1.CHEMICAL CONSTITUENTS OF MARINE SPONGE GEL-LIODES SPINOSELLA IN THE SOUTH CHINA SEA

Three known compounds have been isolated from the marine sponge Gel-liodes spinosella in the South China Sea. The crude ethanol extract was partitioned between ethyl acetate and water, and then between w-butanol and water. The butanol fraction was chromatographed on silica gel column to obtain thymidine(1l) ,phenylacetic acid(2)and L-(-)-3-phenylanine(3). Their structures were determined by UV,IR,1HNMR,and 13CNMR data.

Their structures were determined by UV,IR,MS,1HNMR and 13CNMR data. It first reported about the chemical compounds of the hard coral Pavona frondifera Lamarck.
3.Primary culture of human preadipocyte in a serum-free medium

Objective To establish a method of primary culture of human preadipocyte in a serum-free medium. Methods Collangenase digestion was used to dispart preadipocyte.The cells were identified by microscopy stained by oil red O and determination of the activity of Glycerol-3-phosphate dehydronase(G-3-PDH). Results The primary cultured human preadipocyte proliferated in the aserum-free medium successfully.In the differential serum-free medium the cells turned to be round on the 4 th day.The adipose drops began to cumulate in the cells,and to the most quantity until the 21 st day.Conclusion The human preadipocyte can be primarily cultured and induced to differentiate in serum-free medium,which is the base for researching the effects of hormones and factors to the proliferation and differentiation of preadipocyte.
4.Cytokines regulate the promoter activities of human growth hormone gene in rat pituitary MtT/S cells

Objective:To study the effect of interleukin-11(IL-11),ciliary neurotropic factor (CNTF) and transforming growth factor-β (TGF-β) on the hGH gene promoter activity in rat pituitary MtT/S cells and the interaction with pituitary-specific transcription factor Pit-1.Methods:Stable transformed MtT/S cell line which contains hGH gene promoter -484-30 bp and luciferase reporter gene firstly established,then the concentration of GH in the medium and lysate of MtT/S cells and luciferase activities in MtT/S cells were measured after treatment these cells with the above cytokines,the effects of cytokines on secretion and synthesis of GH,and the promoter activity of the hGH gene were observed.Results:The results showed that IL-11(20 nmol/L),CNTF(10 nmol/L) and TGF-β(5 nmol/L) regulated secretion and synthesis of GH,and the luciferase expression in stable transformed MtT/S cells.IL-11 and CNTF had the stimulatory effect,whereas TGF-β had the inhibitory effect.Neither overexpression of Pit-1 nor inhibiting Pit-1 expression affected the regulatory role of these cytokines.Conclusion:IL-11,CNTF and TGF-β regulate the GH production in pituitary MtT/S cell line by regulating the hGH gene promoter activity.Pit-1 may not be involved in these actions.
5.Stability and safety of mini-implant anchorage in orthodontic treatment

BACKGROUND: Traditional orthodontic anchorage has the disadvantages of discomfort, difficult to control, affecting the appearance, and relying on patient’s close cooperation. Because of short-time clinical research and application, mini-implant anchorage has a great controversy in the orthodontic treatment. OBJECTIVE: To explore the application value of mini-implant anchorage in the orthodontic treatment. METHODS:Eighty cases undergoing the orthodontic treatment at the Center of Ophthalmology and Otolaryngology, Yaan People’s Hospital in China from January 2012 to June 2015 were enrol ed in this study. These patients were equal y randomized into test group and control group, which were subjected to mini-implant anchorage or headgear anchorage, respectively. The treatment was continued for 2 years in the two groups. RESULTS AND CONCLUSION: After the treatment, the successful rate in the test group was significantly higher than that in the control group (P < 0.05); the upper central incisor convex distance difference, inclination angle of the upper central incisor, displacement of the molar, SNA angle, inter-canine width, were significantly better in the test group than the control group (P < 0.05). At weeks 4 and 8 after intervention, there was no difference in the expression of osteoprotegerin in the periodontal tissue between the two groups (P > 0.05), but the rate of adverse reactions was significantly lower in the test group than the control group (P < 0.05). These findings indicate the mini-implant anchorage method can significantly improve the therapeutic efficacy, improve the success rate of treatment, and reduce the incidence of postoperative infections, which has good safety and stability, and has good clinical application value.
6.Change of reference gene glyceraldehyde-3-phosphate dehydrogenase expression during 3T3-L1 adipocyte differentiation

Objective To observe the change of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) gene expression during 3T3-L1 adipocyte differentiation as well as other reference gene expressions.Methods The mRNA expressions of several common reference genes were detected by real time-PCR on day 0,1,3,5,and 7 of 3T3-L1 adipocyte differentiation.Western blot was used to confirm the protein expressions of three common reference genes.Results (1) GAPDH and transferrin receptor(TFRC) mRNA expressions were significantly increased during adipocyte differentiation.GAPDH mRNA level was increased by 5.7,7.6,22.0,and 24.5 folds on day 1,3,5,and 7 after induction of adipocyte differentiation,but no apparent changes of β-actin,α-tubulin,peptidylprolyl isomerase A (PIPA),and 18S mRNA expressions were detected.The expression changes of key transcript factors for adipocyte differentiation such as PPARγ2,C/EBPα,and C/EBPβ were under-estimated by real time-PCR if GAPDH was chosen as the reference gene.Western blotting results showed that the GAPDH protein level increased gradually during adipocyte differentiation,especially on day 5 and 7 after adipocyte differentiation.There were no obvious changes of β-actin and α-tubulin protein expressions.(2) Berberine significantly inhibited mRNA and protein expressions of GAPDH in the process of adipocyte differentiation.GAPDH mRNA levels were reduced by 68.1% and 66.3% on day 5 and 7 after induction of adipocyte differentiation,but with no significant change in other reference genes.Conclusion It is not suitable for GAPDH to be used as an endogenous reference gene during 3T3-L1 adipocyte differentiation.
7.Practice of evidence based nursing program in rehabilitation nursing of ischemic stroke

