1.Whether Guangzhou Reach of Zhujiang River Will be Taken as a Swimming Area:an Investigation of Water Quality of Urban River Surge and a Certain Section

Objective To know the sanitary situation of urban river surge and a certain section of Zhujiang River and whether Guangzhou reach of Zhujiang River will be taken as a swimming region. Methods From July 1 to July 15,2007,the period of swim,the spring-tide water and ebb-tide water at urban river surge and a certain section were sampled and tested. 54 water sample at urban river surge and 40 at a certain section,the items included pH value,turbidity,dissolved oxygen,chemical oxygen demand, amino-nitrogen and heat-resistant coliform. Results The results indicated that pH value and chemical oxygen demand of the water at urban river surge and a certain section were eligible,but amino-nitrogen,dissolved oxygen and heat-resistant coliform were not eligible,the turbidity was high,the water quality did not meet the Hygienic Standard for Swimming Place (GB 9667-1996). 2.Research progresses of Mycobacterium tuberculosis cytochrome P450s as a potential drug target.

Identification and validation of a new target is one of the most important steps for new antituberculosis (TB) drug discovery. Researches have shown that Mycobacterium tuberculosis (Mtb) encodes 20 CYP450 enzymes which play important roles in the synthesis and metabolism of lipid, cholesterol utilization, and the electron transport of respiratory chain in Mtb. With the critical roles within the organism as well as the protein structures of six Mtb CYP450 enzymes being clarified, some of them have been highlighted as potential anti-tuberculosis targets. In this paper, the phylogenetic analysis, the structural features, and the enzymatic functions of Mtb CYPs, as well as the mechanism of interactions with selective inhibitors such as azole antifungal agents for the CYPs have been reviewed and summarized. The druggability of the CYPs has also been analyzed for their further utility as targets in high throughput screening and rational design of more selective inhibitors.

4.Teaching reform of the teaching team of medical examination technology based on the training of application ability

The culture of modern medical talents is the requirements of the times on cultivating talents of Medical Laboratory of Medical College. In order to establish a high level teaching team and im-prove the quality of teaching and personnel training, through the reform of the traditional teacher training way, we created the teacher training mode of multi echelon teachers including department, university, domestic and international training ways. New curriculum teaching system framework was established which was based on theoretical teaching, practical teaching and scientific research. Besides, innovative and pro-gressive practice teaching platform was set up which included basic experiment, clinical skills, professional training, and practice in company. Course of Clinical Examination Skills was opened. The teaching team achieved some results in the above teaching reform.
5.Impact of subclinical hypothyroidism on metabolic risk factors of type 2 diabetes mellitus

Objective To compare and explore impact of subclinical hypothyroidism on metabolic risk factors of type 2 diabetes mellitus.Methods A total of 204 inpatients with type 2 diabetes mellitus.The patients were divide into two groups according to whether complicated with subclinical hypothyroidism:combination of simple type 2 diabetes mellitus (150 cases) and type 2 diabetes mellitus with SCH group(54 cases),two groups of age,gender,duration were no statistical difference.and collecting the clinical data of two groups of patients by Professional people,detection of blood lipids,glycated hemoglobin (HbA1c),uric acid (UA),Oral glucose tolerance test (OGTT),insulin and C peptide releasing test were taken in all groups.The improvement of insulin,C peptide index [HOMR-islet(CP)] and whole body insulin sensitivity index (ISIcomp) were used to estimate insuhn sensitivity,and improvement of insulin,C peptide secretion index [HOMR-isle (CP)] was used to evaluate the function of pancreatic islet β cell.T test was used for statistic alanalysis.Risk factors of type 2 diabetes mellitus with microvascular Complication by using Logistic regression analysis.Results (1) Compared with type 2 diabetes mellitus group,blood pressure,body mass index (BMI),triglycerides(TC),Low density lipoprotein cholesterol (LDL-C),UA,fasting C peptide(C-P0),postprandial 120 min C peptide(C-Pm),HOMA-IR(CP) were increased (P < 0.05),while fasting blood glucose(FPG),bloods glucose in postprandial 120 min (G120),HbA1c,ISI-comp were decreased (P < 0.05).(2) The incidence of diabetic nephropathy was higher in diabetic patents with subclinical hypothyroidism than those without subchnical hypothyroidism,the difference was statistically significant (P < 0.05).(3) Logistic analysis showed that duration,SBP,TSH were the major risk factors of type 2 diabetes mellitus with nephropathy (P < 0.05).Conclusions Type 2 diabetes mellitus patients with subclinical hypothyroidism compared to pure insulin resistance is more obvious,the degree of lipid metabolism disorders more severe.Elevated TSH can predict the occurrence of diabetic nephropathy in type 2 diabetes mellitus.
6.Evaluation of different methods in eliminating Mycoplasma contamination in cell culture of human hepatoma-derived cell line C3A

