1.Quality Standard of Erkening Misture

OBJECTIVE: To establish the quality stangard of Erkening misture.METHODS:Raphanus sativus and Fritillaria thunbergii were identifided by TLC, and the content of the Baicalin in Erkening misture was determinated by HPLC. RESULTS: The TLC spots of Raphanus sativus and Fritillaria thunbergii were distinctive. The linear range of Baicalin was 0.319~2.64 ?g (r=0.999 3). The average recovery was 99.56%, and RSD was 0.83% (n=9). CONCLUSION: This established standard can be used for the quality control of the Erkening misture.
2.Clinical study of laparoscopically assisted vaginal hysterectomy

Objective To investigate the clinical value of laparoscopically assisted vaginal hysterectomy(LAVH).Methods A total of 125 cases of benign uterine diseases were freely divided into two groups: 65 cases was given LAVH(LAVH Group) and 60 cases was given traditional total abdominal hysterectomy(TAH Group).Results The operation time was 127?4.1 min in the LAVH Group and 70?1.7 min in the TAH Group(t=99.669,P=0.000).The blood loss was 80?23.1 ml in the LAVH Group and 190.8?53 ml in the TAH Group(t=-15.339,P=0.000).The incidence of postoperative complications was 30.8%(20/65) in the LAVH Group and 58.3%(35/60) in the TAH Group,with significant difference between the two groups(?~2=9.621,P=0.002).Conclusions LAVH has advantages of micro-invasion,little blood loss,and quick recovery,being an ideal procedure.
3.The tyrosine phosphatase SHP-2 participated in the cell migration induced by IL-1β

Objective To investigate whether the tyrosine phosphatase SHP-2 participate in the cell migration induced by IL-1β.Methods Two different reorganized plasmids transfected into the MCF-7,and achieved two different cell lines:SHP-2-GFP-MCF-7 cell lines.SHP-2C>S-GFP-MCF-7 cell lines.The cell migration was detected by a fluorescence microscope and the expression of E-eadhefin and MMP-9 wee detected by western blotting analysis and RT-PCR.Results The SHP-2-GFP-MCF-7 cell lines showed a higher cell migration ability,and the expression of E-cadherin in the SHP-2-GFP-MCF-7 cell lines was especially lower,neady undetectable.But in this group the expression of MMP-9 was the highest.Conclusion The tyrosine phosphatase SHP-2 participate in the cell migration induced by IL-1β,and maybe produce a marked effect by reinforcing the expression of MMP-9 and reducing the expression of E-cadherin.
4.Na+,K+-ATPase inhibitor induces cell cycle arrest in liver cancer HepG2 cells by regulating expression of DNA damage Mre11/Rad50/Nbs1 complex

Aim To explore the relationship between Mre11/Rad50/Nbs1 ( MRN ) complex focus formation and DNA double-strand breaks( DSBs) caused by cinob-ufagin in human hepatocellular carcinoma HepG2 cells. Methods The Na+,K+-ATPaseα1 subunit expression level in liver cancer tissues was detected by immunohis-tochemistry. After HepG2 cells were treated with 5μmol·L-1 cinobufagin for 6, 12 and 24 h, the drug-in-duced DSBs were assessed by single cell gel electro-phroesis ( SCGE ) , the gene transcription and protein levels of Mrel1, Nbs1, Rad50 and p53 were evaluated by Real time-PCR and Western blot. The cell cycle in parallel was analyzed by flow cytometry. Results The Na+, K+-ATPase α1 subunit expression level in liver cancer tissues was significantly increased compared with the tissue adjacent to carcinoma ( P <0. 05 ) . The 5μmol · L-1 cinobufagin could induce the DSBs in a time-dependent manner (P <0. 05), and it could up-regulate the gene expression levels of Mre11, Nbs1, Rad50 and p53 in HepG2 cells ( P<0. 05 ) . The pro-portions of HepG2 cells in S phase were ( 21. 32 ± 4. 21) % in the control group, and (33. 25 ± 5. 72) %, (56. 72 ± 6. 29) % and (67. 32 ± 9. 42) % in HepG2 cells treated with 5 μmol · L-1 cinobufagin for 6, 12 and 24 h, respectively. The proportions of cells in S phase in cinobufagin groups were significantly increased compared with the control group ( P<0. 05 ) . Conclu-sion Cinobufagin could induce the cell cycle arrest in liver cancer HepG2 cells by activation of Mre11/Rad50/Nbs1 Complex.
5.Determination of Serum Concentration of Vitamin A in Healthy Volunteers and Hepatocirrhosis Patients by HPLC

