1.Influences of Chloropazine, Nimodipine and Their Combination on the Toxic Effects of Cadmium in Liver and Kidney of Mice

The influences of the calmodulin antagonist chlorpromazine (CPZ), and calcium chanmel blocker nimodipine (NLMO) and their combination on cadmium (Cd) poisoning of mice were studied.A series of biochemical parameters including urinary enzyme activities, blood and urine Cd levels, metallothionein (MT) contents in liver and kidney, hepatic ultrastructure and Ca2+ -Mg2- AT Pase activitv in erythrocyte membrane were determined. Animal models for Cd poisoning were established by peritoneal injection of 1/5 LD50 CdCl2. The experimental groups were protected by administration of CPZ, NIMO and CPZ and NIMO in combination I h before the injection of CdCl2. Five days later, samples were collected for analysis. The data showed that CPZ could protect kidney tissue against Cd-induced damage, as the urinary y-glutamyl-traspeptidase (γ-GT) and N-acetyl-β-D-glucosaminidase (NAG) activities were reduced significantly. There was neither evidence of the protective effect of NIMO on kidney tissue nor an indication of a synergistic effecf of CPZ and NIMO.Both CPZ and NIMO showed a considerable protective effect against the decrease in Ca2+ -Mg2+ AT-Pase activity, and a synergistic action was observed. Cd content in blood was reduced significantly by CPZ or the combination of CPZ and NIMO, but elevated by NIMO. Both CPZ and NIMO considerably increased MT contents in livers and kidneys and ameliorated damaged to the hepatic ultrastructures caused by Cd. The results indicated that these inhibitors could protect mice against the toxic effects of Cd in liver and kidney tissues, while CPZ was more efficient than NIMO. The combination of CPZ and NIMO excrted a synergistic action. The protective action of these two drugs might be relevent to the function of MT.
2.Staged target teaching method of neural interventional therapy for ischemic cerebrovascular disease

Objective To strengthen the training effect of the neuro-interventional surgeons using staged target teaching method. Methods A total of 39 neurosurgeons from all over the country were trained at the Department of Interventional Radiology,Xuanwu Hospital,Capital Medical University from March 2012 to March 2013. In the one-year training period,the training was divided into 4 stages (each stage for 3 months). All the learning contents were phased to focus on teaching and set the target at each stage,which had a clear purpose. Questionnaires were sent to the refresher doctors at 6 months and 12 months after they returned to their original work in order to obtain the conditions about their independent work after training. Results All the 39 neurosurgeons achieved their desired goal from theory to practical operation. They mastered the holistic treatment principles of ischemic cerebrovascular disease and were familiar with and mastered the whole brain DSA and stenting techniques. At the fourth stage,as an operator, everyone had finished at least 5 extracranial artery stentings. The recovered questionnaires after the end of training showed that 95. 8%(23/24)neurosurgeons were able to perform whole brain DSA independently and held≥5 surgeries monthly;87. 5%(21/24)neurosurgeons could complete stenting independently (1 patient/month). Conclusion The staged target teaching method is a practical and effective teaching means for special technical training of neurointervention.
3.A retrospective study of Schatzker III tibial plateau--fracture using minimally invasive fixation and bone grafting through medial side.

OBJECTIVETo investigate clinical effects of minimally invasive fixation and bone grafting througn medial side for the treatment of Schatzker III tibial plateau fracture.
METHODSFrom April 2009 to August 2011, 18 patients with Schatzker III tibial plateau fracture were treated with minimally invasive fixation and bone grafting through medial side. There were 15 males and 3 females ranging in age from 64 to 73 years, with an average of (69.75 ± 1.22) years. Sixteen patients were caused by falling down, 2 cases were caused by traffic accident. Operative time and length of incision were be recorded. Clinical and radiological follow-up was performed after operation. Hospital for Special Surgery (HSS) score and Kellgren-Lawrence score were used to evaluate clinical effects.
RESULTSThe mean operative time was (45.32 ± 1.58) min, and the mean length of incision was (5.21 ± 0.65) cm. Postoperative X-ray showed excellent reduction. Eighteen patients were followed up for 10 to 13 months with an average of (11.5 ± 1.35) months. The mean HSS score was 86.51 ± 2.71, 12 cases got excellent results,4 good and 2 fair. Three patients were developed mild osteoarthritis according to the Kellgren-Lawrence system.
CONCLUSIONMinimally invasive fixation and bone grafting through medial side, not only could reduce surgical invasive, but also guarantee early function activities. It has advantages of keeping well after reduction. So it has the favorable future in clinic.

