AIM: To immediately observe the complication of corneal flap after LASIK surgery.
METHODS:A retrospective case series were studies. Totally 2 040 cases ( 4 080 eyes ) from January 2010 to October 2012 in our hospital were collected, the corneal flap was observed using lamp microscope after LASIK within 30min. Corneal flap dislocation, corneal flap strial and intraface debris were examined after operation, the effective treatment and controlled measure should be taken for these complications.
RESULTS: Postoperative complications were corneal flap dislocation 102 eyes (2. 5%), corneal flapstriae 95 eyes (2. 33%), interface debris 105 eyes (2.57%). No failure case was seen. There had no corneal flap-related complications, which seriously impact the visual quality after the surgery.
CONCLUSION: Carefully postoperative examination at the first - time is an effective way to manage some complications of post - LASIK. Thus promoting the diagnosis and treatment of post-LASIK complications.

Objective To study the feasibility of preoperative acute hypervolemic hemodilution for decreasing perioperative blood use. Methods Thirty two patients(ASAI-II) undergoing spinal surgery were randomized into AHH group and control group. After anesthesia 6% HES(200/0.5) was given through internal jugular vein for AHH. The blood volume was increased by about 25%. During the operation, blood loss was replaced by HES in equal volume. Blood was transfused when the Hct dropped below 25%. The HR, MAP, CVP, SPO2, Hb, Hct., Plt, FIB, APTT, PT, amount of blood loss, blood transfused and urine output, volume of drainage 24h after operation and VAS score (0 means no effect, 10 means maximal effect) were observed before AHH, at the end of AHH, at the end of operation, and 24h after the operation. ObjectiveTo study the effects of two cryopreservation methods ( direct vapor nitrogen method and program freezing method) on the motility of post-thaw human sperm.MethodsThe 25 semen samples were taken from 25 donors from Shanghai Human Sperm Bank during May to July 2011,using straw as a frozen carrier, the semen samples contained modified glycerol-yolk-citrate ( GYC ) ( 1 ∶1 ) as cryoprotectant,after the liquefaction, each semen sample was divided into 2 groups,and separately cryopreserved by direct vapor nitrogen method and program freezing method.According to the recommended standardoftheWorldHealthOrganization, semensampleswereanalyzedbeforeandafter cryopreserving.Density,total motility,PR,motility parameters ( including VAL,VSL,VCL,BCF) were determinated by the IVOS sperm analyzer.Semen samples were stained by modified papanicolaou stain and 200 sperms of each sample were counted under microscope by manual analysis to calculate the rate of normal morphology sperm.The paired t test was used to compare the above indictors before and after cryopreserving in one method and after thawing between two methods.Results Before cryopreserving,the total motility,PR,VAL,VSL,VCL,BCF were(91.36±4.96)％,(81.28 ±6.41)％,(59.29 ±9.30) μm/s,(49.80±8.40) μm/s, (83.39 ± 15.70)μm/s, (16.68 ± 3.36)Hz respectively, and after cryopreserving by direct vapor nitrogen method the results were (46.04 ± 20.14)％,(40.22 ± 18.42 )％,(48.00± 11.60)μm/s,(41.11 ±8.50) μm/s,(71.55 ±17.19) μm/s,(15.94 ± 14.30) Hz respectively,the differences of the indictors above were observed with statistic significance except BCF ( t =11.324,11.958,4.266,3.964,3.620,0.258,P of BCF ＞0.05,P of others ＜0.05); and after cryopreserving by program freezing method,the results were(48.05 ± 18.27)％,(42.47 ± 15.62)％,(45.30 ± 6.51) μm/s,(38.08 ± 4.99) μm/s,(64.23 ± 11.75) μm/s,(14.00 ± 2.99) Hz respectively,the differences of the indictors above showed statistic significance (t =10.337,10.874,7.154,5.510,6.494,3.244,P ＜0.05).No significant differences were observed in the revival rate,total motility,PR and normal morphology sperm rate by using different methods (t =0.425,0.364,0.622,1.106,P ＞0.05).The results of VAL,VSL,VCL,BCF in direct vapor nitrogen method were higher than that in program freezing method,when the differences of VSL and VCL were observed with statistic significance(t =2.489,2.822,P ＜ 0.05 ).Conclusions Using straw as carrier is practicable.Direct vapor nitrogen method is easier to operate than program freezing method.Post-thaw sperm cryopreseved by direct vapor nitrogen method has higher motility parameters,thus making this method an ideal one for fast freezing.

