1.Suppression Effect of Di-n-butyl Phthalate in vitro on Superoxide Dismutase of Various Organs of Mice

Objective The present study was conducted to study the oxidation damage effects of DBP on organs of mice and the molecular mechanisms of the effects. Methods The cells of the organs (liver,kidney,testis,lung) were exposed to different concentration of DBP (0?mol/L,5?mol/L,20?mol/L,80?mol/L) for one hour in vitro,and then the superoxide dismutase (SOD) activities of those cells of different organs were measured. Results With statistical significance,the SOD activities in liver,kidney,testis,lung cells were increased gradually with the increase of DBP concentrations. Conclusion The results suggested that DBP could induce the oxidative damage in liver,lung,kidney and testis cells of mice.
2.Preparation optimization of palonosetron hydrochloride oral disintegrating tablets by orthogonal test

Objective To prepare optimization of palonosetron hydrochloride oral disintegrating tablets by orthogonal test. Methods Palonosetron hydrochloride oral disintegrating tablets were prepared with direct compression process.The content of pal-onosetron hydrochloride was determined by HPLC.The formulation was optimized with disintegration time as evaluation indices. Results The optimal formulation(60 mg/tablet)was as follows:L-HPC 12%,mannitol∶SMCC= 2∶1,magnesium stearate 2%, stevia glycosides 3%.The oral disintegrating tablets showed dine appearance and tested better;the disintegration time was 12 sec-onds;the tablets featured a hardness of 3 kg;4 min dissolution rate was 99%.Conclusion The preparation method is simple and reasonable,and the tablets can disintegrate rapidly.
3.Analysis on reference factors for the identification of students from poor families in the university

Objective To know the problems of reference factors,and then provide evidences for scientific and specific identifying index system for students from poor families.Methods Questionnaires were used among 719 students from poor families.Results There were 86.0% families whose earnings were lower than 410 RMB per month.57.2% families depended on agriculture.70.1% students were from the rural areas.Family member number was 4~6 for the majority ( 70.7% ).91.2% ( 94.5% ) fathers'( mothers' ) education levels were under high middle school.64.0% ( 67.3% )fathers ( mothers ) were farmers.The proportion of students having computers was 76.1%.61.2% students' life was not frugal.There were 80.9% students who thought campus cards shouldn't be monitored.Conclusion Family economic situation should be the key indicator.Most of the basic family information could be reference factors.Basic situation of students at school was inappropriate to be reference factors.
4.Analysis on urban-rural differences of reference factors for the identification of students from poor families in university

ObjectiveTo know urban-rural differences of reference factors for the identification of students from poor families in order to provide references for identifying index system.Methods Questionnaires were used among 708 students from poor families.ResultsThere were 78.9% urban and 89.1% rural families whose average earnings were lower than 410 RMB per month.The number of majority urban and rural family (58.8%,75.4% ) was 4 - 6 ; 77.0% ( 86.3% ) urban and 97.3% (97.9%) rural fathers' ( mothers' ) education levels were high school or below; 8.6% ( 23.1% ) urban and 82.3 % ( 85.1% ) rural fathers ( mothers ) were farmers.The proportion of students having computers was 87.2% (urban) and 7 1.9 % (rural) ; 54.9% urban and 63.9% rural students' life was not frugal; 85.2% urban and 79.1% rural students thought campus cards shouldn't be monitored.ConclusionsThere are significant urban-rural differences of reference factors for the idenffication and family residence is the key indicator which should have higher weights.
5.Degradation of earthworm extracts prepared by wet superfine grinding in simulated gastrointestinal environment.