Objective To evaluate the effectiveness of several methods in eliminating Mycoplasma contamination in cell culture of human hepatoma-derived cell line C3A. Methods PCR was performed to detect the Mycoplasmas contamination in cell cultures. The contaminated samples were treated by ciprofloxa-cin, heating, Plasmocure or co-culturing with macrophages. Transmission electron microscope ( TEM) and Q-PCR were used to comparatively analyze the cell morphology and gene expression before and after Plas-mocure treatment. Results Plasmocure succeeded in eliminating Mycoplasma contamination, while cipro-floxacin showed temporary efficacy. Heating and co-culturing with macrophages failed to eliminate Mycoplas-ma contamination. No Mycoplasma contamination in the Plasmocure-treated group was observed under TEM and the expression of ALB, TF and CYP3A4 genes were higher than the genes expressed in the contaminated group (P<0. 01). Conclusion Plasmocure treatment was effective in eliminating Mycoplasma contamina-tion in cell culture. Moreover, the cell morphology and gene expression in Plasmocure-treated group were re-stored to normal.
7.The research of vaccine safety injection and the best disinfection effect of using alcohol

Objective This article is to discuss the best injection time after alcohol disinfection during the prophylactic immunization,provide theoretical support to guarantee the effect of disinfection in practical work,also guarantee the effect of vaccination especially for the vaccination of attenuated live vaccine at the same time reduce the side effect in disinfection.Methods Choosing the vaccination objects as experimental subject,each period contained 40 people,who were named as group A,B,C,D,E,with a total of 200 people.Using sterile cotton swab sampling and agar plate cultivation method,counting bacteria.Selecting vaccine either in liquid or in lyophilized form,counting the time of picking up the vaccine,dissolving it,suction and preparing the injector.Results After alcohol disinfection,there was 1 person and 1 colony growth in group A within 25 s.There were 1 person and 4 colonies growth in group B within 40 s.There were 3 persons and 3 to 12 colonies growth in group C within 60 s,there were 5 persons and 2 to 8 colonies growth in group D within 80 s,there were 8 persons and 3 to 13 colonies growth in group E within 100 s.The difference between group A,B,C and D was not statistically significant.All four groups mentioned above had significant difference compared with group E.The time of whole process for vaccine either in liquid or in lyophilized form was 1.05 min and 29 s.Conclusions During vaccine injection,especially injectable attenuated live vaccine,it is safe time to finish injecting within 25 to 45 seconds.In the operating process of vaccine inoculation,the freeting and drying dosage-form vaccine,the operation sequence must be set as dissolve the seedlings,disinfection,checkup information again and injections.For water dosage-form vaccine,the operation sequence is to disinfection,pulling out seedlings,checkup information again and injections.
8.Construction and expression of recombinant plasmid pEGFPC1uPAR in Pam 212 cells

Objective To construct the eukaryotic expression plasmid of pEGFPC1uPAR gene and explore the effect on the proliferation and invasion ability of Pam 212 cells. Methods The human uPAR cDNA was cloned by PCR, and inserted into the eukaryotic expression plasmid pEGFPC1. After identification of sequencing, the reconstructive plasmid was transformed transiently into Pam 212 cells, then the cell growth and the invasion ability were evaluated. Results The reconstructive plasmid of pEGFPC1uPAR was validated by sequencing. The reconstructive plasmid can promote the growth of Pam 212 cells and enhance the invasion ability. Conclusion The pEGFPC1uPAR plasmid was constructed successfully and uPAR was confirmed to promote the growth and the invasion ability of Pam 212 cells, which lay the foundation for further studies of uPAR in vivo.
9.Construction of experimental course group in molecular medicine based on improving medical students' comprehensive ability

In order to adapt to the requirements of modem medical knowledge and skills for higher medical workers,and to cultivate medical students' molecular medicine quality and comprehensive ability,Weifang Medical University broke the boundaries of disciplines and constructed experimental course group in molecular medicine based on the ideas of curriculum group construction.Molecular medical knowledge was integrated into the teaching process of experimental course group,experimental teaching content system was reasonably integrated and optimized,high quality teaching team was set up,multi-level experimental teaching platform was built and student-centered teaching mode was implemented to explore the experimental teaching approach which helped medical students to form systematic molecular medicine knowledge structure and ability structure.
