1.Effects of FABP-5 gene silencing on the biological characteristics of the cervical carcinoma cell line SiHa

BACKGROUND:Cancer related genes and pathways play a critical role in formation and development of cancer.
OBJECTIVE: To investigate the silencing epidermal fatty acid binding protein 5 (FABP-5) by siRNA interference on biological characteristics of the cervical carcinoma cel line SiHa.
METHODS:Design and synthesis of siRNA interference sequence used to transiently transfect SiHa cels was performed according to FABP-5 mRNA coding sequence. mRNA and protein expressions of FABP-5 were detected by RT-PCR and western blot assay, respectively. Meanwhile, cel cycle, proliferation, invasion and apoptosis were detected by flow cytometry, cel counting bit-8 assay, Boyden chamber assay, and TUNEL staining, respectively.
RESULTS AND CONCLUSION: FABP-5 mRNA and protein expressions, cel growth speed, cel number at S phase (cel cycle was arrested at the G0/G1 phase) and cel invasion were significantly decreased, while the cel apoptosis was significantly increased in FABP-5 siRNA group compared with the negative control and blank control groups. Our findings indicate that the specific silencing FABP-5 gene expression by siRNA interference can inhibit SiHa cervical cel growth and proliferation, but accelerate cel apoptosis. 2.Effects of combined treatment using umbilical cord blood mesenchymal stem cell transplantation and paclitaxel on proliferation, apoptosis, and invasion of A2780 ovarian cancer cells

BACKGROUND:An intensivestudy on ovarian cancer A2780 cels contributes to modify treatment strategies targeting ovarian cancer stem cels and enhance survival rate of ovarian cancer patients.
OBJECTIVE:To investigate the effects of umbilical cord blood mesenchymal stem cels (UC-MSCs) transplantation combined with paclitaxel on proliferation, apoptosis, and invasion of A2780 human ovarian cancer celsin vitro.
METHODS:A2780 human ovarian cancer cels were assigned into three groups: blank control group (routine cel culture),UC-MSCs group (addition of suspension containing 1×106UC-MSCs) and combined treatment group (combined addition of suspension containing 1×106UC-MSCs and 250μL of 1μmol/L paclitaxel solution). CD133+antigen expression, proliferation, apoptosis, and invasion were determined by immunofluorescent staining, flow cytometry, TUNEL staining, and Transwel chamber invasion assay, respectively.
RESULTS AND CONCLUSON:CD133+antigen expression intheUC-MSCs group and combined treatment group was significantly decreased compared withtheblank control group (P< 0.05). Proliferation and invasion abilities were significantly decreased, while apoptosis cel number was significantly increased inthecombined treatment group compared with the other two groups (P< 0.05). These findings indicate that the combined treatment of UC-MSCs transplantation and paclitaxel can inhibit proliferation and invasion and promote apoptosisof A2780 human ovarian cancer cels.
3.Immunohistochemical Studying on Subventricular Zone in Lateral Wall of Lateral Ventricle of Adult Rat

Objective The cells of subventricular zone in lateral wall of lateral ventricle of adult rats were localized. Method Immunohistochemistry. Results Polysialylated neuronal cell adhesion molecule and phosphotyrosine immunoreactive cells were found in subventricular zone, and they have same distributions, however, no any parvalbulin immunoreactive cells were found in subventricular zone. Conclusion neuronal precursor cells in subventricular and expression of phosphotyrosine are associated with proliferation of neuronal precursor cells in subventricular zone.
4.In-vitro Sensitivity of Plasmodium falciparum to the Components of Dihydroartemisinin Compounds in Dongfang City of Hainan Province

[Objective] To examine the sensitivity of P. falciparum to the components of dihydroartemisinin (DHA) and piperaquine (PQ) in Dihydroartemisinin Compounds. [Methods] WHO (World Health Organization) standard micro test and in-vitro test were used. [Results] Survey in the year of 2001 for 61 cases and in 2004 for 28 cases showed that the DHA-resistant rate of P. falciparum in all cases was zero with inhibitory concentration of 50% (IC50) being 6.84 nmol/L and 5.67 nmol/L, and the mean concentration for complete inhibition of schizont formation was 35 nmol/L and 39 nmol/L, respectively. Survey in the year of 2001 for 75 cases and in 2004 for 29 cases showed that PQ-resistant rate of P. falciparum was 22.67% and 17.24% with IC50 being 274 nmol/L and 317 nmol/L, and the mean concentration for complete inhibition of schizont formation was 1 220 nmol/L and 1 269 nmol/L, respectively. [Conclusion] No DHA -resistance exists and PQ-resistance decreases for the treatment of P. falciparum in Dongfang city of Hainan province.
5.Effects of Kunmu decoction on proliferation and apoptosis of fibroblast-like synoviocytes in rheumatoid arthritis

