Chinese Journal of Tissue Engineering Research 2020;25(3):344-349
BACKGROUND: Dual-task walking is more common in daily life, and more challenging than single-task walking. It is more conducive to find potential gait abnormalities in daily life activities. It is widely used to evaluate the changes in motor performance and gait control when attention is distracted. OBJECTIVE: The three-dimensional gait analysis system was applied to compare the difference of gait characteristics under dual-task walking between healthy young people and elderly people so as to provide reference for preventing the elderly people from falling. METHODS: In the youth group, there were 21 cases aged (24.57±2.27) years old and with the height of (1.64±0.08) m. In the elderly group, there were 25 cases aged (62.72±2.39) years old and with the height of (1.60±0.07) m. The two groups of subjects performed the calculation task of “minus 1 (reciprocal) starting from any number within 100” during level walking. Motion Analysis system and Visual 3D software were applied to collect and analyze the gait data. The differences of gait spatiotemporal and kinetic parameters between the two groups were compared when performing dual-task walking. RESULTS AND CONCLUSION: (1) Spatiotemporal parameters: Compared with youth group, both stride length [(1.14±0.09) m vs. (1.20±0.07) m, P < 0.05] and right step length [(0.57±0.04) m vs. (0.60±0.04) m, P < 0.01] were smaller in the elderly group. (2) Kinetic parameters: Compared with the youth group, the left second peak hip abduction moment was higher [(0.94±0.1) Nm/kg vs. (0.86±0.16) Nm/kg, P < 0.01]; the left ankle plantar flexor moment was lower [(1.27±0.11) Nm/kg vs. (1.35±0.15) Nm/kg, P < 0.05]; and bilateral ankle valgus muscle moments were lower [(0.31±0.14) Nm/kg vs. (0.45±0.16) Nm/kg, P < 0.01; (0.38±0.15) Nm/kg vs. (0.51±0.14) Nm/kg, P < 0.01] in the elderly group. (3) These results suggest that the walking automation ability of the elderly group was lower than youth group during dual-task walking. Dual-task walking mainly affects the hip and ankle joints of the elderly, which may be related to the challenge of dual-task walking to the elderly so that they could adapt the corresponding compensatory mode to maintain stability. The characteristics of gait changes in the elderly under dual tasks can be considered as a reference for the prevention of falls.

OBJECTIVETo evaluate the efficacy and safety of the combination therapy of Xipayimaizipizi Capsules and Tamsulo- sin in the treatment of benign prostatic hyperplasia (BPH).
METHODSWe randomly assigned 60 BPH patients to a control and a combination group of equal number, the former aged 62.03 ± 10.19 years with a disease course of 3.24 ± 2.18 years and the latter aged 64.77 ± 10.33 years with a disease course of 4.09 ± 2.63 years. We treated the patients in the control group with Tamsulosin at 0.2 mg qd and those in the combination group with Tamsulosin at 0.2 mg qd plus Xipayimaizipizi at 0.5 g tid, respectively, both for 4 weeks. Then, we obtained the mean frequency of nocturnal urination, maximal urinary flow rate (Qmax), residual urine volume, International Prostate Symptom Score (IPSS) , and quality of life scores (QOL) of the patients, and recorded their adverse reactions.
RESULTSBefore treatment, the nocturnal urination frequency, Qmax, IPSS, and QOL were 3.60 ± 1.81, (10.40 ± 3.53) ml/min, 22.47 ± 8.58, and 4.43 ± 1.50 in the control group, as compared with 3.43 ± 1.61, (10.14 ± 3.43) ml/min, 21.93 ± 8.79, and 4.73 ± 1.31 in the combination group. After 4 weeks of medication, the combination group showed more significant improvement than the control in the nocturnal urination frequency (1.30 ± 1.18 vs 2.27 ± 1.60), Qmax ([13.85 ± 3.15] vs [14.36 ± 3.03] ml/min), IPSS (13.00 ± 1.53 vs 17.20 ± 8.43), and QOL (2.57 ± 1.61 vs 2.93 ± 1.68), all significantly better than the baseline (P < 0.05). The combination therapy achieved remarkable improvement as compared with the control in the nocturnal urination frequency (- [2.13 ± 1.11] vs -[1.73 ± 1.07]), IPSS (- [8.93 ?6.01] vs -[4.80 ± 3.87]), and QOL (- [2.17 ± 1.12] vs -[1.50 ± 1.01]) (P < 0.05), but exhibited no significant differences from the latter in Qmax ([3.72 ± 2.281 vs [3.95 ± 2.53] ml/min) and residual urine volume (- [34.30 ± 37.43] vs - [26.43 ± 30.49] ml) (P > 0.05). Adverse reactions were found in 5 cases in the combination group (16.67%) and 3 cases in the control (10%) , with no remarkable differences between the two groups (P > 0.05).
CONCLUSIONThe combination therapy of Xipayimaizipizi Capsules and Tamsulosin can improve the symptoms of BPH and the patients quality of life of.

