1.Effect of Nerve Growth Factor on Expression of Caspase-3 in Hippocampus of Rats with Seizures Induced by Kainic Acid

Objective To observe the effect of nerve growth factor(NGF) on Caspase-3 expression in hippocampus of epileptic rats induced by kainic acid(KA) and explore the protective effect and mechanisms of NGF on the epileptic neurocytes.Methods Sixty healthy rats aged 21 to 30 days were randomly divided into 3 groups,each of which included 20 rats.Rats in group A were treated with 9 g?L-1 sodium,rats in group B were treated with KA,rats in group C were treated with KA and NGF.Concentration of KA was 2 g?L-1 diluted by 9 g?L-1 sodium;KA was injected into rats in group B and group C,intraperitoneally.Then state of seizure was recorded,and EEG and pathological section were performed.Five rats in each group were sacrificed by perfusion with paraform respectively at 6 hour,on the 1st day,on the 3rd day after injection of KA.Specimen was sampled and made into frozen section.Expression of Caspase-3 in hippocampus of epileptic rats was analyzed by immunohistochemistry.And microscope image measuring and analysis instrument was used to analyze image of expression.Results Symptoms of seizure appeared 30 min after injection of KA.Seizure was proved by detection of EEG and pathological section.Expressions of Caspase-3 detected by immunohistochemistry in group C decreased at every time spot were higher than those in group B,and the defferences between 2 groups had statistically significant(Pa

Objective To study the effect of different approach on myocardial contractile force, angiogenesis and collagen renovation in acute myocardial infarct site by implantation of bone marrow mesenchymal stem cells (MSCs). Methods 32 Gui-zbou Xiang porcine were randomly divided into control group (group C), intra-coronary arterial injection group (group A) ,topical injection group (group T) and intra-coronary arterial + topical injection group( group A + T). 3 ml bone marrow was extracted. MSCs were cultured according to the methods of Wakitani's. After being co-cultured with 5-azacytidine for 24 hours, these cells were labelled with bromodeoxyuridine ( BrdU ). Autologus MSCs were implanted into the acute myocardial infarct site via the distal segment of the ligated left anterior descending artery (LAD) ,topical injection and both. Specimens and muscle strip were harvested in 3 and 6 weeks after MSCs implantation for microvessel count, P Ⅲ NP activity and contractile force measurement. Results The contractile force(48.6±5.9) %, (42.1±6.2 ) %, ( 56.9±5.1 ) %, microvessel count ( 19.6±4.3,17.1±4.0, 23.2±5.5 ) and collagen renovation (4.6±0.5 ) μg/L, ( 5.9±0.7 ) μg/L, ( 3.9±0.3 ) μg/L in the cell implant groups was better than that in the control group(37.9±5.4)% ,13.2±3.8,(8.7±0.8) μg/L,(P <0.01) ,espe-cially in group A + T. Conclusions Implantation of MSCs through intra-coronary arterial plus topical injection of MSCs maybe an ideal approach for infracted myocardium.
4.Change and Significance of Soluble Intercellular Adhesion Molecule-1 in Cerebro spinal Fluid of Bacterial Meningitis Induced by Escherichia Coli in Rabbits

