1.Clinical observation on deep-oblique acupuncture with long needle at Guanyuan (CV 4) for urinary retention.

OBJECTIVETo observe differences of clinical therapeutic effects between deep-oblique acupuncture with long needle and regular perpendicular acupuncture at Guanyuan (CV 4) for urinary retention to explore standard acupuncture method in clinic.
METHODSOne hundred and thirty-two cases of urinary retention were randomly divided into an observation group (78 cases) and a control group (54 cases). Needles with 75 mm in length were deeply and obliquely inserted into Guanyuan (CV 4) in the observation group while needles with 40 mm in length were perpendicularly inserted. Clinical therapeutic effects in two groups were observed.
RESULTSAfter treatment the total effective rate was 80.8% (63/78) in the observation group and 57.4% (31/54) in the control group, which had significant difference between two groups (P<0.05). The curative effect of postpartum retention of urine was obviously superior to that of postoperative and prostate-induced urinary retention in the ebservation group, and the effect of urinary retention due to different etiology in the observation was also superior to that in the control group in the ebservation group [in the observation group total effective rate was 88.6% (47/53) in postpartum retention of urine, 50.0% (3/6) in prostate-induced urinary retention and 68.4% (13/19) in orthopedic postoperative retention of urine while in the control group total effective rate was 69.4% (25/36) in postpartum retention of urine, 20.0% (1/5) in prostate-induced urinary retention and 38.5% (5/13) in orthopedic postoperative retention of urine)].
CONCLUSIONThe deep-oblique acupuncture with long needle at Guanyuan (CV 4) has significant curative effect on urinary retention in clinic, especially for patients with postpartum urinary retention, which could be taken as standard acupuncture method.

3.STAT3 Activation Way Blocking Effects on Colorectal Cancer cells HT29 Growth

Objective:Explore decoy oligonucleotide (ODN) technology targeted blocking STAT3 activation way effects on colorectal cancer HT29 cell growth,to provide experimemt basis for gene therapy of colorectal cancer.Methods:Using decoy ODN in vitro transfect colorectal cancer HT29 cell line,to targeted blocking STAT3 activation way.Fluorescence microscope and flow cytometry detect transfection conditions and efficiency,and MTT method detect the cell growth inhibition,Realtime RT-PCR and Western blotting test STAT3 in cells,the Bcl-xl and Caspase3 mRNA and protein expression.Results:STAT3 decoy ODN can efficient transfection into colorectal cancer cells,and is due to the nucleus.HT29 cell in the STAT3 decoy ODN,growth inhibition rate increased significantly,and mRNA and quantity of protein expression of STAT3 significantly reduce (P<0.05),while caspase3 increase significantly (P<0.05).Conclusion:Named Decoy ODN targeted after blocking STAT3 activation way,which can effectively cut colorectal cancer cells HT29 STAT3 gene expression,and inhibit the growth of the cells.The mechanism may be related to lower the expression of STAT3 and the Bcl-xl and Caspase3.
4.Therapeutic Observation of Centro-square Needling plus Stuck Needling for Injury of Medial Collateral Ligament of Knee Joint

Objective To observe the therapeutic efficacy of centro-square needling plus stuck needling with lifting-thrusting manipulations in treating the injury of medial collateral ligament (IMCL) of knee joint. Methods Seventy-eight patients with IMCL of knee joint were randomized into a treatment group of 40 cases and a control group of 38 cases. The treatment group was intervened by centro-square needling plus stuck needling with lifting-thrusting manipulations, while the control group was by local anesthesia treatment. The therapeutic efficacies were compared between the two groups. Results The total effective rate was 97.5% in the treatment group versus 81.6% in the control group, and the difference was statistically significant (P<0.05). Conclusion Centro-square needling plus stuck needling with lifting-thrusting manipulations is an effective way in treating IMCL of knee joint.
5.Assessment of the results of an investigation on eliminating iodine deficiency disorders in Ya'an city of Sichuan province in 2009

Objective To evaluate the investigation outcome of eliminating iodine deficiency disorders(IDD)in Ya'an city of Sichuan province,and to master the current situation of IDD in the whole city.Methods In 2009,according toCounties'Assessment Guideline in Sichuan for Implementing the Measure to Eliminate Iodine Deficiency Disorders,assessments were conducted.At county level,composite score was given to collected data of the eight counties(districts)and monitoring data of household iodized salt were statistically analyzed.A township (town)was randomly selected by east,west,south,north and center locations,respectively.One primary school was randomly selected in each selected township(town),eighty children aged 8 to 10 in each selected school were checked of their thyroid by palpation(WS 276-2007)and urinary iodine level of20 children among them was measured by As3+_Ce4+ catalytic spectrophotometry(WS/T 107-2006).Thirty fifth-grade students and 10 housewives lived around school were asked to take a questionnaire survey.Results Average score of the 8 counties(districts)was 89.96,ranged from 85.42 to 92.90.The coverage rate of iodized salt of household was 100.00%(2328/2328),rate of qualified iodized salt was 97.98%(2281/2328).and consumption rate of qualified iodized salt reached 97.98%(2281/2328).The goiter rate of children aged 8-10 was 0.91%(33/3620),median urinary iodine was 235.85μg/L,and 1.50%(12/800)of child's urinary iodine was less than 50μg/L.The rate of passing a knowledge questionnaire survey of the fifth-grade students and women was 92.33%(1 131/1225)and 9413%(397/421),respectively.Conclusions Ya'an city in Sichuan province has reached the goal of eliminating iodine deficiency.disorders,but there is a coexist trend of IDD and iodine excess.They should pay attention to the daily consumption of iodized salt in the future;sustained elimination of IDD can be reached only by iodine supplementation scientifically.
6.Current advance in treatment of fungal keratitis by corneal collagen cross-linking

