1.Correlation between polymorphism in the promoter of DNA methyltransferase-3B gene and susceptibility of colorectal cancer

Objective To observe and analyze the correlation between the DNA methyltransferase-3B (DNMT3B) gene single nucleotide polymorphisms (SNP) rs 1569686 and the susceptibility to colorectal cancer (CRC) in Tianjin Han population.Methods A case-control study was performed.89 CRC patients and 94 normal controls were examined,and Sequenom MassArray system was applied to detect the genotype of rs1569686 in DNMT3B gene polymorphisms.The association results were analyzed of the SNP and CRC.Results The frequencies of genotypes TT,GT and GG at SNP rs1569686 were 74.2%,24.7% and 1.1% in CRC patients respectively,which there were no significant differences from that in controls(73.4%,23.4% and 3.2%; P>0.05).Conclusion It suggests that the SNP rs1569686 of DNMT3B gene may not be related to susceptibility of CRC in Tianjin Han population.
2.Effect of 5-HT1A receptors in the hippocampal DG on active avoidance learning in rats.

OBJECTIVETo investigate the effects of serotonin (5-HTIA) receptors in the hippocampal dentate gyrus (DG) on active avoidance learning in rats.
METHODSTotally 36 SD rats were randomly divided into control group, antagonist group and agonist group(n = 12). Active avoidance learning ability of rats was assessed by the shuttle box. The extracellular concentrations of 5-HT in the DG during active avoidance conditioned reflex were measured by microdialysis and high performance liquid chromatography (HPLC) techniques. Then the antagonist (WAY-100635) or agonist (8-OH-DPAT) of the 5-HT1A receptors were microinjected into the DG region, and the active avoidance learning was measured.
RESULTS(1) During the active avoidance learning, the concentration of 5-HT in the hippocampal DG was significantly increased in the extinction but not establishment in the conditioned reflex, which reached 164.90% ± 26.07% (P <0.05) of basal level. (2) The microinjection of WAY-100635 (an antagonist of 5-HT1A receptor) into the DG did not significantly affect the active avoidance learning. (3) The microinjection of 8-OH-DPAT(an agonist of 5-HT1A receptor) into the DG significantly facilitated the establishment process and inhibited the extinction process during active avoidance conditioned reflex.
CONCLUSIONThe data suggest that activation of 5-HT1A receptors in hipocampal DG may facilitate active avoidance learning and memory in rats.

3.Uniform design for in vitro somatic embryogenesis and plant regeneration of Gymnadenia conopsea

Objective Using stem tip of Gymnadenia conopsea in this experiment to investigate somatic embryogenesis and plant regeneration in vitro.Methods Uniform dedign for the most suitable media for embryogenic callus induction and embryogenic cell complex,development of somatic embryo and plant regeneration were screened.Results The results showed that Chaturvedi and Mitra(CM)+6-BA 1.0 mg/L+IAA 2.0 mg/L+2,4-D 0.1 mg/L was fit for embryogenic callus and embryogenic cell complex induction.Percentage was 98%;The medium of development of somatic embryo and plant regeneration was CM+6-BA 1.00 mg/L+IAA 0.10 mg/L.Percentage was 100% and converted into plantlets with shoots and roots after 50 d culture on the same medium.Conclusion The observation of morphostructure and ultrastructure proves the process of somatic embryogenesis of G.conopsea.
4.Study on auditory semantic priming effects in patients with aphasia

