1.Expression and clinical significance of hMLH-1 and hMSH-2 in Han and Uygur in Xinjiang area of sporadic colorectal carcinoma

Purpose To detect the incidence rate, average age and the expression of hMLH-1 and hMSH-2 of sporadic colorectal carci-noma ( SCC) with Han and Uygur patients. Methods The expression of hMLH-1 and hMSH-2 was detected in SCC for 60 cases of Uygur and 196 cases of Han by immunohistochemical method, including 60 Uygur and Han cases normal colorectal mucosa ( NCM) . Results The positive rate of hMLH-1 and hMSH-2 proteins expression in the NCM was 100%. There was a marked difference in the positive rate of hMLH-1 in SCC between Han (93. 4%, 183/196)and Uygur (75%, 45/60) (P<0. 05). There was no significance in the positive rate of hMSH-2 in NCM between Han (94. 4%, 185/196) and Uygur (91. 7%, 55/60) (P>0. 05). The average age of Han and Uygur SCC patients were 65. 64 years, 57. 63 years, respectively, and Uygur SCC cases were more likely to be diagnosed at less 40 years old (P<0. 05). The positive rate of hMLH-1 and hMSH-2 expression in the tubular adenoma was 100%. The positive rate of hMLH-1 and hMSH-2 expression in the tubulovillous adenoma in Uygur and Han were 66. 7%( 2/3 ) and 66. 7%( 2/3 ) , and 74. 2%(23/31) and 90. 3%(28/31), respectively, significantly lower than those of tubular adenoma (P<0. 05). The expression of hMLH-1 was positively correlated with that of hMSH-2 in SCC in Han(rs =0. 737, P<0. 05). The expression of hMLH-1 was positive-ly correlated with that of hMSH-2 in SCC in Uygur(rs =0. 383, P<0. 05). There exists marked difference in the positive rate of hM-LH-1 and hMSH-2 among difference age groups (P<0. 05). Conclusion There is a certain loss of hMLH-1 and hMSH-2 expression in SCC in Han and Uygur Chinese, which is related to adenoma and age. The expression of hMLH-1 in SCC tissue among Uygur pa-tients is not resemble to those of Han patients. The average age of Uygur SCC patients is younger than Han, and the positive rate of hMLH-1 is higher. Combined detection of hMLH-1 and hMSH-2 proteins may be used for judging the severity and prognosis of SCC in Xinjiang, which helps improve patients’ treatment program and rationalize their choices.
2.99Tcm-ciprofloxacin imaging in detecting the secondary infection of severe acute pancreatitis

Objective To evaluate the characteristics of 99Tcm-ciprofloxacin and explore its feasibility in early detection of secondary infectious foci of severe acute pancreatitis (SAP).Methods Ciprofloxacin was labeled with 99Tcm.The labeling efficiency and radiochemical purity of 99Tcm-ciprofloxacin were calculated and its biodistribution in normal pigs was measured.The recruited baby pigs were divided into three groups:normal control group (6), non-infected group (6) and infected group (16).370-400 MBq of 99Tcm-ciprofloxacin was injected into each pig intravenously.SPECT scanning was performed at 0.5, 1,2, 3, 4 and 6 h after administration.The differences of 99Tcm-ciprofloxacin uptake among groups were calculated and the tracer activity ratio of lesion-to-background was recorded at each time point.The diagnostic value of 99Tcm-ciprofloxacin SPECT imaging for the dectection of secondary infection of SAP was assessed using histopathological results as the gold standard.Variance analysis and least significant difference test were used to analyze the data.Results Both the labehing efficiency and radiochemical purity of 99Tcm-ciprofloxacin were over 90% within 6 h.Organs with rich blood supply, such as kidney, liver and spleen were the target organs for the accumulation of 99Tcm-ciprofloxacin; while no significant uptake was found in gastrointestinal tract or normal pancreas tissue of SAP.Rapid plasma clearance and renal excretion were observed.In the infected group, the lesion was visualized at 1 h after administration.The highest radioactivity ratio of lesion-to-background (3.36 ± 0.33) was at 3 h after administration, which was significantly higher than that of the other time point ( F =99.570, P <0.001 ).The sensitivity, specificity, positive and negative predictive values, Youden's index (YI) and Kappa value of 99Tc%ciprofloxacin imaging were 88.2% (15/17), 83.3% (5/6), 93.8% ( 15/16), 71.4% (5/7), 0.715 and 0.667 respectively.Conclusions The biodistribution of99Tcm-ciprofloxacin is suitable for imaging infectious focus of SAP.The optimal imaging time for the detection of secondary infection of SAP is 3 h after administration, with high sensitivity and specificity.
3.Repairing soft-tissue defection of ankle and foot by sural neurovascular pedicle fasciocutaneous reversed island flap.