OBJECTIVE:To establish a method for the concentration determination of vitamin A(VitA)in human serum,and apply it in healthy volunteers and hepatocirrhosis patients. METHODS:After liquid-liquid extraction,serum sample was deter-mined by HPLC. Using VitA acetic ester as internal standard,the separation was performed on Kromasil C18 column with mobile phase consisted of methanol-water (98∶2,V/V) at the flow rate of 1 ml/min. The wavelength was set at 325 nm,and the column temperature was room temperature. The sample size was 20 μl. RESULTS:The linear range of VitA were 0.012 4-3.210 μg/ml(r=0.997 2,n=5),and the limit of quantification was 0.012 4 μg/ml. Inter-day and intra-day RSD ranged 1.66%-2.97%;sample re-coveries were 98.18%-99.56%;extraction recoveries were 89.59%-91.38%. Average blood concentration of VitA were(0.71±0.08)μg/ml in 24 healthy volunteers and (0.28 ± 0.06)μg/ml in 24 hepatocirrhosis patients. There was statistical significance in average blood concentration of VitA between healthy volunteers and hepatocirrhosis patients in different age groups (P<0.05). CONCLU-SIONS:The method is simple,rapid,sensitive and accurate,and can be used for the concentration determination of VitA in serum of healthy volunteers and hepatocirrhosis patients.
6.The effects of preemptive analgesia with nalbuphine on inflammatory factors in elderly patients undergoing ;thoracotomy

Objective To investigate the effects of preemptive analgesia with nalbuphine on inflammatory factors in elderly patients undergoing thoracotomy. Methods Eighty elderly patients with ASA I or Ⅱ undergoing thoracotomy were randomly allocated into control group (group C:n=40) and Nalbuphine group (group N:n=40). Each patient received 20 mg of Nalbuphine intravenously before induction of anesthesia in group N and same volume of normal saline in group C. Plasma levels of IL-10, TNF-α and IL-6 were measured at different time points. The VAS was recorded and the MAP, HR, SpO2 and PaCO2 were monitored. Results Compared with group C, the levels of TNF-α and IL-6 were lower, while IL-10 was higher at T2, T3, T4 in group N(P < 0.05). The VAS, MAP, HR and PaCO2 in group N were lower than those in group C at T2, T3 and T4 (P < 0.05). Conclusion Preemptive analgesia with Nalbuphine can improve the efficacy of postoperative analgesia and effectively reduce inflammatory reaction in the old patients undergoing thoracotomy.
7.MgSO4 inhibited X-rays irradiation induced expression of γ-H2AX in HUVEC

Objective To investigate the influence of MgSO4 with different concentrations on cell survival and γ-H2AX expression in HUVEC irradiated with X-rays.Methods Cell proliferation rate was assayed by CCK-8,γ-H2AX foci formationwas observed with a laser confocal microscope,and γ-H2AX protein expression was detected by flow cytometer and Western blot assay.Results MgSO4 with a concentration of 1.25 mg/ml could improve the survival rate of IHUVEC treated with X-rays (t =-6.34,P < 0.05).After irradiation,γ-H2AX loci approached to the highest level from 30 min to 1 h after radiation and then decreased.MgSO4 significantly reduced the foci formation at 0.5,1,2,6 and 12 h postirradiation (t =12.62,6.36,11.93,5.75,9.43,P < 0.05).Both flow cytometry,and Western blot assays showed that MgSO4 inhibited γ-H2AX protein expression at 0.5,1 and 2 h post-irradiation (t =6.07,5.32,11.85,P < 0.05).Conclusions MgSO4 could improve the survival rate and reduced γ-H2AX expression in HUVEC irradiated with X-ray.
8.The linkage between cell cycle S phase arrest and apoptosis on human hepatocellular carcinoma HepG2 induced by Na~+,K~+-ATPase inhibitors via regulating proteins associated with cell cycle

