This study was aimed to evaluate the clinical effect of 66 yin-deficiency acne cases treated with body constitution adjustment using acupuncture and moxibustion. In this research, 66 acne patients with the body con-stitution of yin-deficiency were selected . These patients were randomly divided into two groups ( 33 every group ) . The acupuncture treatment group was treated by acupuncture and local treatment. And moxibustion treatment group was treated by indirect moxibustion and local treatment. The treatment was given twice a week. And the improvement condition of acne skin lesions and yin-deficiency body constitution were evaluated after 12-week treatment. The results showed that certain curative effects were received in both groups. However, the moxibustion treatment group had higher curative effect compared with the acupuncture treatment group. There were significant statistical differences between two groups ( P < 0 . 05 ) , which meant moxibustion with local treatment had a higher curative effect than acupuncture with local treatment to acne. In the aspect of skin lesion improvement, according to the statistics, the moxibustion treatment group were statistically significant compared with acupuncture group (P< 0.05), which meant more obvious improvement of skin in the moxibustion treatment group. In the aspect of body constitution, the body constitution scores in both groups were significantly declined and the moxibustion treatment group was more obvious. There was statistical significance between two groups (P < 0.05), which meant patients in the moxibustion treatment group obtained a more obvious body constitution improvement. After the treatment, follow-up was given to all patients for 4 weeks. The results showed that both the acne skin lesion con-dition and yin-deficiency body constitution score were decreased and not achieve the recurrence diagnostic stan-dard. It showed that acupuncture and moxibustion are able to activate the self-regulation and disease resistibility of the body. It was concluded that acupuncture and moxibustion not only improve skin lesion of acne patients with the body constitution of yin-deficiency with inner heat , but also regulate yin-deficiency body constitution . Moxibustion had higher efficacy in the treatment of yin-deficiency with inner heat acne than acupuncture.

In this study, polyacrylicacid precipitation alkalescence protein from Momordica charantia L. seeds was studied, and the effect of conditions on experiment was also evaluated. Isoelectric precipitation is achieved by adjusting the pH of a protein solution and is based on that a protein's solubility is at minimum at its pI. The sample was titrated to pH 6.0 with citric acid, and 14.62% proteins were precipitated. With hydrochloric acid to pH 4.0, 32.49% proteins were precipitated. With the acetic acid to pH 6.0 and pH 4.0, 26.17% and 38.72% proteins were precipitated, respectively. In the 1 mL Bitter melon seeds extraction(pH 4.0) adjusted by acetic acid, hydrochloric acid and citric acid, the optimum dosage of PAA (1%) precipiting alkalescency protein (pl 8.65-9.30) was 100 microL, 120 microL and 100 microL, respectively. The respective extraction (1mL) was titrated to pH 5.0, pH 4.0, and pH 3.0 by acetic acid. After isoelectric precipitation, the PAA precipitation protein was performed. When concentration of PAA (1%) was 160 microL/mL, the protein decreased in the supernatant was 33.77% at pH 5.0, and 43.56% at pH 3.0. When concentration of PAA (1%) was 120 microL/mL, the protein decreased in the supernatant was 30.83% at pH 4.0. PAA-Protein complex could redissolve in alkaline conditions (pH > 9.0) and the protein most easilly redissolved when the NaCL was 3.0%. The bitter melon seeds extraction after PAA purification flowed through the Sephadex G-75 columns. The peaks I and II were obtained after 175 min and 300 min, respectively. SDS-PAGE and IEF analysis showed that the molecule weight from peaks I was 30 kD with pI 9.5, peaks II 10 kD with pI 9.3.

Metabolomics, a branch of systems biology, has gained extensive attention and profound achievements in the plant. Although plant metabolomics is to be explored, it has been one of the most effective methods to study the physiological and biochemical process and gene modification in pattern plants. We herein summarized the concept, development, and application of metabolomics and prospected the potentials in the metabolite profiling for plant. Metabolomics provides an omics' methodology to elucidate the whole biological process, identify and quantify the complex components in the plant. A number of metabolites present in the plant are active components of traditional Chinese medicine, and these bioactive components are influenced by the multi-factors such as environment, species, and processing methods etc. Therefore, it is of great importance to analyze a wide spectrum of compositions with diverse chemical characteristics and varied concentration, which is the foundation to quality control, allowing the elucidation of the pharmacological effectiveness, and further exploiting of traditional Chinese medicine.

