1.Optimum Selection Processing Study of Rhizoma Atractylodis Macrocephalae Stir-fried with Bran by Orthogonal Design

Objective To research the best processing method of Rhizoma Atractylodis Macrocephalae stir-fried with bran. Methods L9(34) orthogonal table are decided with three factors:The amount of the adjuvant, heating temperature, heating time. According to the amount of water-soluble extractive, alcohol-soluble extractive and volatile oil, orthogonal design is applied to processing. Results The best processing method is:Rhizoma Atractylodis Macrocephalae were processed by stir-frying with 10 percent of bran for 5 min in a heat of 150 ℃. Conclusion The results can provide basis for establishing the quality standard of this crude drug.
2.Blocking effects of anti-sense peptide of C5a on the adhesion between pulmonary vascular endothelial cell and neutrophil

Objective:To study the blocking effects of anti-sense peptide of C5a on the adhesion between pulmonary vascular endothelial cell(PVEC) and neutrophil resulting from C5a anaphylatoxin.Methods:It was determined by Flowcytometry that the change of adhesion molecule expression on PVEC and the activity of MPO in PMN was determined after adhesion.Results:In response to C5a after interactions with several concentrations of anti-sense peptide R4, the expression of P-Selectin on PVEC decreased significantly and reached the minimum at the concentration of 5 000 ng/ml, and the activity of MPO in PMN reduced by 40% at the concentration of 5 000 ng/ml of anti-sense peptide R4.Conclusion:The results suggested that anti-sense peptide of C5a has significant blocking effects on C5a anaphylatoxin.
3.Acoustic densitometric analysis of phases in normal rabbit liver contrast enhancement using intravenous second harmonic imaging

Objective To investigate phases in normal rabbit liver contrast enhancement with the acoustic densitometry(AD).Methods Ten cases of normal rabbit were examined with second harmonic imaging(SHI).The AD curves and data of parenchyma were compared before and after the portal blood flow was blocked,when the contrast agents were administrated through ear veins.The AD curves and data of parenchyma were also investigated when the contrast agents were injected via ear veins and mesenteric veins.Results The curves of normal rabbit liver parenchyma were single-peak-shaped,looking like "a verticle triangle",when the contrast agent was administrated through ear veins.The curve was divided into 3 sections: ascending,summit,descending,with the value of peak intensity ( 18.4 ? 1.7 )dB.Ascending section indicated the arterial phase,and summit and descending sections the portal phase.The AD curves were quite similar before and after the portal blood flow was blocked,only with the peak value slightly decreased,and the values of peak intensity decreasing by 2 dB.Two peaks were seen in the "Hump-like" AD curves in the parenchyma,with the second peak higher than the first,as the contrast agents were administrated through mesenteric veins again 10-15 seconds after administration via ear veins.Conclusions The arterial phase is short and portal phase is long in normal rabbit liver contrast sonography.The enhancement of parenchyma is caused mostly by contrast from hepatic artery,and the contrast from portal veins only plays a minor role.
4.The preliminary investigation of contrast sonography in lower limb arteries

Objective To investigate the enhancement of color Doppler signals in lower limb arteries by intravenous ultrasound contrast agent.Methods Nineteen cases examined were divided into two groups: Group I consisted of 13 normal cases; group II 6 cases with lower limb artery diseases. The contrast agents were administrated through forearm veins.Results The color Doppler signals and flow width were all increased in normal lower limb arteries after the contrast agent was injected. No color Doppler signals were showed in 4 closed femoral arteries (FA) and popliteal arteries(PA) after enhancement of contrast agent, but the color flow were seen in 3 anterior tibial arteries (ATA) and 1 posterior tibial artery (PTA), which came from lateral vessels in the near end of ATA and PTA. In another 2 cases, a few color Doppler signals were showed in ATA and PTA before contrast admitted, but the color Doppler signals were increased and local color defect were presented after contrast admitted, in which the stenosis of arteries were confirmed by angiography.Conclusions Intravenous ultrasound contrast agent could give rise color Doppler signals in lower limb arteries, and help to make lower limb artery disease diagnosis to a more accurate extent.
5.Influence of basic fibroblast growth factor on proliferation and differentiation of growth plate chondrocytes

BACKGROUND: The proliferative capacity of in vitro cultured chondrocytes is gradually reduced following several passages. Many studies have demonstrated that basic fibrublast growth factor (bFGF) can promote chondrocyte proliferation, and influence its phenotype and differentiation. OBJECTIVE: To observe the influence of bFGF on the proliferation and differentiation of growth plate chondrocytes and to screen the best dosage for in vitro culture of chondrocytes. DESIGN, TIME AND SETTING: For observation and comparison of cells, the experiment was performed at Tissue Engineering Laboratory, Department of Biochemistry, Jinan University Medical School between October 2004 and February 2005. MATERIALS: Twelve New Zealand rabbits were selected for chondrocyte isolation; bFGF was purchased from PeproTech, UK. METHODS: Rabbit growth plate chondrocytes were isolated and cultured in low serum. Different doses of bFGF (0, 1, 2.5, 5, 10, 25, 50 and 100 μg/L) were added. MAIN OUTCOME MEASURES: Cell proliferative multiple was determined using modified MTT method; chondrocyte collagen content was determined using hydroxyproline method; alkaline phosphatase activities were detected by enzyme kinetics. RESULTS: bFGF (5-100 μg/L) could promote chondrocyte proliferation, especially at dose of 25 μg/L. bFGF at dose of larger than 25 μg/L would inhibit collagen production; bFGF at dose of larger than 1 μg/L would inhibit alkaline phosphatase activities. CONCLUSION: bFGF can stimulate growth plate chondrocyte proliferation, and inhibit growth plate chondrocyte differentiation at dose25 μg/L.
6.Evaluation on angiogenesis of breast carcinoma and precancerous lesion with power Doppler imaging and contrast enhanced microvascular imaging

