1.Application of G-CSF in the transplantation of stem cell

Granulocyte colony-stimulating factor(G-CSF) is a specific hematopoietic regulating growth factor of granulocyte lineage.It can be used to treat different kinds of granulocytopenia.Recently a variety of basic and clinical researches reported that G-CSF can mobilize bone marrow stem cells and haemopoietic stem cells in the peripheral blood,which suggested a potential aproach of releasing、enriching、mobolizing、promoting migration and inducing cell differentiation in the stem cell transplantation.The recent application of G-CSF in stem cell transplantation is reviewed in this view.
2.The characteristics of speech,phone and cognition development of children with mild hearing loss and the effect of early intervention

Objective To explore the characteristics of speech,phone and cognition development of infants and toddlers with mild hearing loss and to study the effect of early intervention.Methods Speech and cognition abilities of 59 children with mild hearing loss,aged 8 months to 2 years and 10 months old were assessed and 34 normal hearing children,aged 8 months to 2 years and 9 months old were compared.Eighteen children with hearing loss were fitted with hearing aids and received speech therapy for 6 months.The participants' speech and cognition development were evaluated with Gesell developmental scale.The phone test was adopted to assess the speech sound development in 59 children with mild hearing loss.Results Of the 59 children with mild hearing loss,37 were judged with normal speech development quotients(DQ),8 with abnormal speech DQ,14 suspect of abnormal speech DQ.The hearing loss in cases with abnormal speech DQ was more serious than that in cases with normal speech DQ(P<0.05).The speech DQ and personal-social behavior DQ of children with mild hearing loss were significantly lower than normal children(P<0.01).The speech DQ and personal-social DQ on Gesell developmental scales were significantly higher in 18 children received early intervention,compared with those who did not receive intervention (P<0.01).Six cases with mild hearing loss demonstrated phone errors,but no abnormal manifestations in tone.After early intervention the phone of those with mild hearing loss improved significantly.No significant difference in gross motor,fine motor and adaptive ability was observed between the children with mild hearing loss and those with normal hearing(P>0.05).Conclusion The speech ability of children with mild hearing loss is significantly lower than those with normal hearing.Some mild hearing loss children show phone errors.Mild hearing loss affect the adaptation and communication abilities of mild hearing loss children significantly.Early intervention can significantly improve the speech,phone,adaptation and communication abilities of children with mild hearing loss.
3.Radiosensitivity enhancement of recombimant adenoviral-mediated retinoblastoma 94 gene in human esophageal carcinoma cells

Objective To study the radiosensitivity enhancement effects of recombinant adenovirus mediated retinoblastoma 94 gene on the growth of esophageal carcinoma cells EC109.Methods EC109cells was transfected with recombinant Rb94 gene adenovirus and irradiated by 137Cs γ-rays.The cohorts were divided into groups as blank control,Ad-LacZ,Ad-Rb94,radiation and Ad-Rb94 combined with radiation.Cell inhibition ratio,cell cycle and expression of retinoblastoma protein of EC109 cells were analyzed.Results The growth of EC109 cells transfected with Ad-Rb94,radiation and Ad-Rb94 combined with radiation group was all inhibited.The group of Ad-Rb94 combined with radiation resulted in greater inhibition of cells growth compared with Ad-Rb94 group and radiation group ( F =23.31,P <0.05 ).Cells of G2 phases of EC109 cancinoma cells for Ad-Rb94 combined with radiation group were the highest,which was 50%.The combination of Ad-Rb94 and radiation group resulted in the greatest expression of retinoblastoma protein,which reached 71%,significantly higher than Ad-Rb94 infection and radiation groups ( x2 =8.31,6.73,P < 0.05 ).Conclusions Retinoblastoma 94 gene combined with ionizing radiation can enhance the radiation sensitivity of EC109 cells.