4.Acupuncture with Methadone for heroin withdrawal syndrome: a meta-analysis of randomized controlled trials

Objective:To assess the efficacy of acupuncture plus Methadone in treating heroin withdrawal syndrome.Methods:According to the requirements of evidence-based medicine, the inclusion criteria, exclusion criteria and retrieval strategy were set for original documents. Randomized controlled trials (RCTs) involving acupuncture plus Methadone in treating heroin withdrawal syndrome were retrieved from China National Knowledge Infrastructure Database (CNKI), Wanfang Academic Journal Full-text Database (Wanfang), Chinese Biomedical Literature Database (CBM), Chongqing VIP Database (CQVIP), PubMed, Cochrane Library and EMBASE. According to the Cochrane Handbook for Systematic Reviews of Interventions, each included trial was assessed strictly including risk of bias and quality evaluation. Meta-analysis and descriptive-analysis were conducted using the RevMan 5.3 software.Results:A total of 8 trials involving 931 patients were included. Meta-analysis showed that there was no statistical significance [RR=1.05; 95%CI (0.99, 1.11);P=0.11] in comparing total effective rate between the acupuncture plus Methadone and Methadone alone; the cure rate of acupuncture plus Methadone was higher than that of Methadone alone [RR=1.45; 95%CI (1.19, 1.78);P=0.0003]; the relapse rate of the acupuncture plus Methadone was lower [RR=0.691; 95%CI (0.60, 0.80);P<0.00001]. On the basis of these results, the method recommended by the Grading of Recommendations Assessment, Development and Evaluation (GRADE) was taken to evaluate the quality of the evidence. It showed that the level of the evidence was low and it was weakly recommended.Conclusion:According to the included trials, acupuncture plus Methadone in treating heroin withdrawal syndrome is more effective and better than Methadone alone, but it still needs more high-quality, large sample, and polycentric researches to confirm it.
5.Percutaneous vertebroplasty for symptomatic vertebral hemangiomas

Objective To assess the outcome of percutaneous vertebroplasty for symptomatic vertebral hemangiomas. Methods Five cases with 7 symptomatic vertebral hemangiomas were treated with percutaneous vertebroplasty. Aggressive lesions were treated with absolute alcohol injection in addition. Patients were followed-up and clinical manifestations were observed and CT, MRI and X-ray plain film were compared between before and after vertebroplasty. Results Procedures were successful without complications. Most of the symptoms resolved within 24 hours after vertebraplasty. All patients were followed-up for 12~50 months and free of neurological deficits and symptoms. Imaging follow-up showed no vertebral collapse, nor recurrance of hemangiomas. Conclusion With effective long-term follow-up and quick elimination of symptoms, precutaneous vertebroplasty, added with absolute alcohol injection in aggressive cases, proves to be a safe and effective treatment for symptomatic vertebral hemangiomas.
6.Mechanisms of protection effect of bcl-2 gene transfection on heat-stressed cardiomyocytes.

AIMTo study the mechanisms of protection of bcl-2 gene transfection against heat-stressed cardiomyocytes.
METHODSCardiomyocytes were isolated and cultured. bcl-2 was transfected into cardiomyocytes with Lipofectamine transfection methods. The cardiomyocytes were stressed by heat. The change of H+ -ATPase synthesis activity of cardiomyocytes mitochondria caused by bcl-2 transfection was measured by chemical radiation method. The changes of Caspase 3 activity of cardiomyocytes caused by bcl-2 transfection was measured by fluorometric analysis.
RESULTSbcl-2 transfection could increase the H+ -ATPase synthesis activity of cardiomyocytes mitochondria under heat stress at 41 degrees C and 43 degrees C and could decrease the Caspase 3 activity of cardiomyocytes under heat stress at 41 degrees C and 43 degrees C.
CONCLUSIONThe protection effect of bcl-2 transfection on heat-stressed cardiomyocytes may be associated with preserved H+ ATPase synthesis activity of cardiomyocytes mitochondria and the activity of Caspase 3 of cardiomyocytes.

