1.Correlation between the polymorphism ofβ2-AR and the labor progress after labor analgesia

Objective To investigate the distribution ofβ2-adrenergic receptor (β2-AR)genetic polymorphisms among the included parturient women and observe the relationship between the genetic polymorphisms and labor progress after labor analgesia.Methods A prospective observational study was conducted from March to June in 2014 at Nanjing Maternity and Child Health Care Hospi-tal.A total of 1 65 nulliparous women were enrolled in the study,and finally 1 52 patients were includ-ed after exclusion for several reasons.Theβ2-AR gene polymorphic variations at nucleotides encoding amino acids 1 6 and 27 were detected by polymerase chain reaction-restriction fragment length poly-morphism technique.And the demographic data,rate of oxytocin usage,length of labor progress and the incidence of cesarean section after labor analgesia were observed and recorded.Results The allele frequencies ofβ2-AR genetic polymorphism in the parturient women were dominantly presented in the form of Arg1 6Arg (AA)and Arg1 6Gly (AG)at 1 6 position and Gln27Gln(CC)at 27 position respec-tively.The demographic and background characteristics of the subjects didn’t present any significant differences among the groups with differentβ2-AR genetic polymorphism.And the differences of labor progress among β2-AR 27 genetic polymorphisms parturient women were not significant.However, the length of the first stage and the total labor progress in the β2-AR Arg1 6Arg homozygous were both significantly longer than that in the other two groups (P <0.05),and the incidence of cesarean section was also significantly higher (P <0.05 ).Furthermore,we also found that β2-AR Arg1 6Arg homozygous was related to cesarean delivery after labor analgesia through multivariate logistic regres-sion analysis (OR 2.87,95%CI 2.69-3.09).Conclusion The labor progress of the parturient women with β2-AR Arg1 6Arg homozygous is relatively slower,which maybe an important risk for the nullip-arous women transforming to cesarean delivery after labor analgesia.
2.Clinical effect analysis of intervention treatment for patients with atherosclerotic renal artery stenosis

Objective To evaluate the clinical outcome and relative factors of intervention treatment for atherosclerotic renal artery stenosis in elderly patients.Methods The clinical data of 79 patients diagnosed as atherosclerotic renal artery stenosis by angiography and treated by revascularization were analyzed.Results There were 55(69.6%)successes and 24(30.4%)failures in decreasing blood pressure and 28(35.4%)successes and 51(64.6%)failures in improving renal function after intervention treatment.Predictors of favorable outcome of intervention treatment in decreasing blood pressure were related to lower urine protein,higher glomerular filtration rate,higher systolic and diastolic blood pressure before treatment,lower resistance index(RI)of renal artery,and no complication of cerebral vascular diseases.Predictors of favorable outcome of intervention treatment in improving renal function were related with percentage of angiographic stenosis,category of antihypertension and lower urine protein.The logistic regression analysis showed that the percentage of angiographic stenosis was the most important predictor of intervention treatment for blood pressure control,age and level of serum creatinine before intervention treatment were the most important predictors of intervention treatment for improving renal faction.Conclusion Percentage of stenosis(≥85%),age(133 ?mol/L)can be used as the predictors of therapeutic success for renovascular stenosis in older patients.
3. Erectile dysfunction is associated with subclinical carotid vascular disease in young men lacking widely-known risk factors

This study aimed to gain insight into the underlying pathogenesis of erectile dysfunction in young men under the age of 40 years without widely-known risk factors. Compared with normal controls, patients with erectile dysfunction had increased carotid intima-media thickness, fasting levels of blood glucose and insulin, and homeostatic model assessment index, as well as lower flow-mediated vasodilation and testosterone levels (P < 0.05), though all of these values were within their respective normal range. Multivariate logistic regression analysis identified carotid intima-media thickness, flow-mediated vasodilation, insulin level, and homeostatic model assessment index as significant predictors of erectile dysfunction. Young men with flow-mediated vasodilation <10.65% were 11.645 times more likely to have erectile dysfunction, young men with carotid intima-media thickness >0.623 mm had a 4.16-fold, and young men with homeostatic model assessment index >1.614 had a 5.993-fold greater risk of having erectile dysfunction. In conclusions, in young men with normal results from general clinical screening, an increased carotid intima-media thickness and homeostatic model assessment index and reduced flow-mediated vasodilation were associated with a higher incidence of erectile dysfunction. Erectile dysfunction may appear before the detection of traditional cardiovascular risk factors and may be the earliest clinical sign of subclinical cardiovascular disease.
4.Induction of experimental Graves' disease in Balb/c mice immunized with human thyrotropin receptor ectodomain amino terminus gene