5.Detection of serum hyper sensitive C-reactive protein level in patients with acute ischemic stroke and its clinical significances

Objective To explore the level of hypersensitive C-reactive protein (hs-CRP)in the patients with acute ischemic stroke(AIS)and its clinical significance.Methods The BNⅡhighly sensitive automatical scattering turbidimetric immunity analy-zer was adopted to detect serum hs-CRP level in 64 cases of AIS and contemporaneous 32 individuals with healthy physical examina-tion as the control group.The data of AIS cases were retrospectively analyzed.Results The positive rate of hs-CRP was 53.3% in the AIS group and 9% in the control group with statistically significant difference between them(P <0.05);the overall levels of hs-CRP in the AIS group was in inconformity to the normal distribution,the median was 4.26mg/L,which in the control group was (0.99 ± 0.72 )mg/L,there was statistically significant difference between them (P < 0.05 );the average level of hs-CRP was (5.03±1.51)mg/L in the AIS male group and (2.79 ± 1.09)mg/L in the AIS female group,the difference between them had sta-tostical significance(P <0.05).Conclusion The serum hs-CRP level in the AIS patients is higher than that in the healthy crowd with statistical difference(P <0.05).The age and sex may be the causes influencing serum level of hs-CRP in the AIS patients.The average age in the AIS patients with positive hs-CRP is higher than that with negative hs-CRP.Middle-aged and elderly people with hypertension and diabetes are much more dangerous than healthy people in getting AIS.
6.Relationship between distribution of infected snails and transmission of acute schistosomiasis in hilly regions

Objective To explore the relationship between the distribution of infected snails and transmission of acute schistosomiasis in hilly regions.Methods The data concerning the distribution of the infected snails and acute schistosomiasis in Shitai County,Anhui Province from 1999 to 2008 were collected and analyzed.Results The sehistosome infection rate of human increased as the distance between the settings with infected snails and activity sites of humans shortened.Conclusions Acute infection of schistosome of human is associated with the distance between the settings with infected snails and activity sites of them.Strengthening the measures of snail control in key regions,protecting key populations and carrying out health education for schistosomiasis control are important approaches to control the transmission of acute schistosomiasis in hilly regions.
7.Synergic effect of human IL-21 gene transfer combined with γ-ray irradiation on the growth of cervical carcinoma HeLa cells

Objective To study the combined effect of interleukin-21 gene transfer and ionizing radiation on the growth of cervical carcinoma HeLa cells.Methods Previously constructed Ad-IL-21 gene was amplified by infecting 293A cells and the titer was measured by TCID50 method. HeLa cells were transfected with Ad-1L-21 and then irradiated with 6 Gy 137Cs γ-rays.The cells were divided into 5 groups,including blank control,Ad-LaeZ group,Ad-IL-21 group,radiation group and Ad-IL-21 combined with radiation group (combination group).The cell growth,cell cycle,apoptosis,and the expressions of IL-21 gene and protein in HeLa cells were detected.Results Ad-IL-21 was successfully amplified and the titer of Ad-11.-21 was 9 × 1010 pfu/ml.Compared with Ad-IL-21 group and radiation group,the cell growth of combination group was significantly inhibited at 96 h after transfection ( F =85.26,72.98,P < 0.05 ).The cells in combination group were arrested in G1 phase and decreased at S phase( F =36.69,34.83,P < 0.05),while the cellular apoptosis increased markedly ( F =28.23,25.57,P < O.05 ). The gene expression of 1L-21 in the combination group was 1.54- and 2.43-fold of Ad-IL-21 group and blank control group,respectively (F=22.31,36.65, P < 0.05 ), while the protein expression of IL-21 in the combination group was 1.62-fold and 2.31-fold of Ad-IL-21 group and blank control group,respectively ( F =27.36,35.86,P < 0.05 ).Conclusions Ad-IL-21 gene transfection combined with radiation has synergic effect on the inhibition of cervical carcinoma cell growth.
8.Correlation between the central macular thickness and the visual function in patients with macular edema