In order to investigate the effect of deep brain stimulation on diseases such as epilepsy, we developed a closed-loop electrical stimulation system using LabVIEW virtual instrument environment and NI data acquisition card. The system was used to detect electrical signals of epileptic seizures automatically and to generate electrical stimuli. We designed a novel automatic detection algorithm of epileptic seizures by combining three features of field potentials: the amplitude, slope and coastline index. Experimental results of rat epileptic model in the hippocampal region showed that the system was able to detect epileptic seizures with an accuracy rate 91.3% and false rate 8.0%. Furthermore, the on-line high frequency electrical stimuli showed a suppression effect on seizures. In addition, the system was adaptive and flexible with multiple work modes, such as automatic and manual modes. Moreover, the simple time-domain algorithm of seizure detection guaranteed the real-time feature of the system and provided an easy-to-use equipment for the experiment researches of epilepsy control by electrical stimulation.

In total, 607 bedridden patients aged 60 years and over at Tianlin community hospital and four other hospitals or at home-care in Shanghai were investigated with questionnaire, physical examinations and related laboratory tests. Contents of investigation include general situation, medical conditions, 24-hour recall of diet, intake of nutritional supplement preparation, and so on. As compare to the reference for nutrient intake of the elderly set in the Guide to Diet for Chinese Residents (2007), total nutritional intake was obviously insufficient and three nutrients in diet were imbalanced for most bedridden elderly patients. Intake of most nutrients (including proteins, dietary fiber, calcium, iron, zinc, selenium,copper, magnesium, phosphor, retinol equivalent, vitamin B1 , vitamin B2, vitamin C) was obviously low in bedridden elderly patients, with a larger gap from the requirements of the Dietary Guide.

ObjectiveTo investigate the effect of intrathecal injection of TRPV3-siRNA lentivirus on bone cancer pain(BCP) behaviors in rats.Methods40 female SD rats successfully received intrathecal catheter implantation and without motor dysfunction were randomly divided into 4 groups (n=10 in each group):Sham group (S),BCP group (B),negative control lentivirus group (C) and TRPV3-siRNA lentivirus group (T).Group B,C and T were induced bone cancer pain by intra-right-femur inoculation of Wallker 256 cells,while rats in group S were injected of inactivated cell.Rats in group T were intrathecally treated with 5 μl TRPV3-siRNA lentivirus while rats in group C received 5 μl negative lentivirus on 1~6 d after surgery.All the rats received pain behaviors including paw withdrawal thermal latency(PWTL) and paw withdrawal mechanical threshold (PWMT) at 1 d before BCP and 1,3,6,9,12,15,18 and 21 d after BCP.L4~L6 spinal cords were reserved for RT-PCR and Western Blot.ResultsCompared with group S,PWTL and PWMT of group B were decreased (P<0.05).On the 9th day,PWTL of group T was higher than that of group C((17.52±2.15)s vs (14.36±1.67)s,P<0.05).However,there was no significant difference in PWMT between group T and group C((16.89±1.54)g vs (15.53±1.36)g,P>0.05).The results of RT-PCR and Western blot demonstrated that the expression of TPPV3 in group T was decreased compared with that in group C(P<0.05).ConclusionIntrathecal injection of TRPV3-siRNA lentivirus can inhibit the expression of TRPV3 and thus alleviate symptom of PWTL,but not PWMT.

