Objective To establish a new model of orthotopic-transplatation tumor of human malignant choroidal melanoma. Methods pEGFP-N1, the eukaryotic expressive plasmid of green fluorescent protein (GFP), was transfered into human malignant choroidal melanoma cell line (OCM-1) by liposome lipofectanine, then the cell clones with stable GFP expression were screened out by means of neomycin, fluorescence microscope, and flow cytometer. Two ?l cell suspension of OCM-1 cells with GFP expression with the density of 4.5?107-5.5?107 cells/ml was injected into the subretinal space of right eyes of 40 nude mice (40 eyes) under binocular operating microscope with left eyes as the control ones. The growth of the transplanted malignant choroidal melanoma was observed in vivo using the fluorescence stereomicroscope. The mice were killed at different time after the operation to observe the metastasis of the tumor to optic nerve, brain and other organs including lung, liver, kidney and spleen. Moreover, pathological detection and immunohistochemical staining of GFP was carried out. Results At the postoperative 10 th-12 th days, the growths of the transplanted malignant choroidal melanoma with dilated and distorted blood vessels and neovascularization were observed; at the postoperative 20 th-22 nd days, the melanoma occupied the whole cavity of vitreous body; and at the postoperative 24 th-26 th days, the transplanted tumor grew out of the eye. Metastases of the carcinoma to olfactory bulb, kidney, lung and liver were seen at the failure phase soon after the extra-ocular phase. The histopathological characteristics of the transplanted tumor were similar to those of human, and the results of immunohistochemical staining showed positive expression of GFP in the tumor cells. Conclusion The orthotopic model of malignant choroidal melanoma set up via injection of human malignant choroidal melanoma cells labeled by GFP into the subretinal space of nude mice may provide a new approach to investigate the natural courses of growth and metastasis of malignant choroidal melanoma.

Objective To know the inhibitory effects of 5aza-CdR in combination with HDAC inhibitors on RD-ES cells in culture.Methods This study was to investigate that the HDAC inhibitors (MS-275,Trichostatin-A and depsipeptide)and 5azaCdR inhibition effect on RD-ES cells by MTT.High sensitivity DNA kit was used to detect DNA inhibition rate.RT-PCR method was used to detect the expression of ECAD and TSLC1 for 5aza-CdR joint Depsi on RD-ES cells.Results IC50 of 5aza-CdR for RD-ES cell was 40 ng/ml,for 48 h 50 % DNA inhibition rate,the concentration was approximately 50 ng/ml.IC50 of HDAC inhibitors (MS-275,trichostatin-A,Depsi) for RD-ES cell was 400 ng/ml,2.5 ng/ml and 0.6 ng/ml respectively.The inhibition rate of 5aza-CdR jointed HDAC inhibitors was significantly higher compared with the single used groups (P

OBJECTIVE:To probe into the integrity of package insert for 75 kinds of commonly used antiinfective agents for injection in our hospital and to provide reference for rational use of antiinfective agents for injection. METHODS:According to Drug Description Specification and Management Regulation for Drug Description and Label,the terms included in package insert for antiinfective agents for injection were analyzed statistically. RESULTS:The package inserts for 75 kinds of antiinfective agents for injection were enrolled. The proportion of integral items were high while that of some terms related to clinic were low such as dosage and usage,medication for children,over dosage and skin test. The descriptions of PK,pharmacology,toxicdogy, ADR,et al. were very simple and different. CONCLUSION:It is suggested that items about over dosage and medication for special population should be detailed as well as usage so that package insert can guide rational use of drugs in the clinic.

Objective To investigate the pathological changes of gastric antrum mucosa after Helicobacter pylori eradication.Methods Total 180 cases who suffered from epigastralgia and took the endoscopic examination were randomly divided into two groups,one was Hp eradication group which included 98 cases and given anti-Hp medication treatment,and the other was control group which included 82 cases and given an expectant treatment.At the end of the study,they took reexamination by gastroscope and tests on Hp by Giemsa dyeing & rapid urase detection and on gastric antrum pathological changes by HE dyeing.Results In treatment group,atrophic gastritis as well as intestinal metaplasia decreased significantly after Helicobacter pylori eradication,but in control group,no change of atrophic gastritis was found while intestinal metaplasia aggravated.Conclusion The eradication of Helicobacter pylori is able to decrease atrophic gastritis and intestinal metaplasia.

Objective To study the rationality of biochemistry teaching of liberal arts students and science students in one class and further to improve the quality of teaching.Methods A total of 138 students of seven-year traditional Chinese medicine of Grade 2013 as the objects of study,among whom there were 58 liberal arts students and 80 science students.Questionnaire were surveyed at the end of the teaching of biochemistry,including evaluation of the teaching effect and teaching reform.138 valid questionnaires were dispatched and recovered.And at the same time unified examination was carried out to both liberal arts students and science students.The t test,chi square test or the exact probability method were used to compare the data of two types of students with SPSS 19.0.Results Questionnaire survey:31.03％ (18/58) arts students and 27.50％ (22/80) science students felt that the class hours were not enough.48.28％ (28/58) arts students thought the weak chemical base to be the main factor affecting learning,and 50.00％ (40/80) science students thought that lack of interest was the most important factor.53.45％ (31/58) arts and 47.50％ (38/80) science students would agree that a metabolic reaction of the material was the most difficult problem.51.72％ (30/58) arts students and 42.50％ (34/80) science students preferred blackboard-writing.Test scores:arts and science students got average grades (70.81 ± 9.71) and (77.05 ± 8.46) respectively,and the difference was statistically significant (t=-4.02,P=0.000).Conclusions Survey results and analysis of test scores shows that teaching of liberal arts students and science students in one class is feasible.However,it is necessary to strengthen the students' chemical basis,improve their learning interest and optimize their learning methods,so as to realize the harmonious development of the students of arts and science,and to improve the overall teaching level.

Aim: To prepare diclofenac sodium-layered double hydroxide nanohybrids(DS-LDH) and investigate its properties. Methods: The diclofenac sodium anions were hybridized with LDH by co-precipitation. The synthetic nanohybrids were characterized by various methods, such as laser particle sizer, XRD, IR, and TEM. Paddle method according to the Chinese Pharmacopoeia was used to determine the dissolution. The anti-inflammation of nanohybrids and diclofenac sodium were compared by the effect on auricular tumescence induced by xylene in mice. Results: The acidic negative ion of layered double hydroxides were replaced by the diclofenac anion to form DS-LDH. Release studies showed that the nanohybrids could serve as controlled release systems for DS. Zn-Al-NO3-LDH was proved to have character of neutral pH which is more suitable for orphthalmic preparation. The DS-LDH nanohybrids could keep the effect of anti-inflammation. Conclusion: Layered double hydroxide nanohybrids can be used as sustained-release carrier of anionic drugs. A novel type of drug-inorganic nanohybrids was prepared successfully which has a potential value of clinical application.

Objective To investigate the changes of platelet-leukocyte aggregations(PLA) in patients with acute cerebral infarction and its relationship with C-reactive protein(CRP) and platelet aggregation rate(PAgT).Methods The PLA,CRP and PAgT were measured in 46 patients with acute cerebral infarction(CI group) and in 24 patients without cerebral vascular diseases(control group).Results The platelet-monocyte aggregations(PMA),PAgT and CRP in CI group were((13.00?0.76)%,(59.46?3.07)%,(9.39?1.28) mg/L,respectively,however,there were(6.55?0.29)%,(39.38?5.42)%,(2.37?0.46)mg/L respectively in control group.There were significant differences between the two groups(all(P
