1.Relationship between urinary biopyrrin and coronary atherosclerosis

Objective To study the relationship between urinary biopyrrin level and the degree of coronary atherosclerosis.Methods 220 inpatients with coronary artery disease,which were identified by coronary angiography within 1 year(from April 2006 to April 2007),and 80 healthy examinees (healthy group)were enrolled in the study.The 220 inpatients were divided into three groups according to the involved number and extent of coronary arteries:1 vessel group(group A,n=80),2 vessel group(group B,n=75)and 3 vessel group(group C,n=65).Enzyme linked immunosorbent assay (ELISA)was applied to detecting urinary biopyrrin level for all subjects.Results Compared with that of healthy control group,the urinary biopyrrin in group A,B and C was much higher(P<0.05).There were statistical difference of urinary hiopyrrin level among the different groups r(P<0.05).Conclusion Biopyrrin is significantly associated with the involved number and extent of coronary arteries.

3.Effect of different types of flavonoids on H_2O_2-induced apoptosis in neonatal rat cardiomyocytes

AIM: To investigate the effect of different types of flavonoids, galangin (flavonol), hesperetin (flavonone) and hydroxysafflor yellow A (HYSA, chalcone), on cardiomyocytic injury induced by H2O2, and explore the possible signal pathways involved. METHODS: The cytotoxicity of different flavonoids was determined by MTT assay. Apoptosis rate was determined by flow cytometry (FCM) and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL). Immunohistochemistry was used for detection of Bcl-2, Bax and Caspase-3 protein. RESULTS: It showed that each flavonoid did not have noticeable cytotoxicity at a concentration of 5 ?mol/L by MTT assay. Flavonoids at concentrations of 5, 15 and 30 ?mol/L significantly increased cell viability compared to model group induced by H2O2 (100 ?mol/L). Flavonoids also increased apoptosis rate and neorobiosis rate determined by FCM compared to model group. To study the changes in levels of tumor necrosis factor ? (TNF ?), interleukin 6 (IL 6) and interleukin 1? (IL 1?) produced by the peripheral blood mononuclear cells (PBMC) of patients with congestive heart failure (CHF), and to investigate the effects of captopril, one of angiotensin converting enzyme inhibitors in active form, on production of these cytokines. PBMC from 35 CHF patients and 12 healthy controls were isolated by density centrifugation with Ficoll Hypaque, and were cultured with captopril at 10 -4 ~10 -2 mol/L in the presence of lipopolysaccharide (LPS). After incubation for 24 hours, TNF ? and IL 6 contents in the culture supernatants were measured by an enzyme linked immunosorbent assay (ELISA), and IL 1? by radioimmunoassay (RIA). 5.Internalization and down-regulation of the epidermal growth factor receptor in a human keratinocyte cell line HaCaT

Objective To understand the molecular mechanism underlying the epidermal growth factor receptors(EGFR)signal transduction and its feed-back regulation.Methods Two human keratinocyte cell lines,HaCaT and CHOwt,were cultured and treated with a certain concentration of different ligands,including epidermal growth factor(EGF),heparin-bounding(HB)-EGF,transforming growth factor α (TGFα)and heregulin(HER),for various durations(2,4,8,16,20 hours).After the treatment,cells were collected and protein was extracted.The amount of total and active EGFR was measured by immunoprecipitation and immunoblot assay.The internalization and down-regulation of EGFR were visualized with immunofluorescence and laser seanning confocal microscopy.Results As shown by immunoblot technique,EGF and HB-EGF continuously down-regulated the total amount of EGFRs,whereas TGFα and HER had no significant effect on the degradation of EGFRs.The activation of EGFRs was also attenuated to different extent after long-time treatment with EGF,HB-EGF and TGFα.As indirect immunofluorescenee revealed,in untreated HaCaT and CHOwt cells,EGFRs were essentially located at the plasma membrane,with a little cytosolic distribution;after ten-minute treatment with EGF,EGFRs clustered into patch-like structures which were particularly obvious in HaCaT cells,and translocated into cytoplasmic vesicles resembling endosomes (relatively apparent in CHOwt cells),while the total amount of EGFRs remained constant in these cells.The fluorescence signal from the total EGFRs decreased evidently after four-hour treatment with EGF,indicating a strong reduction in the receptors.Conclusions EGF and HB-EGF,but not TGFα or Heregulin,could down-regulate the amount of total and active EGFRs.There might be different mechanisms for the signal transduction related to EGFRs intemalization and down-regulation between HaCaT and CHOwt cells.
6.Comparison of transrectal ultrasound with transabdominal ultrasound in fat sufferers with bladder tumor

