Objective to observe the correlation between the expression and prognosis of osteosarcoma Insulin-like growth factor receptor(IGF1R),explore the factors affecting the prognosis of patients with osteosarcoma.Methods between January 2005 and April 2014 menstrual standardized comprehensive treatment of 60 cases of patients with stage ⅡB osteosarcoma were retrospectively analyzed.Using immunohistochemical method,the detection of osteosarcoma tissue and IGF1R expression rate in normal tissue adjacent to tumors and to analyze its correlation with survival of patients with osteosarcoma and factors affecting the prognosis.Results IGF1R in the positive expression rate of osteosarcoma tissues was 80.0％ (48/60),expression in normal tissue adjacent to tumor rate was 4/8,has a statistically significant difference between (P =0.032).IGF1R positive expression in osteosarcoma tissues group median disease-free survival time and the median overall survival time were significantly lower than the negative expression group (P =0.023,P =0.040) respectively.IGF1R expression level and pathological necrosis rate of preoperative chemotherapy,the time of first visit to the lung metastasis,and the survival prognosis of patients have significant correlation (P ＜ 0.05),and age,tumor location,tumor size,the serum LDH and ALP levels,is urgent to make a diagnosis and give treatment time,factors such as operation,preoperative chemotherapy times no significant correlation (P ＞ 0.05).Condusion IGF1R positive expression rate in stage Ⅱ B osteosarcoma tissue significantly negatively correlated with the prognosis of patients with osteosarcoma,is the independent prognostic factors of patients with osteosarcoma.

Objective To unmask clues for the investigation of PIH pathogenesis by detecting the altered gene expression profile of placentas from PIH patients. Methods Clinical data and placentas were collected from 42 PIH patients and 22 normotensive pregnancies in seclective cesarean section to construct PIH database from Mar. 2001 to Sep. 2002 in Xi’an district. Suppression subtractive hybridization (SSH) was performed to set the subtractive cDNA library and differential screening was used to identify the positive clones;insertion of positive clones were sequenced by T7 primer method. RT-PCR and immunohistochemistry were employed to confirm the putative gene inhibin A expression. Results A database was set up that consists of pre- and postpartum clinical data and placenta samples of 42 preeclamptic pregnancies and 22 normotensive ones From which,150 features of placental tissue microarray were made. One hundred and three positive clones were isolated by SSH and differential screening. Sequencing and BLAST analysis showed that 90 insertion shared more than 95% homology with sequences in the GenBank/EMBL database. We identified 36 putative genes including pregnancy-specific glycoproteins gene (BC005924),serine protease inhibitor gene (BC012868),VEGFR-1 gene (AF063657),cytokeratine 7 gene(CK7) (AF509887),etc. Inhibin beta A gene was highly expressed in placentas in PIH patients. Conclusion The gene expression profile in PIH placenta was greatly changed;it might be necessary to investigate the role of VEGFR1,serine protease inhibitor,inhibin and CK7 in the pathogenesis of PIH.

OBJECTIVE To analyze the multi-drug resistant genes(genes encoding extended-spectrum ?-lactamases,outer membrane protein gene oprD2,aminoglycoside-modifying enzyme genes,sulfonamides and disinfectant-resistance gene and plasmid-mediated quinolone resistance gene) of Pseudomonas aeruginosa clinical strains.METHODS The drug-resistance to 15 antibiotics was detected by the disc agar diffusion(DAD) and microdilution broth method in 32 P.aeruginosa strains isolated from Jun to Dec 2006.Twenty-eight related drug-resistant genes and outer membrane protein gene oprD2 were examined by PCR method.RESULTS The resistance rate of 32 P.aeruginosa strains to amikacin,ceftazidime,ciprofloxacin,cefepime,cefoperazone/sulbactam,imipenem,meropenem,levofloxacin,piperacillin/tazobactam,aztreonam and piperacillin were 9.4%,25%,59.4%,68.7%,68.8%,78.1%,81.1%,81.3%,84.4% and 94.4%,respectively,and that to others antibiotics were 100%.The detective rate of aminoglycoside-modifying enzyme genes(aac(6′)-Ⅱ,aac(3)-Ⅱ,aac(6′)-Ⅰ,aph(3′)-Ⅵ,ant(3″)-Ⅰ and aac(3)-Ⅰ) were 68.8%,62.5%,21.9%,9.4%,9.4% and 6.3%,and the genes encoding extended-spectrum ?-lactamases(blaFOX,blaIMP,blaVIM and blaDHA) were 37.5% 15.6% 6.3% and 9.3%,respectively.Twenty-two(68.8%) of them were detected in oprD2 and 9(28.1%),but positive in qacE?-sul1 gene.CONCLUSIONS The detective rate of aminoglycoside-modifying enzyme genes,extended-spectrum ?-lactamases genes and the loss rate of outer membrane protein gene oprD2 are high in these 32 multi-drug resistant P.aeruginosa clinical strains.