7.Pharmacokinetics of levofloxacin-methane sulfonic acid in patients with pulmonary tuberculosis

OBJECTIVE To study the pharmacokinetics of levofloxacin-methane sulfonic acid (LF-MSA)in 8 patients with pulmonary tuberculosis after administration 400 mg orally.METHODS The concentrations of LF-MSA in serum were measured by HPLC.The pharmacokinetic paramters were obtained using 3P97 program.RESULTS The results showed that a one-compartment model with 1 st order absorption were fitted for LF-MSA.The average values of tmax was 1.22 h.The concentrations of LF-SA in serum was (4.52±0.72) mg*L-1.The mininal concentrations of LF-AS at 24 hr after a single dose was (0.52±0.26) mg*L-1.The average area under the curve was (48.35±5.38) mg*h*L-1.The elimination half-life was (6.49±1.70) h.The apparent volume of distribution was (77.48±8.33)L.CONCLUSION According to our study,it was suggested that 400 mg twice daily may be given in order to maintain the concentrations of LF-SA in serum around 24 hours above the minimal concentrations of bactericide.
8.Progress of discovery of molecular glues from natural products and their derivatives

Molecular glues are a class of small molecules that induce the formation of protein-protein interactions to confer new biological function or therapeutic effects. As a unique pharmacological modality, molecular glues could target proteins without druggable binding pockets. It exhibits a variety of functions, including regulating signal transduction, stabilization or degradation of targeted proteins, through sticking different proteins together. This review will summarize the development and current status of molecular glues derived from natural products and analogs by illustrating the discovery and interaction mechanism. We hope to present a systematic view, provide valuable clues for researchers and encourage them to explore more efficient and rational molecular glue discovery strategies.
9.Characteristics of neoplasma in 140 127 adults undergoing health check-up

ObjectiveRetrospectively investigate the characteristics of neoplasma of health checkup participants to find ways to improve cancer detection rate.Methods A total of 186 confirmed cancer patients were enrolled in this study.Personal information,including age,gender,andlocation were collected.The contribution of clinical symptoms,physical examination and laboratory test to cancer detection was investigated.ResultsBiopsy confirmed cancers accounted for 0.133%( 186/140 127 ).Lung (0.036% ),liver ( 0.017% ),kidney ( 0.014% ),colorectal ( 0.013% ) and gastric cancer ( 0.011% )were the most commonly found neoplasma.Detection rate of liver cancer in male was significantly higher than that in female (x2 =6.181,P<0.05 ).Asymptomatic cancer was found in 158 adults (84.9% ).Ultrasound showed a significantly higher sensitivity to liver,kidney and thyroid cancer than tumor biomarkers.The detection of lung cancer was improved with the combination of X ray,tumor marker( TM ),clinical symptom and low-dose chest CT scan.In 2008,the detection rate for digest system carcinoma (0.083% ) and lung cancer (x2 =8.538,P<0.05) was significantly increased (x2 =11.792,P<0.05).ConclusionsHealth check-up plays an important role in early detection of cancer.The combined use of physical examination,chest X ray,ultrasound test,TM and CT may improve the detection of cancer.
10.Polarographic catalytic wave of clarithromycin and its applications.

AIMTo develop a new method for the determination of clarithromycin.
METHODSThe catalytic wave of clarithromycin in the presence of K2S2O8 was used for improving the analytical sensitivity. The rapid determination of clarithromycin has been carried out by linear single sweep polarography.
RESULTSThe reduction wave of clarithromycin appeared at ca. -0.79 V (vs SCE) in 0.24 mol x L(-1) KH2PO4-Na2HPO4 (pH 6.81) supporting electrolyte, which was ascribed to the reduction of carbonyl group on C-9 position. In the presence of 0.01 mol x L(-1) K2S2O8, the reduction wave was catalyzed to produce a parallel catalytic wave. The peak current of the catalytic wave was ca. Twenty times higher than that of the corresponding reduction wave. Based on the catalytic wave, a new method for the determination of clarithromycin has been proposed. The peak current of the catalytic wave was rectilinear to clarithromycin concentration in the range of 4.0 x 10(-7)-5.0 x 10(-5) mol x L(-1). The detection limit was 2.0 x 10(-7) mol x L(-1).
CONCLUSIONThe proposed method could be used for the direct determination of clarithromycin in pharmaceuticals and urine without preliminary separation.
