Objective To investigate the effect of radiation on the mitochondrion in adenocarcinoma A549 cells. Methods After A549 cells were irradiated with 0,0.5,3 or 8 Gy of 60Co γrays, mitochondrion membrane potential of A549 cells was detected by JC-1 probe, and ATP activity was measured by ATP kit in a chemiluminescence apparatus. The mitochondria DNA copy numbers was detected by real-time PCR assay. Results At 24 h after radiation, the mitochondrion membrane potential of A549 cells in all the irradiated groups changed significantly (F =243.44,P < 0.05),among which 0.5 or 3 Gy of radiation resulted in a significant increase of mitochondrion membrane potential of A549 cells (t =-10.12,-5.59,P < 0.05).However,the mitochondrion membrane potential of A549 cells exposed to 8 Gy of radiation decreased significantly 24 h after radiation (t =15.22,P < 0.05).The mitochondrion membrane potential of A549 cells in all radiation groups returned to the normal level 48 h after radiation (F =10.36,P < 0.05).24 h after radiation, the level of ATP in A549 cells significantly changed respectively(F =97.08,P < 0.05),similar to the mitochondrion membrane potential. The ATP level in 0.5 and 3 Gy groups increased significantly (t =1.66,7.27,P < 0.05),and the level of ATP in 8 Gy group decreased significantly (t =-8.24,P < 0.05).Furthermore,48 h after both 0.5 and 3 Gy of radiation, the ATP content in A549 cells was still higher than that in untreated A549 cells (t =4.60,8.53,P <0.05).The mitochondria DNA copy numbers in A549 cells increased significantly in all the radiation groups (F =118.00,P < 0.05).Compared with untreated A549 cells the mitochondria DNA copy numbers in A549 cells increased at 0.5 Gy by 12 times(t =0.02,P <0.05),and increased at 3 and 8 Gy by 7 and 10 times, respectively (t =9.68,15.10,P < 0.05).Conclusions High dose of radiation resulted in the decrease of mitochondrion membrane potential of A549 cells, which subsequently affected the production of ATP.However, radiation with moderate and lower dose could lead to the compensatory increase of mitochondrion membrane potential of A549 cells, which promoted the production of ATP.The mitochondria DNA copy numbers compensatory would increase after A549 cells were exposed to radiation within 8 Gy.

369 patients with hypertension living in Xietu community were randomized into 2 groups:intervention group receiving the group-interference-management and control group receiving routine hypertension management services. We found that patients in the intervention group had better self-management behaviors,self-efficacy and health status in comparison with the control group (P<0.05).The systolic and diastolic blood pressure in the intervention group decreased more remarkably than that in the control group (P<0.05).Our result suggested that the community-based hypertension group-interference-management model is more effective to control hypertension than the routine hypertension management services.

Chinese Journal of Experimental Ophthalmology 2011;29(11):983-986
Background Curcumin can eliminate free radical arising in the system and superoxide anion and further inhibit lipid peroxidation.Researches proved that curcumin has resisting effect on oxidative damage of lens, but its effect on the formation of age-related cataract is under clear. Objective This study was to investigate the effect of curcumin on the cataract induced by sodium selenite. Methods Thirty healthy clean SD rats of 12-day-old were selected. All the rats were randomly divided into the normal control group, model group and curcumin group and ten rats for every group. The selenite cataract models were established in the rats of model group and curcumin group by the injection of sodium selenite subcutaneously.Meanwhile,0.005 % curcumin lavaged the rats in the curcumin group daily for two weeks.Lens of the rats were examined under the slit lamp at 4,7,10,14 days after injection in all groups, and the opacification of lens was graded based on the He' criteria. The lenses of rats were obtained at the end of experiment and then the level of superoxide dismutase (SOD),the activities of malondialdehyde (MDA) and glutathione peroxidase(GSH-Px) in rat lens were detected using biochemical assay method. The use of experimental animals followed the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission. Results The lenses were transparent throughout the experimental duration in the blank control group.Compared with model group, the development of the Ⅲ,Ⅳ,Ⅴ grade of cataract was slow down in the curcumin group with the significant difference( P<0.05 ).The content of MDA,SOD and GSH-Px activity in lens appeared considerable differences among 3 groups ( MDA:F =215.42,P<0.01 ; SOD:F =46.83,P< 0.01 ; GSH-Px:F=44.29,P<0.01 ).The level of SOD was significantly reduced in the model group and the curcumin group compered with blank control group ( P < 0.05 ) and that in the curcumin group was higher than the model group ( P<0.05 ).The MDA level in the model group and the curcumin group was obviously rised in comparison with blank control group( P<0.05 ),but that in the curcumin group lowed in comparison to model group( P<0.05 ).GSH-Px activity in the curcumin group was significantly lower than that in blank control group and higher than that in the model group( P < 0.05 ).Conclusions The curcumin can effectively delay rather than the cataract formation induced by sodium selenite in SD rat by improved the resistance of oxidation in the lens.
