1.Investigation on bacteriology and drug resistance in infectious disease in intensive care unit

Objective To investigate the bacterial spectrum and drug resistance of gram-negative bacteria infection in intensive care unit(ICU).Methods A retrospective analysis was conducted on flora isolates from Jan 2003 to Dec 2003 to investigate the bacterial spectrum,bacterial resistance and clinical characteristics.Results The leading causative microorganisms isolated were Acinetobacter spp (22.1%),Pseudomonas aeruginosa(20.1%),and E.coli (16.9%). Gram-negative bacteria exhibited the lowest resistance rates to imipenem/cilastati and Amikacin in all antibiotics currently used.However Pseudomonas aeruginosa had the highest resistance rate to imipenem/cilastatin in all the gram-negative bacteria (12.1%).The respiratory tract was the most common compromised site in ICU.Conclusions The causative bacteria in infection in ICU have a higher resistance to antibiotics currently used including cephalosporins of third-generation.It is critical to adopt specific antibiotics based on bacteriology and susceptibility to prevent the development of drug resistance.
2.Effects of Recombinant Human Growth Hormone on Pathophysiology of Critically Ill Patients

Objective To investigate the role of recombinant human growth hormone (rhGH) in the treatment of patients with multiple organ dysfunction syndrome (MODS). Methods Thirty eight patients with MODS routinely treated with antibiotics and nutrition support were divided into two groups: the rhGH group and control group. The rhGH group was treated by subcutaneous injection of 5 U rhGH for two weeks. Results On the 7th day of treatment, the score of APACHE Ⅱ in the rhGH group was much higher than the control group, the levels of ALT, AST, BUN and Cre did not change much compared with the control group. 4.Differentiation of bone marrow mesenchymal stem cells induced by continuous cell culture

[Abstract ] Objective Bone marrow mesenchymal stem cells(BMSCs) can be induced to the differentiation into vascular smooth muscle cells in many induction conditions.We sought to explore the possibility of the differentiation of mesenchymal stem cells into vascular smooth muscle cells by continuous cell culture in vitro. Methods Rat BMSCs were isolated from the bilateral tibial and femoral bones by the method of whole bone marrow adherence, followed by ex vivo expansion.BMSCs were identified by flow cytometry and three-lineage differentiation.After continuous five days'cell culture of BMSCs, the specific surface antigens of VSMCs were detec-ted by immunofluorescence, western blot and real-time PCR. Results BMSCs expressed CD29、90, in contrast, they did not express CD45、34、49d.After induction of osteogenesis, adipogenesis and chondrogenesis, alizarin red、oil red and alcian blue staining pro-duced a strong reaction in cells.The expressions ofα-SMA、Calponin1、SM-MHC and SM22 in the cells of experimental group were no-tably increased, which indicated that BMSCs were differentiating towards VSMCs. Conclusion In the absence of exogenous stimula-tion, BMSCs can be successfully induced to differentiate into VSMCs by continuous cell culture.
5.Analysis for serological diagnostic results of syphilis among entry personnel in Beijing port during 2012 -2014

