Objective:To explore the expression and role of AQP9 in the models of cultured steatosis hepatocytes.Methods:Steatosis models of hepatocytes was established by adding oleic acid to the growing L-02 cell strain.The expression of AQP9 mRNA was measured with RT–PCR and the protein expression of AQP9 were measured by Western blotting.Lipid droplets in the hepatocytes were observed by oil red staining and the contents of triglyceride,FFA and glycerol in hepatocytes were measured with analyzed kit.Results:Through oil red staining.a few lipid droplets were observed at 24h and steatosis hepatocyte increased greatly at 72h.Triglyceride,FFA and glycerol contents in hepatocytes of model groups also increased,as compared with the control group(P

4.Effects of Qigong Pills Decoction Combined with Chemical Drugs on Serum Hormone Levels of Polycystic Ovarian Syndrome Patients Complicated with Infertility

OBJECTIVE:To investigate the effects of Qigong pills decoction combined with chemical drugs on hormone levels of polycystic ovarian syndrome(PCOS)patients complicated with infertility. METHODS:A total of 98 PCOS patients complicated with infertility selected from gynaecology department of our hospital during Jul. 2015-Jun. 2016 were divided into observation group and control group according to random number table,with 49 cases in each group. Control group was given Clomiphene ci-trate tablet 50 mg orally,qd,for consecutive 5 d;B-ultrasonography for follicle development was monitored at the 11th day of menstruation,and then they were given Chorionic Gonadotropin for injection 2000 U intramuscularly,qod,if there was not domi-nant follicle >10 mm till the diameter of follicle reached 18 mm. Observation group was additionally given one dose of Qigong pills decoction,every day 400 mL after decocted with water,each 200 mL for morning and night,on the basis of control group. After 3 months of treatment,the levels of FSH,LH,PRL,E2,T,DHEA-S and SHBG,the endometrial thickness,the number of mature follicle and pregnancy rate were compared between 2 groups. RESULTS:Before treatment,there was no statistical signifi-cance in the levels of FSH,LH,PRL,E2,T,DHEA-S or SHBG between 2 groups(P>0.05). 3 months after treatment,the lev-els of PRL,T and DHEA-S in 2 groups were decreased and E2,SHBG were increased significantly;the levels of FSH,PRL,T and DHEA-S in observation group were significantly lower than control group,while the levels of E2 and SHBG were significantly higher than control group,with statistical significance (P<0.05). Endometrial thickness of observation group were significantly higher than that of control group;pregnancy rate (18.37%) was significantly higher than control group (4.08%),with statistical significance (P<0.05). CONCLUSIONS:Qigong pills decoction combined with chemical drugs can help to improve pregnancy rate and regulate serum hormone level in the treatment of PCOS complicated with infertility.
5.Protective Effect of Curcumae on Acute Hepatic Injuried Mice Induced with Carbon Tetrachloride

Objective To explore the effect of Curcumae on acute hepatic injuried mice.Methods The model mice of acute hepatic injury were established with intraperitoneal injecting 0.2% carbon tetrachloride (CCl4) 0.2 ml/10mg. 48 mice were randomized into 6 groups: model group, control group, Ganlixin group (25 mg/kg) and three treating groups of Curcumae which were administrated by gavage at the doses of 2.5 g/kg, 5 g/kg, 10 g/kg once everyday.7 d later, all mice were put to death. The liver index, the activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST) in serum, the activities of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) in hepatic tissue were observed. Pathological change was observed under HE staining. Results Compared with the model group, Curcumae could decrease the liver index and the activities of ALT and AST in serum, MDA in hepatic tissue significantly (P<0.05), and increase the activities of SOD (P<0.05). The pathological changes were less in three treating groups than that of the models. Conclusion Curcumae can protect the liver from acute hepatic injuried induced with CCl4 in mice.
6.Correlation study between resistin and the insulin resistance and the urinary albumin excretion rate

ObjectiveTo investigate the correlation between the level of resistin and the insulin resistance(IR) and the incorporation of microamount albuminuria(MA) of type 2 diabetes mellitus (T2DM).MethodsTwo hundred and twenty T2DM patients and 40 normal subjects (control group) were enrolled in this study.Two hundred and twenty T2DM patients were divided into 4 groups,IR group (group A,79 cases),non-IR group (group B,48 cases),IR complicating MA group (group C,51 cases),non-IR complicating MA group(group D,42 cases).The fasting serum resistin was measured by enzyme-linked immunosorbant assay (ELISA),and the fasting plasma glucose (FPG),glycosylated hemoglobin (HbA1c),fasting insulin (FINS) was also determined.ResultsThe level of fasting serum resistin in group A,B,C and D was higher than that in control group [(33.45 ± 1.37),(23.36 ± 1.47),(44.45 ± 1.39),(37.45 ±1.57) μ g/L vs.( 17.44 ± 1.26 ) μ g/L],and there was significant difference among 5 groups (P < 0.01 ).The level of fasting serum resistin in group A was higher than that in group B (P < 0.01 ),and the level of fasting serum resistin in group C was higher than that in group D (P < 0.01 ).The level of fasting serum resistin was both higher in group C and D than that in goup A and B (P< 0.01 ).Correlation analysis showed the fasting serum resistin was positively correlated with body mass index(BMI),FPG,HbA1c and FINS in group A(r =0.35,0.46,0.37,0.49,P <0.05),and the fasting serum resistin was positively correlated with HbA1c,BMI,systolic blood pressure and diastolic blood pressure in group C(r =0.45,0.32,0.37,0.29,P < 0.05 ).ConclusionsSerum resistin may participate in the process of IR and the formation of MA.It may become one of the diagnostic standard of the IR and one of the important index to estimate the MA.
7.Effects of magnesium isoglycyrrhizinate on liver function and liver regeneration in cirrhotic rats after partial hepatectomy