Objective To explore the characteristics of semantic priming effects in Chinese words with different association strength in patients with aphasia by auditory stimulation.Method Stimulus-response word pairs with different association strength including strong,moderate,weak,and no association categories were chosen from word association thesaurus as experiment materials.Both patients with aphasia(n=11)and normal subjects (n=16)were requested to finish an auditory lexical decision task for target words.Semantic priming effects were investigated by means of measuring reaction time(RT)and error rate of each word-pair.Results In patients with aphasia and normal subjects,the mean RTs were significantly shorter in strong,moderate and weak association strength words than in no association strength words(patients with aphasia(1270.20±47.70)ms,(1340.50±266.25)ms,(1429.70±317.07)ms vs(1549.00±325.87)ms,P<0.05 and normal subjects(1140.2±274.48)ms,(1196.50±284.06)ms,(1262.10±274.31)ms vs(1391.20±315.68)ms,P<0.05).In strong,moderate,and no association strength words,the mean RTs were no significant differences between two groups.In the weak association strength words,mean RTs were significantly longer in patients with aphasia than in normal subjects((1429.70±317.07)ms vs(1262.10±274.31)ms,P<0.05).In two groups,mean error rates were significantly less in strong,moderate and weak association strength words than in no association strength words(patients with aphasia:7.73±6.07,4.55±7.23,6.82±8.15 vs 14.09±12.41,P<0.05 and normal subjects:3.44±4.37,2.81±3.64,5.31±5.91 vs 10.94±11.14,P<0.05).However,in strong association strength words,mean error rates were significantly higher in patients with aphasia than in normal subjects(7.73±6.07 vs 3.44±4.37,P<0.05).In moderate,weak and no association strength words,there were no significant differences between two groups.Conclusion The patients with aphasia follow gradient of the association strength words like normal subjects and have semantic priming effects in the strong,moderate association strength words.
5.Clinical observation of gemcitabine combined with S -1capsule on patients with advanced esophageal cancer

Objective To evaluate the efficiency and toxicities of Gemcitabine combined with S -1 cap-sule in the treatment of advanced esophageal cancer .Methods Twenty-nine patients with advanced esophageal cancer were treated with S-1 capsule 80 mg/m2 ,twice daily for 14 days and Gemcitabine (1 000 mg/m2 ) was in-travenously administrated on day 1 and 8.The chemotherapy was repeated every 21 days.After two consecutive treatment circles ,the short term efficacy ,adverse effects and follow -up condition were evaluated .Results One case showed complete remission (CR),11cases showed partial remission (PR),9 cases showed stable disease (SD),8 cases showed progressive disease(PD).The responsive rate(CR+PR)was 41.4%;the illness control rate(CR+PR+SD)was 72.4%.The major adverse events were myelosuppression ranging from grade Ⅰto Ⅲ, hand-foot syndrome,disgusting and the damage of liver function .Some patients displayed the damage of liver function.The median time to progress ( mTTP) was 6.9 months.The median 1-year survival rate was 64.3%. Conclusion The combination of Gemcitabine and S -1 capsule is an effective and well -tolerated method for the patients with advanced esophageal cancer .
6.A case of hydroa vacciniforme-like primary cutaneous CD8-positive T-cell lymphoma

39℃)developed at the progressive stage of this disease.Physical examination showed variously sized,round or oval,atrophic and variola-like scars along with scattered erythematous patches,papules, necrosis and crusts on the face and extremities.The face was edematous,and there were some edematous and erythematous plaques with a necrotic center on the legs and arms.Histological examination revealed a massive infiltration with atypical CD8~+lymphocytes around the vessels and appendages in dermis.A diagnosis of CD8~+cutaneous T-cell lymphoma(CTCL)was made.Glucocorticoid and immunosuppressants were effective in controlling the condition.Up to the time of the writing,there has not been any definite evidence of systemic involvement.

In the pathophysiology experiment teaching,by combining the teaching of prob-lem-based learning with local area network(LAN) teaching,students first carry out a simple ex-periment to ask a question on the observed phenomenon,and then put forward a hypothesis,de-sign experiments to answer questions,and implement the experiment,and finally present experi-mental results.Such experimental design teaching is not only a great way to mobilize the stu-dents’interest in scientific research and learning initiative,but also greatly enhances the effi-ciency of the experiment.Students preliminarily master the basic scientific research program and methods.
8.Development and application of universal and specific diagnostic reagents for human respiratory adenovirus