Objective To study the enhancing effect of ultrasound plus microbubble on small interference RNA(siRNA)transfection in vitro and in vivo. Methods Human vascular epithelial growth factor(VEGF)siRNA with 2'deoxy modification(VEGF 2'-eoxy siRNA,VdsR)was used. The human CNE cells(fromnasopharyngeal carcinoma)line was used for in vitro cell-based experiments and in vivo mouse xenograft model. Two different microbubble agents.BR14 and Levovist.were used together with the RNA transfection reagent RNA-mate.ELISA and RT-PCR assays were used to assess VEGF gene expression.Immunohistochemical staining (IHC)was performed to assess CD31 expression in xenograft tumors. Results VdsR transfection in CNE cells abolished VEGF expression as determined by ELISA experiments. In the first mouse xenograft experiment, ultrasound exposure dramatically enhanced VdsR-mediated tumor inhibition. In the second mouse xenograft experiment, when VdsR was mixed with the microbubble reagents and then injected into xenografts, ultrasoundexposure significantly reduced tumor growth in BR14-mixed VdsR group but not in the Levovist-mixed VdsR group compared to the control.RT-PCR experiments demonstrated that VEGF expression in ultrasound-exposed tumors was significantly lower than that in the control.Meanwhile,VEGF expression in the tumor tissue treated by BRl4-mixed VdsR declined as compared with the controls.Tumor vascular density as measured by CD31 immunostaining was significantly decreased in ultrasound-exposed tumors compared to the control. Conclusions Ultrasound exposure and/or microbubble can significantly enhance delivery and the efficiency of VdsR-mediated anti-tumor effects, and should be a location-specific enhancement approach for siRNA-based anti-cancer therapy.

Objective To investigate the prevalence and features of thyroid nodules in middle and aged patients with type 2 diabetes mellitus(T2DM). Methods High-resolution ultrasonography was used to detect thyroid nodules in 132 cases middle and aged patients with type 2 diabetes and 89 patients without diabetes. The nodule features and its relationships with related indicators in diabetic patients were analyzed. Results The prevalence of thyroid nodules in middle and aged patients with type 2 diabetes was higher than that without diabetes (67.4% vs. 53.9%,P<0.05),and most occurred in 50 to 59 age group (66.7% vs. 42.9%) without dependence on changes in thyroid functions and volumes. In diabetes group, the prevalence of thyroid nodules were 59.5% in male and 81.3% in female (P<0.05),no obvious difference was observed in the size and number of thyroid nodules between male and female, multiple nodules and micronodule (< 1.0 cm) had the higher incidences in both sexes. The prevalence of thyroid nodules was increased with aging, but not with diabetes duration and glycosylated hemoglobin (HbAlc) level (x2 =0.797,P=0.372; x2 =1.078,P =0.229). Conclusions It is common that thyroid nodules combined with diabetes in middle and aged patients, thyroid ultrasound screening and regular following-up of patients aged ≥50 years have important clinical significance.