OBJECTIVETo investigate the time course of calpain activity changes in rat neurons following fluid percussion injury (FPI) under normothermia (37 degrees celsius;) and mild hypothermia (32-/+0.5) degrees celsius;.
METHODSIn vitro cultured rat neurons were subjected to FPI followed by application of mild hypothermia for intervention at different time points, and the changes in intraneuronal calpain activity following FPI and the interventional effect of mild hypothermia on calpain activity were evaluated by UV-spectrophotometry at different time points.
RESULTSRemarkable changes occurred in calpain activity in the neurons following FPI at 37 degrees celsius;, and mild hypothermia produced obvious interventional effect on calpain activity in close relation to the timing of intervention initiation.
CONCLUSIONIntraneuronal calpain activity changes following FPI are involved in the pathological process of cellular injury, and mild hypothermia might offer protection against traumatic brain injury to some extent by regulating calpain activity. The interventional effect of mild hypothermia is associated with the timing of the intervention initiation.

In this study, we investigated the mechanism of linoleic acid-stimulated increase in intracellular calcium concentration ([Ca(2+)](i)) in pancreatic islet β-cells. Pancreatic islet cells were primarily isolated from rats and cultured for the experiments. The cells were loaded with Fluo-3/AM, the indicator of [Ca(2+)](i), and the intensity of Fluo-3 was measured using confocal microscope. The islet β-cells were identified by immunocytochemical staining with insulin antibody after recording. The drugs were given by perfusion system. The results showed that linoleic acid (20 μmol/L) stimulated [Ca(2+)](i) increase with the first peak increase and the following plateau increase. Linoleic acid-stimulated [Ca(2+)](i) increase was partly inhibited by removal of extracellular calcium and by transient receptor potential (TRP) channel blocker, La(3+), and it was totally blocked by exhaustion of intracellular calcium stores and inhibition of phospholipase C. It is concluded that linoleic acid stimulates [Ca(2+)](i) increase in islet β-cells through both extracellular calcium influx via TRP channels and calcium release from intracellular calcium stores.

Journal of Forensic Medicine 2007;23(1):42-43
Autopsy by forensic pathologist is a main mean currently to determine the cause of sudden unexpected death. Retinal examination is important but seldom performed during a forensic autopsy for various reasons. The value of retina examination has not been recognized. With invention of ophthalmic endoscopy and its subsequent application in postmortem retina examination, it has proved to be useful adjunct to determine the cause of death and to estimate the postmortem interval.

Journal of Forensic Medicine 2007;23(5):362-364
Individual response to drugs, toxicants, environmental chemicals and allergens varies with genotype. Some respond well to these substances without significant consequences, while others may respond strongly with severe consequences and even death. Toxicogenetics and toxicogenomics as well as pharmacogenetics explain the genetic basis for the variations of individual response to toxicants by sequencing the human genome and large-scale identification of genome polymorphism. The new disciplines will provide a new route for forensic specialists to determine the cause of death.

Objective To investigate time course changes of intraneuronal calpain activity following fluid percussion injury(FPI)under normothermia(37℃)and mild hypothermia(32+0.5)℃.Methods Rats' neurons produced by external culture and FPI models prepared, mild hypothermia was applied for intervention at different time points and the changes in intraneuronal calpain activity following FPI and the intervention effect of mild hypothermia on calpain activity during different time courses were detected by UV-spectrophotometry. Results Remarkable changes occurred to calpain activity following FPI at 37℃. While the intervention effect of mild hypothermia on calpain activity was different, closely related to the starting time point of intervention. Conclusion The changes of intraneuronal calpain activity following FPI join the pathological process of cellular trauma; mild hypothermia might protect patients with traumatic brain injury(TBI)to some extent through the regulation of calpain activity. And, the intervention effect of mild hypothermia has some correlation with the starting time point of intervention.

Journal of Medical Biomechanics 2016;31(3):E272-E277
Objective To study the effect of mechanical stretch on expression of inflammasome related factors in human periodontal ligament cells (HPDLCs). Methods HPDLCs were subjected to mechanical stretch with a 20% elongation magnitude for 6 h or 24 h, respectively. The mRNA and protein expression levels of IL-1β, Caspase-1 and NLRP3 were detected by real-time quantitative PCR and Western blot. The levels of IL-1β in the cell-culture medium of HPLCs in response to mechanical stretch for 1, 2, 4, 6 h were detected by ELISA, respectively. In control group, HPDLCs were cultured in similar conditions but not subjected with stretch. Results The mRNA and protein expression levels of IL-1β, Caspase-1 and NLRP3 were up-regulated with mechanical stretch for 6 h (P<0.05). The protein expression level of NLRP3 was up-regulated with mechanical stretch for 24 h (P<0.05). Compared with control group, the content of IL-1β in the cell-culture medium of HPLCs was increased significantly in response to mechanical stretch for 4 h and 6 h(P<0.05). Conclusions The expression of NLRP3/Caspase-1/IL-1β related factors in HPDLCs can be induced by 20% mechanical stretch for 6 h.
