1.Effect of sodium aescinate in inducing human breast cancer MCF-7 cells apoptosis by inhibiting AKT, ERK and upstream signal SRC activity.

To study the effect of sodium aescinate in inducing human breast cancer MCF-7 cells apoptosis and its possible mechanism. MTT assay was used to detect the inhibitory effect of sodium aescinate on the proliferation of MCF-7 cells. The morphological changes were observed under inverted microscope. DAPI nuclear staining was used to detect the changes in cell nucleus. Annexin V-FITC/PI flow cytometry was adopted to test the apoptosis rate. Changes in apoptosis-related proteins (PARP, cleaved caspase-8 and pro-caspase-3), cell survival-associated signal molecules (AKT and ERK) and their common upstream kinase SRC was detected by Western blotting. The result showed that after different concentrations of sodium aescinate were used to treat breast cancer MCF-7 cells, they inhibited the proliferation of MCF-7 cells in a dose-dependent manner, induced cell apoptosis (typical morphological changes in nucleus, significant increase in cell apoptosis rate). The expressions of cleaved PARP and caspase-8 increased, while the expression of pro-caspase-3 decreased, which further verified sodium aescinate's effect in inducing cell apoptosis. Sodium aescinate significantly inhibited the phosphorylation of cell survival-related signal molecules (AKT, ERK) and down-regulate the activation of their common up-stream kinase SRC. The findings indicated that sodium aescinate can block signals transiting to downstream molecules AKT, ERK, inhibit the proliferation of breast cancer cell MCF-7 cell apoptosis and induced cell apoptosis by suppressing the activation of SRC.

2.Study on the Characteristics of Nicotine Degradation by Strain DN2 and its Application

The kinetics of nicotine degradation by O.intermedium DN2 and its application in tobacco waste were investigated. The results showed that the optimum temperature of nicotine degradation by O.intermedium was 30 ℃, the pH value was 6.5 and a mount of inoculum was 5 %. Under above conditions, the kinetics of nicotine degradation of initial concentration 500 mg/L was studied. The results indicated that the degradation process of nicotine with no-induced strain DN2 followed inverse S-shaped curve, and degradation process of nicotine with induced cells of DN2 followed Eckenfelder mode. The half life of nicotine degradation was 17.43 h and 4.10 h, respectively. And the results also showed that tolerance of O.intermedium DN2 to nicotine was up to 5000 mg/L when 0.1 % of glucose was added. Nicotine (2 220 mg/L) in extract of tobacco wastes degraded about 95.22 % by strain DN2 in 60 h incubation, indicating that strain DN2 was of application value in treatment nicotine pollution.
3.Clinical analysis of stereotactic radiotherapy combined with Gemzar for treatment of advanced pancreatic cancer

Objective: To observe the efficacy of stereotactic radiotherapy combined with gemzar for treatment of unresectable pancreatic cancer.Methods: Gemzar at dose of 1000mg/m 2,28 days per cycle,drug was given at 1,8,15 days,for two cycles.After 24 hours all patients were treated with stereotactic radiotherapy.Per fraction was 5~7G Y,three times per week.The total dose was 40~48G Y.Results: 13 patients were followed up for one year,symptoms were reduced and median survival was 12 months.Conclusion: Stereotactic radiotherapy combined with gemzar has important valves for unresectabal pancreatic cancer.
4.UPLC fingerprint spectra for discrimination of Aucklandiae radix and Vladimiriae radix.

It's difficult to identify Aucklandiae Radix and Vladimiriae Radix because of their similar composition. In this paper, UPLC method was used to establish their UPLC fingerprint to identify them with the mobile of acetonitrile -0. 05% phosphoric acid water solution by gradient elution at the detection wavelength of 238 nm. Clustering analysis and principal components analysis showed that Vladimiriae Radix was significantly different from Aucklandiae Radix. Eight common peaks and twelve common peaks were defined respectively in Aucklandiae Radix and Vladimiriae Radix herbs by fingerprint analysis. Six of them were identified as syringoside, chlorogenic acid, isochlorogenic acid A, isochlorogenic acid B, costunolide and dehydrocostuslactone by comparing with standard references. There are four peaks in all of Vladimiriae Radix samples and in none of Aucklandiae Radix samples. So UPLC fingerprint can be used to identify these two herbs.

Objective To investigate the expression and function of integrin-iinked kinase (ILK) in the transdifferentiation process of primary tubular epithelial cell( TEC) and its effect on the accumulation of extracellular matrix (ECM) in the aging process. Methods Primary TEC was cultured from kidneys of male Wistar rats aged 3 and 24 months. Then the primary TEC was stimulated by TGF-? at the concentrations of 0, 5, 10 and 20-40/?g/L for 24 h. The TEC expressions of ?-SMA,ILK and F-actin were detected by immunocytochemistry or indirect immunofluorescence. FN or ILK protein expression levels were tested by Western-blot. DIG-labelled cRNA probe of rat ILK in situ hybridization was obtained by transcription in vitro, with which ILK expression level and the location of TEC were detected. Results ?-SMA expression levels were very low in young or aging rat TEC on the base situation, no significant difference was found between the two groups. However, ILK and F-actin levels in 24 months group were higher than in 3 months group. Along with increasing concentration of TGF-?, ?-SMA, ILK and F-actin levels were increased. 6.The clinical study of early systematic nursing intervention on the prevention of venous thrombus of low limbs in ICU patients