OBJECTIVETo explore the clinical application of sural neurovascular pedicle fasciocutaneous reversed island flap to repair soft-tissue defection of the ankle and foot.
METHODSFrom Sep. 1994 to Oct. 2004,29 patients with soft-tissue defects in the ankle and foot were repaired by use of sural neurovascular fasciocutaneous reversed island flap, including 15 cases of traumatic defects, 11 cases of burns and 3 cases of chronic ulcer. The flap area ranged from 5 cm x 7 cm to 12 cm x 20 cm, and the length of pedicle from 5 cm to 12 cm.
RESULTSThe flaps survived totally in 27 cases, the distal necrosed partially and secondary free-skin grafting were further conducted in 2 cases. Twenty-one cases were followed-up for 3 to 60 months,the circulation, color and texture of the flaps were excellent and 2-point discrimination was 10 - 15 mm. The appearance and function of ankle joints were good.
CONCLUSIONThis flap has sufficient blood supply and a high survival rate; It is convenient in design, dissection and without sacrifice of major arteries. So, it is an effective method for the reconstruction of soft-tissue defects in ankle and foot.

4.Analysis on processing mechanism of calamine.

OBJECTIVETo study processing method and mechanism of Calamine.
METHODThermogravimetry analysis method and nano-technology were adopted to analyze and synthesize the components in Calamine, Tetracycline was took as the comparison drug to determine the antibacterial activity of Calamine and its components.
RESULTA part of zinc carbonate in Calamine was decomposed into zinc oxide when processing, and the particle size was smaller than before. The antibacterial activity of Calamine is decided by the content and particle size of zinc oxide, and has nothing with zinc carbonate. The more content and the smaller particle size of zinc oxide, the more powerful antibacterial activity of Calamine.
CONCLUSIONThe content and the particle size of zinc oxide can be the important targets in the processing of Calamine.

5.Biocompatibility of physico-crosslinked regenerated silk fibroin film as tissue engineered cornea

Background Biomaterials for corneal tissue engineering must demonstrate several critical features for potentialutility invivo, includingtransparency, mechanicalintegrity, biocompatibilityand slow biodegradation. Silk film biomaterial had been characterized to meet these functional requirements. ObjectiveThis study was to investigate the feasibility of physico-crosslink regenerated silk fibroin film as tissue engineered corneal scaffold. MethodsHuman corneal epithelial cells(CECs) links were cultured by regular method and CECs in logarithmic phase were than incubated on physico-crosslink regenerated silk fibroin film membrane. The shape of cultured human CECs was observed after 24,48 and 72 hours under the inverted microscope and scanning electron microscope( SEM ) ,and the CECs were cultured on culture plates as controls. The growth state of CECs on regenerated silk fibroin film was observed daily for 7 days by MTT, and cell cycle analysis and the presence of apoptosis of human CECs were examined by flow cytometry after incubation on regenerated silk fibroin film. Regenerated silk fibroin filmCECs (4 mm×3 mm) were implanted into the corneal stroma of the right eyes of New Zealand white rabbits. At the end of 4 and 8 weeks after implantation, the appearance of the ocular surface was examined using slit lamp and corneal neovascular area was measured. Corneal histopathological examination was carried out to assess the degradation of graft materials and immunohistochemistry was performed to detect the expression of CD34 in the corneal tissue after operation. ResultsThe morphology and structure of CECs were identical using the two cultured Methods when observed under the inverted microscope and SEM after 24,48 and 72 hours. No significant difference was found in the A490 value 1,2,3,4,5,6 or 7 days after incubation on regenerated silk membrane and in culture plates ( Fmethod =0. 641 ,P>0.05 ). The apoptosis rates of CECs on regenerated silk membrane or culture plates were 1.8% and 2.0% and the amount of cells in G2/G1 phase was 1. 956 and 1. 945, respectively. Histopathological examination showed that the regenerated silk membrane material degraded and was replaced by regular collagen tissue 2 months after implantation,and the presence of neovascular area and inflammatory cells were less prominent in 2 months than 1 month post-implantation. The expression level of CD34 in corneal tissue was evidently lower 1 and 2 months after operation than the Ad-VEGF165-induced positive control group (P<0. 05), and no significant differences were seen when compared with normal CECs(P>0.05). ConclusionsPhysico- crosslink regenerated silk fibroin film is an excellent biomaterial for tissue engineered corneal scaffold with good biocompatibility.
6.Co-detection of P21, P53 and HSP70 and their possible role in diagnosis of polycyclic aromatic hydrocarbons (PAHs)-related lung cancer.

