1.Evaluation of axillary lymph node metastasis of breast cancer via an ultrasonic multi parameter regression model

Objective To evaluate conventional ultrasound combined with real-time elastography in differential diagnosis of metastatic axillary lymph nodes of breast cancer with a logistic regression model.Methods Conventional ultrasound and real-time elastography were performed in 112 breast cancer patients with 113 axillary lymph nodes.All the cases were confirmed with pathological examinations after surgery. A binary logistic regression model based on ultrasonographic features was developed.A receiver operator characteristic ( ROC) curve was constructed to assess the performance of the model.Results There were 28 nodes with no metastasis and 85 with metastasis in a total of 113 axillary lymph nodes.Three ultrasonographic features including the elastic strain ratio(SR), elastic score and shape were finally selected into the logistic regression model .The correct rate of the logistic regression model for predicting axillary lymph node metastasis was 93.8%, and the area under ROC curve was 0.962.Conclusion The binary classification logistic regression model has a good diagnostic efficacy in the diagnosis of benign and malignant axillary lymph nodes,while real-time elastography can improve the accuracy of conventional ultrasound in the diagnosis of axillary lymph nodes.
2.Establishment of a double-antibody sandwich ELISA for the detection of shiga toxin typeⅡin shiga toxin-producing Escherichia coli infection

Objective To establish a double-antibody sandwich ELISA for the rapid detection of shiga toxin typeⅡ ( StxⅡ) in shiga toxin-producing Escherichia coli ( STEC) infection. Methods A pool of murine hybridomas was used to screen out the optimal antibody pair for the establishment of double-anti-body sandwich ELISA. The established ELISA system was used to detect StxⅡin the culture supernatants of 16 clinical strains of STEC. Specificity and sensitivity of the established ELISA system were also evaluated. Results Two antibodies, S2D8 and S2C6, were successfully screened out, based on which the double-anti-body sandwich ELISA was set up. StxⅡand its variants rather than StxⅠwas detected in the culture super-natants of STEC with a lowest detection limit of 4 ng/ml. Its performance was consistent with that of commer-cial colloidal gold test kit, indicating the characteristics of good specificity and sensitivity. Conclusion The S2D8/S2C6-based ELISA laid a foundation for researches which designates the shiga toxin as a potential can-didate on the diagnosis and therapy of STEC infection.
3.Clinical Observation of Comprehensive Chinese Medicine Therapy for Salvage Treatment of Sudden Deafness

Objective To evaluate the effect on salvage treatment of sudden deafness with comprehensive Chinese medicine therapy. Methods Fifty-three sudden deafness patients failing in conventional western medicine treatment were given comprehensive Chinese medicine therapy, and the salvage treatment followed by oral use of Chinese medicine, intravenous injection of Chinese patent medicine, acupoint injection, ear acupoint plastering, acupoint application, acupuncture, sound healing, and massage. The therapeutic effect and the relief of major accompanying symptoms were evaluated after treatment, and the relationship of disease course with clinical efficacy was also investigated. Results (1) After treatment, 4 cases were cured, 9 were markedly effective, 13 were effective, 27 were ineffective, and the total effective rate was 49.06%. (2) Except for the 2 excluded cases, the major accompanying symptoms such as tinnitus, auricular distress, and vertigo were relieved to various degrees in other cases. ( 3) Twenty-two patients had a course of disease within 14 days, and the total effective rate for them was 72.73%. A total effective rate of 32.26% was achieved in 31 cases with disease course over 14 days. It is indicated that the therapeutic effect for patients with a disease course less than 14 days was superior to that with a disease course over 14 days (P<0.01). Conclusion Comprehensive Chinese medicine therapy exerts inspiring therapeutic effect on salvage treatment of sudden deafness, and earlier intervention results into better effect.