Objective To explore the clinical effect of evidence-based nursing program in rehabilitation nursing of ischemic stroke. Methods Eighty-four patients with ischemic stroke admitted in our department from February 2015 to November 2016 were randomly divided into study group and control group(n=42).The evidence-based nursing program was adopted in the study group and in the control group conventional nursing program was used.In 6 months follow-up,the two groups were compared concerning compliance and daily life ability and hand function. Results The scores on rehabilitation training,life and rest,follow-up visits and the total score of compliance in the study group were significantly higher than those of the control group after 6 months follow-up(P<0.05).The Barthel index(BI)index of the study group was significantly higher than that of the control group,and the score on the nine-hole column was significantly lower than that of the control group(P<0.05). Conclusion Evidence-based nursing can effectively improve the compliance of patients with ischemic stroke and daily life ability and hand function after discharge,so it is worthy of clinical application.
8.Perioperative nursing care of 25 patients underwent total parathyroidectomy with forearm autograft

To summarize key points of nursing care of 25 patients with secondary hyperparathyroidism underwent parathyroid autotransplantation with forearm (tPTX+AT).The integrated mode was adopted during the perioperative period,focusing on the following key points:perioperative management of environment,nursing of vascular access,intro-operative management of sample,nursing management during anesthesia and recovery,management of drawing blood,treatment and nursing care of postoperative hypocalcemia,prevention and nursing care of infection and hemorrhage,wound care in the forearm and monitoring of graft.Compared with the preoperative period,levels of intact parathyroid hormone(iPTH),calcium and phosphorus were significantly decreased during the postoperative period (P<0.01);symptoms were significantly improved after surgery included bone and joint pain,muscular weakness,and insomnia.
9.Expression of RUNX3 in cervical carcinoma and its clinical significance.

OBJECTIVE To explore the role of runt-related transcription factor 3(RUNX3) in the tumorgenesis and progression of cervical carcinoma. METHODS The immunohistochemical staining technique was used to detect the expression of RUNX3 protein in 25 cases of normal cervix, 34 intraepithelia neoplasia (CIN), and 48 cervical carcinomas. SYBR Green I chimeric fluorescence Real-time PCR was applied to detect the expression of RUNX3 mRNA in 10 cases of normal cervix, 24 CIN, and 30 cervical carcinomas. RESULTS The expressions of RUNX3 protein and mRNA in normal cervix, CINI,CINII-III, and cervical carcinoma tissues tended to be down-regulated. There was significant difference among these groups (P<0.05). The expressions of RUNX3 protein and mRNA in the cervical carcinoma tissues were correlated with the histological differentiation, clinical stage, and lymphatic metastasis (P<0.05), but had no relationship with the age, high-risk human papillomavirus infection, and histological classification (P> 0.05). CONCLUSION RUNX3 may function as a tumor suppressor gene in the occurrence and progression of cervical carcinoma.

10.Genotype-phenotype landscape of pituitary adrenocorticotroph hormone adenoma

Objective:Cushing′s disease(CD) is caused by the pituitary adrenocorticotroph hormone(ACTH) secreting adenomas, leading to increased serum cortisol levels and various abnormal metabolic processes. Untreated CD is linked to high mortality, thus it is critical to elucidate its pathogenesis. This study aims to explore the pathogenesis of pituitary ACTH adenomas using whole-genome sequencing analysis.Methods:Fresh tumor tissues and peripheral blood samples were collected in 9 confirmed cases of pituitary ACTH adenomas who underwent surgery. Whole genome sequencing was then performed, followed by analysis and verification of single nucleotide mutations, copy number variation(CNV) and chromosome structure variations.Results:Somatic USP8 mutations(p.Ser718del, p. Ser718Pro, p. Pro720Arg, p. Pro720Gln) were found in 5 patients, with a rate of 55.6%; CNV of USP8 was detected in 1 patient; TP53(p.Cys135Tyr), NF1(p.Val1049Glufs*11) and KMT2C(c.3323+ 1G>A) mutations were identified in 1 patient harboring wild-type USP8. CNV analysis showed a loss of heterozygosity in multiple chromosomes in a wild-type USP8 patient. Structural variations were found in 2 with unknown significance. No germline gene mutations were detected in this study.Conclusion:Somatic USP8 mutations, increased copy number of USP8, variations of tumor-related genes such as TP53 and extensive somatic CNV all contribute to pathogenesis of CD. Chromosomal structure variations may suggest high-risk pituitary ACTH adenomas, and call for frequent follow-up and aggressive treatment.