OBJECTIVETo explore the biomarkers of early diagnosis in patients with polycyclic aromatic hydrocarbons (PAHs)-related lung cancer for the application to detection of occupational lung cancer or related lung cancer.
METHODSWestern dot blotting was used to explore the expression of ras, p53 and heat stress protein 70 (HSP70) in 29 patients with PAHs-related lung cancer (LC), and 28 patients with non-cancerous pulmonary disease, and 30 healthy controls.
RESULTSThe positive detection rates of P21, P53, and HSP70 in LC group (58.62%, 34.48%, 41.38% respectively) were higher than those in non-cancerous pulmonary disease group (14.29%, 7.14%, 10.71% respectively, P < 0.01). The sensitivity of P21, P53 and HSP70 were 58.62%, 34.48% and 41.38% respectively, negative predictive value (NPV) were 68.42%, 78.05% and 63.04% respectively. The co-detection of the three proteins mentioned above produced a sensitivity of 82.76% with a NPV of 78.26% (P < 0.05). Of 18 cases of LC with negative cytology, 13 (72.22%) were found HSP21, P53 or HSP70 positive.
CONCLUSIONSCo-detection of the P21, P53, and HSP70 may be used as the screening marker for diagnosis of PAHs-related lung cancer, and may supplement the diagnostic value of conventional cytology.

7.Effect of Electroacupuncture on Learning and Memory and Expression of Beclin-1 in Rats after Cerebral Ischemic-reperfusion

Objective To observe the efficacy of electroacupuncture at Baihui (GV20) and Shenting (GV24) on learning and memo-ry in rats after cerebral ischemia-reperfusion, and explore the possible mechanism. Methods A total of 45 male Sprague-Dawley rats were randomly divided into sham group (n=15), ischemia group (n=15) and electroacupuncture group (n=15). The left middle cerebral arteries of the latter two groups were occluded with the modified Longa's method for two hours and reperfused, and the electroacupuncture group received elec-troacupuncture at Baihui and Shenting once a day for seven days, then. They were assessed with Longa's score one, three, five and seven days after intervention. They were tested with Morris water maze since four days after intervention, once a day for four days. The cerebral infarction volume was measured with TTC staining seven days after intervention, while the protein expression of Beclin-1 was detected with Western blotting. Results Longa's score improved in the electroacupuncture group compared with that in the ischemia group since three days after intervention (P<0.05). The escape latency decreased and the times cross the area of platform increased in the electroacupuncture group compared with that in the ischemia group at every point (P<0.05). The cerebral infarction volume reduced (F=7.651, P<0.001) and the expression of Beclin-1 decreased (P<0.05) in the elec-troacupuncture group compared with that in the ischemia group.Conclusion Electroacupuncture can improve learning and memory in rats after cerebral ischemia-reperfusion, which may relate with regulation of autophagy network.
8.Large cell calcifying Sertoli cell tumor of the testis: a clinicopathological observation.