Objective To establish a specific,sensitive,simple human respiratory adenovirus detection method to put a good experimental foundation for developing universal and specific diagnostic reagents of human respiratory adenovirus.Methods The nucleotide sequences of 10 serotype of human respiratory adenovirus were obtained from GenBank.Highly consewed five pairs of universal and specific adenovirus primers were designed on the evaluation of multiple sequence alignment of the 10 full genomic sequences with the software DNAMAN 5.2.2,Gene Runner 3.05,BLAST,and to ensure that the polymerase chain reaction(PCR)products were type-specific.NP-40 sample lytic method was employed to prepare the template.The effectiveness,specificity and sensitivity of primers were evaluated.And PCR was carried out to test 64 samples of throat swabs of acute respiratory infection children.The positive PCR products were sequenced directly to identify the adenovirus serotypes.The positive specimens was inoculated on HeLa cells to observe cytopathie effect(CPE)under light microscope,and virus morphology were observed under electron microscope.Results BLAST results indicated that the five pairs of primers were specific adenovirus primers with low homology to the others.The primers were identified as PCR positive fragments obtained by using five pairs of primers to amplify the human adenovirus type 3 DNA,which did not react to the respiratory syncytial virus and Coxsackie virus.the effectiveness and specificity of the primer were thus indicated.PCR sensitive results showed 10-5 dilutin of adenovirus culture and DNA sample could be deteeted which meant the method was sensitive and stable.Two PCR positive specimens were detected in 64 clinical samples.the positive rate was 3.13%(2/64).Using the two PCR positive specimens to inoculate the HeLa cells,the typical adenovirus CPEs of rounded and aggregated,detatched cells were observed under light microscope.And a large number of adenovirus typical particles with characteristic lattice arrangement were observed in the infected cells under electron microscope.PCR product sequencing results showed that these two isolated adenoviruses were typed in human adenovims group B.Conclusions Universal and specific adenovirus PCR primers with the serotype specificity of the PCR products were designed successfully.The PCR primers was sensitive and specific and could be routinely applied in clinical adenovirus diagnosis for respiratory specimens.
9.Clinical outcomes of glutamine supplementation in neonates

Objective To assess the clinical outcomes of glutamine supplementation in neonates.Methods The study was designed as a double-blind randomized controlled trial.Totally 100 infants were randomly divided into 5 groups with 20 infants in each group:control group:standard parenteral nutrition (PN);PNI group:PN supplemented with 0.3 g/ (kg·d) glutamine;enteral nutrition (EN) 1 group:EN supplemented with 0.3 g/ (kg·d) glutamine;PN2 group:PN supplemented with 0.3 g/ (kg·d) glutamine;and EN2 group:EN supplemented with 0.3 g/ (kg·d) glutamine,The primary endpoints were the time to establish full enteral nutrition [defined as receiving 120 ml/ (kg·d) enterally],episodes of gastric residuals,total duration of PN,and mortality.The second endpoints were weight gain and head circumference gain,liver function,renal function,days on ventilator,and length of hospitalization.Results Time to achieve full enteral nutrition,incidence of gastric residuals,duration of PN,weight gain (g/d) ,head circumference gain (cm/w),liver function,renal function,and length of hospitalization were not significantly different among all five groups.Days of mechanical ventilation were significantly decreased in PN1 group and PN2 group when compared with the control group (P < 0.05).Intention-totreat analyses showed that,when compared with the control group,PN1 group:RR:1.053;95% CI:0.952-1.164;ENI group:RR:1.333,95% CI:1.035-1.717;PN2 group:RR:1.053,95% CI:0.952-1.164;EN2 group:RR:1.25,95% CI:1.004-1.556.Conclusions Glutamine supplementation may not be able to decrease the time to achieve full enteral nutrition,the incidence of gastric residuals,and the duration of PN.However,pareneral glutamine supplementation does reduce the days on ventilator in neonates.Also,parenteral glutamine supplementation has no obvious influence on mortality.
10.Therapeutic effects of gensenoside-Rg3 combined with radiotherapy on advanced non-small cell lung cancer

Objective To study the efficacy and toxicity of gensenoside-Rg3 (Rg3) combined with radiotherapy on non-small cell lung cancer ( NSCLC ) at advanced stages (Ⅲ and Ⅴ ).Methods Sixty-three patients with stage Ⅲ or Ⅳ NSCLC were divided randomly into two groups:treatment group ( n =35 ) treated with Rg3 combined with radiotherapy and control group ( n =28 ) treated with radiotherapy alone.The efficacy and side effects were compared after the treatment.Results The response rate ( CR + PR) of the treatment group was 57.14%,significantly higher than that of the control group (32.14%,x2 =3.91,P < 0.05).The median survival time of the treatment group was 14.2 months,significantly longer than that of the control group ( 11.2 months,x2 =2.07,P < 0.05 ).The one-year survival rate of the treatment group was 62.86%,significantly higher than that of the control group (39.29%,x2 =4.40,P <0.05).The incidence rates of side effects of the treatment group were all lower than those of the control group,but there were not significant difference. Conclusions Gensenoside-Rg3 combined with radiotherapy is effective for advanced stage NSCLC,with attenuation and synergistic effects.