Aim To investigate the effect of ouabain and cinobufogenin on cell proliferation,apoptosis and cell cycle on HepG2,and explore their molecular mechanism.Methods The anti-proliferative effect on HepG2 cells was determined by MTT assay.The HepG2 cells were stained with Hoechst 33342,and its morphological changes were observed under fluorescence microscope;The cell cycle was measured by flow cytometry.The Cyclin A1,CDK 2,PCNA and p21~(CIP1) expression levels of HepG2 cells treated with ouabain and cinobufogenin were dectected in mRNA and protein by Real time PCR and Western blot.Results Ouabain and cinobufogenin could inhibit cell proliferation on HepG2 cells,and the inhibitory effects were in time and dose dependent manners.The HepG2 cells treated with ouabain and cinobufogenin showed the typical morphological features of apoptosis.Cell cycle analysis showed that the S phase of HepG2 cells treated with ouabain and cinobufogenin increased significantly compared with the control group.Real-time quantitative PCR and Western blot results showed that ouabain and cinobufogenin could down-regulate Cyclin A1,CDK 2,and PCNA expressions(P<0.05)and up-regulate p21~(CIP1) expression(P<0.05).Conclusion Nα~+,K~+-ATPase inhibitor has the anti-proliferative effect on HepG2 cells and induce apoptosis and S phase arrest.These effects might be related with proteins associated with cell cycle closely.
9.MRI diagnosis of acute spinal cord decompression sickness

Objective To describe MRI findings of acute spinal cord decompression sickness.Methods MRI findings of 5 cases with clinical definite acute spinal cord decompression sickness were retrospectively analyzed.The main clinical informations included underwater performance history against regulations,short-term complete or incomplete spinal cord injury symptoms after fast going out of water,sensory disability and urinary and fecal incontinence,etc.Results Spinal cord vacuole sign was found in all 5 cases.Iso-signal intensity(n=3),high signal intensity(n=1),and low signal intensity (n=1)was demonstrated on T1 WI,and high signal intensity(n=5)was found on T2 WI.Owl eye sign was detected in 3 cases,and lacune foci were seen in 2 cases.Conclusion MRI findings of acute spinal cord decompression sickness had some characteristics,and it was easy to diagnose by combining diving history with clinical manifestations.
10.Investigation and analysis of consciousness of cornea donation in Chengde city

OBJECTIVE: Chengde is a city with many corneal blindness patients In north China. Lacking of corneas donation is the main reason which prevents corneal transplantation. Survey was made by questionnaire in Chengde to evaluate the current situation and the influential factors of corneal donation.METHODS: Survey was made in 3 200 Chengde residents aged 18 years or older, includes outpatients and inpatients of ophthalmology, some undergraduate students and people met accidentally in park, supermarket, station and centre for elders.48.6% are male and 51.4% are female. Self-made questionnaire includes general state, questions about cornea donation and factors influencing cornea donation.RESULTS: Among 3 200 questionnaires, 2 971 were valid. The effective rate was 92.84%. Over 50% people support donating cornea. More than 40% people intend to donate their corneas and support their relatives to donate. Among the factors for not intending to donate cornea, lacking knowledge of cornea donation was the main reason accounting for 42.81%, and worrying about the misusage of donating cornea without corresponding law became the second factor, which accounting for 21.07%. It has no influence on the consciousness of cornea donation by the difference of sex and location between city and countryside.Whereasfession and level of education indeed influence the consciousness of cornea donation, which of the people from 18 to 40 years old was greater than those of over 40, medical workers was greater than those from other fields, the people graduating from secondary specialized school or higher was greater than those graduating under secondary specialized school.CONCLUSION: People in Chengde have a positive attitude towards cornea donation. It is very necessary to enhance the education of cornea donation, establish an easy and smooth way for donation may promote cornea donating. Consummate legislation is also needed for cornea donation.
