Objective To investigate the situation of family intimacy and adaptability, and social avoidance and distress in severe burn patients, and explore the correlation between them, so as to provide basis for intervention. Methods From January, 2013 to June, 2015, 289 patients with severe burn were surveyed with self-designed general condition questionnaire, Family Adaptability and Cohesion Evaluation Scale (FACES Ⅱ) and Social Avoidance and Distress Scale (SADS), and analyed with Pearson correlation analysis. Results The real family intimacy and adaptability were lower in the severe burn patients than in national norm (t>3.830, P<0.01), and the ideal family intimacy, dis-satisfied intimacy level, ideal adaptability and dissatisfied adaptability level were higher in the severe burn patients than in national norm (t>3.857, P<0.01). The SADS score was higher in the severe burn patients than in national norm. The real intimacy, real adaptability and the to-tal score of FACES II negatively correlated with the scores of SADS (P<0.05), while the levels of dissatisfied intimacy and adaptability posi-tively correlated (P<0.05). Conclusion The social avoidance and distress exist in severe burn patients, which may associated with the family intimacy and adaptability, that some interventions may target to.

Objective To study the application value of microvascular density (MVD)in determination of transrectal ultrasound hemodgynamics in the differential diagnosis of prostate cancer and chronic prostatitis, and to provide imageological basis for their differential diagnosis.Methods A total of 6 1 patients suspected of prostate cancer underwent transrectal ultrasound scan and ultrasound guided biopsy.38 cases of prostate cancer and 23 cases of chronic prostatitis were confirmed by pathology and retrospectively analyzed.The peak systolic blood flow velocity (Vs)and blood flow classification of the suspicious lesions were compared and analyzed.The MVD was observed by Weidner method with monoclonal antibody CD34 immunohisochemistry staning. Results The Vs and the blood flow classification of the suspicious lesions in prostate cancer group were significant higher than those in chronic prostatitis group (P<0.05).The MVD in prostate cancer group and chronic prostatitis group were 46.70±13.87 and 34.38±7.28,respectively(P<0.05);the MVD in prostate cancer (C+D)stage and (A+B)stage were 56.99±12.85 and 39.97±10.21,respectively(P<0.05);the MVD in prostate cancer with high Gleason score group and low Gleason score group were 53.79±13.30 and 36.96±7.24,respectively(P<0.05).The Vs and the blood flow classification of the suspicious lesions of prostate cancer had a significantly positive correlation with the MVD (r=0.793,P<0.05;r=0.723,P<0.05).Conclusion The Vs and the blood flow classification of prostate cancer by ultrasound can well reflect the changes of the micrangium in tumor tissue. The Vs and blood supply grade of suspicious lesions in the patients with prostate cancer are significantly higher than those in the patients with chronic prostatitis.They may be useful for the identification of prostate cancer and chronic prostatitis.

A simple spcctrophotometric method for determination of total rare-earth metals in food is presented. Weigh 5 gm of dry sample in a dish. After soaking the sample more than half an hour with 10 ml 3N H2SO4, evaporate to dryness on a hot plate and ash at 600℃. Dissolve the ash in 1 ml 6N HCl, and evaporate to dryness. Dissolve the residue in 2 ml water, and transfer it to a 10 mi-graduated tube. Wash the dish repeatedly with small amount of water. Transfer the washings to the same tube, and dilute to 10 ml with water. Pipette two portions of aliquots of 4 ml sample solution to two 10ml graduated tubes in which 2 ml of HCOOH-NH3 buffer (pH 3.7) is added. After mixing, add 1 ml each of 10% sulphosalicylic acid solution and 5% ascorbic acid solution. Make volume to 8 ml with water. Mix thoroughly. After 10 minutes, add 1 ml 15% alcoholic diphenylguani-dine solution, mix and add 1 ml 0.025% arsenazo Ⅲ &K solution. Mix, and measure the optical density at 680, 660 and 640 nm against a reagent blank solution. Calculate the total rare-earth metals content with a calibration curve. The sensitivity of the method is 0.01 ?g/ml and the detection limit is 0.05ppm (5 gm of sample). The average recovery of added TRExOy is 91.9% (n = 14).

