AIM: To explore the effects of aflatoxin G 1(AFG 1 )on proliferation and TNF-? secretion of human peripheral blood mononuclear cells(HPBM) in vitro. METHODS: The effects of AFG 1 on proliferation of HPBM were analysed with flow cytometric (FCM) DNA analysis and MTT bioassay, while that on TNF-? secretion was detected with ELISA. RESULTS: FCM analysis revealed that 6 h after treatment, proliferation index(PI) of 1000 ?g/L AFG 1 treated HPBM was significantly higher than that of control. 24 h after AFG 1 treatment, stimulating effects on proliferation was found in HPBM treated with AFG 1 at 200 ?g/L and 1 000 ?g/L.Regression analysis showed that PI was postively correlated with the concentrations of AFG 1 in the concentration range from 0 to 1 000 ?g/L( r=0. 5122 and 0.5119 respectively,P

AIM: To investigate the role of angiopoietin-1 (Ang1) and Tie2 receptor in angiogenesis after myocardial infarction through detecting their mRNA expression in normal and infracted myocardium. METHODS: The experiment was conducted in the laboratory of Biochemistry and Molecular Biology, Medical Department of Peking University from April 2006 to April 2007. Forty male SD rats were randomly divided into acute myocardial infarction model group and sham-operation group. The myocardial infarction model was established in the rats of model group through the ligation of left anterior descending artery, while the rats in sham operation group were braided of the left anterior descending artery without ligation. Five rats in both groups were executed at 3, 7, 14, and 28 days after model establishment. RNA was extracted from the same site of left anterior wall, and the polymerase chain reaction was used to semiquantitatively analyze the Ang1 and Tie2 receptor mRNA expression with GAPDH gene as internal control; meanwhile, the immunohistochemistry was used to detect vascular density in and around infarction area. All the treatments for animals were accorded with the animal ethical standards. RESULTS: All 40 rats were included in the final analysis. Both Ang1 and Tie2 receptor were expressed in normal myocardium. In the 28 days after myocardial infarction, Ang1 expression kept at almost the same level without changing, but Tie2 receptor expression was slightly elevated at 3 days, reached peak value at 7 days, and returned to the baseline value at 14 days. The vascular density increased both infarction and peri-infarction area at 7 days after acute myocardial infarction, and did not change with time. CONCLUSION: Tie2 receptor expression is elevated and coincided with angiogenesis after myocardial infarction. It may play a role in the development and stabilization of the blood vessel after myocardial infarction.

ObjectiveTo analyze the relationship between serum total bilirubin coincident with congestive heart failure (CHF) exacerbation and subsequent long-term mortality in patients with CHF.MethodsThe study population consisted of 140 consecutive patients admitted for CHF exacerbation with left ventricular ejection fraction ≤45%.They were divided into 2 groups according to whether death attacked or not in the following 28.5 months. Binary logistic regression analysis was used to investigate independent predictors of death from clinical parameters on admission or within 24 hours. ResultsSerum TBil and B-type natriuretic peptide (BNP) levels on admission were independent predictors of subsequent death after hospital discharge.According to increasing textiles of TBil stratified by the level of 12.8 and 18.2 mmol/L,the patients were divided into 3 groups:lower-level group (TBil ≤ 12.8 mmol/L),moderate-level group (TBil > 12.8 ~ 18.2 mmol/L) and higher-level group (TBil > 18.2 mmoL/L),with the death rates after 28.5 months of 12.2%,17.9% and 38.9%,respectively ( P =0.002 ). Meanwhile, the pulse pressure decreased to (55.5 ± 17.3) mm Hg (1 mm Hg =0.133 kPa),(48.9 ± 13.1) mm Hg and (46.1 ± 13.7)mm Hg,respectively ( P =0.008 ). TBil on admission had significant correlation with echocardiographymeasured left ventricular endo-diastolic diameter ( r =0.34,P =0.000 ) and right ventricular diastolic diameter (r =0.23,P =0.011 ). ConclusionsIncreased TBil coincident with cardiac decompensation predicts a worse long-term death of CHF,presumably through the potential liability to both decompensated RV function and lower cardiac output syndrome occurred simultaneously when HF deteriorates.

Shepp-Logan head phantom is the classical model of simulation computing in Computerized Tomography. This paper proposes a new method--considering 3D Shepp-Logan head phantom as the basic reference model to carry on simulation experiment and algorithms performance evaluating in three-dimension medical image reconstruction field. The paper introduces the design and the implementation of 3D Shepp-Logan head phantom as well the computing of projection data at first, and then describes the simulation process of the three-dimension medical image. The numerical experiment gives the three-dimension medical image reconstruction simulations based on 3D Shepp- Logan head phantom. The result of this experiment indicates the accuracy of model computing and the feasibility of our this method.

Journal of Biomedical Engineering 2008;25(3):708-715
In this paper is presented the research and development of a new myocardial contrast echocardiography (MCE)-based quantitative analysis system for myocardial microcirculation. The system can measure the values of A, beta, A x beta, A-EER (endo-epi ratio of A), beta-EER and A x beta-EER from the signal intensity of real-time 2-D grayscale images and power Doppler images; it can draw the time-intensity curves to indicate the variation of the intensity of microbubbles scattering in subendocardial layer and subepicardial layer with the varying of myocardial segments; and it can estimate the hemodynamic parameters by nonlinear regression analysis. So the MCE software system has brought the quantitative analysis of real-time MCE to success.

This article reports a quantitative analysis software system for myocardial contrast echocardiography (MCE). It can measure the signal intensity of grayscale images and power Doppler images, draw the time-intensity curves of variations on the intensity of microbubbles scattering in subendocardial layer and subepicardial layer with the pulsing intervals, and estimate the hemodynamic parameters by nonlinear regression analysis. This system has been applied to a study on 20 healthy volunteers, and the results suggest that the software has the capacity for bringing the quantitative analysis of MCE to success. The MCE software system conforms to the DICOM standard and can be integrated into PACS.

Objective To investigate the safety and efficacy of second allogeneic hematopoietic stem cell transplantation for the relapsed hematologic malignancies. Methods The data of 25 relapsed patients received the second allogeneic transplantation as a salvage therapy in Institute of Hematology Peking University between October 1999 and March 2010 were analyzed retrospectively. Twenty-four patients relapsed at 8. 8 (1-55) months after the first transplantation, except one received the second transplantation as prophylaxis therapy. They received the second transplantation after 3(0. 3-20) months' therapy. The median time between the 2 transplants was 10. 6(0. 6-59. 0) months. Results Most of the patients were given the conditioning regimen including total body irradiation (TBI, 700-779 cGy) or modified busulfan and cyclophosphamide (BUCY, BU 12 mg). All patients survived more than 30 days and achieved sustained white blood cell engraftment. Sinus obstructive syndrome, irradiation dermatitis and acute myocardial infraction were occurred in 3 patients and recoverable. Until January 31 in 2011, with a median observation period of 9. 1 (2. 0-131. 1) months, 8 patients had been living with a overall survival (OS) of 30.9%.Twelve patients relapsed at a median 4. 4 months and 10 died of it. The other 7 patients died of transplant related complications. The non-relapsed mortality was 35. 1 %. The disease status at the 2nd transplantation was the only factor which effected the OS (P = 0. 009). Conclusions The second allogeneic transplantation is a viable option for patients relapsing after the first transplantation. Reduced intensive conditioning regimen ensures the graft engraftment and reduces transplant related toxicity.
