EGFR-TKI plays an important role in the treatment of non-small-cell lung cancer. However, some researchers find that there are still some patients with primary or acquired resistance to EGFR-TKI. The present known mechanisms of acquired drug resistance finally lead to the re-activation EGFR downstream signa-ling pathways. Liver X receptor agonist has inhibition function to several critical steps of EGFR downstream sig-naling pathways PI3K-Akt-NF-κB,which makes it possible to overcome the drug resistance.

Objective To evaluate the effectiveness of voluntary smoke-free policies in Beijing. Methods With convenient sampling,91 restaurants and 85 restaurants were respectively selected before and after the initiation on voluntarily banning smoking in public eating and drinking places by the catering industry in Beijing, and including a follow-up surveillance on 23 restaurants.Field investigation on smoke-free rules and PM2.5 monitoring in and outside of these restaurants were conducted. Results There were no statistic differences among the proportion of restaurants with different smoking policies(?2=2.60,P=0.28).Univariate analysis showed that the indoor PM2.5 level was positively related with the outdoor PM2.5 level(?=0.51,t=7.01,P

Objective To investigate the activation and role of signal transduction pathway of epidermal growth factor (EGF)-epidermal growth factor receptor (EGFR)-mitogen activated protein kinase (MAPK) in proliferation of human retinal pigment epithelial (RPE) cells. Methods Human RPE cells were stimulated with 0.1%,10% foetal calfserum (FCS) and EGF(0.1, 1, 10, 50 and 100 ng/ml)in 0.1% FCS Dulbeco′s modified Eagle′s medium (DMEM) and in 10% FCS DMEM for 3 days, respectively. Immunohistochemical staining and in situ hybridization were used to observe the expressions of EGFR protein and EGFR mRNA,respectively. Activation of MAPK was detected by immunohistochemical method with specific anti-phosphorylated ERK 1/2 antibody. Results The optimal concentrations of EGF were 10 ng/ml in 0.1% FCS DMEM and 1 ng/ml in 10% FCS DMEM. After 3 days of stimulation with EGF, phosphorylated ERK 1/2 staining was detectable in nucleus of RPE cells, whereas cells presented immunostaining for phosphorylated ERK 1/2 in the cytoplasm before stimulation. Conclusions EGF may improve the expression of EGFR protein and EGFR mRNA of RPE cells, and induced MAPK nuclear translocation in a concentration-dependent manner. EGF-EGFR-MAPK signal transduction pathway may play a key role in RPE cells proliferation, and serum exerts an important acceclerating function in the process.

Objective To investigate the transactivating effect of hepatitis C virus(HCV)core protein on inducible nitric oxide synthase(iNOS)gene promoter and the molecular biological mecha- nisms of HCV pathogenesis. Methods Polymerase chain reaction(PCR)technique was employed to amplify the sequence of iNOS promoter by using HepG2 genomic DNA as template, and the product was cloned into pGEM-T vector. The iNOSp gene was cut from T-iNOSp by KpnⅠand XhoⅠ,and then was cloned into pCAT3-Basic,the constructed vector was named as pCAT3-iNOSp,pCAT3-iN- OSp was transfected into the LO_2 cell line.LO_2 cell was also cotransfected with pcDNA3.1(-)-core and pCAT3-iNOSp by FuGENE 6 transfection reagents. The LO_2 cells transfected with pCAT3-Basic was used as negative control. The activity of CAT in LO_2 cells was detected by an ELISA kit after 48 hours, which reflected the transactivating function of HCV core protein to iNOS gene promoter.Re- sults The expressive vector pcDNA3.1(-)-core and report vector pCAT3-iNOSp had been construc- ted and confirmed by restriction enzyme digestion and sequencing. The expression of CAT in LO_2 cells transfected with pCAT3-iNOSp and peDNA3,1(-)-core was 11 times as higher as that of pCAT3-bas- ic,and 6 times as higher as that of pCAT3-iNOSp.Conclusion It is suggested that HCV core protein can transactivate iNOS gene promoter.

Journal of Guangzhou University of Traditional Chinese Medicine 2001;0(01):-
[Objective] To investigate the characteristics of senile diabetic hip fracture in the peri-operation stage and to summarize the experience for its treatment. [Methods] A retrospective study was conducted in 21 cases of senile hip fracture complicated with diabetic mellitus which had received operation. [Results] No case was dead and no case was infected after operation; the incision was healed well and the patients went through the peri-operation stage safely. [Conclusion] On the basis of effectively controlling the blood sugar level, operation can be taken into consideration and pre-operative and post-operative treatment should also be emphasized.

Objective Using a bioreactor to culture reendothelializing homograft bioprosthetic valve (HBV) in order to ultimately form viable tissue in vitro, so as to provide basic material for animal experiment and clinical application. Methods The reendothelializign HBV was placed in a pulse duplicator system (“bioreactor”), designed and fabricated by us. These grown under gradually increasing nutrient media flow and pressure for additional 14 days: 125 ml/min at 30 mm Hg (days 1-4), 250 ml/min at 40 mm?Hg (days 5-7), 500 ml/min at 50 mm Hg (days 8-10), 750 ml/min at 75 mm Hg (days 11 to 14). Throughout the studies, the flow device was placed in an incubator with 95% humidity and 5% carbon dioxide. The morphologic structure was observed and photographed by stereomicroscope with 0.5% silver nitrate (AgNO_3)staining and SEM on the 7th, 10th and 14th day. The retention rate of the ECs overlaid onto the HBV was evaluated with the percentage (%). Results The reendothelization level on the surface of the leaflets was 92% before pulsatile-flow-cultivation. But the rate of ECs retention on the scaffold exposured to pulsatile flow was only 36%, 23% and 11% on the 7th, 10th and 14th day, respectively. Conclusion The retention rate of ECs on the scaffold was low for pulsatile-flow-cultivation in vitro. Problems, including deficiency of mature stress fibers in the endothelial cells from MSCs deficiency of the adherence and growth factors still need improvement.

Cardiovascular disease, with high morbidity and mortality, has been threatening the health of human beings. Therefore, expecting to find a more effective therapeutic method, a plenty of researchers devote themselves to the study of the cardiovascular disease all the time. Since discovered on the heart, M3 receptor of muscarinic acetylcholine receptor (mAchR, M receptor) became a new starting point of the research of the cardiovascular disease. With more and more investigation, many people found that M3 receptor could protect the heart from kinds of cardiovascular disease, which may make it a new hopeful therapeutic point. So, expecting to give support to the reference and encouragement for the study of disease related to M3 receptor in future, this review expounds M3 receptor on the heart from the main following aspects: the effect on the heart, the influence on the cardiovascular disease and the mechanism of M3 receptor involved.