Objective To study the antigenicity of human thyrotropin receptor(hTSHR)amino terminus (amino acid 29~280)and its association with Graves' disease.Methods Total thyroid RNA was prepared from human normal thyroid tissue.RNA was then reversely transcripted and cDNA was subjected to PCR amplification.PCR product was cloned into pcDNA3.1 and the recombinant plasmid was named pcDNA3.1/hTSHR188~940bp. Balb/c mice were immunized with peDNA3.1/hTSHR188~940bp. The levels of serum thyroxin,anti-TSHR antibody(TRAb)and thyroid stimulating antibody(TSAb)were measured,and the pathological changes of thyroid tissue were also observed.Results A 753 bp fragment encoding hTSHR ectodomain amino end was obtained after PCR amplification.Confirmed by Hind Ⅲ restriction enzyme digestion and DNA sequencing,pcDNA3.1/hTSHR188~940bphad been constructed successfully,with the correct sequence and direction of hTSHR188~940bp.In the Balb/c mice treated with pcDNA3.1/hTSHR188~940bp,elevated TRAb in week 6(0.148±0.018)were observed compared with those at week o(0.106±0.006,P<0.01),and kept a higher level till week 10(0.134±0.011,P<0.01).T4 and TSAb index values were significantly increased in week 10.Serum T4 concentration increased from(41.02±7.97)μg/L in week 0 to(62.20±12.77)μg/L in week 10(P<0.01);TSAb index values rose from 0.864±0.076 at week 0 to 1.392±0.615(P<0.01).Thyroid pathological examination showed that proliferated thyroid follicular epithelial cells and foll icular eapacity increased.Inflammatory cells were occasionally found.Conclusions There are antigen epitopes in hTSHR ectodomain amino acid 29~280,which can stimulate the production of TSAb.And the latter induces hyperthyroidism and Graves' disease like manifestations.It suggests that hTSHR ectodomain amino acid 29~280 is closely associated with Graves' disease,and maybe one of important etiological factors leading to the disease.
5.Correlation between CXCR4,VEGF and the blood metastasis of osteosarcoma

Background and purpose:The chemokine,CXCL12 and its receptor,CXCR4,have recently been shown to play an important role in the metastasis of several kinds of carcinoma. It also has been demonstrated that VEGF regulates both the expression of CXCR4 and invasiveness in cancer cell. Our aim was to study the expression of CXCR4,VEGF in osteosarcoma and the correlation between these two factors and distant metastasis. Methods: The immunohistochemical staining SP method was used to detect the expression of CXCR4 and VEGF in 56 cases of osteosarcoma. We analyzed the correlation between the expression of CXCR4 and VEGF,and the correlation between the expression of CXCR4,VEGF and clinical stage,the level of ALP. The patients were followed up for 2 years. Results:There was signifi cant correlation between the expression of CXCR4 and VEGF in 56 cases(r=5.678,P=0.02). Univariate analysis showed a signifi cant correlation between the expression of CXCR4,VEGF and clinical tumor stage(P=0.026),and no correlation between the expression of these two factors and age,sex and serum ALP level. 31 cases had metastasis in two years in a total of 56 follow-up cases,and the expression of CXCR4 and VEGF was associate with metastasis(P=0.018 and P=0.022,respectively). Conclusion:VEGF can upregulate tumor angiogenesis and promote tumor metastasis to specific organ by increasing expression of CXCR4.The increasing expression of CXCR4 and VEGF is useful to predict metastasis and prognosis of osteosarcoma.
7.Protective effect of epigallocatechin gallate against sperm abnormality in mice.

OBJECTIVETo investigate the protective effect of epigallocatechin gallate (EGCG) on mouse sperm in vivo.
METHODSA total of 64 six-week-old male Kuming mice were randomly divided into eight groups of equal number to be treated with normal saline (negative control), Cyclophosphamide (CP) at 30 mg/kg (positive control), and CP followed by EGCG (experimental) at 20, 40, and 80 mg/kg, respectively, given every other day for 10 days. At 4 and 5 weeks after treatment, the bilateral testes of the mice were harvested for examination of sperm abnormality.
RESULTSEGCG did not increase the rate of CP-induced sperm abnormality in the mice, but reduced it instead with the prolonged time of treatment.
CONCLUSIONEGCG protects mouse sperm in vivo.

