1.Anti-apoptotic actions and mechanisms of ghrelin in cardiomyocytes after hypoxia/reoxygenation injury

Objective:To explore the anti-apoptostic effect of Ghrelin in cardiomyocytes after Hypoxia/Reoxygenation injury and manifest the specific cellular singaling transduction pathway.Methods:Newborn SD rats' hearts were isolated and cardiomyocytes were separated by trypsin digestion method .The primary cardiomyocytes were collected and incubated in vitro and randomly alloted to 4 groups(①Control;②H/R group;③H/R +Ghrelin group;④H/R+Ghrelin+Wortmannin group) after 5 days of cultivation.Hypoxia/ Reoxygenation injury was established by 4 hours of hyopxia and subsquently 1 hour of reoxygenation.Lactic dehydrogenase concentration,cell apoptotic rate and the expression of phosph-Akt were detected by Automatic Biochemistry Analytic Instrument ,Tunel method and Western blotting method respectively.Results:Compared with H/R group and H/R+Ghrelin+Wortmannin group,the LDH concentration in media was significantly reduced and the myocardium apoptotic rate was pretty low in H/R+Ghrelin group.Ghrelin also increased phosph-Akt activity but this effect was abolished by applying Wortmannin simultaneously.Conclusions:Ghrelin provides concrete protection against Hypoxia/ Reoxygenation injury in the primary cultivated rat cardiomyocytes,which manifest dereasing LDH leaking,inhibitting cell apoptosis.Theantiapoptotic effect is partly mediated by the activation of the PI3K /Akt(Phosphatidylinositol-3 kinase/ Akt) pathway.

Pancreatic adenocarcinoma is a severe-invasive, early-metastasis and malignant neoplasm with latent pathogenesis, while its biological behaviors are the highlight of current tumor study.In present study,through reviewing the studies related to expression profile of pancreatic cancer up to now.This artical summarized the achievement, challenge and trend of development of expression profile in pancreatic cancer.In conclusion, exploring the mechanism of carcinogenesis and development of pancreatic cancer based on the breakthrough of expression-profiling studies and establishing ideal models of pancreatic cancer, will promote systemic diagnosis and treatments of pancreatic cancer and do favor to the prognosis of the patients.
3.Study on Microbial Detecting Methods for Cosmetics

Objective To study the viability of S.aureus in common cosmetics and the sensitivities of different detection methods adopted in detecting faecal coliform. Methods A standard method stated in "Hygienic Standard for Cosmetics" and lactose-bile salt medium improved by adding neutralizers were adopted in detecting Faecal coliform. Results The amounts of S.aureus dropped gradually as the prolongation of preservation period and in cosmetics containing or not containing antiseptics were different too. In detecting Faecal coliform, the sensitivity of the improved method was higher than that of the standard method. Conclusion Some ingredients of common cosmetics, especially antiseptics, obviously affected the viability of S.aureus. The lactose-bile salt broths containing Lecithin and Tween 80 were much better than those not containing Lecithin and Tween 80 in detecting faecal coliform.

5.Synergistic inhibitory effects of pirarubicin combined with hyperthermia on proliferation and invasion of gastric cancer cells

Objective To investigate the synergistic effects of pirarubicin (THP) combined with hyperthermia on proliferation and invasive potential of gastrointestinal cancer cells in vitro and to discuss the underlying mechanisms. Methods Gastric cancer cell line MGC-803 was established, and they were divided into 4 groups: control group (37℃), high temperature group (43℃), drug group (37℃+THP), and drug and high temperature group (43℃+THP). After THP and hyperthermia treatment, cell proliferative ability was determined by soft agar clonogenic assay, cell nuclear antigen PCNA was detected by flow cytometry, cyclin E mRNA level was assessed by reverse transcription PCR (RT-PCR). Cell invasion assay was assessed by using transwell chamber model. OBJECTIVE:To study the pharmacokinetics and relative bioavailability of two kinds of preparations of valaciclovir hydrochloride.METHODS:This was a randomized two-way cross-over study.A total of 18 healthy volunteers were randomly assigned to receive single oral dose of test valaciclovir tablets(trial group) or reference valaciclovir tablets(control group),with the plasma aciclovir concentrations determined by HPLC,the pharmacokinetics parameters calculated and the relative bioavailability evaluated using 3p97 program.RESULTS:The main pharmacokinetic parameters test and reference valaciclovir hydrochloride tablets were as follows:AUC0~ 24:(12.85? 4.32) vs.(12.19? 4.63) ?g? h? mL-1;AUC0~ ∞:(14.65? 5.75) vs.(13.27? 5.03) ?g? h? mL-1;Cmax:(3.55? 0.92) vs.(3.71? 0.97) ?g? mL-1;tmax:(1.44? 0.43) h vs.(1.33? 0.37) h;t1/2:(6.23? 2.81) h and(4.55? 1.84) h.The relative bioavailability of the test valaciclovir tablets were(111.01? 23.52) %.CONCLUSION:The reference valaciclovir tablets and the test valaciclovir tablets were bioequivalent.
8. Callus induction and establishment of plant regeneration system of Saposhnikovia divaricate