Objective To investigate the effects of alcohol extract of Kunmu decoction on proliferation and apoptosis of rheumatoid arthritis fibroblast-like synoviocytes (RA-FLS). Methods Synovial tissues were obtained from patients with active RA received joint replacement or arthroscopy. The surface antigen and the amount of apoptotic cells were determined by flow cytometry. The inhibitive effect was detected by MTT assay. Results The CD90+surface antigen of synoviocytes was (94.78 ± 0.98)%. The inhibitive effect on the proliferation in all treatment groups were in a time-and dose-dependent manner. The apoptosis rate was increased in a dose-dependent manner among all dosage alcohol extract groups. Conclusion Kunmu decoction might inhibit proliferation and induce apoptosis of RA-FLS.
6.Integrated fetal diagnosis and postnatal treatment for the transposition of the great arteries

Objective To summarize primary experiences of integrated fetal diagnosis and postnatal treatment for the transposition of the great arteries (TGA). Methods Five fetus were diagnozed as TGA at(28.4 ±4.4) weeks of gestation via fetal echocardiography. The mean age of the pregnant women was (28.4 ±3.0) years old. Delivers were taken by caesarean at (36.5 ±1.8) weeks of gestation. The body weight of neonates was (2468 ±442) grams. All neonates were transported to the department of neonatology and re-checked by echocardiography. 3 cases were TGA with intact ventricular septum, 2 cases were TGA with ventricular septal defect. Prostaglandin and mechanical ventilation were applied if oxygen saturation was lower. The standard arterial switch procedure was performed under cardiopulmonary bypass with moderate hypothermia. Results The mean age for patients at surgery was (9. 0 ±6. 2) days ( ranged 2-19 days). Three neonates survived, 2 preterm neonates died. One with body weight 1770 g, 2 days after operation was died of sudden heart arrest and failure of resuscitation. Another was treated by mechanical ventilation and prostaglandin after delivery and underwent operation at the second postnatal day, the neonate appeared low cardiac output and high serum lactate postoperatively and died in the third day. Conclusion The integrated fetal diagnosis and postnatal treatment for TGA could prevent severe cyanosis and emergency transportation after parturition. The lack of any link in the cooperation among the multidisciplinary team could affect the benefits for the TGA neonates provided by prenatal diagnosis.
7.Effects of delivery classification scale for fetal cardiac disease on the prenatal and postnatal integrated treatment strategies

Objective To explore the effects of delivery classification scale for fetal cardiac disease on the prenatal and postnatal integrated treatment strategies.Methods Delivery classification scale for fetal cardiac disease included:grade Ⅰ,no hemodynamics instability; grade Ⅱ,ducted-dependent lesions,stable hemodynamics anticipated;grade Ⅲ,possibility or likelihood of hemodynamic instability; IMPACT(delivery immediately heart intervention) level,hemodynamic instability is anticipated at separation from placental circulation.During August 2006 to May 2010,a retrospective study of 46 cases of prenatal diagnosis of congenital heart disease and delivery in one cardiac center was taken,in which 33 in grade Ⅰ,9 in grade Ⅱ,4 in grade Ⅲ,and no IMPACT.Results Thirty-nine boys and 7 girls were born at (38.0 ± 1.4) weeks of gestation and had consistent fetal diagnoses of mainly cardiac abnormalities with postnatal screen.Thirteen neonates underwent cardiac intervention within one week after birth with one death,including 2 in grade Ⅰ,7 in grade Ⅱ,4 in grade Ⅲ,of them 1 death.Seven infants including 5 in grade Ⅰ and 2 in grade Ⅱ underwent cardiac intervention with one death.The remaining 26 children in grade Ⅰ had uneventfully outcomes,in which 7 cases of surgical operation,17 cases of interventional therapy,2 cases of spontaneous healing.Conclusion Delivery classification scale for fetal cardiac disease should have some guiding significance for early treatment strategies and could enhance closely integration of prenatal diagnosis and postnatal treatment.The most fetuses in grade Ⅰ need not undergo cardiac interventions in neonatal stage.However,early cardiac intervention for fetuses in grade Ⅱ and Ⅲ should be carried out postnatally with the help of neonatologists.
8.Effects of Antibiotics on Children with Acute Otitis Media