Objective To determine the correlation between central macular thickness (CMT) and the visual function in patients iwht macular edema (ME). Methods The clinical data of 42 eyes of 40 patients with ME which were examined by optical coherence tomography (OCT) and microperimetry (MP-1) wereretrospectively analyzed. In 40 patients (42 eyes), diabetic ME (DME) was in 27 eyes,branch retinal vein occlusion was in 11eyes, and central retinal vein occlusion was in 4 eyes. All of the eyes had undergone OCT,MP-1 and best-corrected visual acuity (BCVA) test. Central macular thickness (CMT) was measured by fast macular scans using OCT. Retinal sensitivity (MS) and fixation patterns were evaluated by Mp-1.The position was chosen :2 disc diameters (DD) temporal to the disc and one third of a DD inferior to the centre of the disc. Results The correlation between CMT and BCVA is not significant (r=-0. 429, P=0. 069) as well as the correlation between CMT and MS (r=-0. 433,P=0. 058). The difference of CMT between the unstable and stable group was significant (F = 3. 262, P = 0. 039). The difference of CMT between the central fixation group and preferred retinal locus (PRL) group was significant (F=3. 173,P=0. 044). Conclusions BCVA and MS have no significant correlation with CMT. When CMT increases, the fixation stability decreases, fixation location, changes, and PRL occurs.
9.Surveillance of influenza viruses attacking children in Beijing during 2009 pandemic influenza A(H1N1)

Objective To investigate the prevalence of influenza virus infections in infants and young children during the pandemic period of 2009 influenza A(H1N1)in Beijing.Methods Throat swabs were collected from children visited the affiliated Children's Hospital to Capital Institute of Pediatrics for influenza-like illness from June 1,2009 to February 28,2010.The specific gene segments of 2009 pandemic influenza H1N1 and seasonal influenza viruses were amplified from samples by real-time RT-PCR recommended by WHO and National Influenza Reference Center of China.Results Out of 4363 clinical samples tested by real-time RT-PCR,the total positive rate of influenza A viruses was 29.3%,including 623(14.3%)identified as 2009 pandemic influenza A(H1N1)and 657(15.1%)influenza A viruses without subtype identity.Among those pandemic influenza H1N1 positive,23 were severe cases with 5 deaths.The ages for 618 pandemic influenza H1N1 infected children with completed information were from 14 days to 16 years.The ratio of male to female wag 1.3:1.Among them,25.2% were patients in age group of 1 to 3 years old and distribution of children in age groups of 3 to 6 years old and 6 to 12 years old were similar(about 30.0%).During the survey period,it appeared only one prevalence wave of pandemic influenza H1N1.The positive rate of pandemic H1N1 increased in September and the peak(36.5%of positive rate)was in November and then declined to 2.7%in February 2010.The data from routine influenza virus surveillance from 20-30 clinical samples collected each week indicated an alternative prevalence of seasonal H3N2,pandemic H1N1 and influenza B during this study period.Respiratory syncytial virus(RSV)became predominant in children after the circulating of pandemic H1N1.Conclusion There was an epidemic of pandemic influenza H1N1 in children in Beijing from June 2009 to February 2010,especially in those of preschool and school aged children.Seasonal influenza viruses and pandemic influenza H1N1 were contributed alternatively.
10.Artificial radial head replacement for the treatment of comminuted fractures of the radial head.

OBJECTIVETo assess the clinical effects of artificial radial head prosthesis replacement for the treatment of comminuted fracture of the radial head.
METHODSFrom June 2011 to June 2015, 25 patients with radial head comminuted fracture were treated with artificial radial head replacement, including 10 males and 15 females, ranging in age form 24 to 61 years old(mean, 40 years old). The functional recovery of the patients suffering from limb and elbow in different periods, the activity degree of the elbow joint and the function of the elbow in the latest follow-up were compared.
RESULTSAll the patients were followed up, and the duration ranged from 12 to 48 months, averaged 26 months. There were no complications such as infection, elbow instability, subluxation of the distal radioulnar joint, and myositis ossificans. The VAS, Broberg and Morrey elbow function score were improved 6, 9 months after operation compared with that 3 months after operation(<0.05). There were significant differences in elbow flexion and extension, rotation activity between injured side and healthy side 3, 6, 9 months after operation(<0.01), but no significant differences between injured side and healthy side at the latest follow-up(>0.05). At the latest follow-up, according to Broberg and Morrey elbow function evaluation criteria, 16 cases got an excellent result, 7 good and 2 poor.
CONCLUSIONSIt can maximize the recovery of elbow joint stability and quicken early functional exercise, prevent and reduce the occurrence of complications by using the artificial radial head replacement therapy to repair comminuted fracture of the radial head. The short-term curative effect is satisfactory, but the long-term effect needs further observation.
