EGFR-TKI plays an important role in the treatment of non-small-cell lung cancer. However, some researchers find that there are still some patients with primary or acquired resistance to EGFR-TKI. The present known mechanisms of acquired drug resistance finally lead to the re-activation EGFR downstream signa-ling pathways. Liver X receptor agonist has inhibition function to several critical steps of EGFR downstream sig-naling pathways PI3K-Akt-NF-κB,which makes it possible to overcome the drug resistance.
3.Diagnosis and Treatment of 6 Children with Anomalous Origin of the Left Coronary Artery from the Pulmonary Artery

Objective To analyze diagnostic experience of children with anomalous origin of the left coronary artery from the pulmonary artery (ALCAPA) and assess its surgical results.Methods Six children with ALCAPA had been diagnosed in our hospital from Mar. 2006 to Oct.2007 aged from 6 months to 10 years old(median 43.5 months).The medical data included common clinical presentations,the findings of electrocardiogram(ECG),the echocardiogram,ECG-gated 64-slice computed tomography,the digital subtraction angiogram (DSA),surgical records and the outcome.Results Diaphoresis,fantod,breathlessness and difficult feeding were commonly presented in 5 cases ALCAPA and the 5 children presented with left ventricular failure in infancy.The predominant ECG findings were Q waves and ST segment changes in the anterolateral chest leads in 4 cases,ST changes in 1 case,and normal ECG in 1 case.Cross-sectional echocardiography showed dilated left ventricles with poor contractility in 4 cases,medium mitral regurgitation in 2 cases and severe mitral regurgitation in 2 cases,aneurysmal dilatation of the right coronary artery(RCA) in 2 cases.CT and DSA examinations showed characteristic changes in these children.The surgical reestablishment of a two-coronary system was performed in 5 cases,direct reimplantation in 2 cases and creation of an autologous extrapulmonary tunnel in 3 cases.Simultaneous mitral annuloplasty was performed in 2 cases.After operation,the symptoms of 5 cases were all improved and none died.Conclusions Understanding the clinical manifestations and lab findings of ALCAPA is helpful to making an early diagnosis of ALCAPA.Early surgical treatment can improve the patients′ outcome.
4.Effects of Extremely Low Frequency Electromagnetic Fields on Lipid Peroxidation of Myocardium in Rats

5.Research about the relationship of tissue factor, tissue factor pathway inhibitor-1, 2 with cancer metastases and thrombosis

Objective To evaluate the effect of tissue factor (TF), tissue factor pathway inhibitor-1,2 (TFPI-1,2) on cancer metastases and thrombosis complicated with cancer. Methods Blood samples from 292 cancer patients were collected and were divided into different teams according to cancer types,complicated with or without thrombosis; TF, TFPI-1, TFPI-2, plasma concentrations were measured by ELISA;Tissue expression of TF, TFPI-1, TFPI-2 were observed by immunohistochemical method. Results Plasma concentrations of TF and TFPI-1 in all kinds of cancers were higher than the control and lung cancer was the highest; TFPI-2 plasma concentrations had no statistics differences among all these teams. Tissue expression of TF of all kinds of cancer were higher than the adjacent tissues, lung cancer was higher than the other types of cancer. There were no statistics differences for TFPI-1 and TFPI-2. Both TF and TFPI-1 plasma concentrations of cancer with-or without-thrombosis were higher than control. TF was even higher in cancer with thrombosis team, TFPI-1 had no statistic difference between these two teams. TFPI-2 concentrations had no differences among all these teams. Conclusion Many kinds of tumor have higher expression of TF, it is expressed with different intensity according to different types of cancer. TFPI-1 has no clear effect in cancer growing and metastases. Unbalance of TF and TFPI-1 in plasma may relate to high coagulation state of cancer and may accelerate the thrombosis formation in cancer.
6.Helical CT Findings and Misdiagnosis of Pulmonary Thromboembolism

Objective To investigate the diagnostic value of helical CT study in pulmonary thromboembolism.Methods A retrospective analysis was made on 16 misdiagnosed cases,studied previously with plain CT scanning.Results All of the 16 cases had positive findings on CT scanning,with sensitivity 100%.Among them,11 cases were misdiagnosed as pleural effusion plus pneumonia;3 pleural effusion;and 2,each of organized pneumonia and peripheral lung cancer.Conclusion The strong evidence of the disease could be given even with plain CT studies.The procedure is simple,safe and reliable,If a patient is suspected with the disease,and if CT is available in the hospital,CT should be the first choice in the study.
7.The metabolic fingerprint of the compatibility of Radix Aconite and Radix Paeoniae Alba and its effect on CYP450 enzymes.

Using a UPLC-MS/MS (MRM) and cocktail probe substrates method, the metabolic fingerprint of the compatibility of Radix Aconite (RA) and Radix Paeoniae Alba (RPA) and its effect on CYP450 enzymes were investigated. These main CYP isoforms include CYP 1A2, CYP 2C, CYP 2E1, CYP 2D and CYP 3A. Compared with the inhibition effect of RA decoctions on CYP450 isoforms, their co-decoctions of RA and RPA with different proportions can decrease RA' inhibition on CYP3A, CYP2D, CYP2C and CYP1A2, but can not reduce RA' effect on CYP2E1. The metabolic fingerprints of RA decoction and co-decoctions with different proportions of RPA in CYP450 of rat liver were analyzed by UPLC-MS. Compared with the metabolic fingerprints of RA decoction, the intensity of diester-diterpenoid aconitum alkaloids decreased significantly, while the intensity of monoester-diterpenoid alkaloids significantly increased in the metabolic fingerprints of co-decoctions of RA and RPA. The results suggest that RA coadministration with RPA increased the degradation of toxic alkaloid and show the effect of toxicity reducing and efficacy enhancing.

8.Effects of focal mild hypothermia on inflammation after traumatic brain injury in rats

Objective To study the effects and mechanism of focal mild hypothennia on brain inflarnmation response during the early stage of tranrntic brain injury(TBI)in rats.Method Traumatic brain injury wag produced in anesthetized rats by using free-falling impact model.Sixty-three male Sprague Dawley rats were randomly divided into three groups:TBI group,focal hypothermia group,and sham group.each group with 7 SD rats.Then all groups Wag further subdivided into three subgroups according to different intervals after TBI.Myeloperoxiduse (MPO)activity assay,immunohistochemical staining for the detennination of intercellular adhesion molecule-1(ICAM-1)expression and the number of ICAM-1 positive blood vessels were counted up in the traumatic brain tissues.All data were analyzed by SNK-q test Using SPSS 10.0 sofeware program.Results The number of ICAM1 positive blood vessels was significantly fewer in the rats of focal hypothermia group than those in the rats of TBI group at different intervals after TBI(P<0.01).Similarly,the MPO activity was significantly lower in the rats of focal hypothermia group than that in the rats of TBI group(P<0.01).Conclusions Focal mild hypothermia could significantly decrease neutrophil infdtration and ICAM-1 expression in the traumatized areas.It might be one of the mechanisms of focal mild hypothermis for TBI.