Objective To study whether the usage of antibiotics and white -blood -cell counts affect the effects of 2~12-year-old children with acute otitis media (AOM ) .Methods A total of 126 children (2~12 years old)with AOM ,from Jan 2013 to Dec 2013 ,were retrospectively analyzed .Usage of antibiotics and white -blood-cell count were recorded at first ,and the relationships between two factors and outcomes of children with AOM were studyed in one week ,one month and three months after treatment ,respectively .Results White blood cell counts were higher than normal in 59 .52% (75/126) 2~12 -year -old children with AOM ,and antibiotics were used in 73 .81% (93 /126) .Ninty three cases were followed -up until 3 month or more .Antibiotics were used in 74 cases ,and watchful waiting were used in the other 19 patients .The effective rates of two groups were 59 .45% and 52 .63% ,75 .68% and 84 .21% ,85 .14% and 78 .95% in one week ,one month and 3 months after treatment ,re‐spectively .There were no statistically significant differences(χ2 values were 0 .295 ,0 .903 ,0 .799 ,P>0 .05) .But the effective rates were signifantly higher in 1 and 3 months than that of in one week (P<0 .05) .There was no sig‐nificant difference (P>0 .05) between one and three months .Whether white-blood-cell count higher than normal or not ,the difference in efficacy was not statistically significant in 1 week ,1 month and 3 months after treatment (P >0 .05) .Conclusion In 2~12-year-old children with acute otitis media (AOM ) ,the final outcomes were inde‐pendent of the usage of antibiotics and white -blood-cell count .
9.Clinical Effect of Modified Qiwei Baizhu Powder for Mesenteric Lymphadenitis in Children with Spleen Deficiency and Dampness Retention

Objective To observe the clinical curative effect of modified Qiwei Baizhu Powder for mesenteric lymphadenitis in children with spleen deficiency and dampness retention. Methods A multi-center randomized and controlled trial was carried out in 150 cases of mesenteric lymphadenitis in children with spleen deficiency and dampness retention. The patients were randomized into Chinese medicine group, western medicine group and combination group, 50 cases in each group. Chinese medicine group was treated with modified Qiwei Baizhu Powder, western medicine group was treated with Cefaclor for Oral Suspension, and the combination group was treated with modified Qiwei Baizhu Powder plus Cefaclor for Oral Suspension. Before treatment and after treatment for 7 days, one month and 3 months, changes in syndrome manifestations and the size of abdominal mesenteric lymph nodes were monitored. The total effective rate and cure rate of the three groups were compared, and the safety of the regimen was also evaluated. Results (1) In Chinese medicine group, 5 patients dropped out, and a total of 45 cases completed the trial; in western medicine group, 3 patients dropped out, and a total of 47 cases completed the trial; in the combination group, 7 patients dropped out, and a total of 43 casescompleted the trial.(2) The total effective rate of Chinese medicine group and combination group after treatment for 7 days, one month and 3 months was significantly higher than that of western medicine group (P < 0.05), and the cure rate after treatment for one month and 3 months was also significantly higher than that of western medicine group (P<0.01). The differences of total effective rate and cure rate were insignificant between Chinese medicine group and combination group(P>0.05) at various time points.(3) After treatment for 7 days, one month and 3 months, the transverse and longitudinal diameters of mesenteric lymph nodes in Chinese medicine group and combination group were shorter than those of western medicine group (P < 0.05) , but the difference was insignificant between Chinese medicine group and combination group (P>0.05). (4) Except for the drop-out cases, all of the children finished the trial. During the treatment, no obvious adverse reaction was found. Conclusion Modified Qiwei Baizhu Powder exerts certain efficacy for the treatment of mesenteric lymphadenitis in children with spleen deficiency and dampness retention.
10.Pre-chemotherapy nursing assessment among adult cancer patients: a best practice implantation project

Objective:To promote evidence-based practice in the pre-chemotherapy nursing assessment among adult cancer patients.Methods:The Joanna Briggs Institute Practical Application of Clinical Evidence System and Getting Research into Practice audit tools were used. The project was conducted in Shanghai Cancer Center of Fudan University from May to October 2018, 12 audit criteria were developed in the program including nursing training, patient medical and allergic history, medical diagnosis, lab data and so on.Results:A baseline audit of pre-chemotherapy nursing assessment among adult cancer patients was conducted, with a sample size of 68 patients and 36 nursing staff, during this stage, the compliance of audit 11 and 12 were 100%. After the implementation of systematic strategies, a follow-up audit involving similar sample as first audit was conducted using the same audit criteria. In the follow-up audit, except criterion 4 and 10, the compliance of the remaining 8 criteria had significantly improved, and χ2 value was 10.29-132.06, P<0.01. The result of history adverse reaction in the follow-up audit showed that among 68 patients, 3 had experienced chemotherapy infusion reactions in the past (The drugs were oxaliplatin, gemcitabine and paclitaxel), 39 had chemotherapy-related symptoms before admission (most of them were relieved at admission), of which the top five were loss of appetite, fatigue, nausea, neurotoxicity and vomiting. Conclusions:The aims of the project were fulfilled. We achieved increased compliance with evidence-based best practice recommended by JBI in most of audit criteria. Further audit will need to be carried out to improve the validity and quality of nursing assessment.
