2.Tissue distribution and excretion of baicalein and its main metabolite in rats by LC-MS/MS

Aim: To study the distribution and excretion of baicalein and baicalin in rats. Methods: An LC/MS method was applied. Chromatographic separation was performed on a C_(18) column( 150 mm ×6.0 mm, 5 μm) with methanol-10 mmol/L ammonium acetate (containing 0. 5% formic acid) as the mobile phase. A trip-quadrupole tandem mass spectrometer was set in positive selected reaction monitoring mode. The sample was extracted with methanol-acetonitrile( 1:1) after the addition of phosphoric buffer solution and luteolin, which acted as the internal standard. The supernatant was evaporated to dryness, and the residual was reconstituted with mobile phase and analyzed. Results: The distribution profiles of the parent drug and its main metabolite showed two peaks between 20-40 min and 8-10 h after oral administration of baicalein, which fit the plasma concentration-time profile of baicalein in rats. At 20 min after the dosing, the concentration levels of baicalein were significantly higher than those of baicalin in stomach, liver and intestines, the converse result occurred in kidney. The excretion results showed that baicalin was the predominant excretion form in bile and urine, while baicalein was the negligible excretion form. There was more baicalein than baicalin in rat feces. Conclusion: Baicalein was absorbed and distributed quickly to various tissues and easily transformed to its metabolite at the same time.

4.Study on the correlation between different treatment methods and postoperative outcome in elderly patients

Objective To analyze the correlation between the diagnosis and treatment and the postoperative death and complication of the elderly patients.Methods A total of 1 932 cases of surgical patients (over 70 years old) were collected from a three-level general hospital 2013-2015.Their medical information of the treatment methods were recorded,including incision category,surgical approach,surgical grade and patient outcome after surgery(death or postoperative complications).Then we analyzed the incidence of death or postoperative complications,as well as the effects of treatment and postoperative outcome on complications or mortality.Results The admission status and incision type were the risk factors of postoperative death in elderly patients.Admission status pollution type,surgical approach and surgical grade were the risk factors of postoperative complications.Conclusion In order to reduce the risk of postoperative death and complications in elderly patients,the effect of admission and incision on the postoperative risk of elderly patients should be evaluated.
5.Effect of NIP regime in treatment of non-small cell lung cancer

This research is to investigate the effect of NIP regime in the treatment of non small cell lung cancer (NSCLC) and its side effects.Thirty two patients with NSCLC (stageⅢ 28 cases, include Ⅲa 6 cases and Ⅲb 22 cases, stageⅣ 4 cases) received chemotherapy regime NIP.Vinorelbine(NVB)25 mg/m 2 V.D,first day and eighth day ;Ifosfomide(IFO) 1.6 g mg/m 2 V.D, from first day to third day; Cisplatin(DDP)25 mg/m 2 V.D,from first day to third day.Four weeks was a cycle, every patients was given 2 cycles. RR was 53.13%(CR 1/32,PR 16/32,SD 4/32,PD 9/32). Main side effect was mylosuppression.NIP regime is effective and safe for patients with advanced NSCLC.
6.The Latest Guideline for Neonatal Jaundice Produced by American Academic of Pediatrics

Jaundice can occur in most neonates.Most jaundice is benign,but beacuse of the potential toxicity of bilirubin,newborn infants with severe hyperbilirubinemia can develop acute bilirubin encephalopathy or kernicterus.In October 2004,American(academic) of pediatrics produced the latest guideline for newborn jaundice aim to reduce the incidence of acute bilirubin encephalopathy or kernicterus and avoid unnecessary costs or treatment.The latest guideline emphasizes the important of successful breastfeeding,time of jaundice developed,assessment for the risk of severe hyperbilirubinemia,close follow-up,and prompt intervention when indicated,provide a framework for the prevention and management of hyperbilirubinemia in newborn infants of 35 or more weeks of gestation.Some new view in the latest guideline may be helpful to us.
9.pRNA: A Safe,Stable and Large Capacity Molecular Vehicle for Gene Therapy

The molecular delivery vectors used in gene therapy need provide the features of safety,stability, efficiency and capacity. The current studies on the structure and action mechanism of pRNA, a packaging RNA of phage?29, showed that pRNA with multiple binding sites can through cell membrane easily and escort exogenous molecules to target cell, without inducing immune reaction. As an ideal nano-scale gene therapy vehicle, pRNA presents a promising application in delivering multiple therapeutic components to detect and treat human diseases.
10.Study on challenge dose of pigeon paramyxovirus type 1 (Chuansha strain).

In order to determine the challenge dose of pigeon paramyxovirus type 1 (PPMV-1) inactivated vaccine (S-1 strain). The virus titer of PPMV-1 E5 allantoic fluid (Chuansha strain) was determined using SPF chicken embryos in this research. After inoculating 30-day-old and 120-day-old pigeons with low-HI antibody against PPMV-1 (HI antibody < or =2) with different doses of PPMV-1 (Chuansha strain), the clinical symptoms and histopathological lesions of the challenged pigeons were examined. The results showed that the minimal lethal dose (MLD) of PPMV-1 (Chuansha strain) was 102.5 ELD50, so we determined that 10(5.5) ELD50, which was 1000 times the MLD, could be taken as the challenge dose in the vaccine efficacy test for PPMV-1 inactivated vaccine (S-1 strain).