Objective: To establish the regeneration system of Saposhnikovia divaricate (Turcz.) Schisck. Methods: The effects of different explants, temperature, medium, and illumination was studied on the induction of S. divaricate. Response surface test design was used to optimize the hormone ratio of callus induction and proliferation. Results: The leaves of S. divaricate were used as the experimental materials, the optimal medium for the callus induction was MS + 6-BA 0.90 mg/L + 2,4-D 1.05 mg/L + KT 0.96 mg/L, with temperature of 25 ℃ and light of 12 h/d. The optimal medium for callus proliferation was MS + 6-BA 0.56 mg/L + 2,4-D 0.48 mg/L + KT 0.40 mg/L, and the subculture cycle was 35 d. The optimal medium for adventitious shoots was MS + NAA 0.6 mg/L + 6-BA 1.0 mg/L. The optimal medium for root growing was MS + NAA 0.4 mg/L + 6-BA 0.1 mg/L. Conclusion: S. divaricate callus was successfully induced and a stable and efficient plant regeneration system was established.
9. Endovascular embolization of an infectious intracranial aneurysm caused by paragonimiasis: A case report

Infectious intracranial aneurysm induced by paragonimiasis is very rare in clinic. Interventional treatment is one of the effective therapies and regular antiparasite therapy is necessary after embolization. This retrospective study summarized one case of intracranial aneurysm caused by paragonimiasis involving the medical history, laboratory examination, imaging findings and treatment strategy, and analyzed the diagnosis and treatment of the disease with literature review.
10. Characteristics of Vancomycin Blood Concentrations after Intraperitoneal Administration and Its Relationship with Efficacy in PDRP Patients with Peritoneal Dialysis-related Peritonitis

OBJECTIVE: To evaluate the relationship between serum vancomycin level and efficacy in peritoneal dialysis-related peritonitis(PDRP) patients by analyzing our hospital′s data. METHODS: Forty-six PDRP patients admitted in our hospital from August 2015 to April 2018 were collected, then divided into three groups by different regimens(1 g q3d, 1 g q4d, 1 g q5d), the probability of target attainment of the first trough concentration and those after several administrations, the characteristics of distribution of vancomycin serum level and the relation with efficacy were analyzed. RESULTS: The first trough concentrations of 1 g q3d, 1 g q4d and 1 g q5d were (10.51±2.79), (6.78±1.58) and (6.68±1.68) mg·L-1 respectively, with statistical difference between 1 g q3d regimen and 1 g q4d, 1 g q5d (P0.01). The trough concentration after the 3rd administration of 1 g q3d regimen was (16.15±4.79) mg·L-1, the trough concentration after the 2nd administration of 1 g q4d regimen was (10.20±2.0) mg·L-1, and the trough concentration after the 2nd administration of 1 g q5d regimen was (9.49±3.24) mg·L-1. The serum vancomycin level was increasing after repeated administration with obvious statistical difference among the three regimens(P0.01). There was no significant difference in the efficacy between concentration10 mg·L-1 and that ≥10 mg·L-1 or concentration <15 mg·L-1 and that ≥15 mg·L-1(P0.05). CONCLUSION: There is significant inter-individual differences of serum vancomycin level in PDRP patients after IP administration, and vancomycin is accumulated in body after repeated administration. It is suggested to monitor the serum vancomycin concentration and the trough concentration kept above 10 mg·L-1.