Objective To investigate the expression of phosphorylated protein kinase B1 (pAkt1) and human telomerase reverse transcriptase (hTERT) in ovarian epithelial carcinoma (EOC) and its relationship with prognosis, and to analyze the correlation between pAkt1 and hTERT expression. Methods 92 patients with EOC in Tangshan Gongren Hospital from January 2012 to December 2016 were selected. The expressions of hTERT and pAkt1 proteins were detected by immunohistochemistry. The relationship between the expressions of pAkt1 and hTERT proteins and their relationship with clinical prognosis were analyzed. Results Of the 92 patients with EOC, 68 cases (73.9 %) had positive expression of pAkt and 52 cases (56.5 %) had positive expression of hTERT. There was no significant correlation between expressions of pAkt1 and hTERT proteins in EOC (r= 0.284, P= 0.633). The expressions of pAkt1 and hTERT proteins were not related with age, tumor pathology type and the International Federation of Gynecology and Obstetrics (FIGO) staging (all P> 0.05), but had significant association with tumor pathology differentiation (χ2= 2.694, P= 0.005; χ2=2.284, P=0.018). The disease-free survival of patients with both pAkt1 and hTERT positive was shorter than that of the other groups (P= 0.013). Conclusion The prognosis of EOC patients with high expression of pAkt1 and hTERT proteins is poor.

To study which subgroup of bone marrow derived cells formed myofibroblasts and the mechanism that transforming growth factor β1 (TGFβ1) regulates the formation of bone marrow derived macrophages into myofibroblasts during renal fibrosis.Chimeric mice were generated by lethally irradiation of C57 mice followed by transfusion of green fluorescent protein (GFP) labeled bone marrow cells.Complete marrow reconstitution was developed until 12 weeks after transplantation.The mice were randomly divided into Sham operation group,unilateral ureteral obstruction (UUO) 3 days group,UUO5 days group,UUO7 days group and UUO7 with TGFβ1 treatment group.Each group had four mice.Flow cytometry was used to evaluate cell components.Compared with Sham operation group the proportions of GFP + CD+14 oα-smooth muscle actin(α-SMA) + cells,GFP + CD+44 CD+105 α-SMA+ cells and GFP+ F4/80+ α-SMA+ cells in each UUO group were progressively increased and the transformation rate in UUO7 day group was the highest.The GFP+ F4/80 + α-SMA+ cells accounted for the largest population.TGFβ1 promoted the transformation of bone marrow derived macrophages into myofibroblasts.Compared with Sham operation group or UUO7 day group,the proportion of GFP+ F4/80+ α-SMA+ cells increased in UUO7 day TGFβ1 treatment group.Compared with Sham operation group (or UUO7 days group) the protein expressions of F4/80,oα-SMA,Collagen Ⅰ increased in UUO7 with TGFβ1 group.Bone marrow derived macrophages are considered as the main type of myofibroblast precursors during the development of renal fibrosis.TGFβ1 regulates the transformation of bone marrow-derived macrophages into myofibroblasts.This process contributes to progressive renal fibrosis and deterioration of renal function.

Purpose:To study the effect of FasL gene on human lung adenocarcinoma cell lines and possibility of ex- ogenous FasL gene for gene therapy of lung cancer.Methods:An adenoviral expression vector with full length cDNA of FasL gene insert was constructed(Ad-FasL) and transfected into A549 cells.The effect of exogenous FasL gene on the growth of A549 cells was examined in vitro and in vivo.Results:Expression of FasL gene in A549 cells was confirmed by FCM and RT-PCR.The in vitro growth of the Ad-FasL transfected A549 cells was significantly inhibited (inhibition rate: 84%) as compared to mock (Ad-LacZ) transfected A549 cells.Colony-forming activity in vitro of the Ad-FasL transfected A549 cells was completely inhibited.The Ad-FasL transfected cells became apoptotic which was confirmed by the appear- ance of pre-G_1 on flow cytometry (FCM).The growth of A549 xenografts in nude mice was retarded by intra-tumol injection of Ad-FasL.Conclusions:FasL gene participates in the induction of cell apoptosis.Its use in gene therapy of cancer is promising.
