4.Advanced liposuction technologies and their potential application in the isolation of adipose-derived stem cells

BACKGROUND:Currently, cosmetic liposuction has developed rapidly, but the liposuction for the isolation of adipose-derived stem cells has not been reported.
OBJECTIVE:By evaluating the quantity and viability of stromal vascular fraction, adipocytes and adipose-derived stem cells obtained from different types of liposuctions, to find the advanced liposuction procedures that are most suitable for use in preparation, preservation and clinical application of adipose-derived stem cells.
METHODS:Authors retrieved PubMed database, NIH clinical trial databaseand CNKI ful text database for relevant articles using the key words of“adipose-derived stem cells, ADSCs, liposuction, SAL, UAL, PAL, WAL”in English and Chinese, respectively. After eliminating the objective-independent articles and repeating reviews, 50 papers were included for further analysis.
RESULTS AND CONCLUSION:Suction-assisted liposuction (SAL), ultrasound-assisted liposuction (UAL), power-assisted liposuction (PAL) and water-assisted liposuction (WAL) are four major types of fat-harvesting technologies that can possibly be applied for adipose-derived stem cells isolation. Accumulative evidence suggests that these advanced liposuctions can obtain high quality of adipose tissue/cells, which are good enough for autologous fat grafting. Adipose-derived stem cells that are isolated from lipoaspirate by WAL, SAL and UAL, can be used as autologous cel-assisted lipotransfer and regenerative therapy. In summary, although each of these four types of liposuction technologies has its own advantages and is feasible to be used as autologous fat grafting, systematic comparative studies for viability, chromosome stability and differentiation potentials of adipose-derived stem cells isolated using each col ecting method are necessary in order to confirm the future perspectives in adipose-derived stem cells isolation, banking and regenerative repair.
5.IMMUNOHISTOCHEMICAL AND HISTOCHEMICAL STUDY OF HUMAN PLACENTAS OBTAINED AFTER ADMINISTRATION OF CHINESE MEDICINAL HERB (CHUAN-XIN-LIAN)INDUCED ABORTION DURING MID-PREGNANCY

12 cases of human placentas were obtained after administration of Chinese medicinal herb (Chuan-Xin-Lian) -induced abortion during mid-pregnancy. Six placentas from curettage induced abortion during mid-pregnancy were used as controls. All the specimens were studied histochemically and immunohistochemically by using antiserum to HCG with PAP method. In most of the specimens from women treated during mid-pregnancy, we observed a decreased immunoreaction of antiserum to HCG and a decrease of RNA content in the syncytiotrophoblasts. Increased activity of acid phosphatase in dccidual cells was observed during mid-pregnancy. These findings indicate that during mid-pregnancy, Chinese medicinal herb (Chuan-Xin-Lian) can reduce the synthesis of HCG in syncytiotrophoblasts and can initiate uterine contractions, so that abortion results. The possibility that the changes of lysosomal enzymes are involved in the initiation of uterine contractions is disscused.
6.Multidrug resistance-associated proteins and their roles in multidrug resistance.

Multidrug resistance (MDR) is a major impediment to the success of cancer chemotherapy. Multidrug resistance-associated proteins (MRPs) play an important role in the process of MDR. As an ATP-binding cassette (ABC) transporter superfamily, MRPs are selective and specific drug efflux pumps. In this paper, physiological characteristics, structural characteristics and resistance profile of MRPs and the associated reversal studies are reviewed.
7.Preparation and percutaneous absorption in vitro of Ruanganxiaoshui Cataplasma

AIM:To study the prepartion of Ruanganxiaoshui Cataplasma and it's percutaneous absorption in vitro as well as dissolvability in vitro. METHODS: In the experiment,orthogonal test was used to optimize the preparation.Meanwhile,with the help of azole and propylene alcohol the percutaneous absorption of ginsenoside Rg_1 from the cataplasma carriers in vitro was determined by Franz's diffusion cell and HPLC,as well as dissolution rate in vitro. RESULTS: The percutaneous absorption rate of ginsenoside Rg_1 through rat skin in vitro was 0.394 ?g/cm~2/h.And the dissolution process fitted to first order model. CONCLUSION: The Cataplasma carriers possess a good drug dissolution property.To add compound penetration enhancers is one of the effective methods for raising percutaneous absorption.
8.Fingerprints of Herba Ephedrae by HPLC

AIM:To study the HPLC fingerprints and to establish a sensitive and specific method for controlling the quality of Herba Ephedrae. METHODS: HPLC method was set up,using CLCphenyl column(5 ?m 4.6 mm?150 mm) with gradient elution,the mobile phase was comprised of acetonitrile and phosphoric acid(0.1%),the UV detection wavelength was at 215 nm,with a flow rate of 1.0 mL/min. RESULTS: There were 17 common peaks in the fingerprints of Herba Ephedrae based on 10 habitats.Sum of peak areas was larger than 90% total area.The similarity was compared and the HPLC fingerprints was set up. CONCLUSION: The quality of Herba Ephedrae can be controlled effectively by HPLC fingerprints.
9.Methods of Difficult Laparoscopic Cholecystectomy

[Objective]To approach the methods of difficult laparoscopic cholecystectomy.[Methods ]The clinical data of 116 patients with Mirizzi syndrome,severe adherence of abdominal cavity and Calot'triangle,liver cirrhosis underwent difficult laparoscopic cholecystectomy since 2004 were retrospectively analyzed.[Results]Except 3 patients treated by abdominal operation,other patients were treated by laparoscopic cholecystectomy successfully.[Conclusion]The rates of open operation midway and complications are not increased in difficult laparoscopic cholecystectomy as long as dealing with them properly.
10.Effects of bufalin combined with doxorubicin on the proliferation and apoptosis of human lung cancer cell line A549 in vitro

Objective:To explore the effects ofbufalin (BUF) combined with doxorubicin (DOX) on the proliferation and apoptosis in human lung cancer cell line A549 in vitro.Methods:Methyl thiazolyl tetrazolium (MTT) assay was used to measure the inhibitory effects of BUF,DOX and their combination on the growth ofA549 cells.Hoechst 33342 staining was used to observe the changes of nucleus.Flow cytometry was used to investigate the apoptosis and cell cycle distribution of A549 cells.Western blot was used to examine the expression of apoptotic protein.Results:BUF and DOX showed inhibitory effect on the A549 cells in a dose and time-dependent manner.Compared with BUF or DOX alone,combination of BUF (1,20,100 nmol/L) with DOX (1.0 μg/mL) could significantly increase the growth inhibition rate ofA549 cells at 24,36,72 h,respectively (all P<0.05).BUF and DOX alone could induce apoptosis,and their combination could significantly increase the apoptosis ratio.In addition,BUF combined with DOX could block the cell stage of A549 cells,keep the cell stage stay in S stage and up-regulate the expression of caspase-3.Conclusion:BUF combined with DOX can significantly inhibit the proliferation ofA549 cells,which might be related to the induction of apoptosis,cell cycle S phase arrest and caspase-3 up-regulation.