Objective To discuss the diagnosis value of transrectal ultrasound and transabdominal ultrasound in fat sufferers with bladder tumor.Methods In own control,transabdominal and transrectal ultrasound were used to examine 87 fat patients with suspected bladder tumor.The tumors were confirmed by cystoscope and pathological section.They were divided into 3 groups according to the size:<10 mm,10-20 mm,and>20 mm,the detection rates of the tumors and blood flow in tumors,the diagnose accordance rates of the tumor stage of the two methods were analyzed.Results Of the 92 lesions pathologically confirmed to be bladder tumors,the diagnose accordance rate of transabdominal ultrasound was 79.3%,missed diagnosis rate was 1 7.4%,misdiagnosis rate was 3.3%,while the diagnose accordance rate of transrectal ultrasound was 95.6%,misseddiagnosis rate was 4.4%,misdiagnosis rate was 0.There was statistical significance on the detection rate of thetumor and blood flow in tumor of the two methods(P<0.05).Also there was statistical significance on the diagnosis accordance rate of the tumor staging on Ta,T1,T2(P<0.05).Conclusions As to detecting bladder tumors in fat sufferers,transreetal ultrasound iS obviously superior to transabdominal ultrasound.
7.Prospective research of ventilator-associated pneumonia caused by the aerocyst continual inflation and clocked deflation

Objective To explore the relationship between the aerocyst continual inflation and clocked deflation and ventilator-associated pneumonia. Methods 60 ICU hospitalized adult patients treated with tracheotomy or trachea canalization for above 24 hours were randomly divided into the tzadi-tional group and the experimental group with 30 patients in each group. After trachea canalization success-fully according to the nursing standard, the experimental group was treated with aerocyst continual inflation, the traditional group gased the ventilator and recorded the time, deflating the gas every 4 hours and inflating again after 5 minutes. The inflation time, nurse management and MOV(minimal occlusive volume)of the two groups were the same. The chest X-rays examination and deep sputum bacilli culture were implemented 12 hours after admission, before pulling out the ventilator and transferring out of ICU to evaluate the inci-dence of ventilator-associated pneumonia according to the infection diagnosis standard. Results The rate of ventilator-associated pneumonia in the experimental group was obviously lower than that of the tradition-al group. Conclusions The rate of ventilator-associated pneumonia reduces obviously in the ICU trachea canalization patients with aerocyst continual inflation.

10.Clinical analysis of 15 cases of malignant ovarian germ cell tumors with lung metastasis

Objective To investigate the clinical characteristics,diagnosis,treatment and prognosis of malignant ovarian germ cell tumors(MOGCT)with lung metastasis.Methods Fifteen patients of MOGCT with lung metastasis treated in Peking Union Medical College Hospital from Jan.1982 to Dec.2010 was retrospectively analyzed.Results(1)Clinical characteristics: the average onset age of these 15 patients is(23 ± 11)years old(6-48 years).The majority of these patients presented with abdominal pain(8/15)or irregular vaginal bleeding(4/15)as their initial symptoms.The primary tumor located in the left ovary in 8 cases,right ovary in 6 cases,and both sides in only 1 case.Metastatic lesions were confined to the lung in 12 patients,while the other 3 patients were found to have multi-site distant metastasis.(2)Diagnosis:all 15 cases included 9 pure non-gestational ovarian choriocarcinoma(NGOC),3 MOGCT containing choriocarcinoma component(one mature teratoma with choriocarcinoma component,one endodermal sinus tumor with embryonal carcinoma and choriocarcinoma components,one choriocarcinoma with dysgerminoma component),2 embryonal carcinoma,one immature teratoma.Only one patient in these 15 cases was correctly diagnosed before surgery.(3)Time of lung metastasis: of 12 MOGCT with choriocarcinoma component,11 patients were found to have lung metastasis in the course of their primary treatment,only 1 had lung metastasis 2 months after the synthetic treatment finished.Three MOGCT patients without choriocarcinoma component were all found to have lung metastasis when tumor relapsed in the advanced stages of the disease.(4)Treatment:all 15 patients received multi-agent chemotherapy combined with surgery.The mean courses of chemotherapy for these patients were 16 courses(5-43 courses).(5)Prognosis:of these 15 cases,complete remission was obtained in 10 patients of NGOC or mixed ovarian germ cell tumors with choriocarcinoma component,3 patients(one NGOC,one endodermal carcinoma and one immature teratoma,respectively)died in the course of treatment as result of tumor progression,2 progressed cases(one NGOC and one endodermal carcinoma respectively)abandoning therapy were lost to follow up.Conclusions MOGCT with lung metastasis are more often to found in NGOC patients.These patients could obtain high complete remission rate after standard multi-agent chemotherapy combined with surgery.The prognosis of MOGCT with lung metastasis containing choriocarcinoma component are better than that of those without containing choriocarcinoma component.