Corneal collagen cross-linking (CXL) is a photochemotherapy for keratoconus, keratectasias and infectious keratitis.Corneal collagen fiber infiltrated with riboflavin, a photosensitizer which generates reactive oxygen species when activated by UVA at 370nm, was induced to form the crosslinks in corneal stroma.Corneal collagen cross-linking alone or combined with medicine therapy could be used to treat fungal keratitis, improve the cure rates, and reduce the complications and the demands of corneal transplantation surgery.The purpose of this paper is to review the basic principle, procedure, laboratory researches, clinical applications and the safety about this treatment.
7.Hypoxia/reoxygenation and lipopolysaccharide induced nuclear factor-κB and hypoxia-inducible factor-1α signaling pathways in intestinal epithelial cell injury and the interventional effect of emodin

Objective To observe pathological process of intestinal epithelial cells subjected to ischemia,ischemia/reperfusion injury and inflammation simulated hypoxia/reoxygenation (H/R) and lipopolysaccharide (LPS) challenged human fetal normal colonic cell (FHC) line in vivo,and to observe the changes when the assaulted intestinal epithelial cells were treated with emodin,in order to explore the possible intervention targets of emodin.Methods Normoxia group:the FHC cells were cultured in 95% air and 5% CO2 at 37 ℃.Hypoxia (H) group:the cells were cultured with a mixed anaerobic gas of 1% O2,5% CO2 and 94% N2 at 37 ℃ for 1,2,3,4 hours.H + LPS group:the cells were cultured in hypoxic condition as H group with simultaneous challenge of LPS (1 mg/L).H/R group:the cells were cultured in hypoxia for 3 hours followed by reoxygenation for 1,2,3 and 4 hours,respectively.H/R + LPS group:the cells were cultured in H/R as H/R group and LPS (1 mg/L) simultaneously.Emodin intervention group:the cells were cultured in H3 h/R2 h + LPS and emodin (20,40,60,80 μmol/L) simultaneously.The variation trends of phosphorylation nuclear factor-κB profilin-o (pIκB-α),phosphorylation NF-κBp65 (pNF-κBp65) and their downstream target gene cyclooxygenase-2 (COX-2),and hypoxia-inducible factor-1α (HIF-1 α) were determined by Western Blot.The morphological changes in intestinal epithelium in different groups were observed using light microscope.The effect of emodin on the proliferation of intestinal epithelial cell was measured by methyl thiazolyl tetrazolium (MTT) assay.Results ① H group:the expressions of pIκB-α,pNF-κBp65 and COX-2 were upregulated,peaking at H1 h (0.350 ± 0.018,1.083 ± 0.054,0.903 ± 0.045),and then they gradually lowered (F value was 3.011,7.247,5.754,P value was 0.013,0.000,0.005,respectively).The expression of HIF-1 α peaked at H3 h (1.511 ± 0.076),but there was no significant difference among different groups (F=1.881,P=0.062).H + LPS group:the expressions of pIκB-α,pNF-κBp65,COX-2,HIF-1α were increased with elongation of duration of hypoxia,and a maximal induction was observed at H3 h (0.504 ± 0.025,1.255 ± 0.063,0.812 ± 0.041,1.209 ± 0.075,F value was 2.683,8.774,9.765,2.432,and P value was 0.011,0.000,0.000,0.026,respectively).H/R group:with the prolonged duration of reoxygenation,the expressions of NF-κB signaling pathway proteins (pIκB-α,pNF-κBp65,COX-2) were decreased and dropped to nadir at H3 h/R4 h (0.712 ± 0.034,1.202 ± 0.048,0.691 ± 0.042,F value was 1.923,6.765,2.719,and P value was 0.063,0.000,0.016,respectively).Compared with H group,HIF-1α was decreased with a prolonged duration of reoxygenation in H/R group,but there was no significant difference in value among different time points (F=1.280,P=0.081).H/R + LPS group:pIκB-o,pNF-κBp65,COX-2,HIF-1α showed no sign of degradation with the prolonged duration of reoxygenation,and their expression increased to maximum analogously at R2-3 h (3.302 ± 0.061,2.315 ± 0.055,2.017 ± 0.043,2.413 ± 0.098,Fvalue was 4.614,1.652,5.970,2.076,and Pvalue was 0.001,0.067,0.000,0.037,respectively).Emodin group:emodin when co-treated with H/R + LPS inhibited the expression of HIF-1o and NF-κB pathways with a dose-effect relationship (P<0.05 or P<0.01).Emodin at the dose of 80 μmol/L showed most marked inhibition (2.599 ± 0.130,1.772 ± 0.089,2.590 ± 0.129,2.518 ± 0.125).However,after treatment of emodin did not show such effect.② After treatment with H/R + LPS,there were morphological changes in cells:vacuoles,deformation and fusion.The speed of cell growth became much slower compared with H group.③ Emodin (20-80 μmol/L) had no significant effect on cell proliferation.Although emodin produced biological effect in this concentration range,it had no cellular toxicity.Conclusions Both hypoxia and inflammation can activate the hypoxia pathway of HIF-1α and the pro-inflammatory pathway of NF-κB,but different stimuli cause varying degrees of activation in these two pathways.In H/R group,both pathways were weakened during reoxygenation.However,in H/R + LPS group,the proteins remained to show a relatively high expression during the process of reoxygenation.This may be related to the pathophysiological mechanism of intestinal ischemia/reperfusion injury:hypoxia/reperfusion injury and LPS act together to destroy the intestinal epithelial cells and induce gut-derived sepsis.Emodin may inhibit inflammation by blocking HIF-1α/NF-κB-COX-2 signaling pathways.
8.Isolation and Identification of Non-fermenting Bacteria and Their Antibiotic Resistance Analysis

