1.Psychological analysis and care for patients with percutaneous coronary intervention surgery

ObjectiveTo investigate the psychological state and care measures for patients undergoing pereutaneous coronary intervention (PCI). Methods300 patients treated with PCI from December 2009 to June 2011 in our hospital were chosen as the research object.The factors arousing psychological disorders for PCI patients were taken for comparison.And then the appropriate nursing measures were discussed. ResultsThe most evident negative emotion of PCI patients was depression.The factors arousing negative emotion included the factors of family members and undesirable environment by many people going in and out. ConclusionsEffective measures can avoid the psychological barriers of PCI patients and improve the quality of postoperative recovery for patients.
2.THE CHINESE DIET BALANCE INDEX REVISED

To revise Chinese Diet Balance Index (DBI) based on the current Chinese Dietary Guidelines and Food Guide Pagoda . Method: Seven food components [(cereals ,vegetables and fruits, bean and dairy , animal food , alcohol ,condiments (salt and edible oil))were selected in combination with food varieties and drinking water to build up Diet Balance Index Revision (DBI-07) .The components were weighed and assigned cut-offs and scores based on the Chinese Food Guide Pagoda. Three scoring methods (LBS-low bound score, HBS-high bound score, DQD-diet quality distance) were used to evaluate the dietary quality of 26300 adults aged 18 to 79 years from 2002 China National Nutrition and Health Survey. Results: The mean LBS, HBS and DQD were 11.5,30.0 and 41.5 respectively. About 78% adult were in moderate or severe deficit of food intake, 35.1% in moderate or severe surplus of food intake, and 43.8% in severe dietary imbalance. Conclusion: DBI-07 can reflect the principles of dietary guidelines, and the scoring method can reflect both deficit and surplus of food intake. DBI-07 can be used to evaluate the dietary quality of individual or the population.
3.A pilot study on phosphorylation of extracellular signal-regulated kinase 1/2-mediated diabetic retinopathy

Background It is well known that diabetic retinopathy(DR) involves in complex pathogenesis.Worse outcome of brain ischemia with hyperglycemia is mediated by mitogen-activated protein kinases (MAPK).However,the effect of phosphorylation of MAPK signal pathway on DR needs further investigation.Objective This study was to explore the the possible role of extracellular signal-regulated kinase1/2(ERK1/2)in DR.Methods Sixty SPF SD rats aged 8-weeks were grouped into the control group and diabetes group.Diabetic models were established by intraperitoneal injection of streptozotocin (STZ) (60 mg/kg) dissolved in citric acid buffer.Blood glucose level was more than 16.7 mmol/L were used as the diabetic group.The equivalent amount of citric acid buffer was injected in the same way in the rats of the control group.The rats were sacrificed and retinas were isolated in 4 weeks and 12 weeks after modeling.The morphology of rat retinas was examined by hematoxylin and eosin staining.The relative expression levels of phosphorylated-ERK1/2 and glial fibrillary acidic protein (GFAP)(absorption,A value) in rat retinas were detected by immunohistochemistry.Results Blood glucose levels of rats were significantly higher than those in the diabetic group compared with the control group at both 4 and 12 weeks after modeling(t=14.174,13.771,both at P<0.05).In addition,the body weight was significantly lower and the drinking was much more in the rats of the diabetic group in comparison with the control group in 12 weeks(t=8.670,18.725,both at P<0.05).Twelve weeks after modeling,the decrease of retinal thickness,swelling of outer plexiform layer and decline of number of retinal ganglion cells,rods and cones were seen under the optical microscope.The relative expression levels of GFAP in the retinas were 3 197.1 ±13.1 and 7 202.0±56.8 in the diabetic group at the 4 and 12 weeks,which were significantly higher than those in the control group (2 152.8 ± 16.1 and 2 337.0±8.6) (t =6.327,16.417,both at P<0.05).In 12 weeks after modeling,the relative expression level of phosphorylated-ERK 1/2 was significantly higher in the diabetic group compared with the control group (2 850.6±2.4 versus 1 274.6± 1.3),showing a significant difference between them (t =12.771,P < 0.05).Conclusions Phosphorylation of ERK1/2 signal transduction pathway is involved in the STZ-induced DR through mediating the activation of Müller cells and inducing the apoptosis of photoreceptor and ganglione cells.
4.Heterozygous CARD9 mutation favors the development of allergic bronchopulmonary aspergillosis.

BACKGROUND:
Previous research demonstrated that a homozygous mutation of g.136372044G>A (S12N) in caspase recruitment domain family member 9 ( CARD9 ) is critical for producing Aspergillus fumigatus -induced ( Af -induced) T helper 2 (T H 2)-mediated responses in allergic bronchopulmonary aspergillosis (ABPA). However, it remains unclear whether the CARD9S12N mutation, especially the heterozygous occurrence, predisposes the host to ABPA.
METHODS:
A total of 61 ABPA patients and 264 controls (including 156 healthy controls and 108 asthma patients) were recruited for sequencing the CARD9 locus to clarify whether patients with this heterozygous single-nucleotide polymorphisms are predisposed to the development of ABPA. A series of in vivo and in vitro experiments, such as quantitative real-time polymerase chain reaction, flow cytometry, and RNA isolation and quantification, were used to illuminate the involved mechanism of the disease.
RESULTS:
The presence of the p.S12N mutation was associated with a significant risk of ABPA in ABPA patients when compared with healthy controls and asthma patients, regardless of Aspergillus sensitivity. Relative to healthy controls without relevant allergies, the mutation of p.S12N was associated with a significant risk of ABPA (OR: 2.69 and 4.17 for GA and AA genotypes, P = 0.003 and 0.029, respectively). Compared with patients with asthma, ABPA patients had a significantly higher heterozygous mutation (GA genotype), indicating that p.S12N might be a significant ABPA-susceptibility locus ( aspergillus sensitized asthma: OR: 3.02, P = 0.009; aspergillus unsensitized asthma: OR: 2.94, P = 0.005). The mutant allele was preferentially expressed in ABPA patients with heterozygous CARD9S12N , which contributes to its functional alterations to facilitate Af -induced T H 2-mediated ABPA development. In terms of mechanism, Card9 wild-type ( Card9WT ) expression levels decreased significantly due to Af -induced decay of its messenger RNA compared to the heterozygous Card9S12N . In addition, ABPA patients with heterozygous CARD9S12N had increased Af -induced interleukin-5 production.
CONCLUSION
Our study provides the genetic evidence showing that the heterozygous mutation of CARD9S12N , followed by allele expression imbalance of CARD9S12N , facilitates the development of ABPA.