Objective:To analyze the results of syphilis specific serological tests among entry personnel in Beijing port during year 201 2 -201 4 in order to provide evidence for the prevention and control of syphilis.Methods:Demographics data and syphilis specific test results of entry personnel were obtained from the information system of Beijing International Travel Healthcare Center and input into Excel 201 6. Database for entry personnel was built up accordingly and was analyzed by IBMSPSS22.0.Results:The number of entry personnel in Beijing port during year 201 2 -201 4 was 1 06 302 person-time.According to the evaluation criteria,71 308 persons were recruited and analyzed,of whom 277 were tested serologi-cally positive and the total prevalence was 0.39% (95% CI:0.34% -0.43%).The prevalence was highest in the persons aged 40 -49 years (0.81 %)and was higher in males (0.50%)than in females (0.23%).The persons from Africa held higher prevalence (1 .27%)than from other five regions.As multivariate logical regression analysis indicated that the risk of syphilis infection was 1 0.38 times more in the persons aged 40 -49 years than in the persons <20 years (95%CI:5.59 -1 9.28),and 1 .85 times more in males than in females (95%CI:1 .40 -2.44).For the people from Africa,South America and Asia,the infection risks were respectively 4.89 times (95%CI:2.97 -8.06),4.82 times (95%CI:2.39 -9.74)and 1 .72 times (95%CI:1 .1 5 -2.59)more than that for the people from North Ameri-ca.Rising trend of the prevalence was observed in both genders by age groups (P value for trend χ2 <0.05).Except for Asia,the prevalences of the people from other five regions were rising slightly by years,however there were no obvious trends (P value for trend χ2 >0.05).Twenty-seven seropositive cases were found within 6 246 entry persons who were tested repeatedly during the research period,9 of which were seroconversion cases so that the cumulative conversion rate was 0.1 4% (9 /6 228).The average conversion time was (6.3 ±3.0)months.There was no statistical significance between the cu-mulative seroconversion rates of the persons with different genders and nationalities (P of Fisher’s exact test >0.05).Conclusion:Serological detection of syphilis for entry personnel in Beijing port and syphi-lis-related health education should be reinforced to reduce the risk of disease importing and spreading, protect susceptible subjects and promote health.
6.Hair roots induction and culture of Withania somnifera and its withanolide A synthesis.

Withanolide A is a biologically active secondary metabolite occuring in roots and leaves of Withania somnifera. In the present study, adventitious roots from leaf explants of W. somnifera were induced for the production of withanolide-A by Agrobacterium tumefaciens strain C58C1 to obtain hair roots. Hair roots induction rate reached 30%. The withanolide A was determined by HPLC in different hair roots lines and different parts of W. somnifera. The average content of withanolide A in all hair roots lines were 1.96 times as high as that in wild-plant, the concentration of withanolide A in hair roots (1.783 mg x g(-1) dry weight) were 1.51 times as high as the roots of wild W. somnifera (1.180 mg x g(-1) dry weight), respectively. It is possible to obtain withanolide A from hair roots culture of W. somnifera.

7.PEI-PMMA cationic nanoparticles as carriers for gene transfer.

AIMTo investigate the properties of cationic nanoparticles composed of poly (ethyleneimine)-g-poly (methyl methacrylate) (PEI-PMMA) as gene delivery carriers and explore the mechanism of PEI-PMMA nanoparticles mediated gene transfer.
METHODSPEI-PMMA nanoparticles were synthesized by free radical polymerization. The morphology of nanoparticles was observed by scanning electron microscopy. The particle size and zeta potential were measured by zeta sizer. The complex between pGL3 plasmid and nanoparticles was analyzed by gel electrophoresis; and PEI-PMMA nanoparticles mediated gene transfer into HeLa cells was observed by confocal laser scanning microscopy.
RESULTSPEI-PMMA nanoparticles are spherical shape and monodispersity. The particle size and zeta potential are 172 nm and +50.3 mV, respectively. When pGL3 plasmid complexed with nanoparticles at N/P ratio of 5: 1 and 20: 1, the particle size of pGL3/nanoparticle complex are 133 and 139 nm and zeta potential is + 21.4 and + 33.7 mV, respectively. pGL3 plasmid complexed with nanoparticles completely at N/P ratio of 5: 1. PEI-PMMA nanoparticles can deliver pGL3 plasmid into HeLa cells by endocytosis and release pGL3 into the cytosol.
CONCLUSIONPEI-PMMA nanoparticles effectively transferred DNA to target cells and it is a promising non-viral carrier for gene delivery.