Objective To investigate the changes and significance of concentration of soluble intercellular adhesion molecule-1(sICAM-1) in cerebrospinal fluid (CSF) of bacterial meningitis(BM) in rabbits. Methods A total of 36 rabbits were randomly divided into m eningitis group, meningitis cefotaxime-treated group and control group. BM indu ced by escherichia coli(Ec) via cerebellomedullary cistern inoculated. Normal sa line was injected in control group. CSF was sampled in different time. The conce ntration of CSF sICAM-1 was detected by ELISA.Results 1.There was a low concentration of sICAM-1 in CSF in 85 percent of normal rabbi ts.2.In meningitis group, there was a sharp rise in the concentration of CSF sIC AM-1 at 6 hours after Ec was inoculated, reached a peak level at 12 hours, and t here was higher concentration of CSF sICAM-1 between 6 and 24 hour than that at 0 hour.3.In meningitis cefotaxime-treated group, the concentration of CSF sICAM -1 at 6 hour and 12 hours was similar to meningitis group, the time that get pe ak level was at 24 hours that at 12 hours after making use of antimicrobial agen t.The peak level was higher than meningitis group. The concentration of CSF sICA M-1 decreased markedly at 48 hours that made use of antimicrobial agent 36 hour s later,but the concentration was still higher than the peak level of meningitis group.Afterwards, with the time of making use of antimicrobial agent lengthened ,the concentration of CSF sICAM-1 decreased gradually.4.Experimental results in dicated by correlating analysis to these data that there was positive correlatio n in the concentration of CSF sICAM-1 with the brain water content.Conclusions sICAM-1 participates in the pathological process of BM, and contributes to the damage of blood brain barrier and the formation of brain edema.There is importan t significance that drugs which can resist the discharge of sICAM-1 will be impl ied. Objective: To evaluate the effect of slow release microcapsules of bFGF on implanted vein graft. Methods: The vein graft stenosis model was made in 24 New Zealand rabbits. Group I (n=8) served as control group, group II (n=8) as blank microcapsule group, group III(n=8) ,each microcapsule contains 1?g bFGF as bFGF group. In group II and III,5 blank microcapsules or bFGF slow release microcapsules were implanted close to the vein graft respectively. Animals were sacrificed 5 weeks later for assessment of reendothelialization, neointimal thickening and for analysis of in vitro vasoreactivity. 8.Estimation of PMI using late postmortem phenomena in the basis of 49 cases.

OBJECTIVE:
To discuss the influencing factors of using late postmortem phenomena to estimate PMI and to provide experience for an accurate estimation.
METHODS:
Forty-nine corpses of late postmortem were collected in Shaoxing City, Zhuji area from 2004 to 2011. The related factors were analyzed including season, scene, estimated PMI, exact PMI, cause of death and main factors effected PMI, etc.
RESULTS:
Of all 49 cases, 20 corpses were outdoor, 11 were indoor and 18 were in water. Thirty-seven cases were successful to estimate PMI and 12 cases were unsuccessful. The main factors affected PMI were infection, poisoning, human destruction and high-pressure electric shock, etc.
CONCLUSION
In general, PMI can be correctly estimated by late postmortem phenomenon. When the cases included infection, poisoning and human destruction, we should estimate PMI with the comprehensive analysis.

9.Studies on triterpenes chemical constituents in rhizome of Alisma gramineum.

OBJECTIVETo investigate the triterpenoids chemical constituents in rhizome of Alisma gramineum.
METHODSilicon gel chromatography and HPLC techniques were employed for isolation and purification of the constituents, and the structures were elucidated by various spectral analyses such as NMR, MS and IR.
RESULT5 protostane-type tetracyclic triterpenoids were isolated and identified as 16,23-oxidoalisol B(1), 11-deoxyalisol C(2), alisol F(3), 16beta-methoxyalisol B acetate (4), 16beta-hydroxyalisol B acetate (5).
CONCLUSIONAll of these compounds were found from A. gramineum for the first time.

10.Identification of cardiomyocytes differentiated from mesenchymal stem cells and study on expression of Nesprin protein

Objective To induce and identify the differentiation of rat bone marrow mesenchymal stem cells(MSCs)into cardiomyocytes in vitro,and observe the expression of Nesprin protein during the differentiation. Methods Rat MSCs were isolated and purified by Ficoll density gradient centrifugation,and adhered for serial subcultivation.Surface-associated antigens of MSCs of the second passage were dedected by flow cytometry.MSCs of the second passage were induced by 10μmol/L 5-azacytidine(5-Aza)to differentiate into cardiomyocytes,and the morphological changes were observed.The expression of Desmin,α-sarcomeric actin and cardiac Troponin I(cTnI) mRNA and protein was detected by RT-PCR,immunocytochemistIv and immunofluorescence staining, and the expression of Nesprin protein was detected by Western blotting. Results The morphology of MSCs induced by 5-Aza was bigger and longer,and the nuclei became bigger,exhibiting more consistent patterns.The expression of Desmin,α-sarcomeric actin and cTnI mRNA and protein of MSCs was positive.Immunofluorescence staining revealed that Nesprin protein positioned in the nuclear membrane,and Western blotting detection demonstrated that the expression of Nesprin protein significantly increased after differentiation(P<0.05).Conclusion MSCs may be successfully induced to differentiate into cardiomyocytes.The expression of Nesprin protein in the differentiated MSCs may significantly increase,indicating Nesprin may play a role in the differentiation from MSCs to cardiomyocytes.