Objective To explore the clinical meaning of early systematic nursing intervention on the prevention of low limbs venous thrombus in ICU patients.Methods Totals of 120 patients in ICU were randomly divided into two groups by entering order number,odd numbers were experimental group(60 cases)and double numbers were control group(60 cases).Experimental group received early systematic nursing intervention while control group received ICU conventional nursing.All patients were measured the lower limb diameter per day,and were received colour Doppler examination to tested their femoral venous blood velocity in first,seventh and fourteen day of ICU,and their femoral venous blood velocity and incidence of low limbs venous thrombus were compared.Results No difference of femoral venous blood velocity in two groups was founded before intervention (P >0.05).After intervention,femoral venous blood peak velocity and average velocity of seventh and fourteen day in experimental group was(52.8 ± 9.6),(52.6 ± 8.8)and(34.2 ± 6.9),(35.3 ± 7.2),respectively,and higher than(41.3 ±7.8),(42.5 ±7.6)and(22.5 ±7.1),(23.7±6.6)in control group,differences between two groups were statistically significant(t =2.724,3.125,2.722,3.114 ;P < 0.05); the incidence of low limbs venous thrombus in experimental group was lower than that of control group,and the difference was statistically significant(x2 =4.904,P < 0.05).Conclusions Adopting early systematic nursing intervention can improve the femoral venous blood velocity,and effectively prevent and reduce the incidence of low limbs venous thrombus of ICU patients.
7.Research of early bundles of care preventing intestinal dysfunction with sepsis

Objective To evaluate the effect of early bundles of care preventing on intestinal dysfunction in patients with sepsis.Methods Totals of 48 cases sepsis patients were randomly divided into intervention group received the early bundles of care,and control group received the routine nursing.Then,the score of intestinal dysfunction,APACHE Ⅱ and detect serum diamine oxidase concentration at 1 d and 7 d after ICU admission were observed.Results The APACHE Ⅱ [(16.20 ± 2.53) vs (24.40 ± 2.68)],intestinal dysfunction score [(1.27 ± 0.18) vs (1.95 ± 0.27)] and serum diamine oxidase concentration [(2.20 ± 0.21)vs (3.26 ± 0.10)mmol/ml] on the 7 d in intervention group were significantly lower than those in control group,with statistically significant differences(t =7.04,-6.59,13.90,respectively;P <0.05).Conclusions The early bundles of care can effectively prevent the occurrence of intestinal dysfunction in patients with sepsis.
8.The clinical research of non-pain catheterization technique used in old male patients

Objective To probe the clinical research of non-pain catheterization technique used inold male patients. Methods The 60 old male patients needed to leave alone the catheter were divided intothe experimental group and the control group stochastically. Using Shntai anesthetic and lubricant to lubricatethe children aerostat catheter (10Fr, diameter is 4.0mm) for persons in the experimental group, while thecontrol group uses convention paraffin wax oil to lubricate the two-chamber aerostat catheter (16Fr, diameteris 6.4mm). We compared the urine road stimulation symptom, the success ratio of disposable intubations,urethra leaves, urine leaves tube, urine road infection and so on. Results The urine road stimulationsymptom in the experimental group is lower than that in the control group obviously (P<0.05), while thesuccess ratio of disposable intubations in the experimental group is higher than that in the control group (P<0.05). In term of these two aspects, there is statistics significance for the two comparing groups. On theother hand, there is no obvious change in urethra leaves, the urine leaves tube and urine road infection, forwhich there is no statistics significance (P>0.05). Conclusions Using Shutai anesthetic and lubricant tolubricate the children aerostat catheter for old male patients can block the urine road stimulation symptom inperiod of setting the catheter, improve the success ratio of disposable intuhatious, change the ill response ofthe traditional leaving alone catheterization brought for old male patients, and reduce patients' pains.

Aim To investigate the effects of dihydromyricetin on the apoptosis in AGZY-83-a tumor cells and its relevant mechanisms.Methods The survival rate of AGZY-83-a cells was assayed by MTT dye reduction.The cellular apoptosis was analyzed by TUNEL method and caspase-3 activity.The intracellular free calcium i was detected to explore the apoptotic mechanism of dihydromyricetin.Results In the MTT assay,dihydromyricetin 50?mol?L-1 significantly inhibited survival ratios of AGZY-83-a cells at dose-and time-dependent manner.It was demonstrated in TUNEL assay that dihydromyricetin could induce the apoptosis of AGZY-83-a cells in a concentration-dependent manner.The examination of Caspase-3 activity indicated that the dihydromyricetin could induce the apoptosis of AGZY-83-a cells,which was dose-dependent activation of Caspase-3 in AGZY-83-a cells.The detection of the intracellular i showed that the average FI of the i could be markedly increased to 20-fold as the basic condition.Conclusions Dihydromyricetin can induce the apoptosis in AGZY-83-a cells,which is associated with the overload of the intracellular i.