Objective To investigate the application of alpha-fetoprotein (AFP),α-L-fucosidase(AFU),5'-nucleotidase(5'-NT) and γ-glutamyltransferase (GGT) in diagnosis of hepatocellular carcinoma(HCC),and to provide the basis for its early and rapid diagnosis.Methods The clinical data of 120 cases with initial diagnosis as hepatocellular carcinoma,150 cases with liver cirrhosis,200 cases with hepatitis were analyzed retrospectively from June 2013 to August 2014,meanwhile,100 healthy people were selected as control group.The serum AFP,AFU,5'-NT and GGT levels were measured by electrochemiluminescence immunoassay,rate method,peroxidase method and enzymatic colorimetric assay,respectively.The results were analyzed by receiver operating characteristic (ROC) curve,while the sensitivity,specificity,AUC were calculated at the same time.Results The levels of serum AFP,AFU,5'-NT and GGT were significantly different among HCC group,liver cirrhosis group,hepatitis group and control group (all P＜0.05).When tested alone,the sensitivities of AFP,AFU,5'-NT and GGT were 58.33 ％(70/120),82.50 ％(99/120),85.00 ％(102/120),78.33 ％(94/120),the specificities were 88.33 ％(106/120),73.33 ％(88/120),88.33 ％(106/120),71.67 ％(86/120),and AUC were 0.655,0.702,0.814,0.754.When four indexes were performed with combined test,the sensitivity was 98.33 ％(118/120),specificity was 96.67 ％(116/120),AUC was 0.975,which were higher than the single detection (all P＜0.05).Conclusion The combined detection of serum AFP,AFU,5'-NT and GGT can greatly increase the sensitivity and specificity for diagnosing HCC and will gain mutual complement advantages,which is of great clinical value for early and rapid diagnosis.

Objective:To investigate specific cellular immune response to the HPV16E7 prophylactic vaccine in mice.Methods:BALB/c mice were randomly divided into 3 groups:experimental group (treated with pcDNA3.1-HPV16E7),control group Ⅰ(treated with pcDNA3.1) and control group Ⅱ(treated with N.S.).Mice were injected (i.m.) pcDNA3.1-HPV16E7,pcDNA3.1 and N.S. one time per week,respectively.After three immunization,the blood samples from eye sockets and the supernatant cultured of spleen cells were taken for measurement IFN-? and the number of CD4 +?CD8 +T-lymphocyte by ELISA and FACS assay.Antigen-specific splenocyte proliferation assay in vitro was detected by MTT method.Results:The splenocytes actively proliferated,the number of CD4 +T lymphocyte and the quantitation of IFN-? in spleen and serum in the experimental group were significantly higher than the control group Ⅰ and Ⅱ.Conclusion:The pcDNA3.1-HPV16E7 DNA vaccine can induce specific cellular immune response in BALB/c mice.

OBJECTIVE:To evaluate the status quo of the use of antiallergic drugs.METHODS:The antiallergic drugs used during 2004～ 2006 in the inpatient pharmacy including the non-surgery department,surgery department and pediatric department of our hospital were analyzed statistically in respect of kinds,consumption sum and DDDs.RESULTS:The antiallergic drugs had a fluctuation in consumption sum and DDDs,showing a reduction in 2006.No significant change was noted in the structure of drug use.CONCLUSION:More and more new antiallergic drugs went on the market,but the application still gives priority to the cheaper kinds.