Objective To investigate the effects of magnesium isoglycyrrhizinate on liver regeneration and hepatic function following partial hepatectomy in thioacetamide induced cirrhotic rats.MethodsForty-five cirrhotic Wistar rats undergoing 2/3 hepatectomy were randomly assigned to control group ( Group A), Group B and Group C starting the day of hepatectomy, rats in Group B were injected 60mg per kg body-weight magnesium isoglycyrrhizina daily intraperitoneally until the day of sacrifice. Rats in Group A recevied same dose of sodium chloride. In Group C, magnesium isoglycyrrhizina was administered daily 3 days before hepatectomy until the rats were sacrificed. Liver function, serum HGF, serum PLA2,BrdU labelling index and percentage of intial liver weight on days 1, 2 and 7 post hepatectomy were assessed. ResultsRats in Group A had significantly lower BrdU labelling index and serum HGF level than Group C ( t = 2. 831, P < 0.05; t = 3.427, P < 0.05 ) and a markedly higher level of serum PLA2 than the other groups on day 1 posthepatectomy ( Group B t = 2. 794, P < 0.05; Group C t = 2. 902, P < 0.05 ).Rats in Group A had a lower BrdU labelling index and a more increased level of serum PLA2 than Group B and Group C on day 2 posthepatectomy ( BrdU labelling index: Group B t = 2. 736, P < 0.05; Group C t =3.083, P<0.05; PLA2: Group B t =2.794, P<0.05; Group C t =2.902, P<0.05), but had no significant difference in HGF level with the other two groups. The three groups were similar in ALT, AST,TP and intial liver weight on days 1,2 after operation. On the 7th day posthepatectomy, rats in Group A had a higher level of AST ( Group B t = 4. 508, P < 0.05; Group C t = 2. 967, P < 0.05 ) and a lower level of TP ( Group B t = 2. 838, P < 0.05; Group C t = 2. 743, P < 0.05 ), lower liver weight than the other two groups ( Group B t = 3.316, P < 0.05; Group C t = 4. 093, P < 0.05) but there was no difference between the three groups in BrdU labelling index, HGF and PLA2 level. Rats in Group C had significantly higher BrdU labelling index and serum HGF level than the rats in Group B on day 1 after hepatectomy( t = 2. 831, P <0.05; t = 2. 836, P < 0.05 ).ConclusionsMagnesium isoglycyrrhizinate inhibits aminopherase release and enhancing liver regeneration in cirrhotic rats after partial hepatectomy.

9.Vacuole formation and endothelial damage in microvessels after ischemia reperfusion

AIM:Damaging of Endothelial cells and adhesion of leukocytes and platelets were studied in rat mesentery microvessels after ischemia-reperfusion.METHODS:The model was made by losing blood and reperfusion from carotid artery in rat. Changes in mesentery microvessels were observed by high magnify microscope.RESULTS:Leukocyte and platelet adhesions were found in venules and co-capillaries 1-3 hours after ischemia-reperfusion. Endothelial cells were edema and vascular walls were thickening. Vacuoles formed in intracytoplasm of some vascular endothelium and some bigger endothelial vacuoles prominenced toward the luminal surface. Vacuoles had the shape of circle and the diameter was 10-30 μm. More vacuoles were found in arterioles, even there were several vacuoles in a arteriole. The biggest vacuole almost occupied 2/3 of vascular lumen.CONCLUSION:Edema and vacuole formation in vascular endothelium showed endothelial cells were damaged seriously after ischemia-reperfusion.
10.Effects of salusins on in vivo mesenteric microcirculation in rats

AIM: To investigate the regulatory effects of salusins on tensility of microvessle in rats. METHODS: Wistar rats were randomly distributed into six experimental groups as follows: (1) normal saline injection-treated control; (2) salusin ?; (3) salusin ?; (4) noradrenaline; (5) noradrenaline+salusin ?; and (6) noradrenaline+Salusin ?. For recording the microcirculation images in the mesentery, the intestinal loop was mounted on the stage of an intravital microscope equipped with a TV camera. Temporal changes in internal diameter of arterioles and venules were measured by computer using the Image-Pro software. RESULTS: Salusin ? (10 -5mol/L) dilated arterioles directly and the dilation up to the peak at 3 min [(25.56?4.30) vs (24.49?4.27)?m, P0.05). Salusin ? has no effects on internal diameter directly and NA-induced contraction of microvessles. CONCLUSION: Salusin ? dilated mesenteric arterioles and inhibited NA-induced contraction of microvessles in rats.
