Objective:To optimize the condition of anthrone-sulfuric acid method to determine the polysaccharide isolated from Bletilla striata by central composite design/response surface methodology.Methods:Dextron T20 was a standard substance, independent variables were the ratio of the polysaccharide and anthrone-sulfuric acid,the time of color hydrolysis and the cooling time at room temperature,dependent variable was the UV absorption,nonlinear mathematic models were used to estimate the relationship between independent and dependent variables.Response surface methodology was used to optimize be condition of determination.Results:The optimum conditions of determination:the volume ratio of the polysaccharide and anthrone-sulfuric acid is 1:2;the time of color hydrolysis is 8.00 min;the cooling time at room temperature is 15.00min.On this condition, there was favorable correlation between the absorption of Dextron T20 and polysaccharide of Bletilla striata.Conclusions: Central composite design/response surface methodology can be used to evaluate the feasibility and applicable conditions of the anthrone-sulfuric acid method to determine the polysaccharide isolated from Bletilla striata using Dextron T20 as a standard substance.

AIM and METHOD To investigate the antibacterial activity (ED 50 value) of cefoperazonesodium and tazobactam sodium (4∶0 5) in vivo using minor modified Karbers method,which will offer some important evidences for clinical trial and therapy RESULTS The ED 50 value of cefoperazone sodium and tazobactam sodium(4∶0 5) and Cefoperazone sodium and sulbactam sodium (1∶1) in vivo protested against zymogenic staphylococcus aureus and Escherichia coli in mice infected by these bacteria were 92 0 mg?kg -1 and 3 4 mg?kg -1 ,164 0 mg?kg -1 and 6 9 mg?kg -1 ,respectively,and 256 5 mg?kg -1 and 38 9 mg?kg -1 in single composed compound of cefoperazone sodium,cefoperazone sodium and tazobactam sodium (4∶0 5) in vivo had more potent antibacterial activity compared with cefoperazone sodium and sulbactam sodium (1∶1) and single composed compound of cefoperazone sodium. The ED 50 value of cefoperazone sodium and tazobactam sodium (4∶0 5) protested against pseudomonas aeruginosa in mice infected was lower than that of Cefoperazone sodium and sulbactam sodium and single composed compound of cefoperazone sodium CONCLUSION Cefoperazone sodium and tazobactam sodium made in China has an markedly therapeutic effect on antibacterial activity protected against zymogenic staphylococcus aureus and Escherichia coli and pseudomonas aeruginosa in mice infected by these bacteria

OBJECTIVE To investigate the TORCH infection of women with childbearing age in Chongqing.(METHODS) Special IgM antibodies were detected by the TORCH-IgM screen test kits in 2067 women with(childbearing) age,among them 1829 were women with normal childbearing and 238 were women with abnormal one.RESULTS The infection rate of Toxoplasma gondii(TOX),Rubella virus(RV),Cytomegalovirus(CMV) in women with abnormal childbearing was significantly higher than those in women with normal one(P(0.05)).CONCLUSIONS The(abnormal) pregnancy is (related) with TORCH infection.So TORCH screen before pregnancy is very important to aristogenesis.

OBJECTIVE To have a research on the detection of Mycobacterium tuberculosis by fluorescence chip of molecular beacon probe.METHODS Fluorescence chip for the detection of M.tuberculosis was designed,based on the better bonding force and specificity of molecular beacon and target molecule.Hybridization efficiency was optimized,which included the concentration and purity of molecular beacon probe,hybridization temperature,hybridization time,the prescription of hybridization solution and the comparing of molecular beacon fluorescence signals of 4 molecules.RESULTS Molecular beacon fluorescence signal approached the highest when the final concentration of Mg was 5 mmol/L and the amplification efficiency of PCR approached the highest.Half extended molecular beacon probe could bind nice with chip when it was middle labeled with quench group.The results of 3 hybridization solutions were as follows: 0.15% SDS better than 0.2% SDS,10?SSC better than 5?SSC.Strong signal of hybridization reaction could be detected when the concentration of Cy3-BDH molecular beacon probe was 15 ?mol/L,the concentration of FAM-DABCLYE probe was 9 ?mol/L and the reaction time was 7 h.The efficiency of fluorescence quenching was higher when the molecule was consisted of Cy3-BDH,other than Cy3-DABCLYE.CONCLUSIONS The design of half extended molecular beacon probe tactfully makes use of the advantages of the high bonding force and specificity of molecular beacon probe on the chip technology.Fluorescence chip of molecular beacon for M.tuberculosis has satisfactory detecting rate,specificity and sensibility.

AIM: To understand the formation of portal hypertension through the change of portal hemodynamics on experiment cirrhosis. METHODS: Carbon tetrachloride was subcutaneously injected in the rat. The changes of the portal hemodynamics in the pathological process of liver tissue were observed. RESULTS: The liver underwent degeneration, necrosis of hepatocytes, and the normal architecture of the liver lobules was replaced by pseudolobule, which consist of regenerative hepatocytes and fibrous septa. The diameter, the blood flow velocity and the blood flow quantity of portal were significantly higher than that in former group (P0 05). CONCLUSION: The changes in hemodynamics of the portal are in accordance with the changes in pathology of liver in the formation of liver cirrhosis.

AIM To investigate the influence on change of SOD and MDA in the blood of rats which had cellular immunoreactive colitis by Fengliao Changweikang Granule (CWKG). METHODS Male Wistar rats were divided into six groups. Numbers of each group were 32. Animal model of rat cellular iumynoreactive colitis was made by TNBS and ethanol mixed liguid enema. Rats were killed at the 1st, 3rd, 7th and 21st day respectively. Gross morphologic observation of intestinal wall was made, SOD vitality and MDA content in blood were determined. Aime To study the anti-C. albicans activity of baicablin,and to explore its anti-C.albicans mechanism preliminarily. Method ① The minimum inhibitory concentration (MIC) and the minimum fungicidal concentration(MFC) of baicalin against C.albicans were determined by microdilution method. ② By means of incorporation experiment of isotope-labelled precursor, the incorporation inhibitation rates of [ 3H]-TdR,[ 3H]-UdR,[ 3H]-leucine were measured after treatment with various concentrations of baicalin respectively. Result ① The MIC and MFC of baicalin (above purity 98%) against C albicans(1?107 cfu?L -1) were both 1.0 g?L -1. The MIC and MFC of baicalin (above 98% purity) against C albicans(2.5?107 cfu?L -1) both 2.0 g?L -1. ② The dose of baicalin of 0.5, 1 and 2 MIC could obviously inhibit incorporation of the precursors and there were significant differences among the inhibition rate of three precursors incorporation (P0.05). Conclusion ① Baicalin had strong anti-C albicans activity in vitro. ② Baicalin(above purity 98%) could obviously inhibit incorporation of [ 3H]-TdR, [ 3H]-UdR and [ 3H]-leucine into C albicans and the incorporation inhibition rate of [ 3H]-UdR was highest among them, suggesting that the anti-C albicans mechanism of baicalin may be involved with the inhibition of DNA, RNA and protein synthesis.
