3.Effects of filter reuse on solute clearance and safety in on-line hemodiafiltration

Objective To investigate the effects of filter reuse on solute clearances, safety and oxidative stress parameters in on-line hemodiafiltration(HDF) . Methods 8 stable maintained uremic patients were treated by post-dilution on-line HDF with first-use or reuse F60 polysulfone filter, respectively. Both blood-side and dialysate-side solute clearances at 20 minute of HDF or during the whole session were measured. Whole blood interleukin-1?(IL-1?) production was monitored before HDF, after HDF, and at 20 minutes of HDF at both venous and arterial lines. Plasma before and after treatment and dialysate were collected for measuring total ascorbic acid, dehydroascorbic acid and total vitamin E. Restults No difference of small molecular substance clearances was observed in blood and dialysate side, while filter reuse group had a significantly higher dialysate side clearance and a significantly lower absorption clearance for ?2-microglobulin. In contrast with stable vitamin E concentration during dialysis, total ascorbic acid level decreased after treatment, with reuse group further inducing a reduced ration of dehydroascorbic acid to total ascorbic acid. No significant changes were found in whole blood IL-1? production within and between each group, so did intra-dialysis symptoms and temperature curves. Reuse filter also caused more albumin loss through high-flux membrane than that of first-use filter. Conclusions Although reuse filter can maintain both small and large molecular weight substance clearance, it increases albumin loss through high-flux membrane. Reuse filter does not stimulate white blood cell to produce more cytokine than the first-use filter, but it increases oxidative stress, and may harm uremic patients in a long run.

Recently,the progress in employing transplanting stem cells to cure injured retina is very fast and has been continuously yielding exciting results.Various sources are used in the studies,including retina-derived cells such as M?ller cells and ciliary body cells,and non-retina-derived cells such as embryonic stem cells and brain-derived stem cells.This review briefly discusses the recent progress of these studies.
5.Clinical value of fractional exhaled nitric oxide level in predicting bronchial hyperreactivity in asthmatic children

Objective To evaluate the clinical value of fractional exhaled nitric oxide (FeNO) level in asthmatic children to predict bronchial hyperresponsiveness by analyzing the correlation between fraction of exhaled nitric oxide and bronchial provocation test.Methods One hundred and fourteen asthma outpatients of Shengjing Hospital were enrolled, FeNO levels, spirometry and bronchial provocation test were measured.Results In the bronchial provocation test,there were 33 positive and 81 negative cases.The positive group had a significantly higher FeNO levels than the negative ones(19.0 × 10-9 vs.16.0 × 10-9 ,p =0.000).By the ROC curve, the best FeNO cut-off value to predict bronchial hyperresponsiveness was 38.5 × 10-9 with high specificity (92.6%) but relatively low sensitivity (36.4%).There was no relationship between methacholine provocative dose causing a 20% fall in FEV1 (PD20-FEV1) and the level of FeNO.Conclusion FeNO level has important predicting value for bronchial hyperreactivity in children with asthma.The level of FeNO > 38.5 × 10-9 has high predictive value in asthmatic children with bronchial hyperreactivity.

8.Improving isoprene production by engineered heterologous mevalonate pathway in Escherichia coli.

Isoprene is an important precursor of synthetic rubber material. In our previous study, metabolic engineered Escherichia coli strain (BW-01) was constructed and used to produce isoprene. Based on the theory of protein budget, using synthetic biology strategies including the increased copy number of genes and rare codons, we regulated the expression of key enzyme to improve isoprene production in Escherichia coli strain. Under shake-flask conditions, isoprene productivity of the engineered strain (BW-07) increased by 73% compared with BW-01, reached 761.1 mg/L. It provides a reference for further studies.

9.Application of intelligent simulator (SimMan) in comprehensive skill training of doctors and nurses

Objective To examine the application effect of intelligent simulator(SimMan) in comprehensive skill training among doctors and nurses.Methods Totally 168 doctors and nurses were chosen as the training object,6 people (4 doctors and 2 nurses) formed a work team.Comprehensive training course was in accordance with 2010 American Heart Association guidelines for cardiopulmonary resuscitation and emergency cardiovascular care science.Real clinical situation was designed by using the third generation SimMan of Norway laerdal company.Trainees were administered two tests before and after training,in which there were two parts containing specialized operation ability and non-technical ability.Overall qualification rate,average mark and treatment time of qualified teams were compared before and after the training.SPSS 16.0 software was used to analyze the data and P <0.05 signifies statistically significance.Results Overall qualification rate,average mark and treatment time of qualified teams were 71.43%,(84.70 ± 3.15),and (449.25 ± 77.0) s respectively before training while 100%,(93.55 ± 3.86) and (323.71 ± 42.26) s after training.There were significant differences in results of two groups before and after training (P =0.002,0.000,0.000).Conclusions Application of the third generation intelligent simulator (SimMan) in comprehensive training will obviously improve training effectiveness and cultivate the non-technical ability.
10.Expression of PRR11 and its effect on proliferation of human osteosarcoma cells

Objective To explore the expression of PRR11(Proline-rich protein 11) in human osteosarcoma and investigate the effect of PRR11 on the proliferation of human osteosarcoma cells.Methods Immunohistochemical staining was applied to detect the PRR11 expression in 75 cases of osteosarcoma and corresponding normal tissues.Western blotting was used to examine PRR11 protein expression levels in osteosarcoma cell lines.We used siRNA to knock down the expression of PRR11 and tested the effects of PRR11 down-regulation on the proliferation in SaOS2 cells.Results PRR11 was overexpressed in osteosarcoma specimens compared to their paired normal tissues,the over expression rate of PRR11 in osteosarcoma and corresponding paracancerous tissues were 76%(57/75) and 9.33%(7/75) with statistical difference(P<0.05).The high expression of PRR11 was correlated with tumor pathological grade and lymphatic metastasis(P<0.05).PRR11 was expressed in 4 osteosarcoma cell lines which were SaOS2,143B,U2OS and MG63 respectively,the expression was highest in SaOS2 cells.Silencing PRR11 inhibited cell growth as compared with control cells(P<0.05).Conclusion PRR11 is overexpression in human osteosarcoma and promotes its progression by enhancing proliferation.The increased expression of PRR11 in osteosarcoma is a new target for treatment and early diagnosis of human osteosarcoma patients.
