1.Analysis residues of perfluorooctane used in ophthalmic operation.

Analyzed the purity of the Perfluorooctane used for Ophthalmic operation using the FID (Flame Ionization Detector) method after the qualitative analysis of that. Research and discussion of the 1--hydrogen perfluorocarbon octane, which is the typical impurity of Perfluorooctane. The standard recovery of the 1--hydrogen perfluorocarbon octane is 93%-97% within the range of linearity, and the RSD (relative standard deviation) is below 2%. The above test method has the advantages of easy operation, stability and accuracy. So it is a good reference support to bring the specification of quality control of the Perfluorooctane which is used for Ophthalmic operation into YY of medical device.

2.Simultaneous determination of three organic fluorides in workplace air by portable gas chromatography-mass spectrometer.

OBJECTIVETo establish a method for rapid determination of organic fluorides in the air of a fluorine chemical plant using portable gas chromatography-mass spectrometer (GC-MS).
METHODSStandard samples of monochlorodifluoromethane, tetrafluoroethylene, and hexafluoropropylene of different concentrations were prepared by static volumetric method with high-purity nitrogen as the diluent gas. The samples were injected into the GC-MS by a hand-held probe. Retention time and characteristic ion were used for qualitative analysis, and the area of selected ion peak was used for quantitative analysis. The standard curves were then created for quantitative determination of the three organic fluorides.
RESULTSThe linear ranges for monochlorodifluoromethane, tetrafluoroethylene, and hexafluoropropylene by the method were 0.39-7.72, 0.45-8.84, and 0.61-12.20 mg/m3, respectively, the average recovery rates for the three concentrations were 102.8%, 96.0%, and 106.5%, respectively, and the average deviations were 2.1%, 5.1%, and 2.4%, respectively.
CONCLUSIONThe portable GC-MS can be used for the simultaneous qualitative and quantitative analysis of monochlorodifluoromethane, tetrafluoroethylene, and hexafluoropropylene in the workplace air, and the method is simple, fast, and accurate.

3.Research on the establishment of standard limits for perfluorooctanoic acid and perfluorooctane sulfonate in the "Standards for Drinking Water Quality(GB5749-2022)"in China.

Perfluorinated compounds, especially Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS), are widely detected in water environments in China. Considering the potential health risks of drinking water exposure routes, PFOA and PFOS have been added to the water quality reference index of the newly issued "Standards for Drinking Water Quality (GB5749-2022)", with limit values of 40 and 80 ng/L, respectively. This study analyzed and discussed the relevant technical contents for determining the limits of the hygiene standard, including the environmental existence level and exposure status of PFOA and PFOS, health effects, derivation of safety reference values, and determination of hygiene standard limits. It also proposed prospects for the future direction of formulating drinking water standards.

5.The influences of inorganic elements in soil on the geolism of Atractylodes lancea.

OBJECTIVETo study the influence of inorganic elements in soil on the geolism of Atractylodes lancea.
METHODThe concentrations of 15 kinds of inorganic elements of regional A. lancea and their correspondingly soils were determined, and the data were analysed with the SPSS10.0 software.
RESULTThe mean concentration of Ni in geoherbs of A. lancea was 3 times higher than that in the non-geoherbs. The accumulation of Ca is high in geoberbs. There is no relationship between the concentrations of inorganic elements in A. lancea and those in their corresponding soil.
CONCLUSIONA. lancea adjusts the concentration of inorganic elements mainly by active absorption, and there are good relationship between the accumulation of Fe and Cr, Fe and Hg, Hg and Cr, As and Pb, Co and Sr.

6.Determination of octafluoropropane content in human albumin micropheres loaded with octafluoropropane.

OBJECTIVETo develop a method for determinating the octafluoropropane (OFP) content in human albumin micropheres loaded with OFP.
METHODSOFP content in the loaded human albumin micropheres was determined by means of gas chromatography coupled with mass spetrum/selected ion monitoring (GC-MS/SIM).
RESULTSThe calibration curve was linear within the concentration range of 40.92-16,368 nmol/L (R(2)=0.9999). The limit of detection was 8.18 nmol/L, and the recovery rate was (100.98-/+1.4)% (204 nmol/L), (102.63-/+2.29)% (2,046 nmol/L) and (97.5-/+2.2)% (8,184 nmol/L), respectively. The intra-day and inter-day RSDs were 0.6%-2.0% and 0.86%-2.3%, respectively.
CONCLUSIONThe method of GC-MS/SIM is accurate, sensitive, precise and applicable for OFP determination in the microspheres.

