2.Advance of molecular imaging in evaluation of RNA interference

Due to its effectiveness and specificity,RNA interference (RNAi) technology is widely used in the treatment of tumor,viral infectious and metabolic diseases.Noninvasively tracing the delivery of therapeutic RNA molecules in vivo and evaluating the therapeutic effects are important to promote the clinical application of RNAi technology.Molecular imaging allows noninvasive visualization of biological processes at the cellular and molecular level in vivo.It can be used to monitor the biodistribution of therapeutic RNA molecules and to assess the effects of the RNAi.This article reviews the advance of molecular imaging in monitoring the delivery of RNAi molecules and evaluating the therapeutic effects of RNAi.
3.The mechanism of chitosan hemostasis and its application

Chitosan have applied extensively in the field of medicine. We review the mechanism of chitosan hemostasis. The effect of chitosan on hemostasis is independent of all known“Cascade-like”coagulation pathways,induding platelets and soluble coagulant factors. Chitosan induce hemostasis by coalesce erythrocytes to one another to form a blood clot. In addition,the use of chitosan in hemostat is indicated in this paper.
4.Expression of IFN-γ,IL-4 and IL-1 7 proteins in peripheral blood of patients with oral lichen planus

Objective:To study the expression of IFN-γ,IL-4 and IL-17 in peripheral blood of patients with oral lichen planus(OLP). Methods:The level of IFN-γ,IL-4 and IL-17 in peripheral blood were detected by ELISA in 60 OLP patients(including 30 erosive ca-ses and 30 non-erosive cases)and 25 healthy volunteers(controls).Data were statistically analyzed by SPSS 13.0.Results:(1 )The level of IFN-γin serum of patients with non-erosive OLP and erosive OLP were significantly lower than that in the controls(P<0.01);while the level of IL-4 in the patients were significantly higher than that in the controls(P<0.01),and the IFN-γ/IL-4 ratio in non-ero-sive OLP and erosive groups were significantly lower than that in the control group(P<0.01);(2)The level of IL-17 in erosive OLP pa-tients was significantly higher than that in the controls(P<0.05),while no statistical difference was seen between non-erosive OLP group and the control group(P>0.05).Conclusion:The down-regulated level of IFN-γand up-regulated level of IL-4,which result in the decrease of IFN-γ/IL-4 ratio,might be one of the immune etiological factors of OLP,while the up-regulated level of IL-17 protein in OLP may be mainly related with the inflammatory processes of OLP.
5.Progresses in the Studies of Structure-activity Correlation and Detection Methods of Medicinal Fungal ?-(1,3)-D-glucans

?-(1,3)-D-glucan is a core structure of many bioactive fungal polysaccharides. It has drawn intensive attentions in recent because of its multiple bioactivities, particularly the immunomodulatory and antitumorous actions. In this review, we introduced the studies of structure-activity correlation of these ?-(1,3)-D-glucans, particularly the contributions of main backbone, branch degree and residues, molecular weight and conformation to the activities. We also summarized the recent progress in the detective methods for both quantitation and qualification using limulus G factor assay,galactosyl ceramide ELISA and etc.
6.Recent advances of the etiology and mechanism of biliary atresia

Biliary atresia ( BA) is a pediatric liver disease characterized by progressive inflammation and fi-brosis of both the extrahepatic and intrahepatic bile ducts. Even though Kasai portoenterostomy increases the survival of children with BA, 80% of patients with BA will eventually require liver transplantation, making this condition the leading indication for pediatric liver transplantation worldwide. Currently,the causes of this disease are largely unde-fined and theories of pathogenesis include viral infection,autoimmune-mediated bile duct destruction,and abnormalities in bile duct development.
7.The effects of using the nursing intervention during the course of cure patients with depression after the stroke

Objective To study the effects of using the nursing intervention during the course of cure patients with depression after the stroke. Methods Divided 127 patients with depression after stroke into the treatment group(63 cases) and the control group(64 cases) randomly,the general nursing intervention and the traditional nursing measures were adopted respectively in the two groups.Compared the effects of treatment between the two groups by the questionnaires. Results Results The effects of treatment in the treatment group were significant better than those of in the control group. Conclusion The general nursing intervention can effective improve the depression of patients with stroke,and then ameliorate their quality of life,shorten the time of recovery.
8.Detection of CK19 mRNA and CK20 mRNA in peripheral blood cells of patients with early cervicalcancer and its clinical significance

Purpose:To study the expression of CK19 and CK2 0mRNA in peripheral blood patients with FRGO Stage ⅠA to ⅡA cervical carcinoma and to investigate its clinical significance. Methods:Using the reverse transcription polymerase chain reacti on(RT-PCR),CK19 and CK20mRNA expression was examined in peripheral blood fro m 250 patients with early cervical carcinoma before operation,50 Patients with benign gynecological tumors and 18 healthy volunteers. In 250 patients,possible correlations between clinical pathological factors were analyzed. Results:The positive expression rates of CK19 and CK20mRNA were 36% and 24% in 250 cervical carcinomas respectively,in comparison with 3.0% an d 0% with benign gynecological tumors and all subjects in healthy volunteers wer e negative; The expression of CK19 and CK20 mRNA were significantly correlated w ith lymph vascular space involvement,but was not associated with prognostic fac tors including stage,differentiation,pathological types ,lymph node metastasis ,bully tumor size . 9.Giving full play to the function of archive administration in the construction of the hospital

The archives of a hospital are a kind of intangible assets of the hospital and archive administration in a hospital constitutes an important part of the hospital's management. It can be seen from the forming process and contents of the archives in a hospital that strengthening archive administration is beneficial to the modernized and scientific management of the hospital, the improvement of medical quality and service level, the spread and application of medical science and technology, and the safeguarding of the interests of both the hospital and patients. To give full play to the function of the archives in the construction of the hospital, it is imperative to attach importance to the fundamental work concerning archives, set up the concept of archive development and application, and improve the quality of the contingents of archive administrators. Only in this way can we meet the needs of the era of knowledge economy.
10.Relationship between the expression of focal adhesion kinase and the sensitivity of HT29 cells to 5-fluorouracil

Objective RNA interference (RNAi) expression vector was constructed to inhibit the focal adhesion kinase(FAK)expression in colon carcinoma HT29 cells, and then the sensitivity of the cells to 5-fluorouracil (5-FU) was determined. Methods One specific pair of oligonucleotides with short hairpin and its negative control sequence were designed and synthesized based on FAK cDNA sequences, then they were inserted into pGenesil-l vector to generate the recombinant plasmids. After identification by the restriction endonuclease and DNA sequencing, the recombinant plasmids were transfected into HT29 cells by lipofectamine TM 2000. The stable transfected cells were selected in a medium containing geneticin G418. The change in FAK expression in HT29 cells before and after RNA interference was detected by reverse transcription and polymerase chain reaction (RT-PCR) analysis and SP immunocytochemistry technique. Sensitivity of HT29 cells to 5-FU was determined by MTT assay. Results The recombinant plasmids coincided completely with the designs in the restriction map and the sequence analysis. After FAK being targeted by RNA interference, immunocytochemistry showed the protein expression of FAK was reduced dramatically, and RT-PCR revealed FAK mRNA expression was down-regulated by 76.94% compared to that of untransfected cells. MTT assay also showed that the sensitivity of HT29 cells to 5-FU in transfected pGenesil-1-FAK vector cells was increased, while IC50 declined remarkably (P