OBJECTIVE To investigate the distribution and resistance of non-fermenting bacteria isolated from(patients) in 2005 and offer a basis for the treatment of bacterial infection.METHODS The isolated bacteria were(identified) with API identified test(API Inc,France) and Kirby-Bauer(K-B) test used for the antibiotics(susceptivity) test.The data were analyzed by using WHONET-5 software.RESULTS Totally 604 strains of non-fermenting bacteria were isolated from the 2908 pathogenic strains.The most common bacteria were Pseudomonas aeruginosa(52.32%),followed by Stenotrophomonas maltophilia(14.07%) and Acinetobacter baumannii((13.74%)).76.32% of non-fermenting bacteria were isolated from the sputum.These bacteria had various(resistances) to all detected antibiotics.CONCLUSIONS Non-fermenting bacteria have high isolation rate and(multi-drug) resistance,so antibiotics should be used correctly under the guidance of antibiotic susceptibility testing.
9.Clinical analysis of 35 cases of adult henoch schonlein purpura

Objective To examine the epidemiological and clinical features of the adults with Henoch Schonlein Purpura (HSP),and improve the diagnosis and therapeutic efficacy of HSP.Methods Thirty five adults with HSP from the hematological department of Tianjin Medical University General Hospital between 1993.9 to 2005.12 were enrolled in this study.Epidemiological,clinical and laboratory data,therapeutic methods and efficacy were investigated.Results Most patients with HSP in our department were male and adolescent.HSP occurred more commonly in the winter.The important predisposing factors were infection and having specific food.Purpuric lesions were the most common presenting signs.Multiple organ involvement was more common.Purpura was present in all case,lower extremities were more commonly involved.Joint involvement was observed in 15 cases(42.9%),abdominal involvement in 20 cases(57.1%),renal involvement in 20 cases(57.1%).Conclusion The adult patients with HSP had more tendenecy to have severe renal damage and multiple organ involvement.
10.Piezoelectric Immunoglobulin Aptamer Biosensor for Determination of IgE

OBJECTIVE A new piezoelectric aptamer biosensor is developed for determination of IgE. The energy converters are 10MHz AT-cut quartz crystals with gold-coated electrodes. The anti-IgE aptamers are immobilized onto the surfaces of crystals gold electrodes by biotin-avidin method. METHODS The standard substance and serum were detected to find the limit of detection and specificity of the biosensor. RESULTS The piezoelectric immunoglobulin aptamer biosensor could complete the detection without nonspecific response. Under the optimized conditions, the experimental results showed that the piezoelectric biosensor had good response to IgE whose frequency shifts were linearly dependent on IgE concentration in different range. The piezoelectric aptamer biosensor had been used to detect IgE in serum, the analytical results given by this method were in satisfactory agreement with those given by chemoluminescence method, its correlation coefficient was 0.9924. CONCLUSIONS Piezoelectric aptamer biosensor for the determination of IgE is of high sensitivity, high specificity, high analysis speed, unnecessary labeling, simple operation, real-time detection, etc. It is suitable for detection of IgE and should be used for clinical detection.
