Objective: To investigate the morphology and growth kinetics of the cells derived from the adult normal and herniated nucleus pulposus tissues. Methods: The cells derived from the adult normal and herniated nucleus pulposus were cultured in vitro. Passage 1,3,5 cells were chosen for light microscope and electron microscope observation. The cell growth rate, MTT value, maximal growth density, cellular mitotic index, cloning efficiency, and other parameters were observed in normal and herniated nucleus pulposus and the results were compared. Results: Compared with adult normal nucleus pulposus cells, the cells isolated from herniated nucleus pulposus were irregularly arranged and lost polarization, with rough endoplasmic reticulum slightly dilated. Under the same cellular density and culture conditions, the growth rate of passage 1 cells from adult normal and herinated nucleus pulposus had no significant difference, but the differences of passage 3 cells(P<0.01) and passage 5 cells (P<0.05) were statistically significant. The maximal growth density, MTT value, cellular mitotic index, and cloning efficiency of passage 1 cells of adult normal and herinated nucleus pulposus were respectively 7.28±0.23 and 7.41±0.17, 0.186±0.015 and 0.173±0.008, (25.36±2.14)‰ and (23.13±2.55)‰, and (52.4±2.8)% and (47.7±2.6)% (all P>0.05); those of passage 3 cells were respectively 6.96±0.19 and 5.48±0.22, 0.154±0.018 and 0.110±0.009, (22.52±2.16)‰ and (16.68±2.35)‰, and (48.7±3.3)% and (40.7±2.5)%; and of passage 5 cells were respectively 2.42±0.13 and 0.98±0.14, 0.085±0.006 and 0.050±0.007, (15.61±1.96)‰ and (9.34±2.12)‰, and (40.2±2.3)% and (28.34±2.9)%. There were significant differences between the parameters of passage 3 and 5 cells(P<0.05). Conclusion: The adult normal and herinated nucleus pulposus cells are different in cellular morphology when cultured in vitro. The continuous proliferation ability of the herniated nucleus pulposus cells is obviously decreased than that of the adult normal control group, and aging is more easily seen in the herniated ones.

Objective To explore the clinical efficacy and safety of closed negative pressure drainage(VSD) in the treatment of elderly patients with abdominal trauma and infection.Methods 82 elderly patients with abdominal trauma and infection were selected into clinical research,they were divided into two groups according to the treatment time number.41 cases of odd numbers were in the control group,they were given conventional dressing change and antibiotic therapy.41 cases of double numbers were in the observation group,they were given VSD therapy and antibiotic therapy.The clinical efficacy and safety after treatment was evaluated.Results The daily volume of the observation group [(48.67 ± 3.87) mL] was significantly increased compared with the control group,the dressing times [(8.59 ±1.04) times],hospitalization time [(18.33 ± 1.50) d] were significantly reduced compared with the control group,the differences were statistically significant(t =8.56,7.88,6.74,all P ＜ 0.05).The VAS score after treatment of the observation group was (3.38 ± 0.51)points,which was significantly lower than those before treatment and of the control group,the difference was statistically significant (t =8.06,7.56,all P ＜ 0.05).The infection control rate of the observation group was 92.68％,which was higher than that of control group,and the incidence of adverse reactions of the observation group was 4.88％,which was lower than that of the control group,the differences were statistically significant(x2 =6.87,7.69,all P ＜ 0.05).Conclusion The effect of vacuum sealing drainage technique in the treatment of elderly abdominal trauma and wound infection is significantly,it can effectively control the infection,reduce the number of dressing,relieve the pain of patients,and has the advantages of simple operation,high safety,which is suitable for promotion and application.

OBJECTIVE:To guarantee the reliability of cold chain drug storage device and the quality of the drugs in the hospi-tal. METHODS:The establishment and effects of drug cold chain monitoring platform in our hospital were introduced as well as re-lated improvement and measures. RESULTS:The cold chain monitoring platform was established in our hospital through equipping with related equipment, constructing platform network, determining monitoring point, setting early warning condition, etc. All-weather and real-time monitoring of temperature and humidity were realized for different districts of the hospital and different cold chain equipments. The platform could provide the information about operating condition of equipment,and complete drug re-frigeration temperature record;equipment failure could be dissolved in time based on early warning message. Through analyzing rea-sons of early warning message,the unqualified equipment was updated in time,which further improve the system and procedure of cold chain monitoring platform. Within 2 years after the implementation of the platform,53 sets of special refrigerator for drug were updated. Monthly early warning message and the rate of refrigeration equipment early warning decreased from 6869 and 100% in Sept. 2014 at the early stage of short message warning function to 1083 and 40% in Feb. 2016,respectively. CONCLU-SIONS:The establishment of drug cold chain monitoring platform guarantees the reliability of cold drug storage condition so as to ensure the quality of drug in the hospital.

Objective To discuss perioperative rehabilitative care for flexor tendon injuries by examples of successful treatment of 30 cases of rupture of flexor tendon pollicis longus at ZoneⅡ.Methods From March 2001 to March 2006,30 cases of rupture of flexor tendon pollicis longus at ZoneⅡwere treated in our department by suture with double loops and crossing six strands before external fixation.Patients were encouraged to do rehabilitation exer- cises for functional recovery under the guidance and supervision of orthopaedists.Results All the patients were followed up for 6 to 16 weeks (mean,8 weeks).According to TAM (Total Active Motion) criteria,12 cases were rated as excellent,10 cases as good,6 cases as fair,and 2 cases as poor.The excellent to good rate was 73.3%.The 22 excellent and good cases could perform daily-life activities with affected fingers,such as writing,holding chopsticks and doing and undoing buttons.No patient experienced rerupture.Conclusion In perioperative rehabilitative care for rupture of flexor tendon pollicis longus at ZoneⅡ,it is important to have correct diagnosis,noninvasive and secure suture,safe external fixation and functional exercises under the guidance of orthopaedists.

During the latest researches on knee ostarthritis,it is discovered that before the morphology changes of cartilage,there were changes of the content of proteoglycan and water and changes of alinement of surface layer collagen fibers in cartilage matrix.By the new technologies of DWI,proton density mapping image,23Na MRI,T2Mapping image and so on,not only the pathological changes of cartilage can be disclosed,the component changes of cartilage matrix can also be detected.Accompany with the progress in the technology of software and hardware,MRI will have more and more prospects in early diagnosis and therapeutic effect evaluation of knee ostarthritis.