The Qinbai Qingfei concentrated pellets by traditional Chinese medicine theoryand party and group, the rats were given the drugs group, comparison of pharmacokinetics parameters changes of baicalin , discusses the rationality of Qinbai prescription. The rats were gavaged monarch drug group (Huang Qincu extract, mainly forbaicalin), and official medicine group, adjuvant group, medicine group and Qinbai group (Quan Fangzu) the content of baicalin equal as the monarch drug group, in the 28 h collection in rat plasma at different time point, application of HPLC determination of baicalin glycosides in rat plasmaconcentration time curve, with 3P97 practical pharmacokinetics program to process the data Based on the data analysis, baicalin in rat plasma of Qinbai group Cmax is 4 times as big as monarch druggroup, AUC is 6 times as big as monarch drug group; the content of baicalin in plasma of rats the highest is Qinbai group, the minister drug group, adjuvant group, medicine group of baicalin in rat plasma content of less than the Qinbai group, but was significantly higher than that of monarch drug group; the medicine group is slightly higher than that adjuvant the content of baicalin in plasma of rats. The pharmacokinetic results show that the measured plasma concentration in rats that Qinbai can significantly increase Cmax and AUC of baicalin, other components of qinbai can promoted the baicalin absorption in vivo. It showed that the reasonable of Qinbai compound compatibility. The minister drug can promote the absorption of baicalin in vivo.

9.Study on the association between SNP 1425G/A in protein kinase Cη gene and genetic susceptibility of cerebral infarction

Objective To explore the association between 1425G/A single nucleotide polymorphism(SNP)of protein kinase Cη gene(PRKCH)and cerebral infarction in Jiangsu Han population.Methods 255 patients with cerebral infarction and 225 controls were recruited in our case-control study.The 1425G/A in PRKCH gene was detected by direct sequencing of PCR products.Data were coded and entered in SPSS Windows(version 13.0).Results The frequencies of the GA+AA genotypes(56.86%)and A allele (36.27%)in cerebral infarct group were significantly higher than those in control group(44.44%and24.67%.χ2=7.377,P=0.007 and χ2=15.104,P<0.01).Further analysis indicated that the genotypes(63.09%)and alleles(40.27%)frequencies were statistically different between lacunar infarction subtype and controls(44.44%and 24.67%;χ2=11.744,P=0.Ol and χ2=20.445,P<0.01).Logistic regression analysis revealed that hypertension,diabetes mellitus.hyperlipidemia and the A allele of 1425G/A polymorphism were independent risky factors for lacunar infarction.Conclusions The SNP 1425G/A in PRKCH is closely associated with cerebral infarction.particularly with lacunar infarction.
10.Comparison of effects of kaempferide and anhydroicaritin on biomineralization of cultured osteoblasts.

This study is to compare the effects of kaempferide and anhydroicaritin on biomineralization of rat osteoblasts (ROB) in vitro. Calvarias were dissected aseptically from newborn SD rats, the osteoblasts were obtained by enzyme digestion and were cultured in MEM containing 10% FBS. The medium was changed every three days, and serial subculture was performed when cells covered with 90% of the dish. Kaempferide and anhydroicaritin were separately added with final concentrations of 1 x 10(-4), 1 x 10(-5), 1 x 10(-6) and 1 x 10(-7) mol x L(-1) under the conditions of osteogenic differentiation. The proliferation was measured by MTT, and the optimal concentration was detected by the ALP activity at the 9th day after osteogenic induction culture. The osteogenic indexes of kaempferide, anhydroicaritin and control group with the optimal concentration were compared. The result showed that the anhydroicaritin at concentration of 1 x 10(-5) mol x L(-1) had significantly promoted the activity of ALP, calcium content and osteocalcin content, increased the number of CFU-F(ALP) and mineralized nodules, enhanced the mRNA level of BMP-2, OSX and Runx-2, which are key genes of osteogenic differentiation, and raised the protein content of collagen-I. However, the kaempferide group had not significantly represented the ability that promoted osteogenic differentiation of ROB. The difference of osteogenic differentiation on ROB between kaempferide and anhydroicaritin was caused by the prenyl group on C-8 of icariin.