Objective To explore the characteristics of semantic priming effects in Chinese words with different association strength in patients with aphasia by auditory stimulation.Method Stimulus-response word pairs with different association strength including strong,moderate,weak,and no association categories were chosen from word association thesaurus as experiment materials.Both patients with aphasia(n=11)and normal subjects (n=16)were requested to finish an auditory lexical decision task for target words.Semantic priming effects were investigated by means of measuring reaction time(RT)and error rate of each word-pair.Results In patients with aphasia and normal subjects,the mean RTs were significantly shorter in strong,moderate and weak association strength words than in no association strength words(patients with aphasia(1270.20±47.70)ms,(1340.50±266.25)ms,(1429.70±317.07)ms vs(1549.00±325.87)ms,P＜0.05 and normal subjects(1140.2±274.48)ms,(1196.50±284.06)ms,(1262.10±274.31)ms vs(1391.20±315.68)ms,P＜0.05).In strong,moderate,and no association strength words,the mean RTs were no significant differences between two groups.In the weak association strength words,mean RTs were significantly longer in patients with aphasia than in normal subjects((1429.70±317.07)ms vs(1262.10±274.31)ms,P＜0.05).In two groups,mean error rates were significantly less in strong,moderate and weak association strength words than in no association strength words(patients with aphasia:7.73±6.07,4.55±7.23,6.82±8.15 vs 14.09±12.41,P＜0.05 and normal subjects:3.44±4.37,2.81±3.64,5.31±5.91 vs 10.94±11.14,P＜0.05).However,in strong association strength words,mean error rates were significantly higher in patients with aphasia than in normal subjects(7.73±6.07 vs 3.44±4.37,P＜0.05).In moderate,weak and no association strength words,there were no significant differences between two groups.Conclusion The patients with aphasia follow gradient of the association strength words like normal subjects and have semantic priming effects in the strong,moderate association strength words.

Objective To evaluate the safety and efficacy of two surgical procedures including retropubic space pathway and obturator foramen pathway in treatment of the old women with stress urinary incontinence (SUI). Methods Intravaginal Slingplasty (IVS) procedure by retropubic space pathway was performed in 52 cases with simple SUI from June 2003 to June 2005. And the modified Transobturator Vaginal Tape Inside-Out (TVT-O) procedure was carried out in 112 cases with simple SUI from October 2005 to October 2008, using self-tailed polypropylene mesh and helical needles. The comparative retrospective study was performed between these two groups. Results The cure rate was 80.8% (42/52) and the improvement rate was 13.5% (7/52) in IVS group. And the corresponding data were 84.8% (95/112) and 11.6% (13/112) in modified TVT-O group. The 3 (5.8%) cases failed in IVS procedure and 4 (3.6%) cases failed in modified TVT-O procedure at 1 month's follow-up. All of the 7 patients received the 'Tong's hammock procedure' and no cases failed at 6 and 12 month's follow-up. One case of vaginal erosion was found in each group although good effect of continence was observed. Meshes were removed in these two patients and then the 'Tong's hommock procedure' was performed which turned out to be good outcome. Bladder injuries were found in 3 cases when route cystoscopy was performed. No significant difference in efficacy was found between these two groups(P＞0. 05). In modified TVT-O group, operative duration, blood loss and medical cost were significantly reduced as compared with IVS group (P＜0. 01). Conclusions Both of the two surgical procedures of modified TVT-O versus IVS are simple, safe and mini-invasive procedures with high efficacy. However, the modified TVT-O procedure is more easy to be adopted by patients due to short operative time and lower medical cost.

Objective To observe the effect of dexmedetomidine on inflammation, oxidative stress and lung injury in lipopolysaccharide( LPS)-induced septic mice.Methods Forty eight male adult BALB/c mice were randomly divided into three groups (n=16): normal control group (Ctrl), sepsis group (Sep), and Dex group.Ae septic mice model was established by LPS 20 mg/kg,and Dex 30μg/kg injected intraperitoneally at 0.5 h after LPS injection.The concentrations of serum IL-6 and IL-10 were detected at 2 h and 6 h after LPS injection while myeloperoxidase ( MPO ) activity and malondialdehyde( MDA) content of lungs were detected and weight dry ratio ( W/D) of lungs was calculated at 6 h after LPS injection.Pathological changes were observed in left lung HE stained with optical microscopy at 6 h after LPS injection. Results Compared with Sep group, the concentrations of serum IL-6 decreased significantly(P <0.05), while the concentrations of IL-10 increased significantly(P<0.05) at 2 h and 6 h after LPS injection in Dex group.MPO activity, MDA content and W/D of lungs decreased significantly(P<0.05) at 6 h after LPS injection in Dex group.The injury to the lung was lightened significantly under optical microscopy in Dex group.Conclusion Dex protects against LPS-induced ALI in septic mice by inhibiting systemic inflammatory response, reducing lung tissue inflammatory infiltration and oxidative stress.

Objective To investigate the effect of noninvasive ventilator therapy on serum C-reactive protein (CRP), endothelin-1 (ET-1) and tumor necrosis factor-α (TNF-α) levels in patients with obstructive sleep apnea hypopnea syndrome (OSAHS) and its clinical significance. Methods One hundred cases of moder-ate and severe OSAHS patients were selected by the method of parallel opening. All of the patients were given health education requirement , quitting smoking and wine , low fat diet and exercise to lose weight and other con-ventional treatment. The patients were randomly divided into the treatment group of 42 cases with noninvasive ventilator treatment , 44 cases treated with conventional treatment , to observe the changes of serum CRP , ET-1 and TNF-α levels and PSG parameters after 12 weeks in two groups. Results Apnea hypopnea index (AHI), oxygen desaturation index (ODI), the lowest oxygen saturation (LSpO2), and the average oxygen saturation MSpO2 in OSAHS patients were significantly improved after treatment (P < 0.01), but the degree of improvement in the two groups after treatment was significantly higher than the control group (P < 0.01). Plasma CRP, ET-1 and TNF-α levels in the two groups after treatment were lower than before treatment (P < 0.05 or P < 0.01), but the treatment group was significantly higher than that of the control group ( P < 0 . 05 or P < 0 . 01 ) . Conclusion Noninvasive ventilator therapy in improving the OSAHS monitoring data of patients with PSG can effectively reduce the serum CRP, TNF-α, ET-1 level, reduce the body′s inflammatory reaction.