OBJECTIVETo investigate the effects of different concentrations of lipopolysaccharide (LPS), tumor necrosis factor α (TNFα), interleukin-6 (IL-6), dexamethasone (Dex), and insulin on the mRNA and protein expressions of GPR54 in the MCF7 cell line in vitro.
METHODSMCF7 breasr cancer cells were cultured and treated with different concentrations of LPS (10 and 20 µg/ml), TNFα (20 and 100 ng/ml), IL-6 (10 and 20 ng/ml), Dex (10(-6) and 10(-7) mol/L), and insulin (0.01 and 0.1 IU/L). Those treated with culture fluid only served as controls. The mRNA and protein expressions of GPR54 were measured by real-time PCR and Western blot, respectively, after 6, 24, 48, and 72 hours of treatment.
RESULTSCompared with the blank con- trol, LPS (10 and 20 µg/ml), TNFα (20 and 100 ng/ml), IL-6 (10 and 20 ng/ml), Dex (10(-6) and 10(-7) mol/L), and insulin (0.01 and 0.1 IU/L) significantly increased the expressions of GPR54 mRNA (P < 0.05) and protein (P < 0.05).
CONCLUSIONLPS, TNFα, IL-6, Dex, and insulin evidently increase the expression of GPR54 in the MCF7 cell line, indicating their influence on the function of gonads by regulating the GPR54 level.

9."Outcomes in patients with ""successful"" recanalization in anterior circulation stroke treated with mechanical thrombectomy: TICI 3 versus TICI 2b"

ObjectiveTo investigate the difference in the outcomes in patients with acute anterior circulation stroke having achieved the Thrombolysis in Cerebral Infarction (TICI) 2b and TICI 3 recanalization after mechanical thrombectomy.MethodsThe patients with acute anterior circulation stroke having achieved successful recanalization after mechanical thrombectomy were enrolled retrospectively.They were divided into TICI 2b (almost complete recanalization) group and TICI 3 (complete recanalization) group.The good outcome was defined as the modified Rankin Scale score 0-2 at 3 months after onset.ResultsA total of 83 patients were enrolled in the study, including 38 patients (45.8%) with TICI 2b and 45 (54.2%) with TICI 3;49 (59.0%) had good outcome, and 34 (40.9%) had poor outcome.The good outcome rate in the TICI 3 group was significantly higher than that in the TICI 2b group (68.9% vs.47.4%;χ2=3.946, P=0.047).After adjusting for age, hypertension, diabetes, baseline systolic blood pressure, triglyceride, intravenous thrombolysis, and ASITN/SIR collateral grades, TICI 3 was an independent predictor for good outcome at 3 months after onset (odds ratio [OR] 3.759, 95% confidence interval [CI] 1.098-12.871;P=0.035), while the higher baseline National Institutes of Health Stroke Scale score (OR 0.820, 95% CI 0.715-0.941;P=0.005) and higher fasting glucose (OR 0.610, 95% CI 0.410-0.906;P=0.014) were the independent predictors for poor outcome at 3 months after onset.ConclusionsThere are difference in the outcomes in patients with successful recanalization in anterior circulation stroke treated with mechanical thrombectomy.The outcome is substantially better in TICI 3 than TICI 2b patients.
10.Asiatic acid induces apoptosis in T98 G human glioblastoma cells by changing autophagy

Aim To investigate the effect of asiatic acid on apoptosis and autophagy in human glioblastoma T98G cells. Methods MTT colorimetry was employed to assay the cellular proliferating activity. The fluores-cence microscope and Hoechst 33258 staining were used to detect the morphological changes. The cell ap-optosis and autophagy were analyzed by flow cytometry with Annexin-V/7-AAD and MDC staining respective-ly. The expressions of associated proteins were detected by Western blot to analyze the mechanism of apoptosis and autophagy. Results MTT assay showed that the growth of T 9 8 G cells was inhibited by asiatic acid ( IC50 =46. 3 μmol · L-1 ) . Annexin V/7-AAD stai-ning and Western blot revealed that asiatic acid in-duced apoptosis in T98 G cells by reducing the expres-sion of Akt, decreasing the mitochondrial membrane potential, and increasing the expression of Caspase-3. MDC staining and Western blot showed that the per-centage of MDC-positive cells was decreased and the expressions of Beclin-1 , LC3-II and Atgs were inhibi-ted by asiatic acid treatment. 5 μmol·L-1 chloroquine was used to up-regulate the expressions of LC3-Ⅱand Beclin-1 . Asiatic acid-inhibited autophagy was blocked and the total apoptotic rate was reduced remarkably. Conclusion Asiatic acid suppresses T98 G cells pro-liferation by inducing apoptosis and inhibiting cell au-tophagy, and the very role of inhibiting autophagy could promote apoptosis to a certain extent.