7.Sampling of exhaled gas after intravenous administration of octafluoropropane-containing human albumin micropheres in dogs.

OBJECTIVETo develop a method for quantitative collection of exhaled gas in anesthetized dogs at given time following intravenous administration of octafluoropropane (OFP)-containing human albumin micropheres for assessing the gas kinetics of OFP.
METHODSOFP-containing albumin micropheres were administered intravenously at 0.4, 0.8 and 1.2 ml/kg, respectively, in anesthetized and ventilated dogs. The exhaled air samples were analyzed by gas chromatography-tandem mass spectrometry (GC-MS-MS).
RESULTSThe correlation curve between the area under curve (AUC) and administered dose was roughly linear (Y=1162.5X-417.38, r square=0.949 9). The total recovery rate was (119.49-/+27.62)% which was not significantly different from the rate of 100% (P>0.05). GC-MS-MS was accurate, sensitive, precise and applicable for OFP determination.
CONCLUSIONThe sampling method is useful for characterizing OFP pharmacokinetics in dogs, and also applicable for studying the pharmacokinetics of other gas-containing drugs.

8.Correlation analysis between prenatal exposure of per-/polyfluoroalkyl compounds and neonatal outcome.

Objective: To investigate the correlation between the prenatal exposure of per-/polyfluoroalkyl substances (PFASs) and the neonatal outcome. Methods: A total of 506 maternal infant cohort samples were collected in Hangzhou, Zhejiang province from 2020 to 2021. The exposure levels of seven PFASs in maternal serum before delivery were detected by solid-phase extraction-ultra performance liquid chromatography tandem mass spectrometry. Multivariable linear regression model was used to analyze the influence of prenatal exposure of PFASs on birth weight, birth length and Apgar score. Results: The maternal age, prenatal body mass index and gestation age were (31.3±4.3) years old, (26.7±3.2) kg/m2 and (265.0±28.3) days, respectively. The birth weight, birth length and scores of Apgar-1 and Apgar-5 were (3.1±0.8) kg, (49.3±2.9) cm, (9.88±0.47) points and (9.99±0.13) points, respectively. PFASs were widely distributed in maternal serum, with the highest concentration of (18.453±19.557) ng/ml, (6.756±9.379) ng/ml and (5.057±8.555) ng/ml for perfluorooctanoic acid (PFOA), perfluorooctane sulfonate (PFOS) and 6∶2 chlorinated polyfluorinated ether sulfonate (Cl-PFESA), respectively. Maternal age, parity and delivery mode were associated with the exposure level of PFASs (P<0.05). Subgroup analysis showed that PFOS had negative effects on birth weight (β=-0.958), birth length (β=-0.073) and Apgar-5 score (β=-0.288) for neonates in the low birth weight (LBW) group. 6∶2 Cl-PFESA and 8∶2 Cl-PFESA inhibited the birth weight (β=-0.926; β=-0.552) and length (β=-0.074; β=-0.045) of newborn in the LBW group. In addition, 4∶2 fluorotelomer sulfonate (FTS) was associated with increased birth weight (β=0.111) and decreased Apgar-5 score (β=-0.030) in the normal weight group. Conclusion: Prenatal exposure to PFASs is associated with birth weight, birth length and Apgar-5 score. It is necessary to continue to pay attention to the impact of PFASs on fetal growth and development through maternal-fetal transmission.

9.Partial liquid ventilation with perfluorocarbon improves gas exchange and decreases inflammatory response in oleic acid-induced lung injury in beagles.

The aim of this study was to determine the effect of partial liquid ventilation (PLV) using a perfluorocarbon (PFC) on gas exchange and lung inflammatory response in a canine acute lung injury model. After inducing severe lung injury by oleic acid infusion, beagle dogs were randomized to receive either gas ventilation only (control group, n = 6) or PLV (PLV group, n = 7) by sequential instillation of 10 mL/kg of perfluorodecalin (PFC) at 30 min intervals till functional residual capacity was attained. Measurements were made every 30 min till 210 min. Then the lungs were removed and bronchoalveolar lavage (BAL) (35 mL/kg) was performed on the right lung and the left lung was submitted for histologic analysis. There was significant improvement in PaO2 and PaCO2 in the PLV group compared to the control group (p < 0.05) which was associated with a significant decrease in shunt (p < 0.05). There was no significant difference in parameters of lung mechanics and hemodynamics. There was a significant decrease in cell count and neutrophil percentage in BAL fluid and significantly less inflammation and exudate scores in histology in the PLV group (p < 0.05). We conclude that PLV with perfluorodecalin improves gas exchange and decreases inflammatory response in the acutely-injured lung.