This is to report the study of degradation of earthworm extracts prepared by wet superfine grinding in simulated gastrointestinal environment. Enzymatic reactions were terminated by adjusting the solution pH or using membrane bioreactor principle. Earthworm protein concentration change was detected by Bradford method, the degraded state of protein was described with SDS-PAGE technology, and the degraded state of small molecule substances was detected by HPLC. The results showed that earthworm protein degraded completely in artificial gastric juice. High molecular weight protein degraded greatly in artificial intestinal fluid, while low molecular weight protein was not significantly degraded. Small molecular substances degradation did not degrade in artificial gastric juice, while they degraded obviously in artificial intestinal fluid, there is even new small molecule substance appeared. Finally it is concluded that the substance that having therapeutic effects in vivo may be some degraded peptide, amino acid and stable small molecules existed in artificial intestinal fluid.
6.Analysis of MYOC gene mutation in a primary open angle glaucoma family from China

Background Primary open angle glaucoma (POAG) is one of the frequent glaucomatous types,and genetic factor participates in pathogenesis and development of the disease.Recently,MYOC mutation was found to be associated with POAG.Objective This study was to describe the clinical and genetic findings in a POAG family from Luoyang,China.Methods This study protocol was approved by Ethic Committee of Affiliated First Hospital of Henan University of Science and Technology.The study adhered to Declaration of Helsinki.A POAG family with 29 members of 5 generations was surveyed and followed-up for 5-year duration.The mode of inheritance was determined by the pedigree analysis.The periphery blood sample was collected form 12 families and 100 health controls for the extraction of genomic DNA under the informed consent.The third exon and its flanking introns of MYOC were amplified,and quantitative real time PCR products were sequenced,and the structure and function of mutated gene were examined by restriction fragment length polymorphism analysis.The predicted effects of the detected variants on the secondary structure of MYOC protein were evaluated using Garnier-Osguthorpe-Robson (GOR) method,and homology analysis of protein was carried out by Blast software provided by National Center for Biotechnology Information (NCBI).Results This POAG family included 29 members of 5 generations,and the clinical data were not clear in 11 family members.Three individuals from 3 generations were determined POAG,another one was ocular hypertension,and 2 were carriers.Pedigree analysis appeared an autosomal dominant inheritance.In 12 subjects included 6 members genetically affected and 6 members with normal phenotype,the heterozygous mutation was found in the third exon of MYOC gene in 6 genetically affected members,which revealed a T→C transition at position 1021 (p.S341P),resulting in a switch of serine (Ser) to proline (Pro).It was a missense mutation abolished a CviKI-1 restriction site that segregated with the affected members.Secondary structure prediction of p.S341P suggested that myocilin protein was misfolded.Analysis of protein homology and switched Ser was conservative amine acid at position 1021 (p.S341P).No similar change was found in the 6 normal families and the normal controls.Conclusions Ser341Pro MYOC mutation is disease-causing factor in the POAG family of Luoyang.The clinical and genetic features of this mutation warrant further investigation.The mutation spectrum of MYOC is expanded to offer a better diagnosis and treatment for POAG patients.
7.Botulinum Toxin A for Post-stroke Spasm under Color Doppler Ultrasound

Objective To evaluate outcome of injection of botulinum toxin A to treat post-stroke muscle spasm under color doppler ultrasound guide. Methods 15 stroke patients following spasm in upper limb were chosen. Under the guidance of ultrasound, botulinum toxin A was accurately injected in biceps, brachialis, deep flexor muscle of fingers, superficial flexor muscle of fingers, musculus flexor carpi ulnarista, thumb-muscle, palmaris longus muscle, musculus pronator teresta, and quadrate pronator muscle. All patients went on rehabilitation training after injection. Upper limb function was evaluated by using modified Ashworth(MAS) and Fugl-Meyer scores before injenction, two weeks and four weeks after injection. Results Recovery of muscle spasm and scores of the upper limb motor function were better than that before injection.Conclusion Under color doppler ultrasound guidance, injectionof the botulinum toxin A can treat post-stroke muscle spasm with accurate location.
8.Drug resistance mediated by outer membrane porin F (OmpF) and its regulatory gene of micF in Shigella strains