Objective To construct,express,purify and identify the gene encodi ng major outer membrane protein of Neisseria gonorrhoeae (Porin I, or PI). Metho ds The gene encoding for PI of N.gonorrhoeae was amplified by PCR and cloned int o expression plasmid pGEX-4T-2 to form pGEX-4T-2/PI recombinants. A high lev el expression of GST-PI fusion protein was obtained in GST gene fusion system (GST:glutathione S transferase). The analysis indicated that the expressed pr otein was present predominantly in the insoluble form. Therefore, the induced pr otein was purified by SDS-PAGE, and bands corresponding to polypeptides of GST-PI fusion protein were excised and subjected to electroelution. A dot immunoch romatographic assay was employed to demonstrate whether the purified protein was gonococcal PI specific. Results The pGEX-4T-2/PI expression recombinants were constructed,expressed,purified and identified successfully. SDS-PAGE analysis and dot immunochromatographic assay suggested that the recombinant GST-PI fusio n protein was a 60 000 molecular weight protein andidentical in size to native PI and reacted with anti-PI monoclonal antibody. Conclusion Our results may lead to a potentiality for further study of diagnosti c kits and vaccine for Neisseria gonorrhoeae.

Objective To analyze the relationship between plasma concentration and efficacy , adverse drug reactions by monitoring vancomycin serum concentrations for appropriately treating the infections caused by methicillin‐resistant Staphylococcus aureus or other gram‐positive cocci .Methods Vancomycin concentration was monitored in the patients with indications for vancomycin therapy .Blood sample was taken after vancomycin was administered for at least 4 doses .The blood sample collected within 30 minutes before dosing was used to determine the trough blood concentration .The samples were taken within 30 minutes to 1 hour after infusion of vancomycin were used to estimate the peak concentration by fluorescence polarization immunoassay .The clinical data were collected at the same time to analyze clinical efficacy and safety .Results Vancomycin trough concentration ranged from 3 .22 mg/L to 50 .79 mg/L in 25 patients ,specifically ,＜ 5 mg/L in 3 cases ,5‐＜10 mg/L in 11 cases ,10‐15 mg/L in 3 cases ,and > 15 mg/L in 8 csaes .Peak concentration ranged from 13 .57 mg/L to 60 .47 mg/L ,specifically ,＜ 25 mg/L in 14 cases ,25‐40 mg/L in 7 cases ,and > 40 mg/L in 4 cases .The infection was cured in 80 .0% (20/25) of the patients .The gram‐positive cocci were eradicated in 87 .5% (21/24) of the patients .The dosage of vancomycin was adjusted in 13 patients according to the results of blood concentration monitoring .Majority of these patients (12/13 ,92 .3% ) were cured .Renal impairment was observed in 4 patients .Conclusions Vancomycin is safe and effective in treatment of methicillin‐resistant Staphylococcus aureus and other gram‐positive bacterial infections . Vacomycin concentration varies from person to person . Serum concentration monitoring is required to achieve best outcomes and the goal of individualized treatment of vancomycin.

Objective To explore the clinical application of single or combined detection of serum Cys-C,5'-NT and AFP in hepatocellular carcinoma (HCC) in order to provide the platform for early diagnosis and prognosis evaluation.Methods The serum levels of Cys-C,5'-NT and AFP from 148 cases of HCC patients (HCC group),135 cases of liver cirrhosis patients (LC group) and 155 cases of hepatitis (hepatitis group) and 100 healthy people (control group) were measured by latex-enhanced immunoturbidimetric assay (LEITD),peroxidase method and electrochemiluminescence immunoassay (ECLI),respectively.Then their differences had been compared.The sensitivity,specificity and Youden's index were calculated and the results were analyzed by receiver operating characteristic (ROC) curve.Results The levels of serum Cys-C,5'-NT and AFP were significantly different in 4 groups.The differences were statistically significant (F =12.35,42.25,58.12,P =0.000).The sensitivity of three indicators for diagnosing HCC was 100 ％,which was higher than that of single or two combined detection (P ＜ 0.05),and the specificity was 78.97 ％.The area under ROC curves of combined detection of three indicators was 0.977,which was also higher than single or two combined detection (P ＜ 0.05).Conclusions The combined detection of serum Cys-C,5'-NT and AFP can greatly increase the sensitivity and accuracy for diagnosing HCC,which has an important clinical value for early diagnosis.Therefore it is worth popularizing and applying on clinic.