5. Progresses of targeted ultrasound contrast imaging in diagnosis and treatment of breast cancer

Breast cancer is one of the common malignant tumors in women, of which the pathogenesis is still unclear. Early diagnosis and treatment can improve the prognosis of patients with breast cancer. With the development of ultrasound molecular imaging technologies, targeted ultrasound molecular imaging technology provides a new method for diagnosis and treatment of breast cancer. The application progresses of targeted ultrasound contrast imaging in diagnosis and treatment of breast cancer were reviewed in this article.
6.Chinese Medicine Monomers Regulate Autophagy of Cervical Cancer Cells: A Review

Autophagy is a lysosome-mediated catabolic process that captures and degrades dysfunctional organelles and useless proteins during cellular stress process, which plays a dual role in cervical cancer. In the early stage of cervical cancer, autophagy inhibits the occurrence and development of cervical cancer by prohibiting the accumulation of oncogenic p62 protein. In the advanced stage of cervical cancer, inhibition of autophagy of cancer cells enhances the sensitivity of cancer cells to chemotherapeutic drugs, thus inhibiting their proliferation. In recent years, the research on Chinese medicine monomers regulating autophagy in the treatment of cervical cancer has attracted extensive attention from scholars at home and abroad. Chinese medicine monomers regulate the autophagy of cervical cancer cells through multiple pathways and multiple targets, so as to increase the apoptosis rate and reduce the resistance of cancer cells to chemotherapeutic drugs. Therefore, this paper reviewed the mechanism of Chinese medicine monomers in inhibiting cervical cancer through autophagy, expecting to find new breakthroughs in the discovery and development of preventive and therapeutic drugs for cervical cancer. By reviewing the literature, it was found that in the early stage of cervical cancer, Chinese medicine monomers activated autophagy to promote apoptosis of cancer cells, and the main mechanism was to increase lysosomal membrane permeability and chemotherapeutic sensitivity and activate intact autophagy flow. In the advanced stage of cervical cancer, inhibition of autophagy reduced the sensitivity of cancer cells to chemotherapy drugs by inhibiting the formation of autophagosomes and autolysosomes. The treatment of cervical cancer by Chinese medicine monomers regulating autophagy has achieved certain effect, but there are few clinical experimental studies and lack of reliable clinical theoretical basis. Therefore, it is essential to carry out more clinical experimental studies on Chinese medicine monomers regulating autophagy to treat cervical cancer, thus finding more reliable theoretical basis for the treatment of tumors.
7.Effect of Huposan Against Endometriosis: A Review

Endometriosis(EMs) is a common chronic inflammatory gynecological disease,affecting about 5%-10% of women of childbearing age worldwide,and has always been a major challenge in clinical treatment. Huposan, derived from the Experiential Prescriptions for Universal Relief (《普济本事方》), has the effects of moving qi, activating blood,expelling blood stasis, and relieving pain. It is often used to treat EMs clinically and has achieved good curative effect. The relevant studies on the treatment of EMs by Huposan were retrieved from databases, such as CNKI,PubMed,Wanfang Data, and VIP for summarizing the mechanisms of action and clinical application of Huposan in the treatment of EMs, aiming to provide ideas and references for the basic research and clinical application of Huposan. As revealed by basic experiments,Huposan could exert therapeutic effects on EMs through resisting cell adhesion by reducing intercellular adhesion molecule 1(ICAM-1)content,decreasing concentrations of matrix metalloproteinase(MMP)-2 and MMP-9 against ectopic endometrial invasion,inhibiting vascular endothelial growth factor(VEGF)expression for antiangiogenesis,inhibiting the expression of tumor necrosis factor-α(TNF-α), interleukin (IL)-6, and IL-1,reversing helper T cell(Th)1/Th2 balance shifts to regulate the body's immune mechanism,and reducing the serum levels of nitric oxide(NO)and nitric oxide synthase(NOS). In clinic practice,Huposan has the effects of reducing ectopic lesions,relieving pain symptoms,reducing serum carbohydrate antigen 125(CA125)content,improving hormone levels,regulating endocrine and immune factors,and reducing postoperative recurrence rate. Huposan plays a therapeutic role in EMs through multiple targets and mechanisms,which is worthy of further exploration and clinical promotion.