OBJECTIVETo investigate the clinicopathological characteristics of large cell calcifying Sertoli cell tumor (LCCSCT) of the testis.
METHODSWe studied a case of LCCSCT by light microscopy, Western blotting and immunohistochemistry, reviewed relevant literature, and analyzed the clinical, morphological and immunohistochemical features, treatment and prognosis of the tumor.
RESULTSThe patient was a 25 years old man. Pathohistologically, the tumor was characterized by a mass of polygonal tumor cells in a tubular and trabecular growth pattern, with abundant acidophilic cytoplasm, enlarged vesicular nuclei, and extensive calcified debris in stroma. The tumor cells were positive for inhibin, S-100, vimentin and alcian blue, but negative for PLAP, SMA, CK, AFP and periodic acid-Schiff (PAS) reaction.
CONCLUSIONLCCSCT is a rare testicular sex cord stromal tumor. Its diagnosis is based on immunohistochemical staining, and it is to be differentiated from other lesions of the testis, including seminoma, Leydig cell tumor, Sertoli cell node, and androgen insensitivity syndrome. For the treatment of LCCSCT, surgical resection often has a good prognosis.

9.Evaluation of the capacity of personal protection on poison emergency items in Chinese disease control and prevention institutes.

OBJECTIVETo analyze the capacity of personal protection on poison emergency items in Chinese disease control and prevention institutes.
METHODSEvaluation analysis based on data obtained from investigating personal protection equipments and professional knowledge quizzing about personal protection in poisoning control of 57 different level centers for disease control and prevention selected from China by a multi-stage stratified sampling.
RESULTSAll 80.70% of the institutes possessed the protection equipments, provincial, municipal, county level institutes were 100.00%, 95.24%, 66.67%, respectively, with significant statistical difference (H = 7.94, P < 0.05). The average level of the type of individual protective equipments in disease control and prevention institutes was (5.42 +/- 4.00) kinds, the average points in category of provincial, municipal, county level institutes were (11.33 +/- 3.67), (6.52 +/- 3.16), (3.47 +/- 3.10) kinds, being statistically significant (F = 17.30, P < 0.05); type difference counts of disease control and prevention institutes in economic development, secondary, less-developed regions were (6.41 +/- 4.03), (3.55 +/- 3.35), (6.45 +/- 4.07) kinds, with statistical significance (F = 3.70, P < 0.05). Protection equipments chiefly possessed were latex gloves, gauze masks and C-protective clothing. Protective clothing and respiratory protective equipments were insufficient evidently. The average points in testing personal protection basic knowledge were (71.39 +/- 12.52) points; there were no differences between different institutes with different economic regions, levels, technical posts and title degrees.
CONCLUSIONCertain advances have been achieved in recent years in personal protection capacity of institutes for disease control and prevention, but far from the actual demands, and maybe no enough effective response on emergency occurred.

10.Mutational analysis of MYO1E in Chinese children with familial steroid-resistant nephrotic syndrome.

OBJECTIVESteroid-resistant nephrotic syndrome (SRNS) with MYO1E mutations has been identified as autosomal recessive focal segmental glomerulosclerosis (FSGS). To date, only two homozygous mutations in the MYO1E gene were reported in three families with FSGS. This study aimed to examine mutations in the MYO1E gene in children with familial SRNS in the Han Chinese ethnic group.
METHODSBetween 2005 and 2010, peripheral blood samples were collected from the probands, their siblings and parents of four families with autosomal recessive SRNS in the Han Chinese ethnic group. Four probands were studied from nine patients. The mutational analysis of MYO1E was performed by polymerase chain reaction and direct DNA sequencing. Fifty-nine healthy volunteers with normal urine analysis were included as controls.
RESULTSTwenty-five MYO1E variants in the prohands from 4 families with SRNS were identified in this study. Among them, 24 variants were found in NCBI dbSNP. One heterozygous mutation IVS21-85G>A was found in the prohand from Family D, whereas it was absent in 59 normal Chinese controls. No splice site change caused by IVS21-85G>A was reported by analysis with NetGene2.
CONCLUSIONSMYO1E mutations are not a major cause of Chinese familial SRNS in this study.
