1.Investigation and study on the happiness index and social support of clinical nurses

Objective To investigate the happiness index and social support of clinical nurses. Methods Inquiry was performed in 60 clinical nurses by Happiness Index scale and Social Support Scale. Results Happiness Index of married and military nurses were higher than that of unmarried and employed nurses. Married nurses gained more social support, objective support and subjective support than unmarried nurses did. Married nurses were also good at making use of support. Conclusions Efforts for enhancing the status of professional nurses in the society and family will improve the nurses' ability of subjective use of the society support.
2.Effects of glucose concentration fluctuation on function of cultured bovine arterial endothelial cells

Objective To explore the effects of glucose concentration fluctuation on function of cultured bovine arterial endothelial cells and underlying mechanism. Methods The thoracic aorta of newborn calf was used for primary endothelial cells culture. Cells were divided into 3 groups and cultured for 48 h : control group ( C, 5.5 mmol/L), constant high glucose group ( HG, 30 mmol/L) and glucose fluctuation ( GF, three circles of 2 h 30 mmol/L followed by 3 h 5.5 mmol/L, 30 mmol/L overnight, repeat the whole procedure on the following day) groups. The membranes fluidity of endothelial cells was detected by fluorescence polarization method. The contents of sorbierite, aldose reductase (AR), sorbitoi dehydrogenase (SDH) and advanced glycation end products (AGEs) were measured. RAGE, eNOS and ET-1 mRNA expressions were detected by semi-quantitative RT-PCR. Results The membranes fluidity of endothelial cells in HG or GF group were significantly decreased compared with the control group( all P <0.01 ) and significantly lower in GF group than those in HG group (all P < 0. O1 ). Sorbierite, AR and AGEs concentrations were significantly higher in HG and GF groups than those in control group ( all P <0. 01 ) and AR and AGEs concentrations were significantly higher in GF group than that in HG group ( all P <0.01 ). SDH of endothelial cells in HG or GF group were decreased compared with the control group and lower in GF group than in HG group ( all P < 0.05). In addition, the mRNA levels of RAGE, eNOS and ET-1 were significantly upregulated compared with the control group( all P < 0.01 ). Conclusions Glucose concentration fluctuation can result in more severe bovine arterial endothelial cells dysfunction than high glucose via activating polyols metabolic pathways, upregulating the expression of AGEs, eNOS and ET-1.Therefore, glucose concentration fluctuation might play a crucial role on macrovascular complications of diabetes.
3.Influence of F protein of hepatitis C virus subtype 1b inhibits on human hepatocellular carcinom HepG2 cell apoptosis

Objective To investigate the effects of F protein of hepatitis C virus subtype lb on the apoptosis of human hepatocellular carcinom HepG2 cells. Methods HepG2 cells were transfected with recombinant plasmid pcDNA3.0-F-EGFP and pcDNA3.0-F-EGFP-HepG2 strain was exposed to Act-D and tumor necrosis factor a (TNFα) treatment in order to induce cell apoptosis with positive control pcDNA3.0-C-EGFP-HepG2, negative control pcDNA3.0-C-EGFP-HepG2 and blank control HepG2.Annexin V-FITC/PI of flow cytometry was performed to determine the number of apoptotic cells. DNA Ladder was used to observe the isolation of apoptotic DNA fragments in the apoptotie cells. Results pcDNA3.0-F-EGFP- HepG2 cell strain showed a much delayed apoptosis as well as obviously lowering the apoptotic rate when compared with the pcDNA3.0-HepG2 strain and HepG2 strain (P<0.001).Conclusion The introduction and expression of extraneous gene (the F gene of hepatitis C virus subtype 1b) could significantly inhibit the apoptosis of HepG2 cells.
4.Analysis of relationship of HBV PreS1 antigen, anti-HBc IgM , DNA load and genotypes in hepatitis B patients

Objective To explore the relationship of HBV PreS1 antigen,anti-HBc IgM,DNA load and genotypes,and the significance for clinical diagnosis and prognostic.Methods Enzyme linked immune-sorbent assay was used to test the HBV serum markers of HB patients; HBV-DNA copies was detected by time fluorescence quantitative PCR; using nested PCR to amplify the S fragment of HBV genome,then sequenc and make blast with HBV standard sequences to ascertain genotypes.Make comprehensive analysis of these indexes.Results 355 serum specimens of acute or chronic HB patients were collected.The positive rates of HBV PreS1-Ag and HBV-DNA in model Ⅰ (positive for HBeAg) were 80.2% and 73.7% respectively,which both higher than other models.The abnormal rate of ALT and AST were higher in PreS1-Ag positive group than negative,as well as in anti-HBc IgM positive group.There are 4 samples is genotype B (2.9%),76 genotype C(55.9%) and 56 genotype D (41.2%).Positive rate of HBeAg and HBV-DNA of genotype C samples were both higher than which of genotype B and D.Conclusion PreS1-Ag and Anti-HBe-IgM indexes are of great value to viral hepatitis B early diagnosis,HBV replication surveillance and prognostic evaluation; the major HBV genotypes in Henan province are C and D,and the positive rate of HBeAg and HBV-DNA were both higher in genotype C HBV infection population than genotype B and D.
5.Serum proteomic patterns may assist the diagnosis of solitary pulmonary nodules.

OBJECTIVETo screen relatively specific biomarkers in serum from lung adenocarcinoma patients by surface-enhanced laser desorption and ionization time of flight mass spectrometry (SELDI-TOFMS), and to investigate the clinical value of SELDI-TOF-MS in differentiation of benign from malignant solitary pulmonary nodules (SPN).
METHODSSerum samples from 71 lung adenocarcinoma patients and 71 healthy volunteers with matched gender, age and history of smoking were analyzed using WCX2 ProteinChip to screen potential biomarkers. 28 patients received surgical treatment among total 53 patients with SPN. The clinical value of SELDI-TOF-MS in differentiation of benign from malignant solitary pulmonary nodules was evaluated by pathological diagnosis.
RESULTSFive highly expressed potential biomarkers were identified with the relative molecular weights of 4047.79 Da, 4203.99 Da, 4959. 81 Da, 5329. 30 Da and 7760.12 Da. The postoperative pathologic diagnosis was lung adenocarcinoma in 24 patients with SPN, validating the clinical value of the 5 potential biomarkers.
CONCLUSIONSELDI-TOF-MS technology is a quick, easy, convenient, and high-throughput analyzing method capable of screening several relatively specific potential biomarkers from the serum of lung adenocarcinoma patients and may have attractive clinic value in differentiation of solitary pulmonary nodules.