Objective To investigate the angiogenesis characteristics of normal breast,simple hyperplasia,atypical hyperplasia,intraductal carcinoma in situ and invasive ductal carcinoma.Methods Twenty cases in normal controls,20 cases with simple hyperplasia,20 cases with atypical hyperplasia,20 cases with intraductal carcinoma in situ and 30 cases with invasive ductal carcinoma were inspected with power Doppler imaging(PDI)and contrast enhanced microvascular imaging(MVI)on the quantity,configuration and distribution of angiogenesis.The microvessel density(MVD)were detected by immunohistochemical technique.ResultsWith the progression of breast cancer,MVD and grades of PDI kept increasing.Meanwhile,the blood vessels became twisted and increased in quantity with expansion into the lesion,the configuration of blood vessels became disordered.However,the changes mentioned above were not obvious in the very early stages.they became significant from the atypical hyperplasia(AH)stage.Conclusions Contrast MVI and PDI may be useful to screen high risk population with high risk of breast cancer due to their detection of changes in angiogenesis of breast carcinoma and precancerous lesions.
7.Status quo of elder abuse and its countermeasures in nursing

The article reviews the current status of elder abuse from the perspective of epidemiology, characteristics of abuse, risk factors and nursing assessment and management, presenting the idea that nurses are playing a major role in the prevention,detection and resolution of elder abuse. It also introduces that nurse can screen out the situation of elder abuse timely by applying the proper assessment tool flexible and respond to the occurrence of elder abuse as well as avoid the elder abuse again by comprehensive application of nursing knowledge and management skills. Some advices about future research and mismanagement in elder abuse being given.
8.Effect of Liu's Massage Technique on Teenagers with Cervical Vertebra Disease

Objective To observe the effect of Liu's massage technique on teenagers with cervical vertebra disease.Methods 65 teenagers with cervical vertebra disease were divided into the trial group (n=35) and control group (n=30) treated with routine physical therapy and cervical vertebra disease gymnastics, but cases in the trial group were added with treatment of Liu's massage technique. All cases of two groups were evaluated with Clinical Assessment Scale for Cervical Spondylosis (CASCS) before and after treatment.Results After treatment, the CASCS scores of all cases improved significantly ( P<0.01), but the effect of the trial group was significantly superior to that of the control group ( P<0.01).Conclusion Liu's massage technique is effective on cervical vertebra disease of teenagers.
9.The screening of C5a antagonist by optic biosensor technique

Objective:To screen the antagonist of C5a anaphylatoxin,and further to study the dinetic characteristics of interaction between C5a and its anti-sense peptides.Methods:The anti-sense peptides were screened that can interact with C5a selectively by means of dinetic analysis,via biosensor technique.Results:There is one piece of anti-sense peptide(R4) being screened from these four synthesized peptides.The dissociation equilibrium constant(K D) between R4 and the immobilized hC5a is 6.62?10 -6 mol/L and the K D between R4 and L2 is 7.02?10 -7 . Conclusion:Based on the results obtained,it was concluded that the optic biosensor is a ideal and powerful tool to facilitate the kinetic analysis of interaction between sense peptide and its anti-sense peptide and to screen antagonist of biologic activity molecule.
10.Mammary dysplasia and abnormal estrogen/progesterone or their receptor expression induced by psychological stress in pregnant rats

Objective To study the effects of psychological stress on breast development and abnormal levels of related hormones or their receptors in pregnant rats .Methods Pregnant Wistar rats were randomly divided into control group and ex-perimental group.The experimental group rats were given unpredictable stress stimuli for 15 days, including noise, restraint, re-versal of day and night , swimming in cold water and tail pinch-induced pain.Gross changes of the breast were observed , the di-ameter and height of breast nipple was measured, and breast coefficient was calculated.DNA and RNA levels in the breast tis-sues were determined by optical density measurement.The estrogen (E2), progesterone (P), growth hormone (GH) and prolac-tin (PRL) levels in plasma and breast tissues were determined by radioimmunoassay.Bmax and Kd of estradiol and progesterone receptors were measured by radolig and bindng assay .Morphological changes of the breast tissues were examined by light micros-copy using HE staining.Results Compared with the control group, the nipple diameter and height, breast weight and breast co-efficient were significantly decreased in the experimental group (P<0.05, P<0.01), the levels of DNA, RNA and RNA/DNA ratio in the breast tissue were significantly decreased (P<0.05, P<0.01), the levels of plasma E2, P and GH were significant-ly decreased (P<0.05, P<0.01), the levels of E2 and GH in the breast tissue homogenate were decreased also (P<0.05, P<0.01), the Bmaxs of E2 receptor and P receptor were decreased, but their Kds increased significantly (P<0.05, P<0.01), and the number of lobules and diameter of acini were significantly decreased (P<0.05).Conclusions Psychological stress can lead to breast dysplasia and abnormal levels of estrogen and progesterone and their receptors in pregnant rats .