Objective To investigate the mechanism of drug resistance mediated by micF gene and outer membrane porin F ( OmpF) in Shigella strains. Methods Shigella strains were isolated from stool samples of patients who presented to the Second Hospital of Tianjin Medical University with acute diar-rhea in 2015. Antibiotic susceptibility test was performed to screen out the multidrug-resistant and non-multi-drug-resistant strains. The ompF gene was amplified by PCR. The micF and ompF genes at transcriptional levels in the two groups of strains were detected by quantitative real-time RT-PCR. Intracellular concentra-tions of ciprofloxacin in the two groups of Shigella strains were measured by automatic microplate reader. Re-sults According to the result of antibiotic susceptibility test, 13 strains that were resistant to 3 or more than 3 antibiotics were classified into the multidrug-resistant group, while the other 8 strains that were sensitive to all antibiotics used in this study or only resistant to 1 or 2 antibiotic were classified into the non-multidrug-re-sistant group. All of the 21 Shigella strains carried the ompF gene. Compared with the non-multidrug-resist-ant strains, the multidrug-resistant strains showed higher expression of micF gene, but lower expression of ompF gene. The differences in micF and ompF genes between the two groups were statistically significant. The result of correlation analysis suggested that there was a negative correlation between micF and ompF genes (r=-0. 244). The intracellular concentrations of ciprofloxacin in multidrug-resistant strains were low-er than those in the non-multidrug-resistant strains (P<0. 001). Conclusion The decreased expression of OmpF was one of the possible mechanisms of multidrug-resistance in Shigella strains. The micF gene was negatively related to the expression of OmpF. Moreover, the decreased intracellular concentrations of cipro-floxacin in multidrug-resistant strains might be related to the decreased expression of OmpF.
9.Clinical study on Honggui capsule combined with chemotherapy for lung cancer

Objective To observe the clinical efficacy of Honggui capsule combined with chemotherapy in the treatment of lung cancer. Methods 61 cases of lung cancer patients were randomly divided into group A of 30 cases, treated with Honggui Capsule and chemotherapy, and group B of 31 cases treated with chemotherapy only by the envelope method. After both groups were treated for 21 days, the clinical effect, life quality improvement, improvement of TCM symptoms and the incidence of toxic and side effects in 2 groups were compared. Results After treatment, there was significant difference between group A and B in the tumor response rate (26.7% vs. 3.2%; χ2=4.927, P=0.026), Karnofsky (23.3% vs. 3.3%; χ2=5.410, P=0.020), and improvement of TCM symptoms (13.3% vs. 0.0%; χ2=4.423, P=0.035). During treatment, there was significant difference between group A and B in bone marrow suppression (33.3% vs. 58.1%; χ2=4.778, P=0.029), and gastrointestinal disturbance (16.7% vs. 58.1%; χ2=11.124, P=0.001). Conclusion Honggui capsule combined with chemotherapy has an enhanced efficacy and a detoxicating effect, besides it improves clinical symptoms and reduces the side effects of chemotherapy.
10.Effect of Low Frequency Ultrasound on Carotid Plaque

Objective To observe the effects of low frequency ultrasound on carotid artery plaque and artery stenosis. Methods 156 patients with carotid atherosclerosis were divided into treatment group (n=80) and control group (n=76). The control group was administered routine medicine, while the treatment group accepted low frequency ultrasound therapy in addition. The size and shape of carotid artery plaque, severity of stenosis and the level of lipid were observed before and after treatment, and the side-effects were recorded. Results The intima-media thickness (IMT), diameter of plaque, plaque score decreased after treatment in both groups, and decreased more in the treatment group than in the control group (P<0.05); while the frequence of moderate stenosis and severe stenosis was less (P<0.05). The levels of low density lipoprotein- cholesterol and total cholesterol decreased in both groups after treatment (P<0.05), and decreased more in the treatment group than in the control group (P<0.05). No serious side-effect was observed. Conclusion Low frequency ultrasound can reduce the atherosclerotic plaques in carotid artery and relieve the stenosis.
