophin in patients with polycystic ovary syndrome and it's relationship with 25-hydroxy vitamin D3

Objective Through the detection of the levels of serum betatrophin,anthropometric and biochemical indices,to determine the alteration of betatrophin levels in patients with polycystic ovary syndrome (PCOS) and the relationship between betatrophin levels and metabolic indexes such as 25-hydroxy vitamin D3 [25 (OH) D3].Methods The study group was composed of 24 women with PCOS and 20 age-natched healthy women as controls.Following general physical examination of the subjects,anthropometrie measurements were performed (height,weight,waist circumference,hip circumference).Body mass index (BMI) and waist-hip ratio were calculated;25 (OH) D3,Ca2+,fasting blood glucose,fasting insulin,triglycerides,high-density lipoprotein cholesterol (HDL-C),low-density lipoprotein cholesterol (LDL-C) were also measured and homeostasis model assessment of insulin resistance (HOMA-IR) were evaluated in all subjects.Serum betatrophin levels were examined with an enzyme-linked immunosorbent assay (ELISA).Using independent sample T test to compare the differences between groups,the linear correlation analysis was performed to study the correlation of betatrophin with anthropometric and biochemical indices,the influencing factors of betatrophin were analyzed by multiple linear regression.Results Circulating betatrophin levels [(0.341±0.034) ng/ml vs.(0.810±0.162) ng/ml,t=4.271,P<0.001] and HDL-C [(1.014±0.321) mmol/L vs.(1.419±0.287) mmol/L,t=2.218,P<0.05] significantly decreased in women with PCOS compared with controls,and BMI [(27.691±4.392) kg/m2 vs.(23.310±4.781) kg/m2,t=-2.073,P<0.05],fasting blood glucose [(5.950±0.411) mmol/L vs.(4.883±0.314) mmol/L,t=-2.142,P< 0.05],HOMA-IR [(4.946 ± 0.741) vs.(3.387± 0.397),t=-2.493,P<0.05],triglycerides [(1.510± 0.848) mmol/L vs.(1.037±0.402) mmol/L,t=-2.223,P<0.05],LDL-C [(3.431±0.479) mmol/L vs.(2.396±0.435) mmol/L,t=-2.433,P<0.05] were obviously increased.Moreover,there was a postive correlation between betatrophin and HOMA-IR (r=0.425,P<0.05) as well as 25(OH)D3 (r=0.577,P<0.05) and Ca2+ (r=0.448,P<0.05),while the betatrophin was negatively related to BMI (r=-0.451,P<0.05),triglycerides (r=-0.454,P<0.05),LDL-C (r=-0.551,P<0.05).Circulating betatrophin levels were higher when 25 (OH) D3 levels were beyond 35 nmol/L in PCOS patients [(0.539±0.092) ng/ml vs.(0.199±0.031) ng/ml,t=3.072,P<0.001],and the multiple linear regression analysis showed that the main factors affecting the levels of betatrophin were BMI (OR=-0.260),HOMA-IR (OR=0.218),25(OH)D3 (OR=0.238),and glycerol (OR=-0.162).Conclusion It is speculated that betatrophin may be a valuable predictive factor of PCOS because betatrophin is closely associated with insulin resistance,lipid metabolism disorders and the lack of 25-hydroxy vitamin D3 in PCOS patients.
8.Relationship between serum HBV DNA levels and hepatic fibrosis markers in chronic hepatitis B.

OBJECTIVETo study the relationship between serum HBV DNA levels and hepatic fibrosis markers in chronic hepatitis B.
METHODSOne hundred and fifty-seven patients with chronic hepatitis B were included in the study, 49 patients among them were diagnosed as early cirrhosis by liver biopsy. Serum HBV DNA levels were determined using fluorescent quantitative PCR, and serum hepatic fibrosis markers including hyaluronic acid (HA), laminin (LN), amino terminal propeptide of type III precollagen (P III P) and type IV collagen (IV-C) were determined by radioimmunity assay. The relationship between serum HBV DNA levels and hepatic fibrosis markers were analyzed. Serum HBV DNA levels and hepatic fibrosis markers of 49 patients with early cirrhosis were compared with those of 108 non-cirrhotic patients.
RESULTSThere was no significant relationship between serum HBV DNA levels and hepatic fibrosis markers in chronic hepatitis B (P>0.05). Patients with early cirrhosis had much higher hepatic fibrosis markers together with lower HBV DNA levels than non-cirrhotic patients (P<0.05).
CONCLUSIONSThere were no significant relationship between serum HBV DNA levels and hepatic fibrosis markers in patients with chronic hepatitis B.

9.A factors study of root resorption after orthodontic treatment.

OBJECTIVETo investigate the factors relating to root resorption after orthodontic treatment.
METHODS96 cases treated with fixed appliances were selected. The panoramic radiographs before and after treatment were examined. The relationship between root resorption after treatment (RRAT) and the variables including sex, age, extraction or nonextraction approach, tooth location, treatment duration and the root resorption before treatment (RRBT) were analyzed by multiple variance analysis and stepwise multivariate linear regression analyses.
RESULTS(1) There were statistically significant differences in root resorption in relation to gender (estimate of RRAT for female: 0.41, that for male: 0.34), extraction (estimate of RRAT for the extraction group: 0.43, that for non-extraction: 0.31), anterior or posterior teeth (estimate of RRAT for the anterior teeth: 0.59, that for the posterior teeth: 0.12). (2) The estimate of RRAT for the upper teeth was 0.40, and that for the lower teeth was 0.37. There were no statistically significant differences. (3) Using multiple regression analysis, age, duration and RRBT were associated with RRAT (R = 0.59, R(2) = 0.35).
CONCLUSIONS(1) Factors including gender, extraction or not and anterior or posterior teeth have influences on the RRAT. More root resorption was found in female or extraction cases. Anterior teeth were more susceptible to root resorption relative to posterior teeth. (2) Age, treatment duration and RRBT can explain approximately 35% of the RRAT. Root resorption aggregated when it was present before treatment. The age and treatment duration were lightly related to the root resorption.

10.Changes of Streptococcus mutans concentration of plaque during fixed appliance treatment.

OBJECTIVETo evaluate the dynamic changes of Streptococcus mutans concentration of plaque during fixed appliance treatment and the effects of two materials of ligation on Streptococcus mutans concentration.
METHODSTwenty-eight patients undergoing fixed appliance treatment were observed. Ligature wire and elastomeric rings were applied on one side of arches, stochastically. The dynamic changes on the quantity and percentage of Streptococcus mutans were observed before and after fixed appliance bonding.
RESULTSStatistically significant increase of the quantity and percentage of Streptococcus mutans was found after fixed appliance bonding, and the percentage of Streptococcus mutans in the plaque around the brackets ligated with elastomeric rings was more than that of ligature wire at the beginning of fixed appliance bonding, statistically.
CONCLUSIONSThe finding suggested that the caries-associated capability of the plaque increased after bonding and there was greater caries-associated capability of the plague on the teeth when elastomeric rings was used than that of the plague when ligature wire was used at treatment beginning. The ligature wire is recommended in the fixed appliance treatment.
